Autosomal STR and SNP characterization of populations from the Northeastern Peruvian Andes with the ForenSeq™ DNA Signature Prep Kit.

Autosomal DNA data from Peru for human identity testing purposes are scarce in the scientific literature, which hinders obtaining an appropriate portrait of the genetic variation of the resident populations. In this study we genetically characterize five populations from the Northeastern Peruvian Andes (Chachapoyas, Awajún, Wampís, Huancas and Cajamarca). Autosomal short tandem repeat (aSTR) and identity informative single nucleotide polymorphism (iiSNP) data from a total of 233 unrelated individuals are provided, and forensic genetic parameters are calculated for each population and for the c

Evaluation of 19 short tandem repeat markers for individualization of Papaver somniferum.

Papaver somniferum, commonly known as opium poppy, is the source of natural opiates, which are used as analgesics or as precursors in the creation of semi-synthetic opioids such as heroin. An increase in opioid addiction in the United States has resulted in high rates of illicit opioid use and overdoses. It has recently been shown that P. somniferum DNA suitable for genetic analysis can be recovered from heroin samples. The development of a comprehensive genetic individualization tool for opium poppy could serve to link cases and strengthen programs such as the Drug Enforcement Administration'

Internal validation of the fully continuous model in EuroForMix for its implementation in routine forensic DNA profiling.

Despite being increasingly more common in the forensic routine, DNA mixtures still present an important roadblock in the analysis of genetic evidence. The International Society for Forensic Genetics (ISFG) recommends the use of the Likelihood Ratio (LR) in the statistical evaluation of the evidence for the contribution of a certain Person of Interest (POI). Numerous mathematical models have been developed for this purpose, which can be classified according to the information included: i) binary models only use the allelic information, ii) qualitative or semi-continuous models also accommodate

The closing kinship window: a continuous identification framework centred on kinship for historical and postconflict mass graves.

Historical and post-conflict mass grave programmes increasingly recover genetic profiles from skeletal remains, yet final identification may remain limited when family reference collection, post-mortem analysis, matching and re-analysis are organised as separate projects. This evidence-informed framework development study presents the Kinship-Centred Continuous Identification Framework for settings in which the possible victim set is open or uncertain. Framework development is defined as the integration of established evidence and operational standards into a time-ordered system with explicit

Standardization of a SNP panel for parentage verification and identification in the domestic cat (Felis silvestris catus).

The domestic cat (Felis silvestris catus) is a valued companion animal throughout the world. Over 60 different cat breeds are accepted for competition by the cat fancy registries in different countries. Genetic markers, including short tandem repeats and SNPs, are available to evaluate and manage levels of inbreeding and genetic diversity, population and breed structure relationships, and individual identification for forensic and registration purposes. The International Society of Animal Genetics (ISAG) hosts the Applied Genetics in Companion Animals Workshop, which supports the standardizati

Developmental validation of the rapidpuncher, an automated laser-based sampling system that eliminates cross-contamination in the forensic analysis of FTA cards.

The rapidPuncher is an advanced, fully automated laser sampling system designed for forensic and diagnostic genetic laboratories, enabling precise disc-shaped sample cutting from FTA cards (by burning a ring) for DNA extraction. Its laser cutting method eliminates physical contact, which solves the long-standing problem of possible cross-contamination that comes with traditional mechanical punching tools. This study provides developmental validation of the rapidPuncher, encompassing evaluations of linearity, cross-contamination risk, and efficacy in STR and mtDNA profiling. Validation tests us

Postmortem Changes in mRNA Expression and Tissue Morphology in Brain and Femoral Muscle Tissues of Rat.

The postmortem interval (PMI), defined as the time elapsed between death and the discovery or examination of the body, is a crucial parameter in forensic science for estimating the time of death. There are many ways to measure the PMI, such as Henssge's nomogram, which uses rectal temperature measurement; livor mortis; rigor mortis; and forensic entomology. However, these methods are usually affected by various conditions in the surrounding environment. The purpose of the present study was to compare molecular genetics and histological changes in the brain and skeletal muscle tissues of SD rat

Knife wound or nosebleed-where does the blood at the crime scene come from?

Secretion analysis is a useful tool in forensic genetics, since it establishes the (cellular) origin of the DNA prior in addition to the identification of the DNA donor. This information can be crucial for the construction of the crime sequence or verification of statements of people involved in the crime. For some secretions, rapid/pretests already exist (blood, semen, urine, and saliva) or can be determined via published methylation analyses or expression analyses (blood, saliva vaginal secretions, menstrual blood, and semen). To discriminate nasal secretion/blood from other secretions (like

DNA Transfer Between Items Within an Evidence Package.

Background/Objectives: Advancements in DNA profiling have made it possible to retrieve intact DNA profiles from increasingly minute biological samples. This increased sensitivity in DNA detection has highlighted crucial considerations to be made when handling and packing items from the crime scene to minimize potential contamination from either direct or indirect transfer of DNA. To investigate potential DNA transfer between items stored within the same evidence package, we conducted a simulation study with items commonly encountered during forensic casework. Methods: Participants were grouped

Inherited Treacher-Collins syndrome in twins after artificial insemination.

We present two cases of the Treacher-Collins syndrome and one case of plagiocephalus in a trigeminus childbirth resulting from artificial insemination. The mother, although affected with the Treacher-Collins syndrome, was not diagnosed before the birth of her children. Genetic counseling did not occur. Medical and forensic problems resulting from modern techniques of reproduction are briefly discussed.

A new multiplex for human identification using insertion/deletion polymorphisms.

Human identification is usually based on the study of STRs or SNPs depending on the particular characteristics of the investigation. However, other types of genetic variation such as insertion/deletion polymorphisms (indels) have considerable potential in the field of identification, since they can combine the desirable characteristics of both STRs and SNPs. In this study, a set of 38 non-coding bi-allelic autosomal indels reported to be polymorphic in African, European, and Asian populations were selected. We developed a sensitive genotyping assay, which is able to characterize all 38 bi-alle

Molecular genetics of sudden cardiac death.

Sudden cardiac death (SCD) is one of the most common causes of death. An important number of sudden deaths, especially in the young, are due to genetic heart disorders, both with structural and arrhythmogenic abnormalities. In recent years, significant advances have been made in understanding the genetic basis of SCD. Identification of the genetic causes of sudden death is important because close relatives are also at potential risk of having a fatal cardiac condition. A comprehensive post-mortem investigation is vital to determine the cause and manner of death and provides the opportunity to

Advances in Aircraft Accident Disaster Victim Identification Using DNA Techniques.

Modern DNA technologies have substantially advanced disaster victim identification. The advances are summarized while stressing privacy, security, and psychological values of victim identification. Identification of victims in fatal aircraft accidents demonstrated the effectiveness of DNA profiling. Initially based on minisatellite or microsatellite loci (i.e., short tandem repeats), DNA analyses are moving toward high-throughput massively parallel sequencing (MPS) that enables analysis of genome-wide single nucleotide polymorphisms, permitting typing of highly degraded samples with increased

Whole exome sequencing of FFPE samples-expanding the horizon of forensic molecular autopsies.

Forensic molecular autopsies have emerged as a tool for medical examiners to establish the cause of death. It is particularly useful in sudden unexplained deaths where the cause of death cannot be determined with a regular medical autopsy. We provide the first study of exome data from formalin-fixed paraffin-embedded samples (FFPE) paired with data from high-quality blood samples in forensic applications. The approach allows exploration of the potential to use FFPE samples for molecular autopsies and identify variants in extensive exome data. We leverage the high uniformity of the hybridizatio

Fatal intoxications and inherited cardiac disorders in the young: where to draw the line?

Sudden cardiac death represents a significant public health concern and is one of the leading causes of early mortality worldwide. The escalating use of illicit drugs, approximately 269 million people in 2018, represents a growing public health. Some of these drugs are stimulants that may have multiple effects on the cardiovascular system including the cardiac rhythm, then substance abuse increases the risk of sudden death. For instance, drugs like cocaine and methamphetamine, may be responsible for myocardial infarction as well as occlusive coronary thrombosis with acute infarction. The conse

The effect of commonly employed forensic DNA extraction protocols on ssDNA/dsDNA proportion and DNA integrity.

The utilisation of massively parallel sequencing (MPS) in forensic DNA analysis is on the rise, driven by the expansion of targeted MPS panels in the market and the introduction of forensic investigative genetic genealogy. The MPS library preparation process, integral to both whole-genome sequencing (WGS) and targeted MPS panel data generation, is largely based on converting double-stranded DNA (dsDNA) into sequencing libraries. In the current study, we examined the effect of seven routinely used forensic DNA extraction methods on the strandedness (single-stranded or double-stranded) and the f

3D forensic facial approximation: Implementation protocol in a forensic activity.

The primary objective of this paper is to report on the successful implementation of forensic facial approximation in a real case in the forensic context. A three-dimensional (3D) facial approximation protocol of the skull was performed with free software, applying techniques in a virtual environment that have already been consolidated in the literature. The skull was scanned with the photogrammetry technique, the digital replica was imported in the Blender software (Blender Foundation, Amsterdam) and individualized model sketches of the face were traced with the MakeHuman software (MakeHuman

Forensic Approaches to Detect Possible Agents of Bioterror.

Many biological agents have been strategic pathogenic agents throughout history. Some have even changed history as a consequence of early discoveries of their use as weapons of war. Many of these bioagents can be easily isolated from the environment, and some have recently been genetically manipulated to become more pathogenic for biowarfare. However, it is difficult to determine accidental outbreaks of disease from intentional exposures. In this review, we examine how molecular tools have been used in combination with forensic research to resolve cases of unusual outbreaks and trace the sourc
