Maize crop yields are substantially diminished by drought during early phases of kernel development. Kernel development is also affected by stresses which limit photosynthate availability, such as high plant density and shade. To advance our understanding of the mechanisms by which these stresses affect kernel development, we subjected potted maize plants in the greenhouse to water stress and shade from 1 to 9 days after pollination and determined evapotranspiration, root growth in deep pots, carbohydrates and ABA in leaves and kernel tissues, and profiled expression of gene transcripts in ker

Consumption of rice (Oryza sativa) is the major dietary source of cadmium (Cd) for populations with rice as the staple. Little is known about the distribution and chemical speciation of Cd in rice grain, which is critical in determining the bioavailability of Cd to humans. We used synchrotron-based techniques for analyses of the speciation and distribution of Cd in rice grain. The majority of the Cd in rice grain was present as Cd-thiolate complexes (66-92%), likely in the form of Cd bound with thiol-rich proteins. The remainder was present as Cd-carboxyl compounds and Cd-histidine. Elemental

Total-Hg (T-Hg) and methylmercury (MeHg) concentrations in rice grains were measured to understand the MeHg accumulation process. Rice plants were cultivated in Hg2+-spiked non-contaminated soils in experimental pots at three different places. Although soil MeHg concentrations in the pots changed significantly and individually during the rice-growing season, T-Hg concentration of brown rice grain was high at high soil MeHg concentration. In addition, there was no significant variation in T-Hg concentration in brown rice grains from individual panicles or among panicles obtained from the same p

Vitamin A deficiency, a global health burden, can be alleviated through provitamin A carotenoid biofortification of major crop staples such as maize (Zea mays) and other grasses in the Poaceae. If regulation of carotenoid biosynthesis was better understood, enhancement could be controlled by limiting beta-carotene hydroxylation to compounds with lower or no nonprovitamin A activity. Natural maize genetic diversity enabled identification of hydroxylation genes associated with reduced endosperm provitamin A content. A novel approach was used to capture the genetic and biochemical diversity of a

Alcohol-soluble endosperm proteins (prolamins) from some cereals (e.g. wheat, barley, and rye) give origin upon proteolytic digestion to biologically-active antinutritional peptides able to adversely affect in vivo the intestinal mucosa of coeliac patients, whereas prolamins from other cereals (e.g. maize and rice) do not. These antinutritional peptides are also able to: (a) prevent in vitro recovery of atrophic coeliac mucosa; (b) to inhibit differentiation of isolated rat fetal and chick fetal intestines; and (c) to interact with undifferentiated cells either agglutinating them or affecting

During the development of disomic additions of rye (Secale cereale L.) chromosomes to wheat (Triticum aestivum L.), two reverse tandem duplications on wheat chromosomes 3D and 4A were isolated. By virtue of their meiotic pairing, the reverse tandem duplications initiated the chromatid type of the breakage-fusion-bridge (BFB) cycle. This BFB cycle continued through pollen mitoses and in the early endosperm divisions, but no clear evidence of its presence in embryo mitoses was found. The chromosome type of BFB cycle was initiated by fusion of two broken chromosome ends resulting in a dicentric o

OBJECTIVE: To quantify the burden of type 2 diabetes mellitus (T2DM) attributable to dietary risks across Asia.
STUDY DESIGN: Using the Global Burden of Disease Study 2023, deaths and disability-adjusted life years (DALYs) of T2DM attributable to seven dietary risks were evaluated across 34 Asian countries from 1990 to 2023.
RESULTS: In 2023, dietary risks accounted for 9.97 million DALYs and 214,810 deaths from T2DM across Asia. A diet low in fruit, covering fresh, frozen, cooked, canned, or dried fruits, was the largest contributor, accounting for 3.60 million DALYs and 85,960 deaths. Regi

Quantitative imaging of plant tissues by laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) is hindered by the lack of matrix-matched calibration standards. Established approaches, such as gelatine or homogenised tissue blocks, do not replicate plant matrices accurately. Here, we introduce a nano-dispenser-based calibration strategy that deposits nanolitre volumes of elemental standards directly onto paraffin-embedded grain sections, exploiting the low endogenous metal content of the endosperm to generate in situ calibration curves. Calibration performance for Mg, Mn, Cu,

Grain size represents a key agronomic trait and a major determinant of barley grain yield potential. We observed that the grain size of Epichloë bromicola-inoculated barley plants were significantly larger than those of the uninoculated barley plants. However, the mechanisms and functions by which the endophyte fungus E. bromicola modulates barley grain size have not been investigated. In this study, we compared grain size between uninoculated (KE-) and E. bromicola-inoculated (KE+) barley plants to elucidate the association between E. bromicola colonization and barley grain size. Quantitative

Orchid seeds are dust-like and lack endosperm, which limits their capacity to support germination using endogenous nutrient reserves. Successful germination therefore depends on the establishment of a compatible symbiotic association with germination-promoting orchid mycorrhizal fungi (OMF). In plant-root symbioses, host-derived small molecules, such as strigolactones and flavonoids, function as early chemical signals that recruit microbial partners and stimulate their growth. However, the chemical constituents that may facilitate fungal recognition, growth, or colonization during orchid seed

Tetrastigma (Miq.) Planch. is one of the most species-rich genera of the economically and agronomically important grape family Vitaceae. It includes ca. 95 species widely distributed in the tropics and subtropics of Asia and Australia. Species of Tetrastigma exhibit great diversity in both vegetative and reproductive characters. Here we inferred a well-supported phylogeny of Tetrastigma based on ten chloroplast DNA regions with an expanded taxon sampling of 72 species and two varieties. Our molecular results support six major clades within Tetrastigma and the relationships among these clades w

The plasma membrane of the cereal aleurone layer is the site of perception of germination signals and release of enzymes to the starchy endosperm. Analysis of membrane proteins is challenging due to their hydrophobicity and low abundance; thus, little is known about the membrane proteins involved in seed germination. A membrane fraction highly enriched for the plasma membrane H+-ATPase was prepared from barley aleurone layers by aqueous two-phase partitioning. Because detergent and salt washes did not efficiently remove soluble proteins from the membrane preparations, an alternative procedure

Understanding the biochemical networks governing seed longevity is essential for germplasm conservation. Integrating germination potential with biochemical profiling elucidates metabolic transitions during decadal cold storage. As phenotypic evidence of ageing, extended cold storage induces a uniform decline in germination energy, reaching a terminal "floor effect" characterised by the complete exhaustion of repair-and-defence strategies, whilst residual germination capacity averages 21%. Metabolic profiling reveals that prolonged storage causes systemic decoupling of the protein-sugar axis, l

BACKGROUND: A. annua (also named Artemisia annua, sweet wormwood) is the main source of the anti-malarial drug artemisinin, which is synthesised and stored in its trichomes. Members of the basic Helix-Loop-Helix (bHLH) family of transcription factors (TFs) have been implicated in artemisinin biosynthesis in A. annua and in trichome development in other plant species.
RESULTS: Here, we have systematically identified and characterised 226 putative bHLH TFs in A. annua. All of the proteins contain a HLH domain, 213 of which also contain the basic motif that mediates DNA binding of HLH dimers. Of

Tomato (Solanum lycopersicum L.) is one of the major cash crops worldwide. The tomato seed is an important model for studying genetics and developmental biology during plant reproduction. Visualization of finer embryonic structure within a tomato seed is often hampered by seed coat mucilage, multi-cell-layered integument, and a thick-walled endosperm, which needs to be resolved by laborious embedding-sectioning. A simpler alternative is to employ tissue clearing techniques that turn the seed almost transparent using chemical agents. Although conventional clearing procedures allow deep insight

BACKGROUND: Greater yam (Dioscorea alata L.) is a major tropical and subtropical staple crop cultivated for its starchy tubers. Breeding of this dioecious species is hampered by its erratic flowering, yet little is currently known on the genetic determinism of its sexual reproduction.
RESULT: Here we used a genome-wide association approach and identified a major genetic barrier to reproduction in yam on chromosome 1, as represented by two candidate genes. A deleterious effect on male fitness could be hypothesized considering the involvement of these two genes in male reproduction and the low

PREMISE OF THE STUDY: The plant fossil record for the neotropics is still sparse and temporally discontinuous. The location and description of new fossil material are fundamental for understanding evolutionary and biogeographic patterns of lineages. A new fossil record of Passifloraceae from the late Eocene of Colombia is described in this study.
METHODS: Plant fossils were collected from a new locality from the Eocene Esmeraldas Formation. Eighteen fossil seeds were selected, described, and compared with fossil and extant angiosperm seeds based on the literature and herbarium collections. Ta

Residual DNA in recombinant protein pharmaceuticals can potentially cause safety issues in clinical applications; thus, maximum residual limit has been established by drug safety authorities. Assays for residual DNA in Escherichia coli, yeast, and Chinese hamster ovary (CHO) cell expression systems have been established, but no rice residual DNA assay for rice expression systems has been designed. To develop an assay for the quantification of residual DNA that is produced from rice seed, we established a sensitive assay using quantitative real-time polymerase chain reaction (qPCR) based on the

BACKGROUND: Wheat gluten has unique nutritional and technological characteristics, but is also a major trigger of allergies and intolerances. One of the most severe diseases caused by gluten is coeliac disease. The peptides produced in the digestive tract by the incomplete digestion of gluten proteins trigger the disease. The majority of the epitopes responsible reside in the gliadin fraction of gluten. The location of the multiple gliadin genes in blocks has to date complicated their elimination by classical breeding techniques or by the use of biotechnological tools. As an approach to silence

Sulfur metabolism and stress responses are fundamental to plant growth and seed development. ETHE1 encodes a mitochondrial sulfur dioxygenase that plays important roles in these processes; however, its function in Jatropha curcas remains unknown. In this study, we report the cloning and in silico characterization of a putative JcETHE1 gene from J. curcas. Bioinformatic analysis revealed that JcETHE1 has an open reading frame of 726 bp encoding a 241-amino-acid protein, which is predicted to localize in mitochondria and possesses a typical sulfur dioxygenase conserved domain. Because the transc
