A gene regulating ergot alkaloid biosynthesis in Metarhizium brunneum.

UNLABELLED: Ergot alkaloid synthesis (eas) gene clusters found in several fungi encode biosynthesis of agriculturally and pharmaceutically important ergot alkaloids. Although the biosynthetic genes of the ergot alkaloid pathway have been well characterized, regulation of those genes is unknown. We characterized a gene with sequence similarity to a putative transcription factor and that was found adjacent to the eas cluster of Metarhizium brunneum, a plant symbiont and insect pathogen. Function of the novel gene, easR, was explored by CRISPR-Cas9-derived gene knockouts. To maximize potential fo

NGT1 Is Essential for N-Acetylglucosamine-Mediated Filamentous Growth Inhibition and HXK1 Functions as a Positive Regulator of Filamentous Growth in Candida tropicalis.

Candida tropicalis is a pathogenic fungus that can cause opportunistic infections in humans. The ability of Candida species to transition between yeast and filamentous growth forms is essential to their ability to undergo environmental adaptation and to maintain virulence. In other fungal species, such as Candida albicans, N-acetylglucosamine (GlcNAc) can induce filamentous growth, whereas it suppresses such growth in C. tropicalis. In the present study, we found that knocking out the GlcNA-specific transporter gene NGT1 was sufficient to enhance C. tropicalis filamentous growth on Lee's plus

Genes potentially relevant in the parasitic phase of the fungal pathogen Paracoccidioides brasiliensis.

Paracoccidioides brasiliensis, a fungal pathogen of humans, switches from a filamentous spore-forming mold in the soil to a pathogenic budding-yeast in the human host. Dimorphism is regulated mainly by the temperature of incubation. Representational difference analysis (RDA) was performed between yeast cells of isolate Pb01 and from isolate Pb4940, the last growing as mycelia at the host temperature. Transcripts exhibiting increased expression during development of the yeast parasitic phase comprised those involved mainly in response to stress, transcriptional regulation and nitrogen metabolis

Loss of Catalase-1 (Cat-1) results in decreased conidial viability enhanced by exposure to light in Neurospora crassa.

Light is one of the most important factors inducing morphogenesis in Neurospora crassa. The reception of light triggers the generation of reactive oxygen species (ROS) including hydrogen peroxide (H(2)O(2)). Catalase-1 (Cat-1) is one of three catalases known to detoxify H(2)O(2) into water and oxygen. We reported that the photomorphogenetic characteristics of mutants in nucleoside diphosphate kinase-1 (NDK-1), a light signal transducer, are severely affected, and NDK-1 interacted with Cat-1 in a yeast two-hybrid assay. To disclose the function of Cat-1, we created a Cat-1 loss-of-function muta

Inducible regulating homologous recombination enables precise genome editing in Pichia pastoris without perturbing cellular fitness.

The methylotrophic yeast Pichia pastoris (also known as Komagataella pastoris) is an ideal host for producing proteins and natural products. Enhancing homologous recombination (HR) is helpful for improving the precision of genome editing, but results in stress to cellular fitness and is harmful for industrial applications. To overcome these challenges, we developed a tetracycline repressor protein (TetR)/tetO2 inducible system to dynamically regulate the HR-related gene RAD52 in P. pastoris. This approach significantly improved the positivity rate of single gene deletion to 81%. Furthermore, i

Expression of the primary biliary cirrhosis antigens in yeast: aspects of mitochondrial control.

The mitochondria of 21 yeast strains were tested for the expression of primary biliary cirrhosis (PBC) specific antigens. The amounts of the antigens in the mitochondrial preparations varied with the strains. Genetic analysis of the strain differences in antigen expression indicated nuclear control which was complex. Those strains expressing the least amounts of antigens exhibited coagulating mitochondria in organellar preparations. Additional evidence relating expression of antigens to the physiological/structural state of mitochondria was that cells grown in the presence of the mitochondrial

Cph1p negatively regulates MDR1 involved in drug resistance in Candida albicans.

The cph1/cph1 efg1/efg1 double mutant in Candida albicans is defective in filamentous growth and is avirulent in a mouse model. We previously reported that Efg1p but not Cph1p is involved in drug resistance by negatively regulating ERG3 in C. albicans. In the current study, we have found that overexpression of CPH1 in Saccharomyces cerevisiae increases susceptibility to the antifungal drug fluconazole. Furthermore, in C. albicans, null mutation of CPH1 increased the expression of MDR1 as well as decreased susceptibility to fluconazole and voriconazole but not to amphotericin B. These findings

Transcriptome profiling of Paracoccidioides brasiliensis yeast-phase cells recovered from infected mice brings new insights into fungal response upon host interaction.

Paracoccidioides brasiliensis is a fungal human pathogen with a wide distribution in Latin America. It causes paracoccidioidomycosis, the most widespread systemic mycosis in Latin America. Although gene expression in P. brasiliensis had been studied, little is known about the genome sequences expressed by this species during the infection process. To better understand the infection process, 4934 expressed sequence tags (ESTs) derived from a non-normalized cDNA library from P. brasiliensis (isolate Pb01) yeast-phase cells recovered from the livers of infected mice were annotated and clustered t

High-Level Mycoprotein Production through Metabolic Regulation of Concanavalin A-Like Lectin/Glucanase Domain-Containing Protein in Fusarium venenatum.

Mycoprotein produced by Fusarium venenatum is emerging as an increasingly widespread edible microbial protein source. However, their development is constrained by low protein content, inefficient substrate conversion, and substantial byproducts like ethanol, leading to considerable carbon loss. This study identifies a new genetic target FvCLG encoding concanavalin A-like lectin/glucanase domain-containing protein that associated with cell wall construction. In the 5 L fermenter, the ΔFvCLG strain FG31 achieved the highest reported protein content (61.34%), representing a 22.41% increase over t

Met32 governs transcriptional control of sulfur metabolic flexibility and resistance to reactive sulfur species in the human fungal pathogen Candida albicans.

UNLABELLED: Although considerable advances have been made in understanding the metabolic machinery that enables bacteria to utilize sulfur sources, our understanding of the corresponding processes in fungi remains comparatively fragmentary. To explore the genetic circuit by which the highly prevalent human opportunistic yeast Candida albicans controls sulfur utilization, we characterized the transcriptional landscape associated with sulfur starvation in this fungus. We identified many desulfonation enzymes that were differentially modulated and showed that Jlp12, a sulfonate/α-ketoglutarate di

Sip4p Regulates the Transcription of Enzymes for Gluconeogenesis in Nakaseomyces glabratus.

To survive in glucose poor niches within the host, the opportunistic pathogen Nakaseomyces glabratus must assimilate alternative carbon sources. In many yeasts, pathways for alternative carbon metabolism are regulated at the transcriptional level by Zn2Cys6 transcription factors, a class of proteins that are unique to fungi. Here, we investigated the transcriptional regulation of alternative carbon metabolism pathways by the Zn2Cys6 transcription factors Cat8p, Sip4p, and Rds2p in the opportunistic pathogen N. glabratus. Phenotypic screens of transcription factor knockout strains grown on alte

Mechanisms and regulation of iron uptake and the role of iron in pathogenesis of Candida albicans.

Candida albicans is a primary pathogen implicated in invasive fungal infections. Through its intricate iron uptake and regulatory systems, C. albicans adeptly adapts to various iron-rich environments, circumventing the growth and virulence restrictions imposed by the host's nutritional immunity and intensifying infection severity. This fungus activates the Sef1-Sfu1-Hap43 iron homeostasis regulatory circuit via iron bioavailability sensors (iron-sulfur cluster assembly system). This activation precisely regulates multiple iron uptake pathways, including the high-affinity iron reduction system,

Mitochondrial iron transporter ClMrs3/4 regulates iron homeostasis to modulate nitric oxide balance facilitating appressorial development in Curvularia lunata.

Iron is indispensable for the vast majority of organisms, and iron homeostasis plays a pivotal role in both the physiology and pathogenesis of fungal pathogens. However, the underlying mechanisms by which iron homeostasis modulates fungal pathogenesis remain to be fully elucidated. We therefore focused on investigating the functions of mitochondrial iron transporter ClMrs3/4 in virulence. We conducted targeted gene deletions, expression analyses, biochemistry, and pathogenicity assays, demonstrating that ClMrs3/4 regulates appressorial development via maintenance of cellular iron balance in Cu

The synergistic effects of ammonium and phosphorus on higher alcohol biosynthesis in Zygosaccharomyces mellis LGL-1 and its application for wampee/grape winemaking.

For wine, higher alcohols are double-edged swords, which benefit flavors in appropriate amount but cause negative effects on aromas as well as side-effects for consumers in excessive amount. In this study, five kinds of ammonium salts were tested to reduce higher alcohols produced by Zygosaccharomyces mellis LGL-1 (LGL-1), among which, (NH4)2HPO4 exhibited most excellent effects with higher alcohols reduced by 32.79 %. Both NH4+ and HPO42- demonstrated individual capacities to reduce higher alcohol levels and transcriptome analysis was employed to investigate the synergistic mechanism. Results

Transcriptional and metabolic modeling analyses of developing Aspergillus fumigatus biofilms reveal metabolic shifts required for biofilm maturation.

UNLABELLED: Aspergillus fumigatus is a filamentous fungus found in compost and soil that can cause invasive and/or chronic disease in humans. Diagnosis and treatment of aspergillosis often occur when A. fumigatus has formed dense networks of hyphae within the lung. These hyphal networks are multicellular, encased in an extracellular matrix, and have reduced susceptibility to contemporary antifungal drugs, similar to bacterial biofilms. A model of these dense hyphal networks observed in vivo can be recapitulated in vitro using a static, submerged biofilm culture. The mechanisms underlying filam

Alternative isoforms and phase separation of Ref1 repress morphogenesis in Cryptococcus.

Cryptococcus neoformans, the causative agent of cryptococcosis and a representative of the Basidiomycota phylum of Fungi, is a valuable model for our understanding of eukaryotic/fungal biology. Negative feedback is a well-documented mechanism across Eukarya to regulate developmental transitions. Here, we describe a repressor of the yeast-to-hypha transition, Ref1, which completes a negative feedback loop driven by the master regulator of hyphal morphogenesis, Znf2, during sexual development. Alternative transcription of Ref1, driven by Znf2, produces a functionally distinct Ref1 isoform. Isofo

Regulators of commensal and pathogenic life-styles of an opportunistic fungus-Candida albicans.

The yeast Candida albicans is primarily a commensal of humans that colonizes the mucosal surfaces of the gastrointestinal and genital tracts. Yet, C. albicans can under certain circumstances undergo a shift from commensalism to pathogenicity. This transition is governed by fungal factors such as morphological transitions, environmental cues for instance relationships with gut microbiota and the host immune system. C. albicans utilizes distinct sets of regulatory programs to colonize or infect its host and to evade the host defense systems. Moreover, an orchestrated iron acquisition mechanism o

The contribution of the White Collar complex to Cryptococcus neoformans virulence is independent of its light-sensing capabilities.

The White Collar complex is responsible for sensing light and transmitting that signal in many fungal species. In Cryptococcus neoformans and C. deneoformans the complex is involved in protection against damage from ultraviolet (UV) light, repression of mating in response to light, and is also required for virulence. The mechanism by which the Bwc1 photoreceptor contributes to virulence is unknown. In this study, a bwc1 deletion mutant of C. neoformans was transformed with three versions of the BWC1 gene, the wild type, BWC1C605A or BWC1C605S, in which the latter two have the conserved cystein

The cause and effect of Cryptococcus interactions with the host.

Upon Cryptococcus neoformans infection of the host lung, the fungus enters a nutrient poor environment and must adapt to a variety of host-specific stress conditions (temperature, nutrient limitation, pH, CO2). Fungal spores enter this milieu with limited nutritional reserves, germinate, and begin proliferating by budding as yeast. Although relatively little is known about the initial stages of infection, recent work has characterized changes that occur upon germination. This program and subsequent yeast-phase proliferation progress in a dynamic environment as host nutrient immunity responds t

Boric acid destabilizes the hyphal cytoskeleton and inhibits invasive growth of Candida albicans.

Exposure of Candida albicans to sub-lethal concentrations of boric acid (BA) restricts the dimorphic fungus to its yeast morphology and prevents the formation of invasive hyphae on solid substrates. Exposure to BA causes a rapid and reversible disappearance of polarisome and Spitzenkörper in growing hyphae. In BA-treated hyphae of C. albicans, actin quickly reorganizes from cytoplasmic cables to cortical patches and cell wall growth switches from an apical to an isotropic pattern. As a result of the cytoskeletal changes, the hyphal tips broaden and directional growth of hyphae ceases in the pr
