Rapidly treating infections with adequate antibiotics is of major importance. This requires a fast and accurate determination of the antibiotic susceptibility of bacterial pathogens. The most frequently used methods are slow because they are based on the measurement of growth inhibition. Faster methods, such as PCR-based detection of determinants of antibiotic resistance, do not always provide relevant information on susceptibility, particularly that which is not genetically based. Consequently, new methods, such as the detection of changes in bacterial physiology caused by antibiotics using f

Aerobic bacteria are continuously fighting potential oxidative stress due to endogenous and exogenous reactive oxygen species (ROS). To achieve this goal, bacteria possess a wide array of defenses and stress responses including detoxifying enzymes like catalases and peroxidases; however until now, the dynamics of the intra-bacterial redox balance remained poorly understood. Herein, we used redox-sensitive GFP (roGFP2) inside a variety of gram-negative bacteria to study real-time redox dynamics immediately after a challenge with hydrogen peroxide. Using this biosensor, we determined the individ

UNLABELLED: Several mechanisms are involved in the bacterial resistance towards antimicrobial agents. The membrane-associated mechanisms of resistance were studied in Escherichia coli strains after incubation with Thymus maroccanus essential oil, its major components (carvacrol and thymol) or with certain antibiotics. The minimum inhibitory concentration (MIC) and the expression of membrane proteins, porins and efflux pumps were determined in wild type and derivative strains. Derivative strains adapted to different compounds displayed a high level of resistance to all tested antibiotics. The M

The incidence of human-pathogenic microorganisms with resistance or even complete insensitivity to broad-spectrum antibiotics is increasing. This poses a serious challenge to infection control in hospitals and to hygiene strategies in clinical areas with critically ill patients, particularly intensive care or transplant units. These microorganisms create problems that are seemingly impossible to solve at present. The management of gram-positive pathogens such as methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant enterococci (VRE) has been facilitated by evidence-based

No novel antimicrobial agents against multi-drug-resistant Gram-negative bacteria have been available to daily clinical practice during the last 5 years. On the other hand, resistance rates and mechanisms of those pathogens are increasing worldwide. Pan-resistant (against which none of the currently available antibiotics is effective) strains of Klebsiella pneumoniae, Acinetobacter baumannii and Pseudomonas aeruginosa have been described. Encouraging is the fact that several novel compounds (some of them with mechanisms of action different to those of the antibiotics commercially available) ar

The wide dissemination of carbapenemase-producing Gram-negatives (CPGNs), including enterobacterial species and non-fermenters, has caused a public health crisis of global dimensions. These organisms cause serious infections in hospitalized patients, and are associated with increased mortality. Cross-transmission is common, and outbreaks may occur in healthcare facilities where the infection control practices are inadequate. CPGNs exhibit extensive drug-resistant phenotypes, complicate therapy, and limit treatment options. Systematic data on therapy are limited. However, regimens combining two

AmpC beta-lactamases, enzymes produced by a range of Gram-negative bacteria and able to hydrolyse penicillins, monobactams, cephalosporins and cephamycins, are not inhibited by beta-lactamase inhibitors. They are spread via plasmids and pose a huge epidemiological and diagnostic problem. The production of AmpC B-lactamases was tested in various Gram-negative bacteria. Of strains isolated from urine of patients diagnosed with urinary tract infection, we selected those that were resistant to third generation cephalosporins and did not produce extended-spectrum beta-lactamases (ESBL). The product

One component of the anti-microbial function of lactoferrin (Lf) is its ability to sequester iron from potential pathogens. To overcome this iron limitation, a number of gram-negative bacterial pathogens have developed a mechanism for acquiring iron directly from this host glycoprotein. This mechanism involves surface receptors capable of specifically binding Lf from the host, removing iron and transporting it across the outer membrane. The iron is then bound by a periplasmic iron-binding protein, FbpA, and transported into the cell via an inner membrane complex comprised of FbpB and FbpC. The

The effect of the effluent from a chicken meat-processing plant on the antibiotic-resistant bacterial profile was investigated in an almost closed water reticulation system. Of the 273 faecal coliform isolates 256 (93%) were resistant to one or more of the eight antibiotics tested. The most prevalent isolates were for the beta-lactam antibiotics ampicillin and cephalothin followed by the sulphonamides sulphatriad and cotrimoxazole. Eleven different resistance patterns were identified with a single pattern, comprising of ampicillin-, cephalothin-, streptomycin-, sulphatriad-, cotrimoxazole- and

The sensitivity to the major antimicrobial agents of consecutively isolated gram-negative rods in intensive care units in Switzerland was investigated. A majority of strains originated from clinical specimens where infection or colonization could not be distinguished from each other. A total of 1024 isolates from 482 patients were tested by a standardized microtiter method. Of the beta-lactam antibiotics, imipenem (92% of bacteria sensitive), ceftazidime (90%), and aztreonam (85%) showed the greatest activity. 94% of the isolates were sensitive to ciprofloxacin, and 93% and 91% to amikacin and

Mixed actinomycotic infections were established in a susceptible weanling mouse model by using combinations of Actinomyces israelii and Eikenella corrodens or A. israelii and Actinobacillus actinomycetemcomitans. Acute lesions caused by either of the gram-negative organisms alone were resolved within a few weeks; however, these organisms persisted up to 3 months in chronic lesions in combination with A. israelii.

BACKGROUND: Sepsis remains a critical global health challenge with high mortality. Rapid distinction between Gram-positive and Gram-negative pathogens is critical for empiric antibiotic selection, yet reliable biomarkers for such pathogen stratification are lacking. Immunoglobulin G (IgG) N-glycosylation modulates inflammatory responses in various diseases, suggesting its potential role in sepsis pathogenesis and prognosis.
METHODS: In this nested case-control study, 180 septic patients (100 Gram-negative, 80 Gram-positive) and 100 healthy controls were enrolled. IgG N-glycosylation was analy

Antimicrobial resistance is a major global health threat, exacerbated by bacterial virulence mechanisms such as quorum sensing (QS) and biofilm formation, which enhance pathogenicity and reduce antibiotic susceptibility. Targeting QS offers a promising antivirulence strategy that may attenuate pathogenic traits without imposing selective pressure. In this study, lawsone was evaluated for anti-QS and antibiofilm activity against Chromobacterium violaceum, Pseudomonas aeruginosa, and Serratia marcescens. Lawsone significantly inhibited QS-regulated virulence factors, with reductions in violacein

VIM-type-producing Gram-negative bacteria (GNB) infections are difficult to treat. This is a retrospective single-center study of 34 patients who received cefiderocol for the treatment of VIM-type-producing GNB infections, including 25 Pseudomonas spp., 7 Enterobacterales, and 5 Achromobacter sp. Primary outcomes were clinical failure (defined as death, lack of clinical improvement, or a switch to another drug) at day 14 and 30-day all-cause mortality. The median age was 59 years (IQR 53.7-73.4), and the median Charlson comorbidity index was 3.5 (IQR 2-5). The main infections were respiratory

BACKGROUND: New strategies are needed to slow the emergence of antibiotic resistance among bacterial pathogens. In particular, society is experiencing a crisis of antibiotic-resistant infections caused by Gram-negative bacterial pathogens and novel therapeutics are desperately needed to combat such diseases. Acquisition of iron from the host is a nearly universal requirement for microbial pathogens-including Gram-negative bacteria-to cause infection. We have previously reported that apo-transferrin (lacking iron) can inhibit the growth of Staphylococcus aureus in culture and diminish emergence
