Non-anticoagulant heparins: glycan-mediated immune modulation and therapeutic applications.

Non-anticoagulant heparins (NAHs) are modified derivatives of heparin that retain many of the biological activities of the parent compound while exhibiting reduced or absent anticoagulant effects. These modifications have unlocked new therapeutic potentials in various diseases, including inflammatory disorders, cancer, sepsis, and viral infections. This review explores the chemical modifications of NAHs, their mechanisms of action, structure-activity relationships, and therapeutic applications. NAHs exert anti-inflammatory, anticancer, antiviral, and vascular protective effects by interacting

[Biomarkers in tear fluid in keratoconus].

This review presents literature data indicating that tear fluid composition in keratoconus reflects the course of metabolic processes in this disease. In keratoconus, tear fluid shows changes in the activity of lysyl oxidase, prolidase, and heparanase, which participate in corneal structure remodeling. Tear fluid also shows enhanced free radical reactions, reduced intensity of energy processes, and disturbances in the protease-inhibitor balance and cytokine status. Despite the absence of visible signs of inflammation, tear fluid changes confirm the role of inflammation in the pathogenesis of k

Novel Therapeutic Targets and Immune Dysfunction in Malignant Pleural Mesothelioma.

Advances in the treatment of malignant pleural mesothelioma (MPM) have been disappointing, despite the apparent need for new therapeutic options for this rare and devastating cancer. Drug resistance is common and surgical intervention has brought benefits only to a subset of patients. MPM is a heterogenous disease with a surprisingly low mutation rate and recent sequencing efforts have confirmed alterations in a limited number of tumor suppressors that do not provide apparent insights into the molecular mechanisms that drive this malignancy. There is increasing evidence that epigenetic regulat

A low androgenic state inhibits erectile function by suppressing endothelial glycosides in the penile cavernous tissue of rats.

BACKGROUND: The endothelial glycocalyx is an important barrier that protects the structure and function of endothelial cells. Androgen deficiency is a common factor that causes structural and functional impairment of endothelial cells. AIM: To investigate changes in the endothelial glycocalyx in the penile corpus cavernosum of the rat with low androgen status and its relationship with erection function. METHODS: Eighteen 10-week-old Sprague-Dawley male rats were randomly divided into 3 groups (n = 6 each): sham operation, castration, and castration + testosterone replacement. The maximum int

High TG/low HDL-C ratio-mimetic lipid stress drives diabetic atherosclerotic endothelial-macrophage dysfunction through glycocalyx disruption and CD36/ABCA1 imbalance.

A high triglyceride/high-density lipoprotein cholesterol (TG/HDL-C) ratio is strongly associated with insulin resistance and atherosclerotic cardiovascular risk, but the cellular mechanisms linking this lipid imbalance to vascular injury remain incompletely defined. This study established a combined hyperglycemic/high TG/low HDL-C ratio-mimetic in vitro model using human coronary artery endothelial cells and THP-1-derived macrophages to investigate endothelial inflammation, glycocalyx disruption, monocyte recruitment, foam-cell formation, oxidative stress, and cholesterol efflux. High TG/low H

A heparin-binding angiogenic protein--basic fibroblast growth factor--is stored within basement membrane.

The basement membranes of bovine cornea are found to contain an angiogenic endothelial cell mitogen, basic fibroblast growth factor (FGF), as determined by heparin-affinity chromatography, immunoblotting, immunofluorescence, and stimulation of capillary endothelial cell proliferation. The growth factor appears to be bound to heparan sulfate and is released from the cornea by treatment with heparin, a hexasaccharide heparin fragment, heparan sulfate, or heparanase, but not by chondroitin sulfate or chondroitinase. These findings indicate that basement membranes of the cornea may serve as physio

Glycosaminoglycans and fucoidan have a protective effect on experimental glomerulonephritis.

Background: The glomerular endothelial glycocalyx is degraded during inflammation. The glycocalyx plays a pivotal role in endothelial function and is involved in many processes including binding of chemokines and cytokines, leukocyte trafficking, and preventing proteinuria. HS-based therapeutics are a promising novel class of anti-inflammatory drugs to restore a compromised endothelial glycocalyx under inflammatory conditions. Recently, we demonstrated that treatment with HS extracted from unstimulated glomerular endothelial glycocalyx (unstimulated HSglx) reduced albuminuria during anti-GBM i

Cardiomyocyte-endothelial cell control of lipoprotein lipase.

In people with diabetes, inadequate pharmaceutical management predisposes the patient to heart failure, which is the leading cause of diabetes related death. One instigator for this cardiac dysfunction is change in fuel utilization by the heart. Thus, following diabetes, when cardiac glucose utilization is impaired, the heart undergoes metabolic transformation wherein it switches to using fats as an exclusive source of energy. Although this switching is geared to help the heart initially, in the long term, this has detrimental effects on cardiac function. These include the generation of noxiou

Acrolein-mediated carbonylation of proheparanase-1 drives endothelial glycocalyx degradation in human immortalized brain microvascular endothelial cells.

The endothelial glycocalyx plays a pivotal role in maintaining blood-brain barrier integrity, and its degradation promotes leukocyte adhesion and transendothelial migration. We previously reported that the degradation of heparan sulfate (HS), a major structural component of the glycocalyx, aggravated inflammation after stroke onset. In addition, acrolein (ACR), an α,β-unsaturated aldehyde generated following stroke onset, promoted HS degradation via activation of proheparanase-1 (proHPSE1), the inactive precursor of heparanase-1 (HPSE1). However, whether this ACR-mediated HS degradation occurs

Updates on the endothelial glycocalyx in heart failure with preserved ejection fraction.

The endothelial glycocalyx (GCX) is a specialized form of extracellular matrix with a dynamic synthesis and degradation balance. It plays a pivotal role in regulating cell signaling, sensing the extracellular environment, and maintaining vascular permeability. In the setting of heart failure (HF), particularly in HF with preserved ejection fraction (HFpEF), systemic endothelial GCX degradation may represent an early pathophysiological mechanism. Indeed, HFpEF results from comorbidities-related proinflammatory states leading to endothelial dysfunction and subsequent myocardial hypertrophy, fibr

Potential Mechanisms of Cancer-Related Hypercoagulability.

The association between cancer and thrombosis has been known for over a century and a half. However, the mechanisms that underlie this correlation are not fully characterized. Hypercoagulability in cancer patients can be classified into two main categories: Type I and Type II. Type I occurs when the balance of endogenous heparin production and degradation is disturbed, with increased degradation of endogenous heparin by tumor-secreted heparanase. Type II hypercoagulability includes all the other etiologies, with factors related to the patient, the tumor, and/or the treatment. Patients with poo

Rational Design and Expedient Synthesis of Heparan Sulfate Mimetics from Natural Aminoglycosides for Structure and Activity Relationship Studies.

Heparan sulfate (HS) contains variably repeating disaccharide units organized into high- and low-sulfated domains. This rich structural diversity enables HS to interact with many proteins and regulate key signaling pathways. Efforts to understand structure-function relationships and harness the therapeutic potential of HS are hindered by the inability to synthesize an extensive library of well-defined HS structures. We herein report a rational and expedient approach to access a library of 27 oligosaccharides from natural aminoglycosides as HS mimetics in 7-12 steps. This strategy significantly

CD138 (syndecan-1) expression in health and disease.

CD138 (syndecan-1, Sdc-1) is a member of the syndecan family that comprises heparan sulfate proteoglycans. CD138 is significant for cell-cell and cell-matrix interactions. In adult human tissues, CD138 is predominantly expressed in epithelial cells and plasmacytes. CD138 immunoexpression is altered in a wide spectrum of benign inflammatory, infectious and fibrotic diseases (colitis, allergic contact dermatitis, fibrosis of various organs, etc) and diabetes mellitus type II. Furthermore, CD138 is involved in molecular pathways that are deregulated during carcinogenesis and are related to cell p

Heparan sulfate regulates ADAM12 through a molecular switch mechanism.

The disintegrin and metalloproteases (ADAMs) are emerging as therapeutic targets in human disease, but specific drug design is hampered by potential redundancy. Unlike other metzincins, ADAM prodomains remain bound to the mature enzyme to regulate activity. Here ADAM12, a protease that promotes tumor progression and chondrocyte proliferation in osteoarthritic cartilage, is shown to possess a prodomain/catalytic domain cationic molecular switch, regulated by exogenous heparan sulfate and heparin but also endogenous cell surface proteoglycans and the polyanion, calcium pentosan polysulfate. Single-Cell Sequencing Analysis and Multiple Machine Learning Methods Identified G0S2 and HPSE as Novel Biomarkers for Abdominal Aortic Aneurysm.

Identifying biomarkers for abdominal aortic aneurysms (AAA) is key to understanding their pathogenesis, developing novel targeted therapeutics, and possibly improving patients outcomes and risk of rupture. Here, we identified AAA biomarkers from public databases using single-cell RNA-sequencing, weighted co-expression network (WGCNA), and differential expression analyses. Additionally, we used the multiple machine learning methods to identify biomarkers that differentiated large AAA from small AAA. Biomarkers were validated using GEO datasets. CIBERSORT was used to assess immune cell infiltrat

Characterization and biological activities of a high-yield galactosylated heparan sulfate from Bursatella leachii egg strings.

Systematic analysis of the gelatinoid of the sea hare Bursatella leachii egg strings showed that the material almost entirely consisted of polysaccharides. Hot water treatment could efficiently and conveniently extract the acidic polysaccharides (BLP) almost completely, with a yield of up to 50 % of the dry weight of the egg strings. Physicochemical analysis revealed that BLP is composed of glucuronic acid, glucosamine, galactose, and sulfate groups in a molar ratio of 1.00: 1.00: 1.12: 2.40, with a molecular weight of 359.7 kDa. The sulfation degree is comparable to heparin and higher than th

Precision Targeting of Heparan Sulfate Proteoglycans Broadly Dysregulates Survival Signaling and Metabolic Adaptation in Pancreatic Cancer.

Treatments for pancreatic ductal adenocarcinoma (PDAC) often fail because of the cancer's ability to adapt its signaling and metabolic pathways to circumvent treatment mechanisms. PDAC hallmarks that promote treatment resistance (increased fatty acid metabolism, survival signaling, desmoplasia, and macropinocytosis) are extensively regulated by heparan sulfate proteoglycans (HSPGs) and their binding partners. The pro-tumor functions of HSPGs are controlled by site-specific cleavage of their heparan sulfate chains by mammalian heparanase; however, cleavage by bacterial heparinase III (HepIII) r

Development of the pig placenta.

Placental insufficiency results in fetal loss, low birth weight, stillbirth, preweaning mortality and poor growth. Placental development begins at conceptus elongation, which is a primary factor controlling the size of the placenta. After elongation, the allantois develops outward from the embryo to establish the allantochorion, which defines the size of the functional placenta. During implantation, chorionic trophoblasts adhere to endometrial epithelial cells. Placental structures known as areolae develop at the openings of the endometrial glands and take up endometrial gland secreted product
