Calcific aortic valve disease is common in older adults and a major cause of aortic stenosis. Despite its clinical impact, there are still no effective drugs that slow disease progression, and treatment largely relies on surgical or transcatheter valve replacement once stenosis becomes severe and symptomatic. Beyond established roles for inflammation and tissue remodeling, recent evidence suggests that chemical modifications of RNA may influence how valve cells shift toward fibro-calcific states. In this study, we compared human valvular interstitial cells isolated from non-calcified valves an

Chemical modifications of RNA molecules, collectively referred to as epitranscriptomics, have emerged as a critical layer of post-transcriptional gene regulation. Among these modifications, N6-methyladenosine (m6A) plays a key role in modulating RNA stability, localization, and translational efficiency without altering the underlying nucleotide sequence. In recent years, epitranscriptomics has been increasingly recognized for its involvement in mammalian reproductive processes. In mammalian oocytes, which undergo an extended period of transcriptional quiescence and rely predominantly on post-t

METTL3 is a key regulator of RNA metabolism, yet its genomic and epitranscriptomic roles in tissue development are largely unexplored. Using embryonic stem cell-derived 3D retinal organoids to model retinal progenitor cell (RPC) differentiation, we integrated transcriptome-wide m6A profiling (GLORI), protein-DNA (chromatin immunoprecipitation sequencing [ChIP-seq] and CUT&RUN) and chromatin accessibility (ATAC-seq) mapping, and targeted m6A engineering (dCas13b-FTO) to dissect METTL3 function. Loss of METTL3 nuclear m6A activity disrupted Rx+ retinal anlage formation in vitro, with dCas13b-FTO

BACKGROUND: The epitranscriptome, particularly N6-methyladenosine (m6A), represents a dynamic layer of post-transcriptional regulation fundamentally implicated in cancer. However, the clinical translation of this knowledge is hampered by profound context-dependency, where the same m6A regulator can exert opposing roles in different tumors. To overcome this barrier, we propose a novel, clinically actionable taxonomic framework that classifies tumors based on their dominant dysregulated m6A component.
METHODS: We synthesized current evidence through systematic reviews of primary research and hi

Ovarian aging leads to follicular atresia and a decline in both the quantity and quality of oocytes, yet its molecular regulatory mechanisms remain incompletely understood. N6-methyladenosine (m6A), one of the most prevalent RNA modifications in mammals, is thought to play a key role in ovarian function and aging. In this study, ovarian tissues from 1–2-year-old and 5–6-year-old Qira black sheep were collected, and an m6A methylation transcriptomic atlas was constructed based on MeRIP-seq and RNA-seq. A total of 660 differential methylation peaks (DMPs) and 2,018 differentially expressed genes

Polyphenism, the capacity of a single genotype to produce discrete phenotypes in response to environmental cues, has long served as a model for understanding developmental plasticity. While classical studies have highlighted hormonal signalling and transcription-factor-driven gene regulatory networks that mediate these environmentally induced shifts, recent advances have uncovered a new layer of gene regulation through chemical modifications of RNA molecules. These RNA modifications, collectively known as epitranscriptomic marks, have been shown to respond to environmental signals and modulate

Approximately 50% of melanoma patients carry a mutation in the BRAF gene, and over 90% of these mutations lead to the substitution of valine 600 with glutamic acid (V600E). Vemurafenib is an FDA-approved kinase inhibitor for BRAFV600E; while the drug elicits effective remission of metastatic melanoma, relapse typically occurs within several months after therapy. Recent studies documented critical roles of reversible modifications in RNA in modulating resistance to cancer therapy. Herein we explored the contributions of epitranscriptomic alterations to vemurafenib resistance by assessing the di

Toxoplasma gondii is an obligate intracellular parasite that can cause serious opportunistic disease in the immunocompromised or through congenital infection. To progress through its life cycle, Toxoplasma relies on multiple layers of gene regulation that includes an array of transcription and epigenetic factors. Over the last decade, the modification of mRNA has emerged as another important layer of gene regulation called epitranscriptomics. Here, we report that epitranscriptomics machinery exists in Toxoplasma, namely the methylation of adenosines (m6A) in mRNA transcripts. We identified nov

Multiple myeloma (MM) remains largely incurable despite major therapeutic advances, underscoring the need to define novel pathogenic mechanisms and druggable targets. Epitranscriptomic dysregulation, encompassing reversible chemical modifications on RNA, has emerged as a post-transcriptional regulatory layer that may contribute to MM biology. This focused review discusses the emerging roles of major RNA modifications and their regulators in MM pathogenesis, bone disease, drug resistance, and immune escape. We summarize representative experimental and translational studies on RNA-modifying enzy

Epitranscriptomic modifications, particularly RNA methylations, have emerged as regulators of gene expression, with their dysregulation acting as a key factor in tumorigenesis and metastatic progression. This review evaluates the therapeutic landscapes of N6-methyladenosine (m6A) and N1-methyladenosine (m1A) modifications in cancer. While the m6A machinery predominantly dictates mRNA turnover and stability, the m1A network is uniquely positioned to drive translational reprogramming, allowing malignant cells to endure severe microenvironmental stress and evade cell death. Despite positional and

Gram-negative bacteria are clinically significant pathogens responsible for life-threatening infections, including respiratory infections. These can be acute or persistent and can exacerbate existing chronic diseases, such as cystic fibrosis, COPD and lung cancer. In this review, we use Pseudomonas aeruginosa as a model organism that demonstrates the molecular complexity of host-pathogen interactions during lower airway infection. Specifically, we focus on RNA modifications and show that they, on the one hand, regulate bacterial fitness and pathogenicity, and on the other control the execution

The growing body of research on cardiac epitranscriptomic factors has underscored their potential biological roles and impact on heart development and disease. While transcriptional and translational regulation of cardiac genes in the developing heart has been extensively studied, the potential roles of posttranscriptional regulation of cardiac mRNAs remain a significant gap in our understanding. In addition, discrepancies between transcriptomes and proteomes in both embryonic and adult hearts are well recognized, further suggesting an emerging regulatory role of the epitranscriptome in cardia

The pathogenesis of metabolic dysfunction-associated steatotic liver disease (MASLD) remains unclear due to the complexity of its etiology. The emerging field of the epitranscriptome has shown significant promise in advancing the understanding of disease pathogenesis and developing new therapeutic approaches. Recent research has demonstrated that N4-acetylcytosine (ac4C), an RNA modification within the epitranscriptome, is implicated in progression of various diseases. However, the role of ac4C modification in MASLD remains unexplored. Herein, we performed acRIP-ac4c-seq and RNA-seq analysis i

BACKGROUND: The epigenetic modifications play important regulatory roles in tissue development, maintenance of physiological functions and pathological process. RNA methylations, including newly identified m1A, m5C, m6A and m7G, are important epigenetic modifications. However, how these modifications are distributed in the transcriptome of vertebrate brains and whether their abundance is altered under pathological conditions are still poorly understood. In this study, we chose the model animal of zebrafish to conduct a systematic study to investigate the mRNA methylation atlas in the brain.

Biomolecular condensates are dynamic membraneless organelles composed of proteins and RNAs that assemble through multivalent interactions and provide cells with powerful means to regulate gene expression in space and time. Different epitranscriptomic marks such as m6A, m1A, and m5C can reshape RNA structure-binding interfaces and multivalency and, in this manner, tuning which transcripts nucleate or partition into specific condensates and influencing their material state. This review summarizes how individual RNA modifications and their associated proteins regulate the formation and function o

Epitranscriptomic modifications have recently emerged into the spotlight of researchers due to their vast regulatory effects on gene expression and thereby cellular physiology and pathophysiology. N6,2'-O-dimethyladenosine (m6Am) is one of the most prevalent chemical marks on RNA and is dynamically regulated by writers (PCIF1, METTL4) and erasers (FTO). The presence or absence of m6Am in RNA affects mRNA stability, regulates transcription, and modulates pre-mRNA splicing. Nevertheless, its functions in the heart are poorly known. This review summarizes the current knowledge and gaps about m6Am

Cancer was initially considered to be an exclusively genetic disease, but an interplay of dysregulated genetic and epigenetic mechanisms is now known to contribute to the cancer phenotype. More recently, chemical modifications of RNA molecules - the so-called epitranscriptome - have been found to regulate various aspects of RNA function and homeostasis. Specific enzymes, known as RNA-modifying proteins (RMPs), are responsible for depositing, removing, and reading chemical modifications in RNA. Intensive investigations in the epitranscriptomic field in recent years, in conjunction with great te

Although aerobic glycolysis is a hallmark of hepatocellular carcinoma (HCC) progression, the underlying epitranscriptomic mechanisms remain poorly understood. Here, we identify METTL15, an N4-methylcytidine (m4C) RNA methyltransferase, as a novel oncogenic driver of this metabolic reprogramming. METTL15 is significantly upregulated in HCC tissues and cell lines, with high expression predicting poor clinical outcomes. Functional assays demonstrate that METTL15 promotes HCC cell proliferation and metastasis both in vitro and in vivo. Mechanistically, METTL15 interacts with c-Myc mRNA and modulat

Retinal ischemia-reperfusion injury (RIR) is the main pathogenic mechanisms of acute glaucoma, diabetic retinopathy, central retinal vein occlusion. As a common post-transcriptional modification of eukaryotic RNAs, N6-methyladenosine (m6A) is associated with the pathogenesis of different diseases, including angiogenesis, through the regulation of RNA metabolism and functions. The aim of this study was to identify the potential relevance of m6A RNA methylation in pathogenesis of RIR. A total of 10,851 mRNAs and 23,270 associated m6A methylation modified peaks were identified in the RIR group. S

INTRODUCTION: Common variable immunodeficiency (CVID) is frequently complicated by duodenal inflammation, but the underlying molecular mechanisms remain poorly understood. While epigenetic alterations have been described in CVID, the epitranscriptome is largely unexplored. We therefore investigated whether RNA N6-methyladenosine (m6A) modifications in duodenal tissue are altered in CVID and whether such changes are associated with the local microbiota or m6A-related enzymes.
METHODS: m6A modification levels were analysed in snap-frozen duodenal biopsies from CVID patients with intraepithelial
