A new Easily Synthesized Coumarin-based Chemodosimeter for Fluorescent Imaging of Copper Ions in Living Cells.

We present a new coumarin-based chemodosimeter 1 that can be easily synthesized and used as a colorimetric and fluorescence sensor for selective detection of Cu2+ in an aqueous solution. Chemodosimeter 1 is visual and fluorescence responses to a variety of different competing metal ions were also investigated. In an aqueous solution (DMSO/H2O = 9/1, v/v, buffered with HEPES pH = 7.4), Chemodosimeter 1 exhibits a selective response to Cu2+ ions that is unique from other cations via diverse color changes and fluorescence enhancement. Chemodosimeter 1 may also be used to detect Cu2+ ions selectiv

Zirconium-Triggered Aggregation-Induced Emission of Copper Nanoclusters for the Sensitive Fluorescent Detection of Acid Phosphatase.

Acid phosphatase (ACP) serves as a vital clinical biomarker for the diagnosis of various pathological conditions. Herein, a sensitive fluorescent platform was developed for ACP detection, leveraging biocompatible Zr4+ ions as a molecular trigger for aggregation-induced emission (AIE) of 4,6-diamino-2-mercaptopyrimidine-capped copper nanoclusters (DAMP-CuNCs). The coordination between Zr4+ and N atoms of the DAMP ligands facilitated the self-assembly of CuNCs into micro-scale aggregates, thereby significantly amplifying the fluorescence intensity. In the presence of ACP, the enzymatic hydrolysi

Fluorescence lifetime mapping of hydrophobic microenvironments via self-assembly of a 7-azaindole probe.

A β-dicyano-substituted derivative of 1,4-diethenylbenzene and 7-azaindole (compound 1) has been synthetized and studied as a fluorescence lifetime probe that is sensitive to hydrophobic environments. This probe displays Aggregation-Induced Emission (AIE) in water/tetrahydrofuran mixtures, resulting in different optical properties in steady-state and time-resolved measurements in hydrophobic and hydrophilic environments. By modulating the equilibrium between free and aggregated species, it is possible to detect microenvironmental changes using fluorescence lifetime measurements. Owing to these

A non-covalently bound fluorescent probe with a larger fluorescence change for imaging Kv7 channels.

BACKGROUND: The dysfunction of Kv7 channels (KCNQ channels) can lead to epilepsy, deafness, neurodevelopmental disorders and mental retardation. Monitoring the expression and function of Kv7 channels is essential for understanding their roles in physiological and pathological process. Some small-molecule fluorescent probes have been reported for labeling Kv7 channels, but these probe showed a little change with only 3- to 8-fold in the restricted rotation state over free rotation state, making them suffer the interference from background fluorescence. Thus, there is a lack of high sensitive pr

Toward anti-bluing shift: A rationally designed photo-induced electron transfer-based polarity responsive lipid droplets-targeting fluorescent probe for real-time monitoring of drug-induced liver injury.

Drug-induced liver injury (DILI) is a significant public health issue due to its unpredictability and potential severity. DILI primarily manifests as hepatic steatosis or phospholipidosis, accompanied with an elevated lipid droplets (LDs) number. Therefore, developing powerful polarity-responsive probes that target LDs is critical for elucidating the precise function of LDs in DILI. Common solvatochromic polarity-sensitive probes often show a significant blue shift in their emission wavelength as the polarity decreases, with some probes exhibiting a shift of over 100 nm, leading to low accurac

The water-soluble red CsPbI3 perovskite quantum dot "on-off-on" fluorescence sensor for the detection of Cu2+ and D-penicillamine.

In this study, a water-soluble perovskite quantum dot (PQDs) based on CsPbI3 "on-off-on" fluorescent probe was successfully constructed. The probe exhibits red fluorescence emission at 660 nm under 465 nm excitation, and the response mechanism of Cu2+-induced fluorescence quenching (OFF) and D-penicillamine (D-PA) triggered fluorescence recovery (ON) is realized. In addition, the dual target detection platform of both copper ions (Cu2+) and D-PA was established. Quantitative analysis showed that the detection linearity range was 0.5-15 μM (LOD = 0.21 μM) for Cu2+ and 0.4-60 μM (LOD = 16.7 nM)

A ratiometric fluorescent probe based on peanut-shell-derived CDs and CuNCs for smartphone-assisted detection of methyl orange.

Methyl orange (MO), a refractory azo dye, can persist in aqueous environments, creating a need for rapid and reliable monitoring methods. In this work, naturally O/N-containing peanut-shell biomass was utilized as an agricultural solid-waste precursor to synthesize oxygen-containing carbon dots (CDs) via a hydrothermal method. The CDs were mixed with D-penicillamine (DPA)/bovine serum albumin (BSA)-stabilized copper nanoclusters (CuNCs) at a controlled volume ratio to form a dual-emission ratiometric fluorescent probe. TEM was used to characterize the nanoscale morphologies, while XPS confirme

A novel dual-lock-dual-key naphthalimide-based fluorescent probe for the detection of H2S and H2O2.

H2S and H2O2 participate in various biochemical reactions as active signaling molecules and maintain cellular homeostasis during oxidative stress. Current methods for detecting H2S and H2O2 mostly rely on their respective single-detection systems, while the research on dual-detection systems for both substances remains less explored. Herein, we rationally constructed a new naphthalimide-based fluorescence probe P1 for the simultaneous detection of H2S and H2O2 using the dual-lock strategy. P1 consists of naphthalimide fluorephore, the disulfide bond-containing ferrocene (first lock) and the bo

Visualization of endogenous hypochlorous acid during ferroptosis based on Rhodamine B fluorescent probe.

Hypochlorous acid (HClO) plays critical roles in ferroptosis process and the specific molecular mechanism related to it is still obscure. To reveal HClO fluctuation in the ferroptosis cells, a fluorescence turn-on probe RDM-577 that can detect intracellular HClO was skillfully engineered. The probe exhibited a substantial fluorescence enhancement in response to HClO (F/F0 = 35), enabling sensitive detection of HClO in both in vitro and in vivo environments. Additionally, it demonstrated remarkable performance characteristics, such as an exceptionally low detection limit (5.8 nM). Consequently,

NADH-Activated Near-Infrared Fluorescent Probe for Efficient Screening of ADH/ALDH2 Dual Agonists for Alcohol Detoxification.

Alcohol-induced liver injury remains a major global health concern, while effective strategies that facilitate alcohol metabolism are still limited. Alcohol dehydrogenase (ADH) and aldehyde dehydrogenase 2 (ALDH2) play central roles in hepatic ethanol metabolism, their catalytic reactions are stoichiometrically coupled to nicotinamide adenine dinucleotide (NADH) production. However, many existing NADH probes suffer from short excitation/emission wavelengths, high background interference, and limited dynamic ranges, restricting their application in enzyme-related screening. Herein, we report a

Construction of viscosity-sensitive RNA fluorescent probes and their application in inflammtation imaging.

Abnormal structure or function of the nucleolus can lead to dynamic imbalances in intracellular viscosity, which is a core pathological feature of major diseases, including inflammation and cancer. However, existing fluorescent probes face challenges in synchronously and accurately detecting nucleolar RNA dynamics and viscosity changes. Therefore, in this study, probes NK-1 and NK-2, which specifically target the nucleolus, were developed and synthesized. Both probes incorporate a donor-π-acceptor (D-π-A) structure with a positively charged quinolinium salt as the core, enabling specific RNA r

Photocaged Fluorogenic Glycoprobes for Light-Activatable Sensing and Imaging of Galactosidase Activities in Live Cells.

The accurate sensing and imaging of enzymatic activities in live cells is key to exploring their intrinsic biological and pathological functions. While previously developed molecular probes are directly activated in the presence of a target enzyme, here, we developed a photocaging strategy for enzymatic substrates to enable tandem light-activatable, fluorogenic enzyme recognition as an approach with improved sensing fidelity. Galactosidases were used as the target enzyme in a proof-of-concept approach. We synthesized α- and β-galactose-conjugated, rhodol-based glycoprobes, in which the C6-posi

Uracil-Peptide-Pyrene Conjugates as Cu2+-Sensitive Probes Targeting DNA, RNA, and Biorelevant Model Membranes.

We report a series of uracil-triazole-pyrene peptidomimetics (2-4) designed to achieve modular geometric control through peptide linkers of varying length and flexibility. All three compounds exhibited submicromolar affinity for ds-DNA, while compound 4 also showed strong binding to ds-RNA, demonstrating the advantage of combining a uracil recognition unit with an extended pyrene aromatic surface compared to the Pyr-Trp reference ligand and the phenanthridine analog 4'. Structural variations strongly affected Cu(II) coordination. Although Pyr-Trp bound Cu(II) approximately 1000-fold more stron

An Organelle-Orthogonal Dual-Response Probe for In Situ Monitoring of Mitochondrial H2O2 and Lipid Droplet Polarity in Ferroptosis.

Ferroptosis, an iron-dependent form of regulated cell death, is characterized by organelle dysfunction and disrupted communication due to reactive oxygen species (ROS). A major obstacle has been the lack of methods for real-time, simultaneous monitoring of key organellar parameters, which has kept the spatiotemporal role of H2O2 in organelle interactions unclear. Here, we designed a dual-function near-infrared fluorescent probe, PSi-NXZD/HCy-H2O2, that enables synchronous, dual-channel tracking of lipid droplet polarity and mitochondrial H2O2 in living cells. Application of this probe unveiled

Unveiling Pathological Lipid Droplet Accumulation of Dopaminergic Neurons in Parkinson's Disease via a Super-Retentive Fluorescent Probe.

Lipid droplets are highly dynamic organelles in nerve cells and are essential for the function of the central nervous system. The abnormal accumulation of lipid droplets in dopaminergic neurons, found in Parkinson's disease (PD) cells and animal models, presents as a promising target for PD diagnosis. However, previously reported fluorescent probes are unsuitable for in vivo long-term tracking of lipid droplets, limiting their applications in animal models of PD. Here, we developed three lipid droplet-targeted probes, named s-CBTA, b-CBTA, and LD-b-PBTA, which share similar chemical structures

Rational design of environment-sensitive fluorescent probes for butyrylcholinesterase and their application in biological imaging.

Butyrylcholinesterase (BChE) is closely associated with Alzheimer's disease (AD), with its expression significantly elevated in the brains of AD patients. This enzyme has emerged as a potential biomarker for monitoring AD progression. Therefore, developing a reliable chemical tool to detect BChE both in vitro and in vivo is of considerable interest. In this study, we rationally designed a series of environmentally sensitive fluorescent probes targeting BChE, based on a highly potent, selective, and reversible BChE inhibitor. Among these, the most promising probe ESP4 demonstrated an ultrafast

In situ NIR-IIb imaging of endogenous H2S signaling for high-resolution abiotic stress visualization in plants.

In vivo analysis of plant stress responses remains a major challenge in precision agriculture, limiting dynamic optimization of crop growth under variable environmental conditions. Fluorescence imaging enables nondestructive tracking of stress biomarkers, but its accuracy is compromised by the low abundance of endogenous signaling molecules, tissue autofluorescence interference, and limited signal penetration depth. Here, we develop a ratiometric near-infrared IIb fluorescent probe for sensitive monitoring of endogenous hydrogen sulfide (H2S). This nanoprobe integrates lanthanide nanoparticles
