Enkephalin-mediated modulation of basal somatic sensitivity by regulatory T cells in mice.

CD4+CD25+Foxp3+ regulatory T cells (Treg) have been implicated in pain modulation in various inflammatory conditions. However, whether Treg cells hamper pain at steady state and by which mechanism is still unclear. From a meta-analysis of the transcriptomes of murine Treg and conventional T cells (Tconv), we observe that the proenkephalin gene (Penk), encoding the precursor of analgesic opioid peptides, ranks among the top 25 genes most enriched in Treg cells. We then present various evidence suggesting that Penk is regulated in part by members of the Tumor Necrosis Factor Receptor (TNFR) fami

Enkephalin related peptides are released from jejunum wall by orally ingested bromelain.

Stem bromelain [EC 3.4.22.32] is a thiol-endopeptidase and orally recommended in traditional medicine due to its analgesic activity, but the mechanisms are not known. Proenkephalin is expressed in the nervous system, but also in the gastrointestinal tract, where it can be assessed by ingested stem bromelain. Here we demonstrated that stem bromelain hydrolyses synthetic proenkephalin fragments after basic amino acid residues flanking the enkephalin sequences. We also observed with in vivo studies that oral administration of bromelain reduced jejunum proenkephalin levels and increased the serum

Influence of leucine-enkephalin on pituitary-ovary axis of the cichlid fish Oreochromis mossambicus.

The present investigation was conducted to elucidate the influence of an opioid peptide, leucine-enkephalin (L-ENK), on the reproductive axis of the tilapia Oreochromis mossambicus. In the first experiment, administration (i.p.) of 25, 100, and 300 μg L-ENK to the stripped female tilapia, for a period of 22 days, resulted in a significantly higher number of stage I follicles compared to those of initial controls and experimental controls, whereas the mean number of stage II and III follicles and serum levels of E2 did not significantly differ among different experimental groups. A significant

Effect of ethanol on hypothalamic opioid peptides, enkephalin, and dynorphin: relationship with circulating triglycerides.

BACKGROUND: Recent evidence has demonstrated that ethanol intake can stimulate the expression and production of the feeding-stimulatory peptide, galanin (GAL), in the hypothalamic paraventricular nucleus (PVN), and that PVN injection of this peptide, in turn, can increase the consumption of ethanol. To test the hypothesis that other feeding-related systems are involved in ethanol intake, this study examined the effect of ethanol on the hypothalamic opioid peptides, enkephalin (ENK), and dynorphin (DYN). METHOD: Adult, male Sprague-Dawley rats were trained to voluntarily drink increasing conce

Lumbar transplantation of immortalized enkephalin-expressing astrocytes attenuates chronic neuropathic pain.

Chronic neuropathic pain is a common symptom in clinical practice and patients with chronic pain are subject to a greatly impaired quality of life. Grafted genetically-modified cells secreting enkephalin have been considered an encouraging treatment for chronic pain. Importantly, the transplanted cell as a therapeutic agent should be reproducible, safe, and controllable. In this study, by combining a tetracycline-controlled (Tet-on) gene expression system and immortalized astrocytes, we attempted to engineer an immortalized astrocyte line carrying the human preproenkephalin gene (IASL/hPPE) un

Changes in brain enkephalin immunostaining after acute carbon disulfide exposure in rats.

Carbon disulfide neurotoxic mechanism in the brain is still not completely clear. In this work, the effect of carbon disulfide exposure in rats on the enkephalinergic neuromodulatory system is described. Caudatus-putamen showed no changes in immunostaining for met-enkephalin when compared with controls. However, a marked reduction in met-enkephalin immunostaining in the central amygdaloid nuclei and the globus pallidus was measured, with a parallel elevation in the lateral septal nucleus and the parietal cortex. It is suggested that enkephalinergic neuromodulatory system could play a role in c

Developmental regulation of leucine-enkephalin expression in adrenal chromaffin cells by glucocorticoids and innervation.

Most catecholaminergic cells derived from the sympathoadrenal lineage of the neural crest contain one or more neuropeptides. Although a great deal is known about the development and regulation of catecholaminergic properties in these cells, relatively little is known about the developmental control of their neuropeptidergic properties. We have investigated the possible role of glucocorticoids and preganglionic innervation in the regulation of leucine-enkephalin (L-Enk) expression in cultures of embryonic and neonatal adrenal chromaffin cells and in mature chromaffin cells in vivo. Exposure of

[The sensitivity of the blood lymphocytes in stomach cancer patients to met-enkephalin, dopamine and serotonin].

Sensitivity of blood lymphocytes to the stimulating effect of met-enkephalin and dopamine was shown to decrease at late stages of the gastric cancer (IIIB-IV) or in the case of metastases to other organs, while to the inhibitory effect of serotonin it increased as compared to early stages (I-IIA). Using specific blockers of opioid, dopamine and serotonin receptors it was shown that these effects were mediated through stimulation of respective type receptors.

Increase of plasma methionine-enkephalin levels in patients at the acute stage of cerebral infarction.

We investigated the participation of endogenous opioid peptide in cerebral ischaemia. In 13 patients with supratentorial infarction, the plasma immunoreactive methionine-enkephalin concentration at the acute stage (30.77 +/- 3.54 pg/ml) was significantly higher than that at the chronic stage (20.37 +/- 2.07 pg/ml) or that of the control subjects (19.64 +/- 1.71 pg/ml). However, it was unrelated to infarct size or patient severity. The increased methionine-enkephalin, which appears to originate from the sympatho-adrenal system or infarcted brain, may play a role in the evolution of cerebral isc

Methionine-enkephalin and thyrotropin-stimulating hormone are intimately related in the human anterior pituitary.

The tissue distribution and function of opioid peptides in humans is incompletely defined. We report here that, unlike that in other species, the human anterior pituitary gland contains high concentrations of methionine-enkephalin (met-enkephalin). The met-enkephalin immunoreactive material was isolated and identified as authentic met-enkephalin by fast atom bombardment-mass spectrometry and Edman degradation sequencing. The met-enkephalin was localized in a large subpopulation of TSH immunoreactive cells (thyrotrophs). No other proenkephalin-derived opioid peptides were found in the pituitary

Effect of aspirin on serotonin and met-enkephalin in brain: correlation with the antinociceptive activity of the drug.

Intravenous administration of acetyl salicylate of lysine, a soluble salt of aspirin, reduced in rats the firing discharge of thalamic neurones, evoked by noxious stimuli. Concomitantly, concentrations of 5-hydroxyindole acetic acid increased, while those of met-enkephalin-like immuno-reactive derivatives were decreased in several areas of the brain. Similar electrophysiological and biochemical responses were obtained by administering tryptophan or 5-hydroxytryptophan plus carbidopa. The effect of aspirin on the evoked firing of the thalamic neurones was counteracted by pretreating the animals

Cardiovascular effects of intraventricular injection of FMRFamide, Met-enkephalin and their common analogues in the rat.

The molluscan neuropeptide, Phe-Met-Arg-Phe-NH2 (FMRFamide), the mammalian opioid peptide met-enkephalin, and their common analogues, met-enkephalin-Arg6-Phe7 (YGGFMRF) and Tyr-Gly-Gly-Phe-Met-Arg-Phe-amide (YGGFMRFamide), were injected into the lateral ventricle of the rat; the cardiovascular effects were studied. FMRFamide caused a rapid, transient elevation in blood pressure accompanied by a great increase in pulse pressure. These effects were followed by secondary increases in blood and pulse pressures. Met-enkephalin produced an initial reduction in blood pressure which was followed by a

Interactions between enkephalin and dopamine in the control of locomotor activity in the rat: a new hypothesis.

D-ala2-met5-enkephalinamide (DALA) was found to be without effect on motility when injected in doses from 10 to 40 micrograms. When ICV injection of DALA was combined with IP injection of amphetamine, DALA markedly enhanced the stimulatory effect of amphetamine. The effects of DALA + amphetamine could be partially antagonized by naloxone. The locomotion-reducing effect of the dopamine antagonist pimozide was not affected by concurrent administration of DALA. These data suggest a complex interaction between opiates and dopamine. It is suggested that the effects of DALA are best explained assumi

Quaternary opiate antagonists lower blood pressure and inhibit leucine-enkephalin responses.

The quaternary opiate antagonists naloxone methylbromide (MB) and naltrexone MB do not cross the blood-brain barrier, and may be used to differentiate peripheral from central nervous system effects of the opioid peptides. When administered intravenously to the conscious, chronically instrumented dog, naloxone MB transiently reduces mean systemic arterial pressure and increases heart rate in a dose-dependent manner over the concentration range from 0.1 to 1.0 mg/kg. The [Leu5]enkephalin response is completely inhibited at naloxone MB doses as low as 0.25 mg/kg, but this inhibition has terminate

[Effect of enkephalins on the energy potential of the rat liver and cerebral cortex during an early period of traumatic shock].

The experiments were performed on 86 white male rats, weighing 180-220 g. The animals with traumatic shock revealed a decrease in ATP concentration and a rise in ADP and AMP levels in the brain cortex and hepatic tissue, resulting in the energy charge reduction. The injection of synthetic leu-enkephalin analog D-ala2-arg6-leu-enkephalin to animals with traumatic shock led to energy charge normalization in the brain cortex and increase in hepatic tissue. The mechanisms of enkephalin effect on the energy metabolism during shock are discussed.

Characterization of a Multiple-Scan-Rate Voltammetric Waveform for Real-Time Detection of Met-Enkephalin.

Opioid peptides are critically involved in a variety of physiological functions necessary for adaptation and survival, and as such, understanding the precise actions of endogenous opioid peptides will aid in identification of potential therapeutic strategies to treat a variety of disorders. However, few analytical tools are currently available that offer both the sensitivity and spatial resolution required to monitor peptidergic concentration fluctuations in situ on a time scale commensurate with that of neuronal communication. Our group has developed a multiple-scan-rate waveform to enable re

Effect of met-enkephalin on chromosomal aberrations in the lymphocytes of the peripheral blood of patients with multiple sclerosis.

Endogenious opiod met-enkephalin throughout previous research manifested cytoprotective and anti-inflammatory effects. Previous research suggests that met-enkephalin has cytogenetic effects. Reducement in the frequency of structural chromosome aberrations as well as a suppressive effect on lymphocyte cell cycle is found. It also reduces apoptosis in the blood samples of the patients with immune-mediated diseases. Met-enkephalin exerts immunomodulatory properties and induces stabilization of the clinical condition in patients with multiple Sclerosis (MS). The goal of the present research was to
