NK Cells In Endocrine Autoimmune Disorders: Current Knowledge and Implications for Therapeutic Strategies.

Natural killer (NK) cells play critical roles in innate immunity and in shaping adaptive immune responses. Recent studies reveal their dual role in autoimmunity, either with a protective role by eliminating autoreactive cells or as contributors to pathogenic inflammation. This review summarizes current understanding of NK cell biology in endocrine autoimmune diseases, focusing on what is known for type 1 diabetes and autoimmune adrenal failure-Addison's disease. We further discuss emerging therapeutic approaches focusing on cytokine-activated (supercharged) NK cells and CAR NK cells, and highl

Regulation of PSAT1 and PHGDH by m6A in endocrine-resistant breast cancer cells.

The N-6-methyladenosine (m6A) modification of mRNA regulates transcript abundance in endocrine therapy (ET)-resistant breast cancer (BCa) cells. We reported that m6A reader HNRNPA2B1 decreased miR-145p and miR-424-5p targeting PSAT1 and miR-34b-5p and miR-876-5p targeting PHGDH, thus stimulating the serine synthesis pathway (SSP) in ET-resistant BCa cells. Here we examined m6A regulation of PSAT1 and PHGDH. We report that siMETTL3 increased miR-145-5p, reducing PSAT1, and miR-34b-5p and miR-876-5p, reducing PHGDH, without affecting HNRNPA2B1 or NFkB and decreasing MYC, known to stimulate PSAT1

Histological distribution of endocrine cells in the pancreas of Javan Mongoose (Urva javanica).

BACKGROUND: The Javan mongoose (Urva javanica) is an endemic wild animal in Southeast Asia, including Indonesia, and is currently classified as an unprotected species. However, intensive hunting may lead to a population decline in its natural habitat, potentially altering its conservation status and posing a threat soon. Despite this concern, the anatomical data of the species remain limited. The digestive system of the Javan mongoose, an opportunistic carnivore, has not been previously documented. While a comprehensive examination of the entire digestive tract would offer broader insights, th

Preparation of Highly Functional Spheroid of Endocrine Cells Based on Thermosensitive Glycol Chitosan.

BACKGROUND: Pancreatic islet transplantation holds great potential as a therapeutic approach for treating type 1 diabetes mellitus (T1D). However, large islets suffer from hypoxia due to the limited diffusion distance of oxygen, leading to cell loss. Therefore, smaller spheroids are needed for better transplantation outcomes. This study aims to develop a method for forming highly functional islet spheroids using glycol chitosan (GC) derivatives, such as N-acetylated glycol chitosan (AGC) and N-hexanoyl glycol chitosan (HGC). METHODS: Thermogelling polymers were produced by performing N-acylat

Impact of endocrine disruptors on key events of hepatic steatosis in HepG2 cells.

Endocrine-disrupting chemicals (EDCs) may contribute to the rising incidence of metabolic dysfunction-associated steatotic liver disease (MASLD). We investigated the potential of 10 environmentally relevant EDCs to affect key events of hepatic steatosis in HepG2 human hepatoblastoma cells. Increased lipid droplet formation, a key marker of steatosis, was induced by PFOA, bisphenol F, DDE, butylparaben, and DEHP, within the non-cytotoxic concentration range of 1 nM-25 μM. Cadmium also induced this effect, but at concentrations impairing cell viability (>1 μM). At non-cytotoxic concentrations, t

Primary culture and endocrine functional analysis of Leydig cells in ducks (Anas platyrhynchos).

Testicular Leydig cells (LCs) are the primary known source of testosterone, which is necessary for maintaining spermatogenesis and male fertility. However, the isolation, identification, and functional analysis of testosterone in duck LCs are still ambiguous. The aim of the present study was to establish a feasible method for isolating highly purified primary duck LCs. The highly purified primary duck LCs were isolated from the fresh testes of 2-month-old ducks via the digestion of collagenase IV and Percoll density gradient centrifugation; hematoxylin and eosin (H&E), immunohistochemistry (IH

Glucagon- and insulin-immunopositive endocrine cells in porcine extrahepatic bile ducts and gallbladder.

INTRODUCTION: Pancreatic β-cells and α-cells have been found in the murine extrahepatic biliary ducts but not in the gallbladder. However, there has been no information reported in the specialized literature about the presence of glucagon- and insulin-expressing endocrine cells in porcine bile ducts and gallbladder. AIM: We aimed to perform an immunohistochemical study to identify glucagon- and insulin-positive cells and their distribution in the porcine extrahepatic biliary ducts and gallbladder. METHOD: The immunohistochemical method was used to detect the presence and distribution of gluc

Expression of Rasd1 in mouse endocrine pituitary cells and its response to dexamethasone.

Dexamethasone-induced Ras-related protein 1 (Rasd1) is a member of the Ras superfamily of monomeric G proteins that have a regulatory function in signal transduction. Rasd1, also known as Dexras1 or AGS1, is rapidly induced by dexamethasone (Dex). While prior data indicates that Rasd1 is highly expressed in the pituitary and that the gene may function in regulation of corticotroph activity, its exact cellular localization in this tissue has not been delineated. Nor has it been determined which endocrine pituitary cell type(s) are responsive to Dex-induced expression of Rasd1. We hypothesized t

Zebrafish ghrelin is expressed in pancreatic endocrine cells and regulated by metabolic state.

Mammalian ghrelin is a stomach-derived peptide that stimulates secretion of growth hormone and food intake. Zebrafish is an excellent model system for forward genetic studies, and many aspects of energy homeostasis characterized in mammals appear to be conserved in the zebrafish. In this study, we investigated the expression and regulation of zebrafish ghrelin by metabolic status. Quantitative RT-PCR revealed that zebrafish ghrelin is highly enriched in anterior gut associated tissues. Using in situ hybridization with adult zebrafish tissues, we found that zebrafish ghrelin mRNA was not expres

Endocrine cells in the human common bile duct in patients with obstructive jaundice.

BACKGROUND/AIMS: There was no data about endocrine cells in the extrahepatic bile duct in secondary cholangitis due to obstructive jaundice. The aim of the present study is to investigate immunohistochemically the endocrine cell types in the lower part of the human common bile duct in biopsy samples, collected during drainage because of complete or incomplete obstruction, caused mainly by stones. We explained the presence of various hormone-producing endocrine cells in this region with the regulation of physiological and pathological processes there. METHODOLOGY: We used light and electron mi

Ngn3(+) endocrine progenitor cells control the fate and morphogenesis of pancreatic ductal epithelium.

During pancreas development, endocrine and exocrine cells arise from a common multipotent progenitor pool. How these cell fate decisions are coordinated with tissue morphogenesis is poorly understood. Here we have examined ductal morphology, endocrine progenitor cell fate and Notch signaling in Ngn3(-/-) mice, which do not produce islet cells. Ngn3 deficiency results in reduced branching and enlarged pancreatic duct-like structures, concomitant with Ngn3 promoter activation throughout the ductal epithelium and reduced Notch signaling. Conversely, forced generation of surplus endocrine progenit

Aflatoxin B1--a potential endocrine disruptor--up-regulates CYP19A1 in JEG-3 cells.

Previous studies have indicated that aromatase (CYP19A1) is involved in the metabolism of aflatoxin B1 (AFB1). We hypothesized that exposure to AFB1 contaminated food during pregnancy could disrupt the normal production of steroid hormones in placenta. We examined the capability of AFB1 exposure to disrupt CYP19A1 expression as a putative endocrine disrupter, and to investigate the metabolism of AFB1 by CYP19A1. JEG-3 cells, as model for placental cells, were exposed alone and in combination to AFB1 and estrogen receptor ligands for 24-96 h. AFB1 (0.3-1.0 μM) induced the expression of CYP19A1

HCO 3--dependent volume regulation in alpha-cells of the rat endocrine pancreas.

Ion transport activity in pancreatic alpha-cells was assessed by studying cell volume regulation in response to anisotonic solutions. Cell volume was measured by a video imaging method, and cells were superfused with either 4-(2-hydroxyethyl)piperazine-1-ethanesulfonic acid-buffered or HCO(3)(-) -buffered solutions. alpha-Cells did not exhibit a regulatory volume increase (RVI) in response to cell shrinkage caused by hypertonic solutions. A RVI was observed, however, in cells that had first undergone a regulatory volume decrease (RVD), but only in HCO(3)(-)-buffered solutions. RVI was also obs

Transdifferentiation molecular pathways of neonatal pig pancreatic duct cells into endocrine cell phenotypes.

Restrictions in availability of cadaveric human donor pancreata have intensified the search for alternate sources of pancreatic endocrine tissue. We have undertaken to assess whether nonendocrine pancreatic tissue, with special regard to ducts, including epithelial cells, and retrieved from neonatal pig pancreata that are used for islet isolation, may under special in vitro culture conditions generate endocrine cell phenotypes. Special care was taken to identify the time-related appearance of molecular and biochemical markers associated with beta-cell specificity, in terms of glucose-sensing a

Expression of K+-Cl- cotransporters in the alpha-cells of rat endocrine pancreas.

The expression of K+-Cl- cotransporters (KCC) was examined in pancreatic islet cells. mRNA for KCC1, KCC3a, KCC3b and KCC4 were identified by RT-PCR in islets isolated from rat pancreas. In immunocytochemical studies, an antibody specific for KCC1 and KCC4 revealed the expression of KCC protein in alpha-cells, but not pancreatic beta-cells nor delta-cells. A second antibody which does not discriminate among KCC isoforms identified KCC expression in both alpha-cell and beta-cells. Exposure of isolated alpha-cells to hypotonic solutions caused cell swelling was followed by a regulatory volume de

Two types of calcium channels in human ovarian endocrine cells: involvement in steroidogenesis.

Nature, regulation, and functional role of ion channels of human ovarian endocrine cells are not well known. In our present study, we show two types of voltage-activated Ca(2+) currents (I(Ca)) in cultured human luteinized granulosa cells (GCs), as assessed by whole-cell patch-clamp experiments. Electrophysiological properties, namely low threshold of activation, pronounced time-dependent inactivation, slow and voltage-dependent deactivation kinetics, insensitivity to SNX-482, and high sensitivity to Ni(2+), defined the predominant I(Ca) as a T-type Ca(2+) current (I(Ca.T)). In 4% of cells a N

[Endocrine cells of the gastric mucosa in patients with duodenal ulcer after vagotomy].

Bioptates of the stomach mucous membrane (SMM) have been investigated in 169 patients suffering from duodenal ulcer (DU). According to the nocturnal gastric secretion test among them there are "hypersecretors" and persons with moderate elevation of acid formation. In conformity with the efficiency of the operative treatment among the patients examined, groups are defined: those with recurrent disease and those recovered after vagotomy. The DU endocrine apparatus undergoes both qualitative and quantitative alterations after vagotomy. When recovery after vagotomy takes place, the number of endoc

Somatostatin-containing and other endocrine cells in the pancreas of the spectacled caiman.

Light-microscopic immunocytochemistry was used to localize 4 major pancreatic hormones in the pancreas of the spectacled caiman, Caiman fuscus. Somatostatin, insulin, glucagon and pancreatic polypeptide were localized by the peroxidase-antiperoxidase complex technique. A relatively large population of somatostatin-containing D cells was present. The D cells were nearly as numerous as the insulin-containing B cells and glucagon-containing A cells which were the most common cell types. All three cell types were commonly intermingled with one another in endocrine cell areas. Pancreatic polypeptid
