Interaction between tramadol and two anti-emetics on nociception and gastrointestinal transit in mice.

BACKGROUND: Clinical studies suggest that tramadol-induced analgesia is partially antagonized by ondansetron. AIMS: To investigate the type of interaction between tramadol and two anti-emetics on antinociception and gastrointestinal transit in mice. METHODS: We assessed the antinociceptive (acetic acid writhing test, plantar test) and antitransit (charcoal meal) effects of tramadol individually, and combined with ondansetron or droperidol in female Swiss CD-1 mice. Isobolograms and analysis of variance were used to determine the type of interaction. RESULTS: In the writhing test, tramadol,

Use of anti-emetics after intragastric balloon placement: experience with three different drug treatments.

BACKGROUND: Tropisetron treatment was compared with alizapride treatment. The secondary aim was to assess whether droperidol supplement would still improve the therapeutic outcome of tropisetron. MATERIALS AND METHODS: A series of 51 obese patients was treated with an intragastric balloon to obtain weight reduction. Patients were divided at random into 3 groups. Each group received a different antiemetic and spasmolytic regimen to control postoperative nausea and vomiting for 24 hours. Statistical analysis of both parameters showed that all 3 populations are comparable and the studied inciden

The efficacy of various classes of anti-emetics in preventing deoxynivalenol-induced vomiting in swine.

The Fusarium mycotoxin deoxynivalenol (DON) is a potent emetic agent. While the basic mechanisms which invoke and mediate emesis are still poorly understood, various neurotransmitters appear to be involved. The action of these transmitters can be blocked by various receptor-specific antagonists. The current study investigated the efficacy of several classes of receptor antagonists to block the emetic effect of DON. Following anti-emetic pretreatment, pigs were administered the toxin (i.v., 80 micrograms/kg, or oral, 300 micrograms/kg) and the onset of emesis was monitored. Certain specific ser

[Combined use of ondansetron and other anti-emetics to control cisplatin-induced nausea and vomiting].

While ondansetron is effective in the control of nausea and vomiting induced by high dose cisplatin, it has to be given in multiple doses and is very expensive. We designed a regimen of combined use of a single dose of ondansetron and dexamethasone to control acute emesis and combined use of metoclopramide, diphenhydramine and valium to control delayed emesis. The results of this regimen was compared with that of the routine treatment regimen using metoclopramide plus diphenhydramine plus valium. A total of 43 patients were rolled in the cross-over study. Effective control of acute emesis was

[Effect of combined use of adrenocortical hormones and domperidone as anti-emetics during cisplatin therapy].

Eighteen patients received repeated administrations of cisplatin in relatively large amounts (55-75-100 mg/m2) given systemically by drop infusion. As antiemetic treatments, the following were scheduled: on the day before; prednisolone 30mg (3 X p.o.), immediately before; methylprednisolone 500mg (i.v.), and 3 hours after administration of cisplatin ; methylprednisolone 500mg (i.v.) and domperidone 60 mg (suppo.). Domperidone was given twice a day for one week. Nausea, vomiting and anorexia were studied objectively for two weeks. At a dose of 75 mg/m2 of cisplatin, the occurrence and the durat

[Effectiveness of anti-emetics for the prophylaxis of cisplatin-induced delayed emesis: a systematic review].

We performed a systematic review of the effectiveness of anti-emetics for prophylaxis of cisplatin-induced delayed emesis using meta-analysis. We selected 12 reports of randomized controlled trials from MEDLINE (1966-2003. 4) and The Cochrane Library Issue 1, 2003. Nine of these reports were evaluated as high quality and the others as low quality according to the evaluation criteria of Jadad et al., and only the high-quality reports were subjected to meta-analysis. The statistical results obtained from all 12 reports were also compared with those obtained from the 9 reports of high quality. Co

Nasal In Situ Hydrogels: A Boon For Anti-Emetic Drug Delivery.

Nasal route presents an alternative avenue for achieving local or systemic drug effects owing to its non-invasiveness, expansive surface area, rapid drug absorption, and circumvention of first-pass effect. It offers a promising route for brain targeting too, as it bypasses the blood-brain barrier, thus facilitating direct nose to brain drug delivery. However, traditional routes of administering anti-emetic drugs, primarily oral or parenteral, suffer from drawbacks like poor patient compliance and low drug bioavailability. The nasal route is a favorable alternative, albeit limited by the short

Monkey Feeding Assay for Testing Emetic Activity of Staphylococcal Enterotoxin.

Staphylococcal enterotoxins (SEs) are unique bacterial toxins that cause gastrointestinal toxicity as well as superantigenic activity. Since systemic administration of SEs induces superantigenic activity leading to toxic shock syndrome that may mimic enterotoxic activity of SEs such as vomiting and diarrhea, oral administration of SEs in the monkey feeding assay is considered as a standard method to evaluate emetic activity of SEs. This chapter summarizes and discusses practical considerations of the monkey feeding assay used in studies characterizing classical and newly identified SEs.

Zingiber officinale (ginger) as an anti-emetic in cancer chemotherapy: a review.

Despite significant advances and development of novel anti-emetics, nausea and vomiting (emesis) is a major side-effect of cancer chemotherapy. At times, severe nausea and vomiting may also lead to reduction in adherence to the treatment regimen, and this will concomitantly affect the patient's survival. The rhizome of Zingiber officinale, commonly known as ginger, is globally an important spice. It has been used for centuries in the Indian, Chinese, Arabic, Tibetan, Unani, and Siddha systems of traditional medicine to treat nausea and vomiting induced by different stimuli. Preclinical studies

Characterization of emetic Bacillus weihenstephanensis, a new cereulide-producing bacterium.

Cereulide production has until now been restricted to the species Bacillus cereus. Here we report on two psychrotolerant Bacillus weihenstephanensis strains, MC67 and MC118, that produce cereulide. The strains are atypical with regard to pheno- and genotypic characteristics normally used for identification of emetic B. cereus strains. MC67 and MC118 produced cereulide at temperatures of as low as 8 degrees C.

Enhanced schistosomicidal efficacy of tartar emetic encapsulated in pegylated liposomes.

The aim of the present study was to evaluate the ability of liposomes to improve the efficacy of tartar emetic (TA) against established Schistosoma mansoni infection. TA was used as a schistosomicidal drug model and both conventional liposomes (CL) and long-circulating pegylated liposomes (LCL) were evaluated. In the first experiment, TA, either free or encapsulated within CL or LCL, was given intraperitoneally (i.p.) as a single dose of 11 mg Sb/kg to mice experimentally infected with S. mansoni. Only the group treated with LCL showed a significant (55%) reduction in the worm burden, compared

Bacillus cereus produces most emetic toxin at lower temperatures.

Seven emetic toxin-producing strains of Bacillus cereus were examined for toxin production in Skim Milk Medium at incubation temperatures ranging from 10 to 50 degrees C. Minimum and maximum growth temperatures were found to be 12 and 46 degrees C, respectively. At 12 and 15 degrees C, levels of toxin production were significantly higher (P < 0.01) than that observed at 30 degrees C, while no toxin was produced above 37 degrees C. Increased levels of sporulation were observed at increased temperatures, and no correlation was found between levels of sporulation and toxin production (R(2) = 0.08

Dose-related antagonism of the emetic effect of morphine by methylnaltrexone in dogs.

Opioids administered to produce analgesia cause unwanted emesis in patients (incidence 20%-30%, depending on situation). Tests in animals show that quaternary narcotic antagonists like methylnaltrexone (MNTX) do not affect the analgesic potency of morphine, but such compounds have not been examined for their potential to antagonize morphine-induced emesis. To determine the effects of MNTX on emetic response, we assigned 85 dogs to one of 11 groups challenged with morphine alone or morphine and various doses of MNTX IM or i.v. Antagonism of the emetic response was dose related: MNTX, 0.25 mg/kg

Surgical and pharmacological dissociation of cardiovascular and emetic responses to intragastric CuSO4.

Under control (intact and laparotomized) conditions, arterial pressure of urethan-anesthetized ferrets rose significantly in response to intragastric copper sulfate (CuSO4; 10 ml of 5 mg/ml). CuSO4 was emetic 75-90% of the time, and a cardiovascular response always immediately preceded and accompanied emetic responses. The cardiovascular response was significantly reduced or abolished by hexamethonium (0.35 mg/kg) pretreatment combined with atropine methyl bromide (1 mg/kg). Addition of the alpha 2-receptor antagonist 2,3-dichloro-alpha-methylbenzylamine HCl (DCMB, 10 mg/kg) and the beta-recep

The abdominal visceral innervation and the emetic reflex: pathways, pharmacology, and plasticity.

In recent years the role of the area postrema in the emetic reflex has been predominant and the involvement of the abdominal visceral innervation has tended to be overlooked. This paper attempts to redress the balance reflex by reviewing aspects of the existing literature and complementing this with original studies from the ferret. In view of the widespread use of the ferret in studies of emesis and particularly in the characterization of the antiemetic actions of 5-HT3 receptor antagonist, the opportunity is taken to assess the suitability of this species for studies of emesis. It is conclud

A prospective study of drugs and pregnancy. II. Anti-emetic drugs.

In a prospective study of anti-emetic drug use during 6 376 pregnancies, the previously demonstrated negative correlation between the use of such drugs and miscarriage was confirmed. It is probably completely due to less frequent and possibly less severe morning sickness complaints in pregnancies ending with a miscarriage. A slight positive correlation is found between the use of anti-emetics and infant malformation. A detailed analysis showed that this was mainly due to a high incidence of drug use (mainly promethazine) among women giving birth to infants with congenital dislocation of the hi
