Effects of Cynanchum auriculatum Royle ex Wight ethanol extract on hyperglycaemic symptoms and gut microbiota in type 2 diabetes mellitus mice.

Chronic hyperglycaemia accompanied by impaired pancreatic β-cell function defines Type 2 diabetes mellitus(T2DM), a disorder linked to progressive complications. This study identified flavonoids, phenols and prenol lipids as the major bioactive constituents of Cynanchum auriculatum Royle ex Wight ethanol extract (CRE) and investigated its therapeutic potential on the gut microbiota(GM) in T2DM mice. After a four-week intervention, CRE markedly ameliorated glycemic control, liver function and insulin resistance. It effectively improved key indicators, encompassing body weight, fasting blood glu

Lactobacillus rhamnosus FSJ13 alleviates pancreatic fibrosis in mice via fibroblast growth factor 21.

Chronic pancreatitis (CP) is a progressive fibro-inflammatory condition with a multifactorial etiology, leading to diminished quality of life and increased mortality rates. The incidence and prevalence of chronic pancreatitis are increasing every year, and no definitive cure is currently available. Lactobacillus rhamnosus (L. rhamnosus) has demonstrated potential in alleviating fibrotic diseases. However, its role in CP remains unexplored. This study demonstrated that CP in mice is associated with gut dysbiosis, characterized by a significant decrease in the abundance of butyrate-producing bac

Environmental and Intestinal Phylum Firmicutes Bacteria Metabolize the Plant Sugar Sulfoquinovose via a 6-Deoxy-6-sulfofructose Transaldolase Pathway.

Bacterial degradation of the sugar sulfoquinovose (SQ, 6-deoxy-6-sulfoglucose) produced by plants, algae, and cyanobacteria, is an important component of the biogeochemical carbon and sulfur cycles. Here, we reveal a third biochemical pathway for primary SQ degradation in an aerobic Bacillus aryabhattai strain. An isomerase converts SQ to 6-deoxy-6-sulfofructose (SF). A novel transaldolase enzyme cleaves the SF to 3-sulfolactaldehyde (SLA), while the non-sulfonated C3-(glycerone)-moiety is transferred to an acceptor molecule, glyceraldehyde phosphate (GAP), yielding fructose-6-phosphate (F6P).

The Role of Oral Microbiota in Intra-Oral Halitosis.

Halitosis is a common ailment concerning 15% to 60% of the human population. Halitosis can be divided into extra-oral halitosis (EOH) and intra-oral halitosis (IOH). The IOH is formed by volatile compounds, which are produced mainly by anaerobic bacteria. To these odorous substances belong volatile sulfur compounds (VSCs), aromatic compounds, amines, short-chain fatty or organic acids, alcohols, aliphatic compounds, aldehydes, and ketones. The most important VSCs are hydrogen sulfide, dimethyl sulfide, dimethyl disulfide, and methyl mercaptan. VSCs can be toxic for human cells even at low conc

Comparative in vitro fermentations of cranberry and grape seed polyphenols with colonic microbiota.

In this study, we have assessed the phenolic metabolism of a cranberry extract by microbiota obtained from the ascending colon and descending colon compartments of a dynamic gastrointestinal simulator (SHIME). For comparison, parallel fermentations with a grape seed extract were carried out. Extracts were used directly without previous intestinal digestion. Among the 60 phenolic compounds targeted, our results confirmed the formation of phenylacetic, phenylpropionic and benzoic acids as well as phenols such as catechol and its derivatives from the action of colonic microbiota on cranberry poly

Bacteriology and antimicrobial susceptibility of gram-positive cocci isolated from pus specimens of orofacial odontogenic infections.

We recently reported the beta-lactamase production and antimicrobial susceptibility of anaerobic gram-negative rods isolated from pus specimens of 93 orofacial odontogenic infections. In this report, we determine the bacteriology and antimicrobial susceptibility of bacteria other than anaerobic gram-negative rods, mainly gram-positive cocci, isolated from the same specimens. Streptococcus constellatus and Peptostreptococcus micros were frequent isolates from all types of infection examined. Peptostreptococcus prevotii, Corynebacterium species, and Eubacterium species were recovered only from d

Lythrum salicaria L. herb and gut microbiota of healthy post-weaning piglets. Focus on prebiotic properties and formation of postbiotic metabolites in ex vivo cultures.

ETHNOPHARMACOLOGICAL RELEVANCE: Herb of purple loosestrife (Lythrum salicaria L. from Lythraceae family) (LSH) was used in Europe since ancient times till early-20th century in the therapy of diarrhea and dysentery in human and veterinary medicine. Post-weaning diarrhea is a main problem affecting global piglet production, which leads to significant economic losses because of increased morbidity and mortality, reduced average daily gain, and high antibiotic consumption. Post-weaning diarrhea has various causes, all of which have been linked to imbalances of intestinal microbiota. The aim of th

Are intestinal bacteria involved in the etiology of rheumatoid arthritis? Review article.

Observations in bowel-related joint diseases give support to this hypothesis. In Crohn's disease and ulcerative colitis, the bowel wall inflammation is complicated in about 20% of the patients by joint inflammation. Bowel infection by Salmonella, Shigella and Yersinia can provoke joint inflammation and supports an etiological link between bowel bacteria and arthritis. The arthropathic properties of the most abundant group of intestinal bacteria, i.e. the obligate anaerobic bacteria, were studied in an animal model. Cell wall fragments (CWF), with peptidoglycan as the major component, from some

Causal relationship between gut microbiota and constipation: a bidirectional Mendelian randomization study.

BACKGROUND: Constipation is a prevalent gastrointestinal disorder affecting approximately 15% of the global population, leading to significant healthcare burdens. Emerging evidence suggests that gut microbiota plays a pivotal role in the pathogenesis of constipation, although causality remains uncertain due to potential confounding factors in observational studies. This study aims to clarify the causal relationships between gut microbiota and constipation using a bidirectional Mendelian Randomization (MR) approach, which helps to overcome confounding issues and reverse causality. METHODS: Uti

Lactobacillus gasseri LG-G12 Alleviates High-Fat Diet-induced Obesity through Gut Microbiota-Bile Acid Axis.

The role of probiotics in weight management remains controversial. In this study, we found that Lactobacillus gasseri LG-G12 exhibits gut colonization capacity and metabolic regulation potential in vitro. In vivo, LG-G12 alleviated high-fat diet-induced weight gain and visceral fat accumulation in mice. Fecal targeted metabolomics identified bile acid metabolism as a key pathway. Furthermore, LG-G12 might exhibit bile salt hydrolase and hydroxysteroid dehydrogenase activities, contributing to a bile acid profile characterized by low cholic acid and high ursodeoxycholic acid levels. Theabrownin derived from Pu-erh tea enhances intestinal barrier integrity and microbiota balance via the ST6GalNAc6-STn-MUC2 axis in inflammation.

Ulcerative colitis (UC) involves chronic intestinal inflammation, epithelial barrier disruption, and microbial dysbiosis. Theabrownin (TB), a major bioactive compound derived from Pu-erh tea, has demonstrated anti-inflammatory properties, yet its role in intestinal barrier protection and underlying mechanisms remain unclear. In this study, TB (1 g per kg BW) was administered to dextran sulfate sodium salt (DSS)-induced colitis mice, leading to a marked alleviation in body weight loss and colon injury. TB significantly enhanced goblet cell function and mucus layer integrity, thereby supporting

Xanthoceras sorbifolium Bunge Leaves Ameliorate Type 2 Diabetes Mellitus by Modulating Glucolipid Metabolism Through the Gut Microbiota-Metabolite Axis.

Xanthoceras sorbifolium Bunge leaves (XBL), traditionally consumed as herbal tea, have attracted increasing attention as potential functional food ingredients for managing type 2 diabetes mellitus (T2DM). This study investigated the anti-diabetic effects of an aqueous XBL extract in T2DM rats induced with a high-fat, high-sucrose diet combined with streptozotocin. XBL administration significantly improved glycemic control, insulin sensitivity, lipid profiles, and pancreatic and renal histopathology. Integrated 16S rRNA sequencing and untargeted fecal metabolomics revealed the modulation of key

Exploring substrate cofeeding for enhanced acetogenic C1 bioconversion with AneVO, a low-cost anaerobic parallel bioreactor platform.

Acetogenic bacteria are attractive biocatalysts for biochemical production from sustainable single-carbon (C1) feedstocks. The major challenge is their energy-constrained anaerobic lifestyle, which results in slow growth and low productivity. To overcome this limitation, here, we investigate substrate cometabolism in the acetogen Eubacterium limosum, cofeeding either carbon monoxide or glucose as a dopant alongside the primary C1 substrate, methanol or formate. To increase experimental throughput, we developed AneVO, a parallel minibioreactor system that enables benchtop anaerobic batch and fe

Minimal residual disease negativity in multiple myeloma is associated with intestinal microbiota composition.

Patients with multiple myeloma (MM) who achieve minimal residual disease (MRD) negativity after upfront treatment have superior outcomes compared with those who remain MRD+ Recently, associations have been shown between specific commensal microbes and development of plasma cell disorders. Here, we report the association between intestinal microbiota composition and treatment outcome in MM. Microbiota composition of fecal samples collected from 34 MM patients after induction therapy and at the time of flow cytometry-based bone marrow MRD testing was determined by 16S ribosomal RNA sequencing. Bacteremia following surgical dental extraction with an emphasis on anaerobic strains.

Our aim was to investigate bacteremia caused by surgical extraction of partly erupted mandibular third molars. From 16 young adults, bacterial samples were taken from the third-molar pericoronal pocket and post-operatively from the extraction socket, and blood samples were drawn from the ante-cubital vein up to 30 min after surgery. Of the subjects, 88% had detectable bacteremia-50% 1 min after the incision, 44% immediately after extraction. The respective percentages at 10, 15, and 30 min were 44%, 25%, and 13%. Blood cultures contained 31 species (74% anaerobes), with 3.9 +/- 2.6 species iso

Black-pigmented gram-negative anaerobes in endodontic infections.

Necrotic dental root canal infections are polymicrobial infections dominated by anaerobic bacteria. The number of different species in one canal is usually low, approx. 4-7 species. The species isolated most frequently belong to the genera Prevotella, Porphyromonas, Fusobacterium, Peptostreptococcus, Eubacterium and Streptococcus. The frequency of isolation of black-pigmented Gram-negative anaerobes in endodontic infections varies from 25% to > 50%. Pr. intermedia is the most commonly found pigmented species, followed by Pr. denticola and two Porphyromonas species, P. gingivalis and P. endodon

Methanol Metabolism in the Cytochrome and Menaquinone-Containing Acetogen Moorella thermoacetica.

Methanol derived from pectin degradation is common in oxic and anoxic sediments and is used by various aerobic and anaerobic microorganisms such as acetogenic bacteria or methanogenic archaea as a carbon and energy source. Acetogens use the Wood-Ljungdahl pathway to disproportionate methanol to CO2 and acetate. Methanol metabolism has been studied primarily in mesophilic acetogens such as Acetobacterium woodii and Eubacterium callanderi, that do not have cytochromes or quinones. Moorella thermoacetica, the model organism for elucidation of acetogenesis, is a thermophilic, cytochrome- and quino
