Investigation of the putative immunodominant T cell epitopes in coeliac disease.

BACKGROUND: Coeliac disease (CD) is an enteropathy mediated by gluten specific T cells which secrete interferon gamma (IFN-gamma) when stimulated by gluten peptides presented by HLA-DQ2 or DQ8 molecules. Residues 62-75 of alpha(2) gliadin have been proposed as the immunodominant epitope in the majority of CD patients. Deamidation by tissue transglutaminase (tTG) of the glutamine (Q) at position 65 to glutamic acid (E) is essential for T cell stimulation. AIMS: To investigate the antigenicity of this peptide and to establish whether its T cell activating properties can be downregulated by the

Tau epitopes in spinal cord neurofibrillary lesions in Chamorros of Guam.

We examined spinal cord sections from Guamanian Chamorros with or without amyotrophic lateral sclerosis or parkinsonism-dementia complex using immunohistochemistry and antibodies to epitopes that span the length of tau to characterize the tau epitope profile of neurofibrillary tangles in these spinal cords. Most (16/20) spinal cords, including some from Chamorros without documented clinical disease, contained tangles with a tau epitope profile similar to the tangles found in the forebrain and brain stem of these patients.

Thyrotropin receptor epitopes recognized by graves' autoantibodies developing under immunosuppressive therapy.

Abnormal modulation of the immune system is a prerequisite for the expression of Graves' disease. Thus, when hyperthyroidism developed in a renal transplant recipient under long term immunosuppression with cyclosporine A and prednisone, we carefully evaluated the basis for her hyperthyroidism and her state of immunosuppression. Immunosuppression was confirmed by finding markedly deficient lymphocyte responses to common mitogens. Lymphocyte phenotype frequencies were those previously found in Graves', i.e. elevated frequencies of CD3/DR, CD5/26, and CD3/25 lymphocytes. There was also reversal o

Identification of murine protective epitopes on the Porphyromonas gingivalis fimbrillin molecule.

Fimbriae from Porphyromonas gingivalis are believed to play an important role in the pathogenesis of periodontal diseases. The aim of the present study was to identify the fimbrial protective T-cell epitopes in CBA/J mice. A truncated protein corresponding to amino acids 1 to 198, PgF1-198, was generated and allowed us to demonstrate that the N terminus of the protein contains T-cell epitopes. With synthetic peptides, an immunodominant sequence was identified between amino acids 103 and 122. The corresponding peptide, PgF-P8, induced T-cell proliferation after in vitro restimulation of in vivo

Immunoreactive epitopes on an expressed recombinant flagellar protein of Borrelia burgdorferi.

A recombinant Borrelia burgdorferi flagellin protein expressed in Escherichia coli is bound by a murine monoclonal antiflagellin antibody (H9724) and by antibodies in the sera of patients with Lyme disease. Immunoreactive epitopes on the flagellar protein were identified by immunoblot analysis of antibody binding to expressed truncated flagellar proteins. The epitope recognized by the murine monoclonal antibody is within the central heterologous region of the flagellar protein (amino acids 90 to 266). However, antiflagellin antibodies in the sera of patients with Lyme arthritis bound an epitop

Immunoblotting of Creutzfeldt-Jakob disease prion proteins: host species-specific epitopes.

Creutzfeldt-Jakob disease (CJD) is a rare dementia that is generally found in older people and is caused by unusual infectious pathogens or prions. Using rabbit antisera raised against hamster scrapie prion proteins (HaPrPSc), we identified by immunoblotting human CJD prion proteins (HuPrPCJD) in the brains of 14 patients dying of CJD. Extracts from 6 of the patients were transmitted to mice after prolonged incubation. The rabbit antisera raised against HaPrPSc also reacted with the mouse CJD prion proteins (MoPrPCJD) found in the brains of these experimentally infected mice. When mice were im

A T-Cell Epitope-Based Multi-Epitope Vaccine Designed Using Human HLA Specific T Cell Epitopes Induces a Near-Sterile Immunity against Experimental Visceral Leishmaniasis in Hamsters.

Visceral leishmaniasis is a neglected tropical disease affecting 12 million people annually. Even in the second decade of the 21st century, it has remained without an effective vaccine for human use. In the current study, we designed three multiepitope vaccine candidates by the selection of multiple IFN-γ inducing MHC-I and MHC-II binder T-cell specific epitopes from three previously identified antigen genes of Leishmania donovani from our lab by an immuno-informatic approach using IFNepitope, the Immune Epitope Database (IEDB) T cell epitope identification tools, NET-MHC-1, and NET MHC-2 webs

Second-line treatment after docetaxel, cisplatin and 5-fluorouracil in metastatic squamous cell carcinomas of the anus. Pooled analysis of prospective Epitopes-HPV01 and Epitopes-HPV02 studies.

BACKGROUND: Squamous cell carcinoma of the anus (SCCA) is a rare disease often diagnosed at a localised stage. For locally advanced recurrence or metastatic disease, DCF (docetaxel, cisplatin, 5-fluorouracil) demonstrated high efficacy and became one of the standard regimens. However, there is no standard of care in the second line. PATIENTS AND METHODS: In the Epitopes-HPV01 and Epitopes-HPV02 prospective trials, 115 patients with advanced SCCA were treated with a DCF regimen in the first line. In these studies, second-line data were registered per protocol. RESULTS: After a median follow-u

Replacing the decoy epitope of PCV2 capsid protein with epitopes of GP3 and/or GP5 of PRRSV enhances the immunogenicity of bivalent vaccines in mice.

Porcine circovirus type 2 (PCV2) is the causative agent of postweaning multisystemic wasting syndrome (PMWS), porcine dermatitis and nephropathy syndrome (PDNS), and reproductive failure and causes economic losses in the domestic swine industry. The decoy epitope (169-180 amino acid (aa)) of the PCV2 capsid (Cap) protein is an immunodominant epitope and diverts the immune response away from protective epitopes. The mixed infection of PCV2 and porcine reproductive and respiratory syndrome virus (PRRSV) is one of the most common co-infections in the pig industry and shows more severe clinical sy

Frequent associations between CTL and T-Helper epitopes in HIV-1 genomes and implications for multi-epitope vaccine designs.

BACKGROUND: Epitope vaccines have been suggested as a strategy to counteract viral escape and development of drug resistance. Multiple studies have shown that Cytotoxic T-Lymphocyte (CTL) and T-Helper (Th) epitopes can generate strong immune responses in Human Immunodeficiency Virus (HIV-1). However, not much is known about the relationship among different types of HIV epitopes, particularly those epitopes that can be considered potential candidates for inclusion in the multi-epitope vaccines. RESULTS: In this study we used association rule mining to examine relationship between different typ

Genetic Variation of HPV53 and the Identification of T-Cell Epitopes.

Human papillomavirus type 53 (HPV53) is one of the most prevalent HPV genotypes in China, frequently detected in cervical intraepithelial neoplasia and cervical cancer, yet remains outside the coverage of all currently available prophylactic vaccines and is relatively understudied. This study performed a comprehensive analysis of HPV53 clinical infection profiles, genomic diversity, and T-cell epitopes to inform therapeutic vaccine development. Clinical analysis of 158 HPV53-positive patients showed that infections were most prevalent in women aged 40-59 years, with persistent infection identi

Epstein-Barr virus gp350 neutralizing epitopes define distinct sites of vulnerability.

Epstein-Barr virus (EBV) is a widespread human herpesvirus linked to several malignancies, yet no vaccine has been approved. Its major surface glycoprotein, gp350, mediates attachment to B cells via CR2 binding, but gp350-based vaccines have shown limited protection, raising questions about its neutralizing epitopes and mechanisms. Here, we isolated human monoclonal antibodies from a phage-display library derived from EBV-seropositive donors. Several antibodies showed potent neutralization, including 1A12, which conferred robust protection in a humanized mouse model by markedly reducing viral

Anti-idiotypic antibodies as molecular mimics of conserved influenza hemagglutinin epitopes.

Broadly neutralizing antibodies (bNAbs) provide insights for developing universal influenza vaccines through their ability to neutralize diverse viral strains. However, designing antigens that reliably elicit bNAb responses remains challenging due to the difficulty in targeting conserved epitopes with a single antigen. We developed a method using anti-idiotypic antibodies (IDs) to generate antigens that induce cross-reactive antibodies against diverse influenza A and B hemagglutinins. Our approach leverages IDs that mimic the conformational epitopes targeted by bNAbs. Structural analysis revea

Broadly conserved protective epitopes on the lyme disease vaccine antigen, OspA.

Lyme disease, caused by the spirochete, Borrelia burgdorferi sensu latu (Bbsl), is a tickborne infection of increasing incidence in North America, Europe and Asia. While vaccines based on Outer surface protein A (OspA) have proven highly efficacious at blocking Bbsl tick-to-human transmission, the high degree of antigenic variability among the major OspA serotypes (ST) has made the development of a broadly cross protective vaccine difficult. Recent profiling of protective human monoclonal antibodies (mAbs) has suggested the existence of conserved epitopes situated within OspA's central β-sheet

Discover Potential New Epitopes through Post-Translational Modification in Sjögren's Disease.

Sjögren's syndrome (SjD) is a chronic autoimmune disorder in which the immune system attacks the glands that produce tears and saliva, leading to symptoms such as dry eyes and dry mouth. If left untreated, SjD can also cause inflammation and damage to other parts of the body, including the skin, lungs, kidneys, and nervous system, and increase the risk of developing lymphoma. The human leukocyte antigen (HLA) class II molecule HLA-DR3 is strongly associated with SjD. To investigate how post-translational modifications (PTMs) influence the presentation of SjD-associated autoantigens by HLA-DR3,

Automated Glycan Assembly of Porphyromonas gingivalis Lipopolysaccharide Epitopes for Vaccine Design.

Porphyromonas gingivalis (P. gingivalis) is the major cause of chronic periodontitis and is associated with systemic diseases, such as rheumatoid arthritis. A better understanding of the interplay of the human immune system and P. gingivalis lipopolysaccharide (LPS) may result in novel treatment and prevention strategies. Twelve LPS fragments of P. gingivalis were synthesized using automated glycan assembly, with the help of remote benzoyl ester participation to introduce 1,2-cis-glycosidic linkages. A deep learning model for predicting systemic lupus erythematosus-associated epitopes.

BACKGROUND: The accurate prediction of epitopes associated with Systemic Lupus Erythematosus (SLE) plays a vital role in advancing our understanding of autoimmune pathogenesis and in designing effective immunotherapeutics. Traditional bioinformatics methods often struggle to capture the intricate sequence patterns and high-dimensional signals characteristic of epitope data. Deep learning presents a compelling alternative, with its ability to perform automatic feature learning and model complex dependencies inherent in biological sequences. This study proposes a hybrid deep learning architectur
