Resistance reversion after Haemonchus contortus replacement in a sheep flock: monitoring of benzimidazole and levamisole resistance-associated polymorphisms and impact on lamb survival.

BACKGROUND: Haemonchus contortus is a highly prevalent gastrointestinal nematode whose control is hindered by anthelmintic resistance. Worm replacement (WR), where a resistant parasite population is substituted for a susceptible one, was performed through an artificial infection of nematode-free ewes with a susceptible H. contortus isolate. The infected ewes were kept in a nematode-free paddock (total replacement—TR) or in a paddock naturally infested with resistant helminths (partial replacement—PR) and compared to untreated ewes kept in a naturally infested paddock (control—C). Lambs born in

In vitro synergy analysis of the combination of Citrus aurantium bergamia (Risso & Poiteau, 1826), ivermectin, and nitroxinil against gastrointestinal nematodes of sheep.

Nematodes are diverse parasites affecting humans and animals, with resistance to anthelmintics becoming a major concern due to prolonged drug use. Essential oils (EOs) from plants offer a promising alternative for parasite control. This study evaluated the preclinical efficacy of Citrus aurantium bergamia essential oil (BEO) combined with nitroxynil (NTX) or ivermectin (IVM) against Haemonchus contortus and Trichostrongylus spp., using the larval migration inhibition test (LMIT). Gas chromatography-mass spectrometry identified (+)-limonene (50.5 %), linalyl acetate (21.0 %), and linalool (6.9

Effect of three feeding levels on the pathogenesis and establishment of Haemonchus contortus in parasite-naïve Pelibuey hair sheep lambs during their first infection.

This study evaluated the effect of three feeding levels on the pathogenesis and establishment of H. contortus upon the first infection of parasite-naïve Pelibuey hair sheep lambs. Forty-two 6-month-old hair sheep lambs (24 ± 4 kg) raised parasite free from birth were used. The lambs were assigned to 3 groups (n = 14), and each was fed a diet designed for different daily weight gain (DWG): 75 g/d (Diet 1), 125 g/d (Diet 2) and 200 g/d (Diet 3). After four weeks of diet adaptation, 10 lambs/group were infected with 450 L3H. contortus/kg BW (infected), and 4 lambs/group were kept parasite-free (N

Is the Famacha© system always the best criterion for targeted selective treatment for the control of haemonchosis in growing lambs?

Targeted selective treatment (TST) aims to aid in the control of gastrointestinal nematodes in small ruminants, identifying animals that require treatment with the purpose of preserving the Refugia population and delaying the buildup of anthelmintic (AH) resistance. The objective of this study was to evaluate the effectiveness of both productive and clinical indicators in the selective treatment of gastrointestinal parasites in growing lambs. Two experiments were carried out between 2015 and 2016. Experiment 1: lambs (n = 70) exclusively raised on pasture were evaluated from the ages of 60 day

A detection of benzimidazole resistance-associated SNPs in the isotype-1 β-tubulin gene in Haemonchus contortus from wild blue sheep (Pseudois nayaur) sympatric with sheep in Helan Mountains, China.

BACKGROUND: Benzimidazole (BZ) resistance is an increasingly serious problem due to the excessive use of this anthelmintic for controlling Haemonchus contortus, which is one of the major gastrointestinal nematodes infecting small ruminants worldwide. Three known single nucleotide polymorphisms (SNPs), F167Y (TAC), E198A (GCA) and F200Y (TAC), in the isotype-1 β-tubulin gene of H. contortus are associated with BZ resistance. Comprehending the spread and origins of BZ resistance-associated SNPs has important implications for the control of this nematode. RESULTS: Twenty-seven adult H. contortus

Complement and Coagulation Cascade Activation Regulates the Early Inflammatory Mechanism of Resistance of Suckling Lambs Against Haemonchus contortus.

Haemonchus contortus is a highly pathogenic blood-sucking nematode from the abomasum of small ruminants. To develop effective control strategies, it is essential to understand the initial mechanisms involved in host resistance to this parasite. In this study, we used computational tools to analyze the complement and coagulation pathways generated from RNA sequencing of abomasal tissue from resistant (Santa Ines) and susceptible (Ile de France) young sheep artificially infected with H. contortus. Thirty-two differentially expressed genes annotated to the ovine genome were associated with the co

Reducing fecal egg count through selective breeding alters dorper lamb response to Haemonchus contortus in an artificial challenge trial.

Infection by gastrointestinal nematodes (GIN), particularly Haemonchus contortus, can be detrimental to sheep health and performance. Genetic susceptibility to GIN varies between breeds, with those lacking high levels of natural resistance often requiring frequent anthelmintic treatment when facing parasitic challenge. Genetic technology can serve as a tool to decrease GIN susceptibility via selection for sheep with reduced fecal egg count (FEC) estimated breeding values (EBVs). However, the physiological changes that result from implementation of this strategy are not well described. Addition

Efficacy of a Haemonchus contortus vaccine under field conditions in young alpacas.

Anthelmintic resistance to Haemonchus contortus creates increasing management challenges with small ruminants and camelids. The commercial vaccine, Barbervax®, contains H11 and H-gal-GP antigens, derived from gut mucosal membrane enzymes of H. contortus involved in digesting blood. Antibody neutralization of these antigens causes failure of H. contortus to digest blood, resulting in parasite death. H11 and H-gal-GP are considered "hidden" antigens, meaning the host immune system does not encounter these proteins under natural infection. Therefore, repeat immunization is required to maintain pr

The anthelmintic potential of Bacillus thuringiensis to counter the anthelmintic resistance against Haemonchus contortus.

Haemonchus contortus (H. contortus) has developed resistance to nearly all available anthelmintic medications. Hence, alternative strategies are required to counter anthelmintic resistance. The present study investigated the anthelmintic potential of Bacillus thuringiensis (B. thuringiensis) against H. contortus. Bacterial spp were identified by conventional methods and confirmed by PCR; In addition, PCR amplification of the bacterial 16S rRNA gene detected B. thuringiensis at 750 base pairs (bps). The amplified products were sequenced, and the sequence data were confirmed using the Basic Loca

Abomasal RNA-seq reveals a strong local cellular response in suckling lambs with resistance against Haemonchus contortus.

Santa Ines (SI) and Ile de France (IF) sheep are known to be resistant and susceptible to Haemonchus contortus infection, respectively. Several studies have shown some genes as potential biological markers for sheep resistance against gastrointestinal nematodes using molecular tools, including transcriptomic analysis. In this study, we sequenced the polyadenylated RNA of the abomasal tissue of SI and IF suckling lambs to identify mucosa-specific transcript alterations between breeds artificially infected with H. contortus. Naïve SI (n = 4) and IF (n = 4) lambs were artificially infected every

Portable near-infrared spectroscopy: A rapid and accurate blood test for diagnosis of Haemonchus contortus infection and for targeted selective treatment of sheep.

Haemonchus contortus is the most prevalent and important gastrointestinal nematode (GIN) in small ruminants. Since it reduces the packed cell volume (PCV), causing anemia, early diagnosis can be used for targeted selective treatment (TST) of sheep, reducing antiparasitic drug use and anthelmintic resistance. This study aimed to predict PCV values through near-infrared spectroscopy (NIRS) and to develop a classification and diagnosis model of H. contortus infection using PCV values, eggs per gram of feces (EPG) counts and mean daily weight gain (DWG). A total of 1728 spectra were collected from

Identification of excretory and secretory proteins from Haemonchus contortus inducing a Th9 immune response in goats.

Th9 cells have been shown to play crucial roles in anti-parasite immunity, pathogenic microbe infection, and allergy. Previous studies have demonstrated that Haemonchus contortus excretory and secretory proteins (HcESPs) induce the proliferation of Th9 cells and alter the transcriptional level of IL-9 as well as its related pathways in the Th9 immune response after infection. However, the exact molecule(s) in HcESPs inducing the Th9 immune response is not yet known. In this study, flow cytometry, co-immunoprecipitation (Co-IP) and shotgun liquid chromatography tandem-mass spectrometry (LC-MS/M

Association between FAMACHA scores and fecal egg counts in Katahdin lambs.

The FAMACHA system was introduced to the U.S. just over 10 yr ago to allow selective deworming of lambs with anemia associated with and retard the development of anthelmintic resistance. The FAMACHA system was initially developed as a predictor of packed cell volume (PCV), but correlations between FAMACHA and fecal egg counts (FEC) have also been reported. It is important to understand factors that influence FAMACHA scores among farms to improve management of gastrointestinal nematodes. The objectives of this study were therefore to quantify associations between FAMACHA scores, FEC, BW, and ag
