The expression of MDP-1, a component of Drosophila embryonic basement membranes, is modulated by apoptotic cell death.

Using a novel monoclonal antibody we have studied the expression of a large proteoglycan-type molecule in Drosophila embryos. This molecule is secreted exclusively by migratory, embryonic hemocytes/macrophages and was therefore named MDP-1 for Macrophage-Derived Proteoglycan-1. Expression of MDP-1 begins late during hemocyte differentiation, after these cells have left their birthplace in the head mesoderm. At this time, macrophages are engaged in extracellular matrix deposition and the phagocytosis of cell debris generated by apoptotic events in various parts of the embryo, in particular from

Mineral phase in shell repair of Manila clam Venerupis philippinarum affected by brown ring disease.

The mineral phase of shell repair in the Manila clam Venerupis philippinarum affected by brown ring disease (BRD) was characterised at various scales and at various stages of shell repair by confocal Raman microspectrometry and scanning electron microscopy. Spherulitic and quadrangular aragonite microstructures associated with polyene pigments were clearly observed. Von Kossa staining showed that at the beginning of shell repair, hemocytes are filled with insoluble calcium carbonate salts in all fluids and then are transported toward the extrapallial fluids and the repair sites. Our analyses s

A possible role for an enkephalinergic system in the internal defense mechanism of Biomphalaria alexandrina exposed to Schistosoma mansoni.

The present study documents that in Bioniphalaria alexandrina coordinated responses to Schistosoma mansoni infection are modulated by receptor-mediated opioid signals. Rather comprehensive tests in susceptible and resistant snails have demonstrated: I- the presence of an endogenaus opioids in the snail hemolymph (in particular, Leu-enkephalin-like material). II- in vitro treatment of snail hemocytes with synthetic Leu-enkephalin analogue (DADLE) resulted in the modulation of cellular adherence, and phagocytic activity. III- the addition of Naloxone, either alone or in combination whith DADLE,

Tissue- and time-dependent deployment of Toll- and Imd-associated immune responses in Manduca sexta larvae.

Little is known about how the Toll and Imd pathways are deployed across tissues and over time in insects outside the Drosophila lineage. To address this, we performed a systematic analysis of immune gene responses in Manduca sexta using deep RNA-seq of fat body, hemocytes, and midgut after aseptic wounding or microbial injection, identifying 1895 genes strongly induced by injury or infection. Immune challenge triggered extensive transcriptional reprogramming, including induction of pattern recognition receptors, serine protease cascades, cytokines, antimicrobial peptides (AMPs), and core immun

LKB1 regulates autophagy through AMPK/TOR signaling pathway to alleviate the damage caused by Vibrio alginolyticus infection.

LKB1 (liver kinase B1) is a key upstream kinase of AMPK and plays an important role in various cellular activities. While the function and mechanism of LKB1 have been widely reported in the study of tumor, there are few reports on its role in bacterial infectious diseases, especially in shrimp. In the present study, molecular characterization revealed that LvLKB1 has an open reading frame (ORF) of 1266 bp encoding 421 amino acids with a molecular weight of about 48 KDa, including the kinase region, N-terminal regulatory domain and C-terminal regulatory domain. LvLKB1 in hepatopancreas and hemo

Shrimp Shapes a Nitrite Tolerance Trait via Regulating Autophagy and Apoptosis.

Nitrite is a significant toxic substance that causes acute toxicity in aquatic animals. However, the molecular mechanisms underlying nitrite tolerance remain largely unidentified. Here, we investigated the physiological and transcriptomic profiles of nitrite-sensitive and nitrite-tolerant families of the white shrimp Penaeus vannamei under nitrite stress. Exposure to a lethal concentration of nitrite for 72 h caused higher mortality, higher nitrite accumulation, and more severe physiological damage in both the hemolymph and hepatopancreas of nitrite-sensitive families than in nitrite-tolerant

Organization of hematopoietic tissue in the intermolt lobster, Homarus americanus.

Hematopoiesis in the American lobster Homarus americanus, as in most decapod crustaceans, occurs in a thin tissue covering the dorsal surface of the foregut. This tissue is composed of loosely attached, ovoid lobules containing the hematopoietic precursors and maturing hemocytes. Release of hemocytes into the dorsal hemocoel is accomplished by rupture of a portion of the connective tissue capsule covering the lobule. Cross sections of the lobules contain between 6 and 40 hematopoietic cells, of which approximately 90% constitute stages in granulocyte maturation and 10% are intermediates in hya

Biomonitoring of manganese metal pollution in water and its impacts on biological activities of Biomphalaria alexandrina snail and larvicidal potencies.

Metal pollution has many dangerous environmental and human health consequences due to the bioaccumulation in the tissues. The present study aims to measure the bioaccumulation factor of the manganese (Mn) heavy metal in Biomphalaria alexandrina snails' tissues and water samples. The current results showed the concentration of Mn heavy metal in water (87.5 mg/l) and its bioaccumulation factor in Helisoma duryi tissue was higher than that in tissues of Physa acuta and B. alexandrina snails. Results showed that 87.5 mg/l Mn concentration had miracidicidal and cercaricidal activities. Also, this c

Immune-physiological aspects of synergy between avermectins and the entomopathogenic fungus Metarhizium robertsii in Colorado potato beetle larvae.

The interaction between the entomopathogenic fungus Metarhizium robertsii and natural avermectin metabolites of the actinomycete Streptomyces avermitilis were investigated on Colorado potato beetle larvae. A synergy in the mortality of larvae was detected after simultaneous treatment with half-lethal doses of avermectins (commercial name actarophit) 0.005% and fungus (5×105conidia/ml). The treatment with avermectins led to rapid fungal colonization of the hemolymph. The defense strategies of insects infected by fungus and treated with avermectins and untreated insects were compared to investig

Tissue-Specific Trade-Offs Between Biomineralisation and Antioxidant Responses in Magallana gigas Infected with Boring Sponges Pione vastifica.

Infestation by boring sponges poses a serious problem for Pacific oyster Magallana gigas (Thunberg, 1793) aquaculture. This study aimed to assess the effect of Pione vastifica sponge infestation on the oysters' capacity for shell repair, antioxidant defence status, and hemocyte functional state. We analysed the expression of VEGF pathway genes and biomineralisation enzymes, molecular chaperones (Hsp70, Hsp90), growth arrest and DNA damage gene (Gadd45α), antioxidant enzyme activity and lipid peroxidation levels in the hemolymph and various mantle parts (central and outer-edge). Beauvericin modulates immune responses in the silkworm, Bombyx mori: insights from cellular and transcriptomic analyses.

Despite the well-documented cytotoxic effects of beauvericin (BEA), its specific interactions with the insect immune system remain inadequately explored. This study systematically investigated the impact of BEA on the immune response of the silkworm, Bombyx mori. Larvae were exposed to BEA (0.5-10 μM) to determine the LC50 and evaluate key immune responses: hemocyte counts, viability, phagocytosis, nodulation, encapsulation, and PO activity, alongside transcriptomic analysis by RNA-Seq. BEA exhibited an LC50 of 5.23 μM in silkworm larvae. While it suppressed hemocyte viability, it paradoxicall

Functional Characterization of a Novel FADD from Anodonta woodiana Reveals Its Regulatory Roles in Apoptotic Cascades and Antibacterial Immunity.

Fas-associated death domain (FADD) is a core conserved adaptor in death receptor-induced apoptotic signaling pathway that plays a crucial role in apoptosis and innate immunity in vertebrates. However, the function of FADDs in invertebrates, especially in bivalve mollusks, remains elusive. In the present study, a novel molluscan FADD homolog (AwFADD) was identified and functionally characterized from the bivalve Anodonta woodiana. The AwFADD protein consists of 232 amino acids and harbors two conserved functional domains: an N-terminal death effector domain (DED) and a C-terminal death domain (

Organ Tropism of Angiostrongylus vasorum Larval Stages in Infected African Giant Snails (Lissachatina fulica).

Angiostrongylus vasorum is a metastrongyloid lungworm causing severe cardiovascular disease in domestic and wild animals. During its heteroxenous life cycle, A. vasorum requires obligate gastropod intermediate hosts. Little is known about A. vasorum larval organ tropism and development in gastropod intermediate hosts. Thus, the aim of this study was to analyze in vivo development of A. vasorum larval stages in experimentally infected African giant snails (Lissachatina fulica). Adult L. fulica (n = 26) were orally infected with A. vasorum-L1 and thereafter continuously euthanized. Gastropod org

High individual variability in the transcriptomic response of Mediterranean mussels to Vibrio reveals the involvement of myticins in tissue injury.

Mediterranean mussels (Mytilus galloprovincialis) are sessile filter feeders that live in close contact with numerous marine microorganisms. As all invertebrates, they lack an adaptive immune response and how these animals are able to respond to a bacterial infection and discriminate it from their normal microbiome is difficult to understand. In this work, we conducted Illumina sequencing of the transcriptome of individual mussels before and after being infected with Vibrio splendidus. The control mussels were injected with filtered seawater. We demonstrate that a great variability exists amon

Species-specific mechanisms of cytotoxicity toward immune cells determine the successful outcome of Vibrio infections.

Vibrio species cause infectious diseases in humans and animals, but they can also live as commensals within their host tissues. How Vibrio subverts the host defenses to mount a successful infection remains poorly understood, and this knowledge is critical for predicting and managing disease. Here, we have investigated the cellular and molecular mechanisms underpinning infection and colonization of 2 virulent Vibrio species in an ecologically relevant host model, oyster, to study interactions with marine Vibrio species. All Vibrio strains were recognized by the immune system, but only nonvirule

Identification and functional characterization of thioredoxin-related protein of 14 kDa in Oncomelania hupensis, the intermediate host of Schistosoma japonicum.

Oncomelania hupensis is the unique intermediate host of the blood fluke Schistosoma japonicum, which causes schistosomiasis. In snails, highly toxic reactive oxygen species (ROS) can be continually generated by hemocytes in response to foreign particles or pathogens, and may be involved in damaging and eliminating digenean larvae. Thioredoxin-related protein of 14 kDa (TRP14) is a member of the Trx superfamily, and plays an important role in the scavenging of ROS. This study was designed to identify and characterize TRP14 from O. hupensis (OhTRP14), and investigate the involvement of OhTRP14 i

Size control of the Drosophila hematopoietic niche by bone morphogenetic protein signaling reveals parallels with mammals.

The Drosophila melanogaster larval hematopoietic organ, the lymph gland, is a model to study in vivo the function of the hematopoietic niche. A small cluster of cells in the lymph gland, the posterior signaling center (PSC), maintains the balance between hematopoietic progenitors (prohemocytes) and their differentiation into specialized blood cells (hemocytes). Here, we show that Decapentaplegic/bone morphogenetic protein (Dpp/BMP) signaling activity in PSC cells controls niche size. In the absence of BMP signaling, the number of PSC cells increases. Correlatively, no hemocytes differentiate.

Parasitic suppression of feeding in the tobacco hornworm, Manduca sexta: parallels with feeding depression after an immune challenge.

The parasitic wasp, Cotesia congregata, suppresses feeding in its host Manduca sexta. Feeding suppression in the host coincides with the emergence of the wasps through the host's cuticle. During wasp emergence, host hemocyte number declined, suggesting that the host mounts a wound/immune response against the exiting parasitoids and/or resulting tissue damage. Eliciting a different type of immune response by injecting heat-killed Serratia marcescens also resulted in a decline in feeding and a reduction in hemocyte number. Both the emerging wasps and the bacteria induced an increase in hemolymph
