Cytauxzoon felis cytochrome b gene mutation associated with atovaquone and azithromycin treatment.

BACKGROUND: Atovaquone and azithromycin (A&A) with supportive care improve survival rates in cats with cytauxzoonosis. Resistance to atovaquone via parasite cytochrome b gene (cytb) mutations occurs in other Apicomplexan protozoans but is not described in Cytauxzoon felis. OBJECTIVE: To serially characterize the C. felis cytb sequences from a cat that remained persistently infected after A&A treatment. ANIMAL: A cat with naturally occurring C. felis infection. METHODS: Case report of the anemic cat persistently infected with C. felis before, during and after A&A treatment. Cytauxzoon felis

Domestic cats seropositive for Felis catus gammaherpesvirus 1 are often qPCR negative.

Felis catus gammaherpesvirus 1 (FcaGHV1) is a newly described virus that infects domestic cats. To identify FcaGHV1 antigens, we developed an immunofluorescent antibody assay by expressing FcaGHV1 open reading frames (ORFs) in feline cells and incubating fixed cells with sera from FcaGHV1-positive cats. Of the seven ORFs tested, ORF52 and ORF38 had the strongest, most consistent antibody responses. We used recombinant ORF52 and ORF38 proteins to develop two FcaGHV1 ELISAs. These assays were used to detect reactivity in cats previously tested by qPCR for FcaGHV1 in blood cell DNA. Results indic

Molecular detection of Rickettsia felis in different flea species from Caldas, Colombia.

Rickettsioses caused by Rickettsia felis are an emergent global threat. Historically, the northern region of the province of Caldas in Colombia has reported murine typhus cases, and recently, serological studies confirmed high seroprevalence for both R. felis and R. typhi. In the present study, fleas from seven municipalities were collected from dogs, cats, and mice. DNA was extracted and amplified by polymerase chain reaction (PCR) to identify gltA, ompB, and 17kD genes. Positive samples were sequenced to identify the species of Rickettsia. Of 1,341 fleas, Ctenocephalides felis was the most p

Geographic association of Rickettsia felis-infected opossums with human murine typhus, Texas.

Application of molecular diagnostic technology in the past 10 years has resulted in the discovery of several new species of pathogenic rickettsiae, including Rickettsia felis. As more sequence information for rickettsial genes has become available, the data have been used to reclassify rickettsial species and to develop new diagnostic tools for analysis of mixed rickettsial pathogens. R. felis has been associated with opossums and their fleas in Texas and California. Because R. felis can cause human illness, we investigated the distribution dynamics in the murine typhus-endemic areas of these

Prevalence of Trichinella nativa in lynx (Felis lynx) from Alaska, 1988-1993.

Lynx (Felis lynx) carcasses were collected during the 1989 to 1990 through 1992 to 1993 trapping seasons in Alaska (USA). Seven areas were represented. Tongue samples were removed from 1,065 carcasses. Specimens were examined for the presence of Trichinella nativa larvae by means of enzymatic digestion. Overall prevalence was 21%. Both prevalence and number of larvae per gram of host tissue were directly related to age of the host. Age-specific prevalence ranged from 4% for kittens up to 59% for lynx 5 yr of age and older. For infected lynx, intensity ranged from 0.27 larvae per gram of host t

Olfactory flow in the sea catfish, Ariopsis felis (L.): Origin, regulation, and resampling.

The olfactory epithelium of the sea catfish, Ariopsis felis, is found on a pinnate array of lamellae (the olfactory rosette) housed within a nasal chamber. The nasal anatomy of A. felis suggests an ability to capture external water currents. We prepared models from X-ray micro-computed tomography scans of two preserved specimens of A. felis. We then used dye visualisation and computational fluid dynamics to show that an external current induced a flow of water through a) the nasal chamber and b) the sensory channels of the olfactory rosette. The factors responsible for inducing flow through th

Fatal infection caused by Cytauxzoon felis in a captive-reared jaguar (Panthera onca).

Fatal infections with Cytauxzoon felis are common in domestic cats, and jaguars (Panthera onca) are believed carriers of this protozoan. Fatal C. felis infections have never been described in jaguars before. Thus, this study describes such an infection in a 5-month-old captive-reared jaguar that presented hyporexia and died at 26 days after the first clinical signs. During necropsy, samples were taken from several tissues, some were fixed in 10% formalin and process for histopathological diagnosis, and some samples were used for DNA extraction, amplification via PCR and sequencing of the poten

The Cat Flea (Ctenocephalides felis) Immune Deficiency Signaling Pathway Regulates Rickettsia typhi Infection.

Rickettsia species are obligate intracellular bacteria with both conserved and lineage-specific strategies for invading and surviving within eukaryotic cells. One variable component of Rickettsia biology involves arthropod vectors: for instance, typhus group rickettsiae are principally vectored by insects (i.e., lice and fleas), whereas spotted fever group rickettsiae are exclusively vectored by ticks. For flea-borne Rickettsia typhi, the etiological agent of murine typhus, research on vertebrate host biology is facilitated using cell lines and animal models. However, due to the lack of any st

Increased Helicobacter felis colonization in male 129/Sv mice fails to suppress gastritis.

Development of the pathologies associated with Helicobacter pylori infection, most seriously gastric adenocarcinoma, are a consequence of chronic inflammation, which both host and pathogen go to some lengths to minimize. Recently, we presented evidence that H. pylori can suppress the development of inflammation in its immediate microenvironment in the gastric mucosa of 129/Sv mice. We have now extended this study by showing that H. felis, a gastric colonizing Helicobacter closely related to H. pylori, does not possess the same ability to suppress Helicobacter-induced gastritis in mice. Differe

Immunisation against Helicobacter felis infection protects against the development of gastric MALT Lymphoma.

UNLABELLED: The formation of mucosa-associated lymphoid tissue (MALT) in response to Helicobacter pylori infection is closely associated with the development of primary gastric MALT lymphoma. AIM: To examine whether immunisation against Helicobacter felis can protect against development of MALT lymphoma. RESULTS: The majority of control infected mice demonstrated MALT formation (13/15) and five developed lymphoma. Fifteen immunised mice were protected against bacterial challenge, of which only five had evidence of MALT formation and none developed lymphoma. Interestingly, of the four mice in

Heartworm (Dirofilaria immitis) disease and glomerulonephritis in a black-footed cat (Felis nigripes).

A 6-yr-old, 1.36-kg, intact female black-footed cat (Felis nigripes) was presented to the Veterinary Medical Teaching Hospital, University of Florida, with a history of depression, lethargy, and anorexia. Cardiac dysfunction and renal failure were diagnosed on the basis of antemortem and postmortem findings. At necropsy, heartworms (Dirofilaria immitis), glomerulonephritis, and endometritis were present. The glomerulonephritis could have been immune mediated and may have been associated with the heartworm infection or the chronic endometritis or both. Heartworm disease should be included in th

Helicobacter felis gastritis in gnotobiotic rats: an animal model of Helicobacter pylori gastritis.

The gastric spirillum Helicobacter felis, originally isolated from the cat stomach, colonizes the stomachs of germfree rats. Studies were designed to examine the pathological and serological responses of germfree rats inoculated orally with H. felis. At 2 weeks postinoculation, the gastric mucosa of germfree rats had lymphocytes and eosinophils scattered in small foci throughout the subglandular region of the antrum. Small numbers of lymphocytes were present in the subglandular portion of the antral mucosa that focally extended through the lamina propria towards the luminal surface. Eight week

A Felis catus proteomic dataset and PeptideAtlas resource to explore the proteome of the domestic cat.

Domestic cats (Felis catus) are one of the most common companion animals, and the health status and well-being of cats is becoming increasingly important. Several aspects of feline biology are vastly understudied relative to the importance of cats in society. These include robust methods to identify and measure proteins, which are critical for fundamental understanding and disease diagnosis. A searchable proteome map for cats is not available yet and here we measured immortalized feline cells, various tissues and serum using data-dependent acquisition (DDA) mass spectrometry to provide a found

The use of selected electrophoretic techniques to assess the health of domestic cats (Felis catus).

INTRODUCTION: Electrophoretic analytical techniques provide extremely important information about an animal's clinical condition. They are recommended in every case, including in screening tests of animals showing no concerning clinical symptoms. Such tests can detect subclinical conditions, such as inflammation, antigen stimulation or certain forms of cancer. The aim of the study was to determine the suitability of native serum protein electrophoresis and the comet assay for assessing the health status of cats. MATERIAL AND METHODS: Electrophoresis was performed on serum samples from 125 cat

Molecular survey of hemotropic mycoplasmas and piroplasmids in cats (Felis catus) in Central Taiwan.

Data on feline hemotropic mycoplasmas and piroplasmids in Taiwan are limited. Therefore, the aim of this study was to investigate the prevalence of hemotropic mycoplasmas and piroplasmids in cats in central Taiwan, as well as to conduct a phylogenetic analysis of the detected pathogens. In total, blood samples of 213 cats were collected from three regions of central Taiwan: Taichung, Yunlin, and Nantou. Polymerase chain reaction (PCR) was used to amplify the 16S rRNA gene fragments of hemotropic mycoplasmas and the 18S rRNA gene fragments of piroplasmids. Risk factors were also assessed, and p

Subtype Characterization and Zoonotic Potential of Cryptosporidium felis in Cats in Guangdong and Shanghai, China.

: Cryptosporidium felis is an important cause of feline and human cryptosporidiosis. However, the transmission of this pathogen between humans and cats remains controversial, partially due to a lack of genetic characterization of isolates from cats. The present study was conducted to examine the genetic diversity of C. felis in cats in China and to assess their potential zoonotic transmission. A newly developed subtyping tool based on a sequence analysis of the 60-kDa glycoprotein (gp60) gene was employed to identify the subtypes of 30 cat-derived C. felis isolates from Guangdong and Shanghai.
