Stigmatic papilla cells secrete jasmonic acid to promote pollen germination through calcium signaling.

Double fertilization begins with signal communication between pollen grains and the pistil stigma, a process that includes pollen adhesion, hydration, germination, and penetration. While several molecular regulators governing pollen hydration and penetration have been characterized, the molecules that mediate pollen germination on the stigma remain largely unelucidated. In this study, we identified jasmonic acid (JA), a maternal factor secreted by stigmatic papilla cells, which facilitates pollen germination by elevating cytosolic Ca2+ levels in pollen grains. We found that pollen grains exhib

Comparative Evaluation of Germination Methods on the Nutritional and Sensory Profile of Coix.

Coix has gained significant research interest for its medicinal and nutritional value, yet its characteristic bitterness limits food applications. To enhance its utilization in the food industry, this study examined the impact of conventional water immersion, ultrasound-assisted immersion (10, 15, and 20 min) and heat-treated immersion (40, 50, and 60 °C) on the nutritional profile and taste properties of Coix. Germinated Coix showed reduced starch, fat, and phytic acid content, but increased protein, γ-aminobutyric acid (from 38.05 to 58.00-116.90 mg/100 g, p < 0.05), dietary fiber, soluble s

Role of Stress Response Genes in Resistance and Germination of Bacillus subtilis Spores.

Bacterial adaptation to hostile environments depends on the coordinated expression of stress response genes. When adverse conditions persist and nutrients become limiting, sporulating species may initiate sporulation as a last-resort survival strategy. However, sporulation under such conditions may alter the resistance and germination properties of the resulting spores. In this study, we investigated whether stress response regulators that facilitate vegetative cell adaptation to temperature and/or salinity changes during growth can influence the properties of Bacillus subtilis 168 spores. To

Chloride Salts and Light Modulate Germination and Early Seedling Performance in Dittrichia viscosa.

Soil salinity is one of the main environmental factors limiting plant germination and establishment, particularly in Mediterranean ecosystems. This study evaluated the germination, recovery, and early development of Dittrichia viscosa exposed to NaCl, KCl, CaCl2, MgCl2, and an equimolar mixture (0-360 mM) under a 12 h light-dark regime and continuous darkness. In the absence of salt, final germination was similar under both regimes, although it was faster in the dark. Increased salt concentration reduced and delayed germination, depending on the type of salt and the light regime used. KCl main

Unconventional vesicle transport driven by formin-actin-myosin working system during pollen germination.

Directional transport of massive secretory vesicles to the germination site is a key process for pollen germination, but the molecular mechanism remains unclear. Here, we find that Myo11C proteins, Myo11C1 and Myo11C2, act redundantly. This functional redundancy is evidenced by impaired vesicle tethering efficiency and aberrant F-actin organization in myo11c1 myo11c2 double mutant. Myo11C1 unexpectedly colocalized with AtFH5-located secretory vesicles (AtFH5-SVs), rather than with actin filaments. Simulated modeling and truncation mutation analyses show that Myo11C binds actin filaments nuclea

Nitrate attenuates abscisic acid signaling via NIN-LIKE PROTEIN8 in Arabidopsis seed germination.

Abscisic acid (ABA) suppresses Arabidopsis (Arabidopsis thaliana) seed germination and post-germinative growth. Nitrate stimulates seed germination, but whether it directly regulates ABA signaling and the associated underlying molecular mechanisms remain unknown. Here, we showed that nitrate alleviates the repressive effects of ABA on seed germination independently of the nitric oxide (NO) pathway. Moreover, nitrate attenuates ABA signaling activated by ABSCISIC ACID INSENSITIVE3 (ABI3) and ABI5, two critical transcriptional regulators of the ABA pathway. Mechanistic analyses demonstrated that

Exogenous GA3 Promotes Germination by Reducing Endogenous Inhibitors in Sainfoin (Onobrychis viciifolia) Seeds.

Endogenous inhibitors can inhibit seed germination, and GA3 can promote seed germination. Whether GA3 can affect the changes in endogenous inhibitors has not been clarified. In order to study the effect of GA3 on the endogenous inhibitors in sainfoin (Onobrychis viciifolia) seeds, the systematic separation method and gas chromatography-mass pectrometry (GC-MS) method were used to determine the endogenous inhibitors using cabbage (Brassica rapa var. glabra Regel) as a bioassay model to validate the inhibitory activity in sainfoin seeds, and then the optimal concentration of GA3 was determined t

The pre-mRNA splicing modulator pladienolide B inhibits Cryptococcus neoformans germination and growth.

Cryptococcus neoformans is an opportunistic fungal pathogen responsible for life-threatening infections, particularly in immunocompromised individuals. The limitations of current antifungal therapies due to toxicity and the emergence of resistance highlight the need for novel treatment strategies and targets. C. neoformans has an intron-rich genome, and pre-mRNA splicing is required for expression of the vast majority of its genes. In this study, we investigated the efficacy of a human splicing inhibitor, pladienolide B (PladB), as an antifungal against C. neoformans. PladB inhibited the growt

Strategies for effective high pressure germination or inactivation of Bacillus spores involving nisin.

UNLABELLED: The major challenge in employing high pressure (HP) at moderate temperature for sterilization is the remarkable resistance of bacterial spores. High isostatic pressure can initiate spore germination, enabling subsequent inactivation under mild conditions. However, not all spores could be triggered to germinate under pressure at temperatures ≤80°C so far. In this study, germination treatment combinations were evaluated for Bacillus spores involving moderate HP (150 MPa, 37°C, 5 min), very HP (vHP, 550 MPa, 60°C, 2.5 or 9 min), simple and complex nutrient germinants [L-valine, L-alan

Relationship between Protein Digestibility and the Proteolysis of Legume Proteins during Seed Germination.

Legume seed protein is an important source of nutrition, but generally it is less digestible than animal protein. Poor protein digestibility in legume seeds and seedlings may partly reflect defenses against herbivores. Protein changes during germination typically increase proteolysis and digestibility, by lowering the levels of anti-nutrient protease inhibitors, activating proteases, and breaking down storage proteins (including allergens). Germinating legume sprouts also show striking increases in free amino acids (especially asparagine), but their roles in host defense or other processes are

Inactivation Effect of Germination Combined with Cold Plasma Treatment on Bacillus licheniformis Spores.

Food spoilage, primarily caused by spore-forming bacteria, has become a critical concern since it results in substantial economic losses within the food industry. Past investigations have successfully identified Bacillus licheniformis as the main bacterium responsible for spoilage in roast chicken. In this study, we screened a new sterilization combination from 16 germinants and 4 cold plasma conditions, respectively. Among them, the combination of "A"GFNa-1 (composed of 60 mmol/L L-alanine, 10 mmol/L D-glucose, 10 mmol/L D-fructose, and 1 g/L NaCl) with cold plasma treatment (packed with 100%

Gasotransmitter H2S accelerates seed germination via activating AOX mediated cyanide-resistant respiration pathway.

Hydrogen sulfide (H2S) has been witnessed as a crucial gasotransmitter involving in various physiological processes in plants. H2S signaling has been reported to involve in regulating seed germination, but the underlying mechanism remains poorly understood. Here, we found that endogenous H2S production was activated in germinating Arabidopsis seeds, correlating with upregulated both the transcription and the activity of L-cysteine desulfhydrase (EC 4.4.1.28, LCD and DES1) responsible for H2S production. Moreover, seed germination could be significantly accelerated by exogenous NaHS (the H2S do

Allelopathic effect of Artemisia argyi on the germination and growth of various weeds.

Allelopathy means that one plant produces chemical substances to affect the growth and development of other plants. Usually, allelochemicals can stimulate or inhibit the germination and growth of plants, which have been considered as potential strategy for drug development of environmentally friendly biological herbicides. Obviously, the discovery of plant materials with extensive sources, low cost and markedly allelopathic effect will have far-reaching ecological impacts as the biological herbicide. At present, a large number of researches have already reported that certain plant-derived alle

Pulses' germination and fermentation: Two bioprocessing against hypertension by releasing ACE inhibitory peptides.

Angiotensin-Converting Enzyme (ACE) is one of the main blood pressure regulators in the renin-angiotensin system leading to hypertension. Hypertension is known as the modern world disease which increases the risk of serious human health problems. Synthetic drugs and some natural compounds could treat this disease by binding to ACE and reducing its activity. Pulses, one of the legumes group, that are the rich in protein sources in the human diet, have several bioactive compounds with ACE inhibitory (ACE I) properties. However, several processes need to break down proteins and improve ACE I acti

The Anthelmintic Ingredient Moxidectin Negatively Affects Seed Germination of Three Temperate Grassland Species.

In animal farming, anthelmintics are regularly applied to control gastrointestinal nematodes. There is plenty of evidence that also non-target organisms, such as dung beetles, are negatively affected by residues of anthelmintics in faeces of domestic ungulates. By contrast, knowledge about possible effects on wild plants is scarce. To bridge this gap of knowledge, we tested for effects of the common anthelmintic formulation Cydectin and its active ingredient moxidectin on seed germination. We conducted a feeding experiment with sheep and germination experiments in a climate chamber. Three wide
