Down-regulation of homeobox genes MEIS1 and HOXA in MLL-rearranged acute leukemia impairs engraftment and reduces proliferation.

Rearrangements of the MLL (ALL1) gene are very common in acute infant and therapy-associated leukemias. The rearrangements underlie the generation of MLL fusion proteins acting as potent oncogenes. Several most consistently up-regulated targets of MLL fusions, MEIS1, HOXA7, HOXA9, and HOXA10 are functionally related and have been implicated in other types of leukemias. Each of the four genes was knocked down separately in the human precursor B-cell leukemic line RS4;11 expressing MLL-AF4. The mutant and control cells were compared for engraftment in NOD/SCID mice. Engraftment of all mutants in

[Involvement of WT1 gene expression in regulation of P53 and MDM2 proteins function in acute lymphoblastic leukemia].

Using sensitive techniques such as reverse transcription polymerase chain reaction (RT-PCR) the expression of WT1 gene in acute lymphoblastic leukemias (ALL) is indicated. High level of mRNA WT1 was only observed in ALL cases with leukemic cells characterized by P53- and MDM2-positive staining in cytometric analysis. The overexpression of P53 protein has not been induced by P53 gene abnormalities and MDM2 protein synthesis was independent from respective gene amplification. The data suggest that WT1 may play a distinct role in the pathophysiology of acute leukemias. It can regulate the functio

Possible role of intragenic DNA hypermethylation in gene silencing of the tumor suppressor gene NR4A3 in acute myeloid leukemia.

Expression of the tumor suppressor gene NR4A3 is silenced in the blasts of acute myeloid leukemia (AML), irrespective of the karyotype. Although the transcriptional reactivation of NR4A3 is considered to have a broad-spectrum anti-leukemic effect, the therapeutic modalities targeting this gene have been hindered by our minimal understanding of the transcriptional mechanisms regulating its expression, particularly in human AML. Here we show the role of intragenic DNA hypermethylation in reducing the expression of NR4A3 in AML. Bisulfite sequencing analysis revealed that CpG sites at the intrage

Investigation of the Expression Levels of miR-383, miR-381, and miR-141 miRNAs as Diagnostic Biomarkers in Acute Myeloid Leukemia.

BACKGROUND: Acute myeloid leukemia (AML) is a clonal illness that is different in both medical and biological traits. Nonetheless, the genetic basis of this malignancy remains inadequately elucidated. MicroRNAs (miRNAs) are small noncoding RNAs that modify the expression of target mRNAs at both transcriptional and translational levels. In the past decade, miRNAs have evolved a unique mechanism in gene regulation, exhibiting varying expression throughout myeloid differentiation. This investigation examines the serum levels of miR-383, miR-381, and miR-141 in AML and evaluates their diagnostic p

Aberrant Expression of Non-Coding RNAs in Pediatric T Acute Lymphoblastic Leukemia and Their Potential Application as Biomarkers.

Less than 5% of the DNA sequence encodes for proteins, and the remainder encodes for non-coding RNAs (ncRNAs). Among the members of the ncRNA family, microRNAs (miRNAs) and long non-coding RNAs (lncRNAs) play a pivotal role in the insurgence and progression of several cancers, including leukemia. Thought to have different molecular mechanisms, both miRNAs and lncRNAs act as epigenetic factors modulating gene expression and influencing hematopoietic differentiation, proliferation and immune system function. Here, we discuss the most recent findings on the main molecular mechanisms by which miRN

The Chronic Lymphocytic Leukemia Epigenome: A Powerful and Stable Framework for Understanding Biological and Clinical Heterogeneity.

This review explores how epigenetic information, and in particular DNA methylation-based "epitypes," enhances our understanding of chronic lymphocytic leukemia (CLL) biology and prognosis. Epitypes correlate with B-cell maturation stages and outperform traditional biomarkers like immunoglobulin heavy chain variable region mutation status in predicting outcomes. The stability and specificity of CLL epigenetic patterns offer insights into disease history, risk stratification, and treatment response, even amid evolving therapeutic landscape. Emerging epigenetic signatures, including proliferative

MECOM is a master repressor of myeloid differentiation through dose control of CEBPA in acute myeloid leukemia.

The transcription factor MECOM, located at 3q26, is essential for hematopoietic stem cells in healthy individuals. Enhancer translocations, due to 3q26 rearrangements, drive out-of-context MECOM expression in one of the most aggressive subtypes of acute myeloid leukemia (AML). Aberrantly expressed MECOM is essential for the survival and immature phenotype of these leukemia cells. Direct depletion of MECOM using an endogenous auxin-inducible degron immediately upregulates expression of CEBPA, which encodes a transcription factor required for neutrophil development and is frequently mutated in o

Aberrant expression of CD19 as a marker of monocytic lineage in acute myelogenous leukemia.

Immunophenotypic analysis plays a critical role in the diagnosis and classification of acute leukemia. Certain characteristic immunophenotypic patterns have emerged that aid in the classification of acute myelogenous leukemia (AML). We describe a unique pattern of expression of CD19, a B cell-associated cell surface antigen, in cases of AML. We reviewed 59 cases of de novo AML to determine the pattern of CD19 expression in cases of AML using three different CD19 monoclonal antibodies, including B4 (Lytic), B4 89B (Coulter, Miami, Fla) and SJ25-C1 (GenTrak, Plymouth Meeting, Pa). We confirmed t

Chromosome translocations: good genes gone wrong.

Cytogenetic analysis of tumor cells has revealed that recurring chromosome abnormalities are present in many tumors. In the leukemias, lymphomas, sarcomas, these abnormalities are frequently translocations or less often inversions which are closely associated with particular morphologic subtypes of these tumors. Rearrangements involving chromosome band 11q23 are common in acute leukemia, both lymphoblastic and myeloid (monoblastic), and are less common in lymphoma. Although several different genes have been cloned from 11q23 translocation breakpoints, the great majority involve the MLL (myeloi

Analysis of microRNA and gene networks in human chronic myelogenous leukemia.

Molecular biologists have identified a number of genes and microRNAs (miRs) associated with chronic myelogenous leukemia (CML). However, their underlying mechanisms in CML remain unclear. In the present study, three regulatory networks of genes and miRs were constructed to elucidate the underlying mechanisms of CML. The first network was the experimentally validated network of miRs and genes. The second was the dysregulatory network of CML, consisting of dysregulated genes and miRs, contributing to the pathogenesis of CML. The third was the CML‑associated network, consisting of CML‑associated

Transcriptomic analysis of non-leukemic cell subsets in azacytidine-responsive AML highlights pathways associated with adhesion, platelet aggregation, and angiogenesis in mice and humans.

BACKGROUND: Hypomethylating agents (HMAs), such as 5-azacytidine (AZA), are valuable treatment options for patients with acute myeloid leukemia (AML). Despite providing significant extensions in survival when used alone or in combination with BCL-2 inhibitors, resistance and eventual relapse is observed. Reported mechanisms of these outcomes are inconsistent when focusing on leukemic populations within bone marrow, indicating a need for studies on the impact of HMAs on non-leukemic cells in the blood and other tissue compartments. METHODS: Whole blood and spleens from vehicle- or AZA-treated

Overexpression of GATA1 confers resistance to chemotherapy in acute megakaryocytic Leukemia.

It has been previously shown that acute myeloid leukemia (AML) patients with higher levels of GATA1 expression have poorer outcomes. Furthermore, pediatric Down syndrome (DS) patients with acute megakaryocytic leukemia (AMKL), whose blast cells almost universally harbor somatic mutations in exon 2 of the transcription factor gene GATA1, demonstrate increased overall survival relative to non-DS pediatric patients, suggesting a potential role for GATA1 in chemotherapy response. In this study, we confirmed that amongst non-DS patients, GATA1 transcripts were significantly higher in AMKL blasts co

Gene expression profiling of ATL patients: compilation of disease-related genes and evidence for TCF4 involvement in BIRC5 gene expression and cell viability.

Adult T-cell leukemia/lymphoma (ATL) is an aggressive and fatal disease. We have examined 32 patients with smoldering, chronic, lymphoma and acute leukemia using Affymetrix HG-U133A2.0 arrays. Using the BRB array program, we identified genes differentially expressed in leukemia cells compared with normal lymphocytes. Several unique genes were identified that were overexpressed in leukemic cells, including TNFSF11, RGS13, MAFb, CSPG2, C/EBP-alpha, and TCF4; 200 of the most highly overexpressed ATL genes were analyzed by the Pathway Studio, version 4.0 program. ATL leukemia cells were characteri

Effect of Gambogic acid on the regulation of hERG channel in K562 cells in vitro.

Overexpression of human ether-à-go-go (eag) related gene (hERG) has been found in a broad range of human leukemia cell lines and primary human leukemia. The block of hERG protein might be a potential therapeutic strategy for leukemia. Gambogic acid (GA) has recently exhibited marked anti-tumor potency on solid tumors of various derivations. Here, we investigated the anti-leukemia effects of GA and its relation to the regulation of hERG in K562 leukemia cells in vitro. K562 cells were treated with various concentrations of GA (0.125-8.0 micromol/L) for 0-72 h. MTT assay was used to evaluate the

Let-7 Family microRNAs Regulate the Expression of the Urokinase-Receptor in Acute Myeloid Leukemia Cells.

The urokinase-receptor (uPAR) exerts multiple functions supporting most cancer hallmarks. Increased uPAR expression is associated with an unfavorable prognosis in several cancer types, including hematologic malignancies. We previously reported that three oncosuppressor microRNAs (miRNAs) can target the 3'untranslated region (3'UTR) of the uPAR mRNA and that uPAR mRNA is a competitive endogenous RNA (ceRNA) able to recruit oncosuppressor miRs, thus impairing their activity. We now show that uPAR mRNA can also be targeted by oncosuppressor members of the let-7 miRNA family in acute myeloid leuke
