High-dose melphalan with 6-hydroxydopamine-purged autologous bone marrow transplantation for poor-risk neuroblastoma.

Long-term results are presented of 28 patients who were diagnosed with neuroblastoma at more than 12 months of age and who received melphalan 180 mg/m2 (n = 6) or 240 mg/m2 (n = 22) to consolidate remissions of Stage IV disease or to control refractory disease. Twenty-four patients also received dianhydrogalactitol 180 to 240 mg/m2, and 11 received total body irradiation 450 to 600 cGy. Autologous bone marrow transplantation (ABMT) was performed with marrow that was unpurged (n = 2) or purged ex vivo (n = 26) with 6-hydroxydopamine (6-OHDA) 20 micrograms/ml plus ascorbate 200 micrograms/ml. Cytotoxic effects of 6-hydroxydopamine, merocyanine-540 and related compounds on human neuroblastoma and hematopoietic stem cells.

6-Hydroxydopamine(6-OHDA) and Merocyanine-540(MC-540) have been used clinically for purging of neuroblastoma cells prior to autologous bone marrow transplantation. Both substances were found to be more toxic against neuroblastoma cells than against hematopoietic stem cells. The more pronounced cytotoxic effects of 6-OHDA against neuroblastoma cells were not caused by its selective uptake; the rapid autooxidation at physiological pH leads to the formation of H2O2 already in the incubation medium. Cytotoxic effects were not detected in short-time test systems (4 hour chromium-51 release assay) b

Assessment of purine-dopamine interactions in 6-hydroxydopamine-lesioned rats: evidence for pre- and postsynaptic influences by adenosine.

Lesch-Nyhan syndrome involves disorders of both purine and dopamine metabolism. Neonatal lesioning of dopaminergic neurons with 6-hydroxydopamine (6-OHDA) has been proposed as a rodent model of the dopamine deficiency in this childhood disorder. In the present studies, the functional interaction between purines and dopamine was examined in adult rats which received 6-OHDA lesions either as neonates or as adults. Even though dopamine levels were decreased by at least 92%, both neonatal- and adult-6-OHDA-lesioned rats had normal hypoxanthine-guanine phosphoribosyltransferase function and purine

Properties of rotational behaviour produced by methylxanthine derivatives in mice with unilateral striatal 6-hydroxydopamine-induced lesions.

The present investigation was undertaken to study effects of methylxanthine derivatives on rotational behaviour produced by dopamine receptor stimulating drugs and properties of methylxanthine-induced rotation in mice with unilateral 6-hydroxydopamine (6-OHDA)-induced lesions of the striatum. L-Dopa 10mg/kg, i.p., produced contralateral turning which lasted for about 40 min. When L-dopa 10mg/kg was given to mice in combination with theophylline 25 mg/kg, total turns for 2 hr were significantly higher than those of L-dopa and theophylline alone. Caffeine and theophylline in doses of 25 and 50 m

The effect of depletion of brain dopamine by 6-hydroxydopamine on tolerance to the anorexic effect of d-amphetamine and fenfluramine in rats.

The effects on food intake of repeated administration of d-amphetamine and fenfluramine were examined in rats adapted to a 4 hr/day feeding schedule. Control rats given daily injections of amphetamine before food access rapidly developed a partial tolerance to the food intake reducing effect of amphetamine. However, rats given a 6-hydroxydopamine (6-HDA) treatment which produced a large selective depletion of dopamine in brain failed to display tolerance to amphetamine-induced anorexia after either 10 or 23 days of amphetamine injections. Although higher than normal doses of amphetamine were r

Decreased intoxicating effect of ethanol in rats after 6-hydroxydopamine-induced degeneration of ascending dopamine pathways.

Selective lesion of ascending dopamine pathways was made by injecting the neurotoxin 6-OHDA (8 microgram/4 microliter) bilaterally close to the nigro-striatal dopamine pathway of 18 male Long Evans rats. Similar injections of the vehicle were given to 10 control rats. Two months after the operation intoxication was measured in a tilting-plane test after an injection of ethanol (2 g/kg, IP). Ethanol impaired the performance of the 6-OHDA-treated rats significantly less than that of the controls. This finding suggests a role for the central dopamine neurons in the intoxicating effect of ethanol.

The effect of intraventricular 6-hydroxydopamine on the content of oxytocin and vasopressin in the hypothalamus and pituitary gland of water-deprived rats.

The effect of intraventricular 6-hydroxydopamine on the content of oxytocin and vasopressin in the hypothalamus and pituitary gland of water deprived rats. Acta Physiol. Pol., 1977, 28 (6): 497-504. Rats received one infusion of 200 microgram 6-hydroxydopamine with 25 microgram of ascorbic acid into the lateral cerebral ventricle. After 57 days some rats were deprived of water for 4, 8 or 12 days. Then, the animals were sacrificed by decapitation. Oxytocin was determined in extracts from the posterior pituitary lobe and hypothalamus by the method of Van Dongen and Hays, while the vasopressin c

Exploring Cognitive and Affective Outcomes Following Bilateral 6-Hydroxydopamine Lesions of the Substance Nigra or Striatum in Rats.

The majority of animal models of Parkinson's disease (PD) focus on motor symptoms that are induced by unilateral injections of such neurotoxins as 6-hydroxydopamine (6-OHDA) in nigrostriatal dopaminergic pathways. However, motor changes that are induced by unilateral 6-OHDA injections may interfere with the identification of cognitive and affective dysfunctions induced by dopaminergic neurodegeneration. To select an appropriate method for studying nonmotor symptoms of PD and potential neuroprotective treatments, the present study compared behavioral effects of bilateral 6-OHDA infusions direct

Pathogenic mechanism underlying parkinsonism induced by neurotoxicants (MPTP and 6-hydroxydopamine) and α-synuclein: a unifying hypothesis.

The mechanism underlying the selective loss of dopaminergic neurons in Parkinson's disease (PD) is still not understood at present. MPTP, an illicit drug contaminant, can selectively induce parkinsonism in humans and animals which is very similar to idiopathic PD. Like MPTP, 6-hydroxydopamine (6-OHDA) is another neurotoxicant also capable of selectively inducing parkinsonism in animal models. In this paper, a unifying hypothesis is proposed, which offers a plausible explanation for the pathogenic mechanism of parkinsonism induced by MPTP and 6-OHDA. This hypothesis has three core elements. (i)

The Natural Neuroprotective Compounds Used in the 6-Hydroxydopamine- Induced Parkinson's Disease in Zebrafish: The Current Applications and Perspectives.

BACKGROUND AND OBJECTIVE: Parkinson's disease (PD) is a neurodegenerative disorder characterized by the degeneration of the dopaminergic neurons in the substantia nigra pars compacta, resulting in the loss of dopamine in the striatum, leading thus to the PD classic movement symptoms: resting tremor, rigidity, and bradykinesia/akinesia. Furthermore, Levodopa's efficacy declines with long-term use, generating serious motor complications. Neuroprotection implies the use of different agents exhibiting various neuroprotective strategies to prevent brain degeneration and neuron loss. The present rev

NOD2 promotes dopaminergic degeneration regulated by NADPH oxidase 2 in 6-hydroxydopamine model of Parkinson's disease.

BACKGROUND: In Parkinson's disease (PD), loss of striatal dopaminergic (DA) terminals and degeneration of DA neurons in the substantia nigra (SN) are associated with inflammation. Nucleotide-binding oligomerization domain-containing protein (NOD)2, one of the first discovered NOD-like receptors, plays an important role in inflammation. However, the role of NOD2 has not been elucidated in PD. METHODS: NOD2 mRNA and protein expression in the SN and the striatum of C57BL/6 mice treated with 6-hydroxydopamine (6-OHDA) was measured. We next investigated the potential contribution of the NOD2-depen

[Synchrotron radiation-based FTIR microspectroscopy study of 6-hydroxydopamine induced Parkinson's disease cell model].

SH-SY5Y cell line treated with 6-hydroxydopamine (6-OHDA) is a classical Parkinson's disease (PD). In the present study, synchrotron-based Fourier transform infrared (FTIR) microspectroscopy was used to analyze the biochemical composition of SH-SY5Y cell line treated with 6-OHDA. The detailed spectral analyses show the significant changes in cellular compositions such as lipids, proteins and nucleic acids in SH-SY5Y cells treated with 6-OHDA compared to control SH-SY5Y cells. As a result, the unsaturation levels of phospholipids decrease in SH-SY5Y cells treated with 6-OHDA compared to control

Alterations of NMDA receptor binding in various brain regions among 6-hydroxydopamine-induced Parkinsonian rats.

The N-methyl-d-aspartate (NMDA) system closely interacts with the dopaminergic system and is strongly implicated in the pathophysiological mechanisms and therapeutic paradigms of Parkinson's disease. This study aims to systematically investigate the changes of NMDA receptors in a wide range of brain structures 3 weeks after unilateral medial forebrain bundle lesion by 6-hydroxydopamine (6-OHDA). NMDA receptor distributions and alterations in the post-mortem rat brain were detected by [(3)H] MK-801 binding autoradiography. In the 6-OHDA-induced Parkinsonian rat model, nigrostriatal dopaminergic

Exercise partly reverses the effect of maternal separation on hippocampal proteins in 6-hydroxydopamine-lesioned rat brain.

Animals subjected to maternal separation stress during the early stages of development display behavioural, endocrine and growth factor abnormalities that mirror the clinical findings in anxiety/depression. In addition, maternal separation has been shown to exacerbate the behavioural deficits induced by 6-hydroxydopamine (6-OHDA) in a rat model of Parkinson's disease. In contrast, voluntary exercise reduced the detrimental effects of 6-OHDA in the rat model. The beneficial effects of exercise appeared to be largely due to compensation in the non-lesioned hemisphere. The aim of the present stud

Destruction of dopaminergic neurons in the midbrain by 6-hydroxydopamine decreases hippocampal cell proliferation in rats: reversal by fluoxetine.

BACKGROUND: Non-motor symptoms (e.g., depression, anxiety, and cognitive deficits) in patients with Parkinson disease (PD) precede the onset of the motor symptoms. Although these symptoms do not respond to pharmacological dopamine replacement therapy, their precise pathological mechanisms are currently unclear. The present study was undertaken to examine whether the unilateral 6-hydroxydopamine (6-OHDA) lesion to the substantia nigra pars compacta (SNc), which represents a model of long-term dopaminergic neurotoxicity, could affect cell proliferation in the adult rat brain. Furthermore, we exa

6-hydroxydopamine-mediated release of norepinephrine increases faecal excretion of Salmonella enterica serovar Typhimurium in pigs.

Salmonella enterica serovar Typhimurium is an animal and zoonotic pathogen of worldwide importance. In pigs, transport and social stress are associated with reactivation and spread of Salmonella Typhimurium infection. The stress-related catecholamine norepinephrine (NE) has been reported to activate growth and virulence factor expression in Salmonella; however the extent to which NE contributes to stress-associated salmonellosis is unclear. We studied the impact of releasing NE from endogenous stores during Salmonella Typhimurium infection of pigs by administration of 6-hydroxydopamine (6-OHDA

Effect of subthalamic nucleus lesions in a 6-hydroxydopamine-induced rat parkinsonian model: behavioral and biochemical studies.

OBJECT: The purpose of this study was to determine whether subthalamic nucleus (STN) ablation caused by kainic acid can restore dopaminergic neurotransmission and improve motor deficits in a 6-hydroxydopamine (6-OHDA)-induced hemiparkinsonian model. METHODS: The authors investigated behavioral changes in rats displaying parkinsonian symptoms (6-OHDA-lesioned rats) after an STN lesion was created using kainic acid. They also measured levels of dopamine and its metabolites following tissue dissection. The results of this study showed that STN ablation led to behavioral improvement in parkinsoni

Fate of (D-Ala2)-deltorphin-I-like immunoreactive neurons in 6-hydroxydopamine lesioned rat brain.

The use of a polyclonal antiserum specific to C-terminal tetrapeptide amide of (D-Ala2)deltorphin-I, a naturally occurring amphibian skin opioid peptide, has already demonstrated the presence of immunoreactive neurons in rat midbrain. Double immunostaining identified these neurons as a subpopulation of the mesencephalic dopaminergic neurons that were also tyrosine hydroxylase-immunopositive and calbindin-D28kD- negative, namely, the neurons predominantly affected in Parkinson disease. We followed the fate of these neurons after a monolateral injection of 6-hydroxy-dopamine into rat brain. Almo
