Effects of fatty acid supplements on feed intake, and feeding and chewing behavior of lactating dairy cows.

Saturated and unsaturated fatty acid supplements (FS) were evaluated for effects on feed intake, meal patterns, and chewing behavior. Eight ruminally and duodenally cannulated cows were used in a replicated 4 x 4 Latin square design experiment with 21-d periods. Treatments were control and a linear substitution of 2.5% fatty acids from supplemented saturated FS (SAT; prilled, hydrogenated free fatty acids) for partially unsaturated FS (UNS; calcium soaps of long-chain fatty acids). All rations contained identical forage and concentrate components including 37.2% forage and 13.5% cottonseed. Dr

The time course of erythrocyte membrane fatty acid concentrations during and after treatment of non-human primates with increasing doses of an omega-3 rich phospholipid preparation derived from krill-oil.

BACKGROUND: A commonly used measure to reflect the intake of the long-chain omega-3 fatty acids EPA and DHA is the omega-3 index, defined as the sum of EPA + DHA as % of total fatty acids in erythrocyte membrane. When the omega-3 index changes it follows that the relative fractions of other fatty acids in the membrane are also changed. In the present study, increasing doses of a preparation of omega-3 rich phospholipids extracted from krill oil were administered orally to non-human primates for 12 weeks and the time course of EPA, DHA and 22 other fatty acids in erythrocytes was determined bi-

Effects of fatty acids on the growth and composition of Malassezia pachydermatis and their relevance to canine otitis externa.

Malassezia pachydermatis is one of the many infective agents involved in canine otitis externa. Two major phenotypes (large and small colony types), which had different fatty acid compositions, were found among strains of M pachydermatis isolated from canine ears or skin. The large-colony type contained palmitic, stearic, oleic and linoleic acids, whereas the small-colony type contained myristic, palmitic, stearic, linoleic acids and five other unidentified fatty acids. Exogenous saturated fatty acids in the medium had no effect on the growth of a large colony strain but oleic and linoleic aci

The role of cholesterol in chronic blepharitis.

Chronic blepharitis has been a difficult disease to define either microbiologically or biochemically. Sterols from meibomian secretions of normal subjects and patients were analyzed, and important differences were observed. Based on analyses of these secretions, two significantly different (P less than 0.001) types of normal subjects were found, those with and those without cholesterol esters [Norm(CP) and Norm(CA), respectively]. All patients' secretions contained cholesterol esters. Evidence was obtained which suggests that oxysterols may control the ester cholesterol accumulation. Furthermo

Interference by naturally occurring fatty acids in a noncellular enzyme-based aromatase bioassay.

Natural product drug discovery efforts frequently utilize noncellular screening assays. Fatty acids are commonly found in natural product extracts, and some have been shown to interfere with noncellular assays. Several pure fatty acids were tested using a noncellular aromatase assay, with the unsaturated analogues showing strong inhibitory activity, while the saturated analogues were inactive. Unsaturated fatty acids were further tested against SK-BR-3 hormone-independent human breast cancer cells that overexpress aromatase and were found to be inactive. In natural product screening efforts, e

Low serum fatty acid levels in pregnancies with fetal gastroschisis: A prospective study.

Fetal gastroschisis is a paraumbilical abdominal wall defect with herniation of abdominal organs. The underlying cause of the disease remains unknown; however, studies suggest that nutritional factors may play a role in its development. This prospective case-control study explored the association of serum fatty acid levels of pregnant women and occurrence of gastroschisis. Gastroschisis group comprised 57 pregnant women with fetuses with gastroschisis, and the control group comprised 114 pregnant women with normal fetuses. Serum fatty acids levels were compared between the groups for the overa

[Fatty acid and lipid peroxidation in human atherosclerosis].

Plasma fatty acids and lipid peroxidation were studied in human atherosclerosis. Analysis of fatty acids in 16 controls and 32 hyperlipidemic patients showed, in the latter, a decrease in saturated fatty acids, especially palmitic and stearic acids, and an increase in unsaturated fatty acids, especially arachidonic acid. Compared to hyperlipidemic patients without arterial injury, patients with arterial injury exhibit a significant increase in malonaldehyde (MDA). In the former, MDA concentrations are significantly increased compared to controls. Therefore, peroxidation of unsaturated fatty ac

Use of Fatty Acid Metal Salts for Preventing Maillard Reaction-Driven Browning of Lecithins.

We previously reported that fluid soybean lecithin (SL) undergoes heat deterioration by the newly reported pseudo-Maillard rearrangement reaction. To inhibit heat deterioration, SLs were treated with metal silicates, such as magnesium silicate and calcium silicate. When soybean fatty acid was added to SL before treatment with calcium silicate, the color index after heating improved significantly as the acid value increased from 10 to 35 mg KOH/g. To elucidate the role of soybean fatty acid, calcium silicate treatment was carried out by adding several fatty acids to SL. Although saturated fatty

Fatty acid cytotoxicity to organ-cultured bovine lenses.

High sensitivity of cultured bovine and human lens epithelial cells to unsaturated free fatty acids has been reported in previous papers. Here we show that micromolar concentrations of unsaturated free fatty acids also impair lens cells during organ culturing, a system which resembles the in vivo situation much better than cell cultures do. This added weight to the assumption that fatty acid cytotoxicity might be causally related to senile cataract as well as to cataracts associated with systemic diseases.

Stimulation of chloride transport by fatty acids in corneal epithelium and relation to changes in membrane fluidity.

The effect of altering cell membrane lipids on ion transport across isolated corneas was studied. Corneas mounted in Ussing-type chambers showed a rapid increase in short-circuit current following treatment with a variety of unsaturated fatty acids of varying chain length and unsaturation. Measurements of membrane fluidity which utilize immunofluorescence labelling of membrane proteins showed corneal epithelial cell membranes to be significantly more fluid following linoleic acid treatment. Uptake studies indicate rapid incorporation of [14C]linoleic acid into corneal cell membranes. Highly un

Oleate hydratase from Staphylococcus aureus protects against palmitoleic acid, the major antimicrobial fatty acid produced by mammalian skin.

Oleate hydratases (OhyAs) belong to a large family of bacterial proteins catalyzing the hydration or isomerization of double bonds in unsaturated fatty acids. A Staphylococcus aureus gene (Sa0102) is predicted to encode an OhyA. Here, we recombinantly expressed and purified SaOhyA and found that it forms a homodimer that requires FAD for activity. SaOhyA hydrates only unsaturated fatty acids containing cis-9 double bonds, but not fatty acids with trans-9 double bonds or cis double bonds at other positions. SaOhyA products were not detected in S. aureus phospholipids and were released into the

[Changes in the free fatty acid composition of rat salivary glands induced by chronic administration of isoproterenol].

The major constituent free fatty acid in rat parotid and submandibular glands was palmitic acid (about 40% of the total), followed by saturated and unsaturated fatty acids with 18 carbon atoms and arachidonic acid. The composition was changed by chronic administration of isoproterenol, with an increase of linoleic acid and a decrease of arachidonic acid coinciding with changes in the acyl composition of phospholipids. With respect to the metabolism of unsaturated fatty acids, delta 6-desaturation activity converting linoleic acid to gamma-linolenic acid was detected using [14C]linoleoyl-CoA in

Profiles of the fatty acids in the plasma membrane of human brain tumors.

Several studies demonstrated that certain fatty acids have specific effects on tumor cells. n-3 series fatty acids (alpha-linolenic acid, eicosapentaenoic acid and docosahexaenoic acid) may suppress the carcinogenesis, whereas n-6 series fatty acids (arachidonic acid, linoleic acid) may exert tumor promoting effects. In this study, 19 patients with various brain tumors and 12 control brain tissues were studied. n-3, n-6, n-9 unsaturated fatty acids and certain saturated fatty acids levels were measured in the plasma membrane of tumor or control brain tissues by capillary gas chromatography. Trapping of 2,7-dichlorodibenzo-p-dioxin in aqueous solution by enzymatic reaction of fungal manganese peroxidase in the presence of polyunsaturated fatty acids.

In the presence of polyunsaturated fatty acids, including cis-4,7,10,13,16,19-docosahexaenoic acid (DHA), 2,7-dichlorodibenzo-p-dioxin (DCDD) was treated with manganese peroxidase (MnP) from white rot basidiomycete Phanerochaete sordida YK-624. After incubation with MnP, DCDD could not be extracted from the reaction mixture with n-hexane and was trapped in the water layer. DCDD was released by alkalification of the water layer. DCDD was also trapped after treatment with lipoxidase, which produces hydroperoxides from unsaturated lipids. The amounts of thiobarbituric acid-reactive substances pro

Sperm membrane fatty acid composition in the Eastern grey kangaroo (Macropus giganteus), koala (Phascolarctos cinereus), and common wombat (Vombatus ursinus) and its relationship to cold shock injury and cryopreservation success.

Marsupial spermatozoa tolerate cold shock well, but differ in cryopreservation tolerance. In an attempt to explain these phenomena, the fatty acid composition of the sperm membrane from caput and cauda epididymides of the Eastern grey kangaroo, koala, and common wombat was measured and membrane sterol levels were measured in cauda epididymidal spermatozoa. While species-related differences in the levels of linolenic acid (18:3, n-6) and arachidonic acid (20:4, n-6) were observed in caput epididymal spermatozoa, these differences failed to significantly alter the ratio of unsaturated/saturated

Alterations in circulating fatty acids and the compartmentation of selected metabolites in women with breast cancer.

The presence of the tumor in women with breast cancer provokes a profile of biochemical change characterized by hypoglycemia, hyperuremia and high levels of free fatty acids and ketone bodies in plasma. The total circulating levels of amino acids and lactate are slightly higher in patients with breast cancer. Moreover, alterations in the circulating levels of free and total fatty acids are associated with enhanced levels of total free fatty acids and significantly lower levels of esterified arachidonic acid. This profile may indicate a state of moderate catabolic activation in breast cancer pa

The Effect of Phosphorus Concentration on the Co-Production of Fucoxanthin and Fatty Acids in Conticribra weissflogii.

The production of fucoxanthin and fatty acids in Conticribra weissflogii has been examined, but the role of elements like phosphorus in their mutualistic interactions is not well understood. To fill this gap, our study utilized potassium dihydrogen phosphate (KH2PO4) as a source of phosphorus to examine its impact on the synthesis of fucoxanthin and fatty acids in C. weissflogii. Our findings revealed that at a phosphorus concentration of 10 mg L-1, the cell density (9.5 × 105 cells mL-1), carotenoid concentration (1.67 mg g-1), fucoxanthin concentration (0.91 mg L-1), and fucoxanthin content
