Requirement for etoposide in the treatment of pregnancy related hemophagocytic lymphohistiocytosis: a multicenter retrospective study.

BACKGROUND: Hemophagocytic lymphohistiocytosis (HLH) is a rare severe clinical syndrome. HLH manifesting during pregnancy has been paid much attention in recent years. Despite the specificity of pregnancy-related HLH, there has not been any consensus regarding its treatment. According to a previous study, corticosteroid/IVIG is the mainstream therapy; however, the efficacy is controversial. Etoposide is an important agent in the HLH-94 regimen; nevertheless, its use is limited because of possible toxicity to the fetus. METHODS: In this study, we summarized 13 cases from 4 medical institutions

Redefining the role of etoposide in first-line treatment of peripheral T-cell lymphoma.

Peripheral T-cell lymphomas (PTCLs) have an aggressive biological course and poor clinical outcomes. Despite producing somewhat less-than-satisfactory results, the combination of cyclophosphamide, doxorubicin, vincristine, and prednisone (CHOP) remains the de facto standard in PTCL treatment. Addition of etoposide to CHOP backbone to overcome such unsatisfactory results has yielded contradictory information. We aimed to thoroughly analyze the impact of incorporating etoposide into first-line treatment. Using merged data from the Korean National Health Insurance Service and National Cancer Regi

[Thymic cancer effectively treated by combination chemotherapy of carboplatin and etoposide with concurrent radiotherapy].

A 63-year-old man was admitted to our hospital for acute heart failure. A chest CT scan revealed a large anterior mediastinal mass and pericardial effusion. Percutaneous needle biopsy showed that the mass was an advanced thymic cancer (squamous cell carcinoma). The patient was treated by combination chemotherapy of carboplatin and etoposide with concurrent radiotherapy (44 Gy). There was no severe toxicity except for grade 4 neutropenia. After 3 courses of chemotherapy, the mass showed an approximately 81% reduction in tumor size and disappearance of the pericardial effusion. Finally, the thym

Combination chemotherapy with cisplatin, carboplatin, and etoposide in advanced malignancy: a phase I trial.

Cisplatin and carboplatin are platinum-based chemotherapeutic agents with broad antitumor activity and significantly different toxicity profiles. They are commonly used in combination with etoposide (VP-16) in chemotherapeutic regimens. We conducted a phase I trial using the combination of cisplatin, carboplatin, and etoposide in advanced malignancy, aimed at delivering a higher dose intensity of active platinum species while taking advantage of their nonoverlapping toxicities. Etoposide was added because of its synergistic action with platinum compounds. The initial chemotherapy regimen consi

Higher in vivo protein binding of etoposide in children compared with adult cancer patients.

Etoposide is bound to plasma albumin (94%). Previous studies have revealed altered protein binding of etoposide in cancer patients. This has clinical implications since only the free fraction is considered pharmacologically active. We have studied the etoposide protein binding in 11 children (eight acute lymphocytic leukemia, two malignant histiocytosis, and one oligodendroglioma; age 1-17 years) and 46 adult patients (28 acute myelocytic leukemia, eight lymphoma, one multiple myeloma, and nine small cell lung cancer; age 38-81 years). All patients were treated with etoposide 50-200 mg/m2 i.v.

[An advanced invasive thymoma that responded remarkably to neoadjuvant chemotherapy with carboplatin, adriamycin, and etoposide].

A 23-year-old man was admitted to the hospital with hoarseness caused by recurrent nerve palsy. The chest X-ray film and CT scan on admission showed a large mass in the antero-superior part of the mediastinum and a small round mediastinal mass adjacent to right inferior pericardium. Examination of a specimen aspirated from the mediastinal mass revealed thymoma. The tumor was considered to be inoperable because of the pericardial dissemination, and the patient was treated with carboplatin, Adriamycin, and etoposide. After 4 courses of chemotherapy, remarkable regression of both tumors was obser

Assessing the impact of aprepitant on response to dose-intensive cyclophosphamide, etoposide, and cisplatin (DICEP).

BackgroundDespite evidence demonstrating the effectiveness of aprepitant for chemotherapy-induced nausea and vomiting (CINV), its use in stem cell transplant settings across Canada is not standard. While pharmacokinetic data exists, the clinical significance of cytochrome P450 3A4 (CYP 3A4) inhibition of cyclophosphamide by aprepitant is unclear. Reduced activation of cyclophosphamide may reduce the effectiveness of dose-intensive cyclophosphamide, etoposide, and cisplatin (DICEP).ObjectivesTo compare response rates to DICEP in patients with Hodgkin lymphoma (HL) and diffuse large B-cell lymph

Repurposing the Antibacterial Activity of Etoposide─A Chemotherapeutic Drug in Combination with Eggshell-Derived Hydroxyapatite.

Drug repurposing has been gaining increasing interest recently due to the reduction in development cost and reduced development timelines. Here, we report the antibacterial activity of the anticancer drug etoposide investigated in combination with the eggshell-derived hydroxyapatite (EHA). Hydroxyapatite (HA) is a well-known bioactive material with enhanced osteoconductivity and possesses superior drug delivery properties. In the present work, we have synthesized etoposide-loaded EHA by the wet precipitation method. The physicochemical characterization of the samples confirmed the composition
