The Met243 sulfonium ion linkage is responsible for the anomalous magnetic circular dichroism and optical spectral properties of myeloperoxidase.

The heme group of myeloperoxidase shows anomalous optical properties, and the enzyme possesses the unique ability to catalyze the oxidation of chloride. However, the nature of the covalently bound heme macrocycle has been difficult to identify. In this work, the electronic and magnetic properties of the heme groups in oxidized and reduced forms of wild-type and Met243Thr mutant myeloperoxidase and wild-type lactoperoxidase have been investigated using variable-temperature (1.6-273 K) magnetic circular dichroism (MCD) spectroscopy along with parallel optical absorption and electron paramagnetic

Inherently distorted heme as a novel tool for myoglobin-based oxygen carrier.

The two types of artificial myoglobins (Mbs) containing inherently distorted α-ethyl-2,4-dimethyldeuteroheme and undistorted 2,4-dimethyldeuteroheme were prepared to examine the influence of nonplanar heme deformation on the reactivity of Mb. In ferrous deoxy proteins, the paramagnetic proton nuclear magnetic resonance spectra showed that the deformed heme caused a 3.2-ppm lower-field shift of the proximal histidine signal, indicating a larger iron displacement from the heme plane upon the nonplanar deformation. The Mb with the nonplanar heme exhibited a markedly lower oxygen affinity as compa

Electronic structure of six-coordinate iron(III) monoazaporphyrins.

The electronic structures of six-coordinate iron(III) octaethylmonoazaporphyrins, [Fe(MAzP)L 2] (+/-) ( 1), have been examined by means of (1)H NMR and EPR spectroscopy to reveal the effect of meso-nitrogen in the porphyrin ring. The complexes carrying axial ligands with strong field strengths such as 1-MeIm, DMAP, CN (-), and (t)BuNC adopt the low-spin state with the (d xy ) (2)(d xz , d yz ) (3) ground state in a wide temperature range where the (1)H NMR and EPR spectra are taken. In contrast, the complexes with much weaker axial ligands, such as 4-CNPy and 3,5-Cl 2Py, exhibit the spin trans

Evaluation of Mesoporous Silica Nanoparticles as Carriers of Triarylmethyl Radical Spin Probes for EPR Oximetry.

In vivo measurement and mapping of oxygen levels within the tissues are crucial in understanding the physiopathological processes of numerous diseases, such as cancer, diabetes, or peripheral vascular diseases. Electron paramagnetic resonance (EPR) associated with biocompatible exogenous spin probes, such as Ox071 triarylmethyl (TAM) radical, is becoming the new gold standard for oxygen mapping in preclinical settings. However, these probes do not show tissue selectivity when injected systemically, and they are not cell permeable, reporting oxygen from the extracellular compartment only. Recen

Unveiling the mystery of chlorine radical in water by a simple methanol-based spin trapping.

Both hydroxyl radical (•OH) and chlorine radical (Cl•) are recognized as significant reactive species in aquatic and atmospheric sciences. While •OH can be readily detected with the conventional spin trap, 5,5-dimethyl-1-pyrroline N-oxide (DMPO), Cl• is often considered difficult to detect by electron paramagnetic resonance (EPR) technique and has therefore been largely underexploited. In this work, we clarify the complicated reactions of Cl• with DMPO, which pose challenges for the direct detection of Cl•. To overcome this limitation, we take advantage of the differences in the products forme

Structural Flexibility of Tau in Its Interaction with Microtubules as Viewed by Site-Directed Spin Labeling EPR Spectroscopy.

Tau is a microtubule-associated protein that belongs to the Intrinsically Disordered Proteins (IDPs) family. IDPs or Intrinsically Disordered Regions (IDRs) play key roles in protein interaction networks and their dysfunctions are often related to severe diseases. Defined by their lack of stable secondary and tertiary structures in physiological conditions while being functional, these proteins use their inherent structural flexibility to adapt to and interact with various binding partners. Knowledges on the structural dynamics of IDPs and their different conformers are crucial to finely decip

Imaging free radicals in organelles, cells, tissue, and in vivo with immuno-spin trapping.

The accurate and sensitive detection of biological free radicals in a reliable manner is required to define the mechanistic roles of such species in biochemistry, medicine and toxicology. Most of the techniques currently available are either not appropriate to detect free radicals in cells and tissues due to sensitivity limitations (electron spin resonance, ESR) or subject to artifacts that make the validity of the results questionable (fluorescent probe-based analysis). The development of the immuno-spin trapping technique overcomes all these difficulties. This technique is based on the react

DNA cleavage and detection of DNA radicals formed from hydralazine and copper (II) by ESR and immuno-spin trapping.

Metal ion-catalyzed oxidation of hydrazine and its derivatives leads to the formation of the hydrazyl radical and subsequently to oxy-radicals in the presence of molecular oxygen. Here, we have examined the role of Cu(2+)-catalyzed oxidation of hydralazine in the induction of DNA damage. Neither 5,5-dimethyl-1-pyrroline-N-oxide (DMPO) nor dimethyl sulfoxide (DMSO) was effective in inhibiting hydralazine-Cu(2+)-induced DNA damage. Singlet oxygen did not appear to participate in this DNA cleavage. The one-electron oxidation of hydralazine also leads to the formation of DNA radicals as confirmed

Characterization of the disordered-to-α-helical transition of IA₃ by SDSL-EPR spectroscopy.

Electron paramagnetic resonance (EPR) spectroscopy coupled with site-directed spin labeling (SDSL) is a valuable tool for characterizing the mobility and conformational changes of proteins but has seldom been applied to intrinsically disordered proteins (IDPs). Here, IA₃ is used as a model system demonstrating SDSL-EPR characterization of conformational changes in small IDP systems. IA₃ has 68 amino acids, is unstructured in solution, and becomes α-helical upon addition of the secondary structural stabilizer 2,2,2-trifluoroethanol (TFE). Two single cysteine substitutions, one in the N-terminus

Libration motion of guest spin probe molecules in organic glasses: CW EPR and electron spin echo study.

Echo-detected (ED) EPR spectra of nitroxide spin probes dissolved in glassy materials provide evidence that guest molecules in these media undergo fast librational motion. Theory of spin relaxation of a librating molecule is presented. The mean squared amplitude, <alpha2>, of this motion which can be derived from continuous wave (CW) EPR spectral splitting is found to depend linearly on temperature in the low temperature region. This may be ascribed to thermal harmonic vibrations. The slope of the linear dependence varies from glass to glass and seems to correlate with the strength of the inte

pH-dependent phase transition of chlorpromazine micellar solutions in the physiological range.

The effects of pH and drug concentration on aggregation properties of chlorpromazine-HCl (CPZ) are examined. The critical micelle concentration (cmc) changes from 0.2 mM at pH 7.3 to 2 mM at pH 5.6 as estimated from the stearic acid spin label solubility measurements. For concentrations above the cmc CPZ micelles undergo a concentration-, temperature- and pH-dependent transition leading to phase separation. This phase transition is followed by a sudden increase of light scattering. The phase diagram pH vs. concentration is obtained by observation of the cloud point for concentrations ranging f

Spectral changes of lignin peroxidase during reversible inactivation.

The heme environment of lignin peroxidase (LiP) has been investigated by electronic absorption and electron paramagnetic resonance (EPR) spectroscopy. Native LiP was a pentacoordinate, high-spin ferric iron with a high-spin absorption band at 634 nm and g values at 5.86 and 2.07 in the EPR spectrum. Upon thermal inactivation, calcium ions were released from the enzyme and the Soret absorption decreased and red-shifted about 2 nm, the high-spin absorption band at 634 nm disappeared, and a low-spin absorption band appeared at 532 nm. The EPR spectrum and the temperature dependence of electronic

Molecular Cocrystal Packing Suppresses Hopping-Driven Decoherence of Excitonic Spin Qubits.

Molecular excitonic spins have garnered significant interest for quantum technologies because they can be initialized into addressable, multilevel quantum states through spin-selective intersystem crossing or singlet fission. However, excitonic spin coherence is difficult to maintain above liquid helium temperatures due to typical crystal packings, which promote decoherence through exciton hopping between magnetically inequivalent sites. Here, we engineer donor-acceptor cocrystals where molecular packing in isolated π-stacks of magnetically equivalent molecules suppresses hopping-induced decoh

Magnetic Polaron States in Photoluminescent Carbon Dots Enable Hydrogen Peroxide Photoproduction.

Photoactivation of aspartic acid-based carbon dots (Asp-CDs) induces the generation of spin-separated species, including electron/hole (e- /h+ ) polarons and spin-coupled triplet states, as uniquely confirmed by the light-induced electron paramagnetic resonance spectroscopy. The relative population of the e- /h+ pairs and triplet species depends on the solvent polarity, featuring a substantial stabilization of the triplet state in a non-polar environment (benzene). The electronic properties of the photoexcited Asp-CDs emerge from their spatial organization being interpreted as multi-layer asse

Orthogonal spin labeling and Gd(III)-nitroxide distance measurements on bacteriophage T4-lysozyme.

We present the first example of chemoselective site-specific spin labeling of a monomeric protein with two spectroscopically orthogonal spin labels: a gadolinium(III) chelate complex and a nitroxide radical. A detailed analysis of the performance of two commercially available Gd(III) ligands in the Gd(III)-nitroxide pulse double electron-electron resonance (DEER or PELDOR) experiment is reported. A modification of the flip angle of the pump pulse in the Gd(III)-nitroxide DEER experiment is proposed to optimize sensitivity.

Detection and removal of contaminating hydroxylamines from the spin trap DEPMPO, and re-evaluation of its use to indicate nitrone radical cation formation and S(N)1 reactions.

A previous report that the spin trap 5-diethoxyphosphoryl-5-methyl-1-pyrroline-N-oxide (DEPMPO) allows DEPMPO radical cation formation to be detected via the production of a carbon-centred radical adduct (assigned as the cis-hydroxyethyl species, formed by an intramolecular process) is shown to be incorrect. Rather, this and other paramagnetic species arise from the facile oxidation of trace hydroxylamine impurities present in commercial DEPMPO samples. As a result, techniques for the detection and elimination of such hydroxylamine impurities from DEPMPO solutions were developed and are descri

Molecular Mechanism for the Unprecedented Metal-Independent Hydroxyl Radical Production from Thioureas and H2O2.

The most well-known hydroxyl radical (•OH)-generating system is the classic iron-mediated Fenton reaction. Thiourea has been considered as an efficient •OH scavenger and is frequently used to study the role of •OH in various biochemical and medical research studies. Here we found that the highly reactive •OH can be produced from thiourea and H2O2 through a metal-independent pathway, as measured by electron spin resonance (ESR) secondary radical spin-trapping and fluorescent methods. The major reaction intermediates from thiourea/H2O2 were identified as formamidinesulfenic acid and formamidines

EPR investigation of UV light effect on calcium carbonate powders with different grain sizes.

This study is based on investigation of calcium carbonate powders with different grain sizes exposed to UV light. Calcium carbonate is widely used in many branches of industry, e.g. as a filler for polymer materials; therefore, knowing its properties, among them also its reaction to UV light, is essential. Samples of powdered calcium carbonate with average grain sizes of 69 and 300 nm and 2.1, 6, 16, 25 µm were used in this investigation. Measurements were performed at room temperature using EPR X-band spectrometer, and they have shown the additional signals induced by the light from Hg lamp.
