The potential of anti-glutamic acid decarboxylase antibodies to support a diagnosis of autoimmune diabetes mellitus.

OBJECTIVE: Anti-glutamic acid decarboxylase (anti-GAD) antibodies are a frequently used diagnostic marker for autoimmune forms of diabetes mellitus (DM), namely, type 1 diabetes mellitus (T1DM) and latent autoimmune diabetes in adults (LADA). We sought to provide insight into a unique diagnostic application of anti-GAD antibodies in patients potentially misdiagnosed with type 2 diabetes mellitus (T2DM). METHODS: We present a case series of patients who had a change in diagnosis from T2DM to autoimmune DM that was supported by positive anti-GAD antibodies. Patients were identified via a retros

In-gel protein digestion using acidic methanol produces a highly selective methylation of glutamic acid residues.

Mass-tolerant open search methods allow the high-throughput analysis of modified peptides by mass spectrometry. These techniques have paved the way to unbiased analysis of post-translational modifications in biological contexts, as well as of chemical modifications produced during the manipulation of protein samples. In this work, we have analyzed in-depth a wide variety of samples of different biological origin, including cells, extracellular vesicles, secretomes, centrosomes and tissue preparations, using Comet-ReCom, a recently improved version of the open search engine Comet-PTM. Our resul

Intravenous methylprednisolone or immunoglobulin for anti-glutamic acid decarboxylase 65 antibody autoimmune encephalitis: which is better?

BACKGROUND: Patients positive for anti-glutamic acid decarboxylase 65 (GAD65) antibodies have attracted increasing attention. Their clinical manifestations are highly heterogeneous and can be comorbid with tumors. Currently, there is no consensus on the therapeutic regimen for anti-GAD65-associated neurological diseases due to the clinical complexity, rarity and sporadic distribution. We reported six anti-GAD65 autoimmune encephalitis (AE) patients who received intravenous methylprednisolone (IVMP) or immunoglobulin (IVIG) or both. Then, we evaluated the therapeutic effect of both by summarizi

Viral Vector-Mediated Gene Transfer of Glutamic Acid Decarboxylase for Chronic Pain Treatment: A Literature Review.

Chronic pain is long-lasting nociceptive state, impairing the patient's quality of life. Existing analgesics are generally not effective in the treatment of chronic pain, some of which such as opioids have the risk of tolerance/dependence and overdose death with higher daily opioid doses for increasing analgesic effect. Opioid use disorders have already reached an epidemic level in the United States; therefore, nonopioid analgesic approach and/or use of nonpharmacologic interventions will be employed with increasing frequency. Viral vector-mediated gene therapy is promising in clinical trials

Cortical Gene Expression After a Conditional Knockout of 67 kDa Glutamic Acid Decarboxylase in Parvalbumin Neurons.

In the cortex of subjects with schizophrenia, expression of glutamic acid decarboxylase 67 (GAD67), the enzyme primarily responsible for cortical GABA synthesis, is reduced in the subset of GABA neurons that express parvalbumin (PV). This GAD67 deficit is accompanied by lower cortical levels of other GABA-associated transcripts, including GABA transporter-1, PV, brain-derived neurotrophic factor (BDNF), tropomyosin receptor kinase B, somatostatin, GABAA receptor α1 subunit, and KCNS3 potassium channel subunit mRNAs. In contrast, messenger RNA (mRNA) levels for glutamic acid decarboxylase 65 (G

Glutamic acid decarboxylase levels in the cochlear nucleus of rats with acoustic trauma-induced chronic tinnitus.

Tinnitus is the perception of phantom sounds, a phenomenon believed to be due to abnormal neuronal activity in auditory regions of the CNS such as the brainstem cochlear nucleus (CN). One possible mechanism for the abnormal neuronal activity in the CN, supported by recent animal studies, is a decrease in GABAergic inhibition. One possible explanation for this is a decrease in the enzyme responsible for the synthesis of GABA, glutamic acid decarboxylase (GAD). In this study, we used immunohistochemistry to analyse the levels of GAD in the dorsal and ventral CN of rats that had been exposed to n

Kinetics of lethal factor and poly-D-glutamic acid antigenemia during inhalation anthrax in rhesus macaques.

Systemic anthrax manifests as toxemia, rapidly disseminating septicemia, immune collapse, and death. Virulence factors include the anti-phagocytic gamma-linked poly-d-glutamic acid (PGA) capsule and two binary toxins, complexes of protective antigen (PA) with lethal factor (LF) and edema factor. We report the characterization of LF, PA, and PGA levels during the course of inhalation anthrax in five rhesus macaques. We describe bacteremia, blood differentials, and detection of the PA gene (pagA) by PCR analysis of the blood as confirmation of infection. For four of five animals tested, LF exhib

Humoral immunity against glutamic acid decarboxylase and tyrosine phosphatase IA-2 in Lambert-Eaton myasthenic syndrome.

Some beta-cell-specific autoantigens also are present in the central nervous system. Furthermore, stiff man syndrome, an autoimmune neurological disease, is frequently associated with diabetes and shares with this one an anti-GAD and IA-2 humoral immunoreactivity. We wondered whether these autoantibodies could be found in other neurological diseases with a present or supposed autoimmune origin. So, anti-GAD65 (GAD65A) and anti-IA-2 (IA-2A) autoantibodies were assayed in various neurological diseases. There was a higher prevalence of such antibodies in Lambert-Eaton myasthenic syndrome (LEMS) (

Dopamine receptor blockade increases dopamine D2 receptor and glutamic acid decarboxylase mRNAs in mouse substantia nigra.

To study the influence of dopaminergic activity on the expression of dopamine D2 receptors and glutamic acid decarboxylase in substantia nigra, mice were treated daily for several days with an irreversibly acting dopamine D1 and dopamine D2 receptor antagonist N-ethoxycarbonyl-2-ethoxy-1,2-dihydroquinoline (EEDQ) or with a selective irreversible D2 dopamine receptor antagonist fluphenazine-N-mustard. Mice were killed 24 h after the last injection. Dopamine D1 and dopamine D2 receptors were determined by receptor autoradiography, and dopamine D1 and dopamine D2 receptor mRNA and glutamic acid d

De novo mutation in the COL4A5 gene converting glycine 325 to glutamic acid in Alport syndrome.

Southern blot analysis of the COL4A5 gene in a 6 year old Italian Alport patient (proband VIZ) showed the loss of an MspI site that was present in the mother and control DNAs. PCR amplification and DNA sequencing revealed a single G-->A nucleotide change. The mutation results in substitution of a glutamic acid for a glycine residue at position 325 in the triple helical region of the alpha 5(IV) chain.

Glutamic-acid-decarboxylase-and parvalbumin-like-immunoreactive structures in the olfactory bulb of the human adult.

This study examines the distribution and morphological characteristics of glutamic-acid-decarboxylase-like (GAD)- and parvalbumin-like (PA)-immunoreactive structures in the olfactory bulb of the human adult. GAD-immunoreactive somata occurred in the glomerular layer, the external granule cell layer, the more superficial portion of the external plexiform layer, and the internal granule cell layer. The cells were small- to medium-sized. Demonstration of lipofuscin pigment revealed the presence of unpigmented as well as pigmented neurons, thus suggesting the existence of two subpopulations of GAD

GABA and benzodiazepine-induced modification of [14C]L-glutamic acid release from rat spinal cord slices.

The spontaneous and potassium-evoked release of [14C]-label from rat spinal cord slices preloaded with [14C]L-glutamic acid and its modification by GABA and related drugs, such as flurazepam, was studied as a possible indirect measure of presynaptic inhibition and of the ability of benzodiazepines to augment it. GABA (100 microM) reduced the spontaneous release of [14C]-label (glutamate) provided that GABA metabolism was blocked by amino-oxyacetic acid (AOAA), but failed to reduce the potassium-evoked release of glutamate, although muscimol (10 microM) had some effect. In contrast, flurazepam

Visual Disturbance and Elevated Glutamic Acid Decarboxylase Antibody Levels as Manifestations of Central Nervous System Lymphoma.

Various neurologic manifestations are observed in the course of hematologic malignancies. Clinical neurologic symptoms and manifestations of malignancies may result from the malignancy itself, inflammatory responses, or paraneoplastic syndromes. Therefore, differential diagnosis on the basis of clinical presentation is challenging. This is the first study to report a patient with central nervous system lymphoma and elevated glutamic acid decarboxylase antibody levels who presented with uveitis masquerade syndrome (UMS), followed by progressive consciousness change, mimicking autoimmune encepha

Anti-Glutamic Acid Decarboxylase 65 (Anti-GAD65) Autoimmune Encephalopathy: A Case of Diagnostic and Treatment Challenge.

Anti-glutamic acid decarboxylase 65 (anti-GAD65) autoimmune encephalopathy is an uncommon but increasingly recognized cause of cognitive impairment and refractory seizures. We report a 69-year-old woman with a background of insulin-treated diabetes mellitus, hypertension, hypercholesterolemia, and prior hyperthyroidism, who presented with acute altered consciousness and cognitive dysfunction. Initial investigations excluded metabolic, infectious, vascular, and paraneoplastic causes. Cerebrospinal fluid (CSF) analysis showed mildly elevated protein without pleocytosis, and MRI brain demonstrate

Optimization and Characterization of Poly-γ-Glutamic Acid Production From a Locally Isolated Bacillus cereus ARM24.

For identification of this new local poly-γ-glutamic acid (γ-PGA) bacteria and partial purification of γ-PGA, the bacterium was studied and identified as Bacillus cereus. Sixty isolates of Bacillus spp. were collected from 28 sources (agricultural soil, local market vegetables, and fruits). Preliminary screening was performed on selective plates; 37 strains were selected on the basis of their ability to form a slimy mucoid colony. All isolates were studied for morphological and microscopic characters and identified by biochemical tests; a test for the production of γ-PGA was conducted with spe

Solvent-Driven Control of Poly-l-Lysine and Poly-l-Glutamic Acid Properties and Multilayer Growth.

Despite the widespread use of cosolvents to modulate polypeptide conformation, their solvent-specific role in coupling polypeptide charge regulation in solution along with interfacial multilayer growth and mechanics remains poorly understood. Here, we systematically investigate how three common cosolvents, namely ethanol, urea, and dimethyl sulfoxide (DMSO), alter the solution behavior of poly-l-lysine (PLL) and poly-l-glutamic acid (PGA) across a broad pH range (3.0-11.0) at controlled ionic strength. We also examine how the cosolvent and the pH effects govern PLL and PGA layer-by-layer assem

Glutamic Acid at Position 168 Is a Constitutive Activator of Tank Binding Kinase 1 Catalytic Function.

TANK binding kinase 1 (TBK1) is serine/threonine protein kinase member of the inhibitor of nuclear factor-kB kinase family, with links to the etiology of familial as well as idiopathic Amyotrophic Lateral Sclerosis. It contributes to several regulatory cellular processes such as autophagy, inflammation and apoptosis. Reduction or loss of TBK1 kinase activity is associated with increased risk of ALS, and so understanding the molecular basis of this activity is an important research priority. In this current study, the role of the E168 residue, located adjacent to the active site of TBK1, has be

A Paper Strip-Based Photoinduced Electrogenerated Chemiluminescence Platform with CTF/PMo12 Heterojunction-Sensitive Glutamic Acid Detection.

Glutamic acid (GA), a crucial neurotransmitter, plays a significant role in brain function, muscle health, and various neurological disorders. Elevated GA levels have been associated with conditions such as stroke, Parkinson's disease, epilepsy, and Alzheimer's disease. Accurate detection of GA levels is paramount for both diagnostic and research purposes. Herein, we develop a photoinduced electrogenerated chemiluminescence (PECL) test strip fiber paper coated with a covalent triazine framework (CTF)/H3PMo12O40 polyoxometalate (PMo12) heterojunction. Under blue-light irradiation, photogenerate

Plasmodium falciparum Glutamic Acid-Rich Protein-Independent Polyclonal Antibodies Inhibit Malaria Parasite Growth in Human Erythrocytes.

Plasmodium falciparum glutamic acid-rich protein (PfGARP) is a recently characterized cell surface antigen encoded by Plasmodium falciparum, the causative agent of severe human malaria pathophysiology. Previously, we reported that the human erythrocyte band 3 (SLC4A1) serves as a host receptor for PfGARP. Antibodies against PfGARP did not affect parasite invasion and growth. We surmised that PfGARP may play a role in the rosetting and adhesion of malaria. Another study reported that antibodies targeting PfGARP exhibit potent inhibition of parasite growth. This inhibition occurred without the p
