Identification and Thermal Stability Modification of a Zearalenone Lactone Hydrolase ZHD30L.

In this study, a previously uncharacterized zearalenone lactone hydrolase, ZHD30L, from Knufia peltigerae was identified and characterized, and semi-rational design was employed to enhance its thermostability. The results indicate that the optimal reaction conditions for this enzyme are 40 °C and pH 9.0, under which it catalyzes the ring-opening of the ZEN lactone to generate the non-toxic product HZEN. The mutant with five combined sites Mut5, (E11T-A127V-I180K-H235P-V156I), was obtained through semi-rational design combined with multiple rounds of iterative superposition and negative elimina

[Semi-rational evolution of ω-transaminase from Aspergillus terreus for enhancing the thermostability].

ω-transaminase (ω-TA) is a natural biocatalyst that has good application potential in the synthesis of chiral amines. However, the poor stability and low activity of ω-TA in the process of catalyzing unnatural substrates greatly hampers its application. To overcome these shortcomings, the thermostability of (R)-ω-TA (AtTA) from Aspergillus terreus was engineered by combining molecular dynamics simulation assisted computer-aided design with random and combinatorial mutation. An optimal mutant AtTA-E104D/A246V/R266Q (M3) with synchronously enhanced thermostability and activity was obtained. Comp

Advances in Membrane-Bound Catechol-O-Methyltransferase Stability Achieved Using a New Ionic Liquid-Based Storage Formulation.

Membrane-bound catechol-O-methyltransferase (MBCOMT), present in the brain and involved in the main pathway of the catechol neurotransmitter deactivation, is linked to several types of human dementia, which are relevant pharmacological targets for new potent and nontoxic inhibitors that have been developed, particularly for Parkinson's disease treatment. However, the inexistence of an MBCOMT 3D-structure presents a blockage in new drugs' design and clinical studies due to its instability. The enzyme has a clear tendency to lose its biological activity in a short period of time. To avoid the en

Influence of commonly used excipients on the chemical degradation of enalapril maleate in its solid state: The role of condensed water.

The physicochemical stability of enalapril maleate was investigated in the presence of fourteen different excipients divided into four different classes. The extent of a drug-excipient interaction was investigated by following the chemical stability using HPLC. It was found that there is a certain order in the stability of enalapril maleate. Enalapril maleate remained most stable in the presence of: disaccharides > celluloses > starches > superdisintegrants. The amount of degradation can be related to the excipient characteristics. A material with a higher water sorption capacity and lower cry

Formaldehyde regulates tetrahydrofolate stability and thymidylate synthase catalysis.

Tetrahydrofolic acid and formaldehyde are key human metabolites but their physiologically relevant chemistry is undefined. Our NMR studies confirm formaldehyde as a product of tetrahydrofolic acid degradation but also reveal their reaction regulates the stability of tetrahydrofolic acid. These observations identify a novel non-enzymatic feedback mechanism regulating formaldehyde and folate metabolism that has important implications for folate-targeting chemotherapy in cancer and other diseases.

Preparation of antimicrobial biopolymer particles through co-precipitation of hen egg white lysozyme and poly(hydroxy butyrate).

Co-precipitation of poly(hydroxy butyrate), a biobased and biodegradable polymer produced through bacterial fermentation, and hen egg white lysozyme, an enzyme with antimicrobial activity, is a viable route towards non-covalent enzyme immobilization. Ultrasonication during the co-precipitation process results in spherical particles with a well-defined size distribution. The amount of immobilized enzyme can be controlled through variation of its concentration in the antisolvent (aqueous) phase. The presence and density of the enzyme bound to the particles have a decisive influence on the intern

Efficient biomanufacturing of vitamin E succinate in non-aqueous media via synergistic computational evolution and immobilization of lipase.

Biocatalysts exhibit broad application prospects in industrial production. However, the inherent instability of natural enzymes in non-aqueous media severely limits their practical utilization. Using Candida rugosa lipase as a model, a synergistic evolution strategy integrating ancestral sequence reconstruction, free energy optimization and surface hydration shell modulation was developed in this study. The optimal mutant CM5 exhibited simultaneously enhanced stability and activity, showing a 4.78-fold increase in tolerance to 65% (v/v) dimethyl sulfoxide, a 1.67-fold extension in half-life at

Decorating channel walls in ordered macroporous ZIF-8 with hydrophilic PEG to immobilize lipase for efficient chiral resolution.

The development of efficient immobilization support for the enhancement of enzyme activity and recyclability is a highly desirable objective. Single-crystalline ordered macro-microporous ZIF-8 (SOM-ZIF-8), has emerged as a highly effective matrix for enzyme immobilization, however, the inherent hydrophobic nature limits its further advancement. Herein, we have customized the immobilization of the Pseudomonas cepacia lipase (LP) in the modification-channels of SOM-ZIF-8 by functionalizing the inner surface-properties with polyethylene glycol (PEG) (LP@SOM-ZIF-8-PEG), and significant enhancement

Boosting catalytic efficiency of lipase by regulating amphiphilic microenvironment through reversible addition-fragmentation chain transfer polymerized modifications on polyacrylonitrile fiber.

Lipases are increasingly attracting attention in green and sustainable biodiesel production. Currently, the research emphasis lies in immobilizing unstable lipase onto carriers to enhance its performance. Polyacrylonitrile fiber (PANF) is considered to be a promising material for lipase immobilization due to its excellent properties. In this study, functional carriers with regulated surface hydrophobicity were obtained by loading functional groups on PANF via reversible addition-fragmentation chain transfer (RAFT) polymerized modification, and Candida rugosa lipase (CRL) was covalently immobil

Distinctive activation of β-galactosidase by carboxymethylated β-glucan in vitro and mechanism study: Critical role of hydrophobic and electrostatic interactions.

β-galactosidase (lactase) is commercially important as a dietary supplement to alleviate the symptoms of lactose intolerance. This work investigated a unique activation of CMP (carboxymethylated (1 → 3)-β-d-glucan) on lactase and its mechanism by comparing it with carboxymethyl chitosan (CMCS), an inhibitor of lactase. The results illustrated that the secondary and tertiary structures of lactase were altered and its active sites exposed after complexation with CMP, and dissociation of lactase aggregates was also observed. These changes favored better accessibility of the substrate to the activ

Structural and functional characterization of a P-III metalloproteinase, leucurolysin-B, from Bothrops leucurus venom.

Leucurolysin-B (leuc-B) is an hemorrhagic metalloproteinase found in the venom of Bothrops leucurus (white-tailed-jararaca) snake. By means of liquid chromatography consisting of gel filtration on Sephracryl S-200, S-300 and ion-exchange on DEAE Sepharose, leuc-B was purified to homogeneity. The proteinase has an apparent molecular mass of 55kDa as revealed by the reduced SDS-PAGE, and represents approximately 1.2% of the total protein in B. leucurus venom. The partial amino acid sequence of leuc-B was determined by automated Edman sequencing of peptides derived from digests of the S-reduced a

Synthesis and properties of vinyl monomer/enzyme conjugates. Conjugation of L-asparaginase with N-succinimidyl acrylate.

Monomer conjugation of an enzyme followed by copolymerization with free monomer is a useful method of enzyme immobilization. L-asparaginase was conjugated with N-succinimidyl acrylate. Analysis of the conjugated enzyme via isoelectric focusing showed that a molar ratio of 9.5 free monomers per enzyme was needed during the conjunction for each vinyl group bound. Only 3% of the enzyme activity was lost per vinyl group added, and conjugation of an average of four monomers per enzyme thermally destabilized the enzyme only at temperatures above 50 degrees C. Activity of the enzyme at physiological

SH3-like domains from Leuconostoc citreum ABK-1 alternansucrase enables efficient enzyme immobilization.

Enzyme immobilization is generally used to improve enzyme stability and reusability for industrial applications. In this study, a novel non-covalent immobilization strategy was developed using the SH3-like domains of alternansucrase from Leuconostoc citreum ABK-1 (LcAlts). SH3-like domains contain hydrophobic surface, suggesting its ability to interact with hydrophobic materials. Based on this property, LcAlts was successfully immobilized onto Phenyl Sepharose™ beads through hydrophobic interactions without the addition of anti-chaotropic salts. The immobilized enzyme exhibited strong binding

Immobilization of β-Agarase onto Ni-NTA Magnetic Nanoparticles for Efficient Production of Functional Neoagarooligosaccharides.

AgaDcat is a promising β-agarase for the production of functional neoagarooligosaccharides (NAOSs), but its limited thermal stability and poor reusability as a free enzyme increase enzyme consumption and production cost during repeated agarose hydrolysis. In this study, β-agarase AgaDcat was immobilized on nickel-nitrilotriacetic acid magnetic nanoparticles (Ni-NTA-MNPs) through His-tag/Ni-NTA affinity interactions. The successful preparation of Ni-NTA-MNPs and the immobilization of AgaDcat were verified by SEM, EDS, VSM, FT-IR, XRD, and TGA. Following optimization, the immobilized enzyme exhi
