The Emerging Role of Cellular Mechanical Memory In Aqueous Humor Outflow Dysfunction In Glaucoma.

Emerging evidence suggests that cells can retain information about previous mechanical environments and subsequently exhibit altered phenotypes even after the original mechanical stimulus is removed. This phenomenon is known as cellular mechanical memory that occurs in three phases: memory acquisition, storage, and recall. The literature supports a model in which mechanical experiences become encoded through interconnected cytoskeletal, nuclear, metabolic, and epigenetic mechanisms. Mechanical memory is increasingly recognized as a central regulator of cell fate and tissue remodeling, as well

High-salt diet in macrophage-associated metabolic disorders: Mechanisms and therapeutic implications.

High-salt diet (HSD) has emerged as a prevalent environmental factor that exacerbates chronic inflammation and insulin resistance in obesity-associated type 2 diabetes (T2D) by modulating macrophage polarization, metabolic reprogramming, and epigenetic imprinting. Current evidence demonstrates that HSD activates p38/mitogen-activated protein kinase (MAPK), nuclear factor kappa-B (NF-κB), and NOD-like receptor family pyrin domain containing 3 (NLRP3) inflammasome signaling pathways, by which it drives macrophage polarization toward a proinflammatory M1 phenotype while inducing a glycolysis-domi

The biological memory buffer hypothesis: can programmable nanocarriers shape cellular memory to improve the durability of mRNA vaccines and gene therapies?

INTRODUCTION: Messenger RNA vaccines and gene therapies enable rapid and programmable biological intervention, but their therapeutic durability remains variable. Existing nanomedicine research primarily evaluates delivery efficiency, targeting, RNA protection and early protein expression, which do not fully explain why some biological effects persist while others rapidly decline. METHODS: This Hypothesis and Theory article presents a narrative conceptual synthesis of peer-reviewed literature on lipid nanoparticle delivery, mRNA vaccinology, RNA therapeutics, genome editing, trained innate imm

Impaired CpG Demethylation in Common Variable Immunodeficiency Associates With B Cell Phenotype and Proliferation Rate.

Common Variable Immunodeficiency (CVID) is characterized by impaired antibody production and poor terminal differentiation of the B cell compartment, yet its pathogenesis is still poorly understood. We first reported the occurrence of epigenetic alterations in CVID by high-throughput methylation analysis in CVID-discordant monozygotic twins. Data from a recent whole DNA methylome analysis throughout different stages of normal B cell differentiation allowed us to design a new experimental approach. We selected CpG sites for analysis based on two criteria: one, CpGs with potential association wi

Associations between mechanical stress and epigenetic modifications in outer retinal layer remodeling in high myopia.

High myopia is characterized not only by a progressive increase in refractive error but also, more critically, by its significant association with retinal degenerative diseases, posing a considerable public health concern. An increasing amount of research illustrates the role of several epigenetic regulatory mechanisms-including DNA methylation, modifications of histones, non-coding RNAs, and additional epigenetic elements-in the onset and advancement of high myopia. This condition primarily results from the pathological elongation of the eye axis, which alters the mechanical stress on the fun

HDAC inhibitor treatment restores transcriptome, chromatin accessibility, and memory deficits in a mouse model for Down syndrome.

Down syndrome (DS) is associated with widespread molecular alterations, including aberrant gene expression, which not only affects chromosome 21 genes but perturbs the whole transcriptome. The genome-wide nature of this alteration suggests that it might be epigenetically mediated, which may also offer therapeutic opportunities given the reversibility of epigenetic modifications. Here, we show that decreased global acetylation level in the hippocampus of the trisomic mouse model Ts65Dn is associated with decreased chromatin accessibility at gene promoters in hippocampal neurons. Strikingly, pha

[Epigenetics in Parkinson's Disease].

The genetic information encoded in the DNA sequence provides a blueprint of the entire organism. The epigenetic modifications, in particular DNA methylation and histone modifications, determine how and when this information is made available and define the specific gene transcription pattern of a given cell. Epigenetic modifications determine the functional differences of genetically identical cells in multicellular organisms and are important factors in various processes from embryonic development to learning and memory consolidation. DNA methylation patterns are altered by environmental cond

Tissue-resident memory T cells in atopic dermatitis: mechanisms of residual inflammation, relapse, and therapeutic persistence.

BACKGROUND: Atopic dermatitis (AD) is a chronic, relapsing inflammatory skin disease in which disease flares frequently recur at previously affected anatomical sites. This distinctive clinical pattern suggests the presence of residual inflammation and long-lasting local immune memory that persists beyond apparent clinical remission. OBJECTIVES: To synthesize current evidence on the role of tissue-resident memory T (TRM) cells in the pathogenesis, chronicity, and relapse of AD, and to discuss how TRM biology may explain disease recurrence after treatment withdrawal and inform future therapeuti

Epigenetic Clock Trajectories and Brain Health in Midlife.

BACKGROUND: Accelerated biological aging can be assessed with DNA methylation (DNAm)-based epigenetic clocks. Research suggests that greater DNAm is associated with faster cognitive decline and risk of Alzheimer disease (AD) and other dementias. However, most studies have relied on single-time-point measurements of clocks, rather than evaluating dynamic changes over time. We examined the association between 15-year epigenetic aging trajectories and brain health outcomes in midlife. METHODS: We analyzed 2,833 middle-aged adults (mean baseline age 40 years, 59% female and 44% Black) with ≥ 3 Du

Unveiling the Therapeutic Promise of Epigenetics in Vascular Cognitive Impairment and Vascular Dementia.

Vascular dementia (VaD) is a progressive neurodegenerative disease characterized by cognitive decline and memory deficits. Despite its significant prevalence and impact, the pathophysiology of VaD remains poorly understood, and current treatments are limited to symptom management. Emerging evidence highlights the importance of lifestyle-associated risk factors in VaD, emphasizing the role of gene-environment interactions, particularly in the realm of epigenetics. While preclinical studies using animal models have provided valuable insights into epigenetic mechanisms, the translatability of the

Increased epigenetic alterations at the promoters of transcriptional regulators following inadequate maternal gestational weight gain.

Epigenetic modifications are thought to serve as a memory of exposure to in utero environments. However, few human studies have investigated the associations between maternal nutritional conditions during pregnancy and epigenetic alterations in offspring. In this study, we report genome-wide methylation profiles for 33 postpartum placentas from pregnancies of normal and foetal growth restriction with various extents of maternal gestational weight gain. Epigenetic alterations accumulate in the placenta under adverse in utero environments, as shown by application of Smirnov-Grubbs' outlier test.

Epigenetic modification and therapeutic targets of diabetes mellitus.

The prevalence of diabetes and its related complications are increasing significantly globally. Collected evidence suggested that several genetic and environmental factors contribute to diabetes mellitus. Associated complications such as retinopathy, neuropathy, nephropathy and other cardiovascular complications are a direct result of diabetes. Epigenetic factors include deoxyribonucleic acid (DNA) methylation and histone post-translational modifications. These factors are directly related with pathological factors such as oxidative stress, generation of inflammatory mediators and hyperglycemi

Epigenetics in women's health care.

Epigenetics refers to structural modifications to genes that do not change the nucleotide sequence itself but instead control and regulate gene expression. DNA methylation, histone modification, and RNA regulation are some of the mechanisms involved in epigenetic modification. Epigenetic changes are believed to be a result of changes in an organism's environment that result in fixed and permanent changes in most differentiated cells. Some environmental changes that have been linked to epigenetic changes include starvation, folic acid, and various chemical exposures. There are periods in an org

Hyperglycaemia-induced metabolic stress and epigenetic imprinting in the inflammatory pathogenesis of diabetic neuropathy.

Diabetic neuropathy (DN), a major microvascular complication of diabetes mellitus, results from a complex interplay among oxidative stress, inflammation, and persistent epigenetic modifications. Hyperglycemia-induced mitochondrial dysfunction increases reactive oxygen species (ROS), which activate redox-sensitive inflammatory cascades, including NF-κB, JAK/STAT, and the NLRP3 inflammasome. These pathways amplify cytokine release and neuronal sensitisation, while reciprocal feedback between ROS and inflammation mediated by Nrf2 suppression further perpetuates nerve damage. Damage-associated mol

Monozygotic twins discordant for common variable immunodeficiency reveal impaired DNA demethylation during naïve-to-memory B-cell transition.

Common variable immunodeficiency (CVID), the most frequent primary immunodeficiency characterized by loss of B-cell function, depends partly on genetic defects, and epigenetic changes are thought to contribute to its aetiology. Here we perform a high-throughput DNA methylation analysis of this disorder using a pair of CVID-discordant MZ twins and show predominant gain of DNA methylation in CVID B cells with respect to those from the healthy sibling in critical B lymphocyte genes, such as PIK3CD, BCL2L1, RPS6KB2, TCF3 and KCNN4. Individual analysis confirms hypermethylation of these genes. Anal

Epigenetic control of the Epstein-Barr lifecycle.

Epstein-Barr virus (EBV) infects 95% of adults worldwide, causes infectious mononucleosis, is etiologically linked to multiple sclerosis and is associated with 200 000 cases of cancer each year. EBV manipulates host epigenetic pathways to switch between a series of latency programs and to reactivate from latency in order to colonize the memory B-cell compartment for lifelong infection and to ultimately spread to new hosts. Here, we review recent advances in the understanding of epigenetic mechanisms that control EBV latency and lytic gene expression in EBV-transformed B and epithelial cells. W

OxLDL-mediated immunologic memory in endothelial cells.

Trained innate immunity describes the metabolic reprogramming and long-term proinflammatory activation of innate immune cells in response to different pathogen or damage associated molecular patterns, such as oxidized low-density lipoprotein (oxLDL). Here, we have investigated whether the regulatory networks of trained innate immunity also control endothelial cell activation following oxLDL treatment. Human aortic endothelial cells (HAECs) were primed with oxLDL for 24 h. After a resting time of 4 days, cells were restimulated with the TLR2-agonist PAM3cys4. OxLDL priming induced a proinflamma

Development of innate immune memory by non-immune cells during Staphylococcus aureus infection depends on reactive oxygen species.

INTRODUCTION: The mechanisms underlying innate immune memory (trained immunity) comprise epigenetic reprogramming of transcriptional pathways associated with alterations of intracellular metabolism. While the mechanisms of innate immune memory carried out by immune cells are well characterized, such processes in non-immune cells, are poorly understood. The opportunistic pathogen, Staphylococcus aureus, is responsible for a multitude of human diseases, including pneumonia, endocarditis and osteomyelitis, as well as animal infections, including chronic cattle mastitis that are extremely difficul

Targeting the cytokine-epigenetic axis: a new paradigm and prospects for disease treatment.

Cytokines, as key signaling molecules, are involved in the regulation of physiological and pathological processes such as inflammation, immunity, and cell growth. Epigenetic mechanisms, including DNA methylation, histone modifications, chromatin remodeling, and non-coding RNAs, enable precise control of gene expression without changing the DNA sequence. Recent studies have revealed that cytokines interact with epigenetic regulation to form a dynamic and complex "cytokine-epigenetic axis". Through metabolic reprogramming and regulation of epigenetic enzyme activity, this axis affects gene expre

Epigenetic modifications in type 2 diabetes mellitus.

Type 2 diabetes mellitus (T2DM) develops through the interplay of insulin resistance and progressive beta-cell dysfunction. Beyond genetic predisposition, epigenetic regulation provides a mechanistic link between environmental exposures (e.g., overnutrition, physical inactivity, inflammation) and durable changes in gene expression in pancreatic islets and insulin-sensitive tissues (skeletal muscle, adipose tissue, and liver). Accordingly, this review aims to synthesize current evidence on the epigenetic mechanisms underlying T2DM and its complications, with a particular focus on epigenetically
