The lectin-gold technique was used to reveal Helix pomatia lectin (HPL)-binding sites in human kidney glomeruli. HPL-binding sites were associated mainly with podocyte foot process bases, and extended over the lamina rara externa of the glomerular basement membrane. In addition, kidneys from blood group A1 donors showed labeling of capillary endothelial cells and erythrocytes. After neuraminidase digestion of the sections prior to incubation with HPL-gold complexes, HPL-binding sites appeared on previously negative regions of the glomerulus. This increase in labeling was particularly striking

Valvular heart disease (VHD), a prevalent age-associated condition associated with high mortality, is primarily treated by prosthetic valve replacement. Bioprosthetic heart valves (BHVs) are increasingly preferred in clinical practice owing to their favorable hemodynamic performance and compatibility with transcatheter delivery. However, current glutaraldehyde crosslinking fails to confer antithrombotic, antioxidant, and immunomodulatory properties, leaving BHVs prone to thrombosis, oxidative stress, inflammation, and calcification, all of which accelerate structural valve deterioration (SVD)

Mycoplasma hyopneumoniae (Mhp) is the primary etiological agent of porcine enzootic pneumonia and establishes infection through close association with the respiratory ciliated epithelium. Classical models of Mhp colonization have focused mainly on specific interactions between adhesins, particularly P97, and ciliary receptors. Although this framework has provided important insights into ciliary attachment, increasing evidence indicates that Mhp adhesion involves multiple surface proteins and host ligands, including sulfated glycosaminoglycans (GAGs), fibronectin, plasminogen (Plg) and extracel

INTRODUCTION: To study post injury metabolite activity and glycocalyx shedding in older adults (OAs) compared to younger adults (YAs). Changes in the metabolome and the glycocalyx after injury are increasingly understood in YAs, but not in OAs.
METHODS: We performed a pilot study (2023-2024) at our level I trauma center. Plasma samples were obtained within 60 min of presentation and stratified by age (OAs ≥ 60 y and YAs < 60 y) and compared. Extracts were analyzed using liquid chromatography and mass spectrometry. Enzyme-linked immunosorbent assay testing measured glycocalyx biomarkers, synde

Acute cardiovascular events driven by atherosclerosis primarily originate from thrombosis triggered by vulnerable plaque rupture or endothelial erosion. Endothelial barrier destabilization-characterized by glycocalyx impairment, intercellular junction disassembly, and abnormal cytoskeletal tension-is a core upstream pathological stage that promotes atherogenic lipoprotein leakage, inflammatory cell infiltration, and matrix degradation. Hemodynamics, primarily through wall shear stress (WSS), shape the spatial distribution and plaque phenotypes of atherosclerosis; notably, low or oscillatory sh

The glycocalyx is a proteoglycan-rich layer present on the surface of all mammalian cells and is particularly prevalent on endothelial cells lining the vasculature. The glycocalyx is thought to affect leukocyte migration by masking adhesion molecules and reducing leukocyte adhesion to the endothelium. Leukocyte recruitment is a key driver of inflammatory diseases, including psoriasis. Here, we found that leukocytes had the glycocalyx component heparan sulfate on their cell surface and that it was lost in response to psoriasis-like skin inflammation. In contrast, endothelial heparan sulfate was

BACKGROUND: Current therapeutic strategies for chronic obstructive pulmonary disease (COPD) remain insufficient to halt disease progression or prevent acute exacerbations. This study aimed to investigate vascular endothelial glycocalyx components (Syndecan-1, Heparan Sulfate, and Hyaluronic Acid) as biomarkers for detecting disease severity and predicting acute exacerbations (AECOPD).
METHODS: 100 patients with COPD and 50 healthy volunteers from the Third Affiliated Hospital of Zhejiang Chinese Medical University. The serum glycocalyx (SDC-1, HS, and HA) levels of the subjects were detected,

Although neurogenic pulmonary edema (NPE) often occurs after aneurysmal subarachnoid hemorrhage (SAH), the mechanism of NPE progression after SAH remains unclear. This study investigates whether pulmonary endothelial glycocalyx (PEG) impairment accompanies NPE after SAH. Accordingly, SAH was induced by endovascular perforation in male mice. The lung tissues of the mice were removed 24 h after SAH induction. The degree of pulmonary edema and lung injury, and the extent of PEG injury were assessed. Water content of lung tissue by the wet/dry method in the SAH group was significantly increased co

OBJECTIVE: To examine whether plasma glycocalyx levels in patients with severe intracranial arterial stenosis (ICAS) are associated with the prediction of new cerebral infarctions following endovascular treatment.
METHODS: 105 patients with ICAS who underwent endovascular treatment were enrolled from September 2020 to June 2021. Plasma glycocalyx components were detected. The condition of the cerebral artery was obtained by high-resolution nuclear magnetic resonance, and risk factors for new cerebral infarction postendovascular treatment in patients with ICAS were analyzed using both univaria

The blood-brain barrier (BBB) glycocalyx is the dense layer of glycans and glycoconjugates that coats the luminal surface of the central nervous system (CNS) vasculature. Despite being the first point of contact between the blood and brain, not much is known about the BBB glycocalyx. Here, we performed a multi-omic investigation of the BBB glycocalyx which revealed a unique glycan landscape characterized by enrichment of sialic acid, chondroitin sulfate, and hyaluronan. We found that the BBB glycocalyx was thicker than glycocalyces in the peripheral vasculature and that hyaluronan was the majo

The endothelial glycocalyx (EG) is the thin sugar-based lining on the apical surface of endothelial cells. It has been linked to the physiological functioning of the microcirculation and has been found to be damaged in critical illness and after acute care surgery. This review aims to describe the role of EG in severely injured patients undergoing surgery, discuss specific situations (e.G. major trauma, hemorrhagic shock, trauma induced coagulopathy) as well as specific interventions commonly applied in these patients (e.g. fluid therapy, transfusion) and specific drugs related to perioperativ

The endothelial glycocalyx depends on Exostosin-1 (Ext1)-mediated heparan sulfate synthesis; however, its endothelial cell-autonomous role in acute inflammation remains unclear. Therefore, we examined the role of Ext1 during acute inflammation. In this study, endothelial-specific Ext1-deficient mice (Ext1flox/flox and VE-cadherin-Cre) were generated, and their response to endotoxin challenge was investigated using transcriptomics and imaging. Transcriptomic profiling revealed downregulation of heparan sulfate- and glycocalyx-related genes, with upregulation of genes associated with immune acti

The development of microcirculation imaging devices has significantly advanced our comprehension of the capillary environment's dynamics. Early research suggested that erythrocytes did not contact the vessel's inner surface due to the Fåhraeus effect, implying the presence of a covering on the endothelial cell surface. Subsequent electron microscopy studies revealed this layer to be a complex part of the vessel wall, now known as the endothelial glycocalyx (EG). The EG is a network of proteoglycans and glycoproteins bound to the endothelial membrane, incorporating soluble molecules from the en

BACKGROUND: Microvascular dysfunction plays a central role in organ dysfunction during septic shock. Endothelial glycocalyx (eGC) damage could contribute to impaired microcirculation. The aim was to assess whether several eGC-damaged biomarkers are associated with microvascular dysfunction in resuscitated septic shock patients.
METHODS: This cross-sectional study included resuscitated septic shock patients (N = 31), and a group of healthy individuals (N = 20). The eGC damage biomarkers measured were syndecan-1 (SDC-1), soluble CD44 (CD44s), hyaluronic acid (HYAL) in blood sample; sulfated gly

Advancing age increases cardiovascular disease risk, in part, because of impaired glycocalyx thickness and endothelial dysfunction. Glycocalyx-targeted therapies, such as Endocalyx Pro, could improve both glycocalyx thickness and endothelial function in older adults; however, this has yet to be tested. We hypothesized that Endocalyx Pro supplementation would increase glycocalyx thickness and endothelial function in older adults. Twenty-three older adults aged 66 ± 7 yr (52% female) were enrolled in a randomized, double-blind, placebo-controlled, parallel-arms study to investigate the effect of

BACKGROUND: Diabetes mellitus is a chronic disease which is detrimental to cardiovascular health, often leading to secondary microvascular complications, with huge global health implications. Therapeutic interventions that can be applied to multiple vascular beds are urgently needed. Diabetic retinopathy (DR) and diabetic kidney disease (DKD) are characterised by early microvascular permeability changes which, if left untreated, lead to visual impairment and renal failure, respectively. The heparan sulphate cleaving enzyme, heparanase, has previously been shown to contribute to diabetic microv

BACKGROUND: Cerebral malaria (CM) is a rare, but severe and frequently fatal outcome of infection with Plasmodium falciparum. Pathogenetic mechanisms include endothelial activation and sequestration of parasitized erythrocytes in the cerebral microvessels. Increased concentrations of glycosaminoglycans in urine and plasma of malaria patients have been described, suggesting involvement of endothelial glycocalyx.
METHODS: We used lectin histochemistry on postmortem samples to compare the distribution of multiple sugar epitopes on cerebral capillaries in children who died from CM and from nonmal
