Upregulation of miR-128 Mediates Heart Injury by Activating Wnt/β-catenin Signaling Pathway in Heart Failure Mice.

Cardiac hypertrophy contributes to heart failure and is pathogenically modulated by a network of signaling cascades including Wnt/β-catenin signaling pathway. miRNAs have been widely demonstrated to regulate gene expression in heart development. miR-128 was routinely found as a brain-enriched gene and has been functionally associated with regulation of cardiac function. However, its role and molecular mechanisms that regulate cardiac hypertrophy remain largely unclear. Adeno-associated virus serotype 9 (AAV9)-mediated constructs with miR-128 or anti-miR-128 were generated and delivered to over

Interventricular septal defects from penetrating injuries of the heart: a report of 12 cases and review of the literature.

In the past 7 years, 1967 through 1973, 12 patients who had interventricular septal defects from penetrating stab wounds of the thorax were treated at Detroit General Hospital. In 10, the defects were confirmed by cardiac catheterization. Four of the 12 patients were symptomatic and required surgery. Two of these patients had pulmonary-to-system flow ratios less than 2:1. Both these patients had left ventricular dysfunction as a result of coronary artery and myocardial damage from the penetrating trauma to the left ventricle. One of the 4 patients had an injury to the tricuspid valve; this is

Burn-induced subepicardial injury in frog heart: a simple model mimicking ST segment changes in ischemic heart disease.

To mimic ischemic heart disease in humans, several animal models have been created, mainly in rodents by surgically ligating their coronary arteries. In the present study, by simply inducing burn injuries on the bullfrog heart, we reproduced abnormal ST segment changes in the electrocardiogram (ECG), mimicking those observed in ischemic heart disease, such as acute myocardial infarction and angina pectoris. The "currents of injury" created by a voltage gradient between the intact and damaged areas of the myocardium, negatively deflected the ECG vector during the diastolic phase, making the ST

An Addition of U0126 Protecting Heart Grafts From Prolonged Cold Ischemia-Reperfusion Injury in Heart Transplantation: A New Preservation Strategy.

BACKGROUND: Ischemia-reperfusion injury (IRI) is the major cause of primary graft dysfunction in organ transplantation. The mitogen-activated protein kinase/extracellular signal-regulated kinase (ERK) signaling pathway plays a crucial role in cell physiological and pathological processes including IRI. This study aims to investigate whether inhibition of ERK signaling with U0126 can prevent prolonged cold IRI in heart transplantation. METHODS: Rat cardiac cell line H9c2 cells were treated with U0126 before exposure to hypothermic hypoxia/reoxygenation (H/R) conditions. The effect of U0126 on

The ACE2 expression in human heart indicates new potential mechanism of heart injury among patients infected with SARS-CoV-2.

A new type of pneumonia caused by a novel coronavirus SARS-CoV-2 outbreaks recently in China and spreads into many other countries. This disease, named as COVID-19, is similar to patients infected by SARS-CoV and MERS-CoV, and nearly 20% of patients developed severe condition. Cardiac injury is a prevalent complication of severe patients, exacerbating the disease severity in coronavirus disease 2019 (COVID-19) patients. Angiotensin-converting enzyme 2 (ACE2), the key host cellular receptor of SARS-CoV-2, has been identified in multiple organs, but its cellular distribution in human heart is no

N-acetyl-seryl-aspartyl-lysyl-proline treatment protects heart against excessive myocardial injury and heart failure in mice.

Myocardial infarction (MI) in mice results in cardiac rupture at 4-7 days after MI, whereas cardiac fibrosis and dysfunction occur later. N-acetyl-seryl-aspartyl-lysyl-proline (Ac-SDKP) has anti-inflammatory, anti-fibrotic, and pro-angiogenic properties. We hypothesized that Ac-SDKP reduces cardiac rupture and adverse cardiac remodeling, and improves function by promoting angiogenesis and inhibiting detrimental reactive fibrosis and inflammation after MI. C57BL/6J mice were subjected to MI and treated with Ac-SDKP (1.6 mg/kg per day) for 1 or 5 weeks. We analyzed (1) intercellular adhesion mol

Cardiac Opioid System and Its Role in Heart Failure and Ischemia-Reperfusion Injury.

Opioid receptors and peptides act as modulators of cardiac and vascular function. These effects are mediated not only by central mechanisms but also via a local cardiac opioid system, which has gained increasing attention for its potential role in cardiovascular disease and perioperative medicine. In ischemia-reperfusion injury, opioids have been associated with cardioprotective effects in preclinical models and have also been investigated clinically. In contrast, opioid administration in heart failure has been linked to adverse outcomes, including higher in-hospital mortality and an increased

Clozapine-Associated Myocarditis Presenting With Acute Heart Failure: Inflammation Before Injury.

Clozapine is the most effective antipsychotic medication for treatment-resistant schizophrenia but is associated with rare and potentially life-threatening myocarditis. Early clinical manifestations are often nonspecific and may delay diagnosis. We report a man in his 30s who developed fever and palpitations three weeks after initiation of clozapine therapy. Laboratory testing demonstrated a marked elevation in C-reactive protein that preceded the development of cardiac biomarker elevation and new left ventricular systolic dysfunction. Transthoracic echocardiography demonstrated an ejection fr
