alpha-Phenyl-N-tert-butylnitrone attenuates excitotoxicity in rat striatum by preventing hydroxyl radical accumulation.

Various in vitro experiments have indicated that oxygen-derived free radicals may contribute to excitotoxic neuronal death. In the present study we induced excitotoxicity in rat striatum by perfusing glutamate at a high concentration through a microdialysis probe. We observed an increased formation of hydroxyl radicals (.OH) during the perfusion of the excitotoxin and an extensive striatal lesion 24 h after the insult. The spin trap, alpha-phenyl-N-tert-butylnitrone (PBN), attenuated both hydroxyl radical levels and the volume of the lesion. This result suggests that the neuroprotection may be

Cardiac microdialysis of salicylic acid to detect hydroxyl radical generation associated with sympathetic nerve stimulation.

We examined the relationship between norepinephrine (NE) and hydroxyl free radical (OH) generation on cardiac nerve stimulation. Salicylic acid in Ringer's solution (0.5 nmol microliter-1 min-1) was infused directly through a microdialysis probe to detect the generation of hydroxyl radicals (OH) as reflected by the formation of dihydroxybenzoic acid (DHBA) in the myocardium of anesthetized rats. Sympathetic nerve stimulation increased the release of NE and the formation of DHBA. A positive linear correlation between the release of NE and the formation of 2,3-DHBA (R2 = 0.982) or 2,5-DHBA (R2 =

Sensitive assay of hydroxyl free radical formation utilizing high pressure liquid chromatography with electrochemical detection of phenol and salicylate hydroxylation products.

Evidence is presented for a sensitive method useful for the detection of hydroxyl free radical generation in various systems. The methodology employs high pressure liquid chromatography with electrochemical detection (LCED) for the quantification and identification of the hydroxylation products from the reaction of OH with both phenol and salicylate. A detection limit of less than 1 pmol for the hydroxylation products has been achieved with electrochemical detector responses linear over at least three orders of magnitude. Detection and quantitation of the hydroxylation products obtained and fo

Hydroxyl radical generation by the tetracycline antibiotics with free radical damage to DNA, lipids and carbohydrate in the presence of iron and copper salts.

Tetracycline antibiotics caused the degradation of carbohydrate in the presence of a ferric salt at pH 7.4. This degradation appeared to involve hydroxyl radicals since the damage was substantially reduced by the presence of catalase, superoxide dismutase, scavengers of the hydroxyl radical and metal chelators. Similarly, the tetracycline antibiotics in the presence of a ferric salt greatly stimulated the peroxidation of liposomal membranes. This damage, which did not implicate the hydroxyl radical, was significantly reduced by the addition of chain-breaking antioxidants and metal chelators. O

Hydroxyl radical chemistry in advanced oxidation processes: a cross-system mechanistic framework and catalyst design perspectives.

Hydroxyl radicals (˙OH) are widely recognized as the most powerful oxidizing species in advanced oxidation processes (AOPs) and play a central role in the degradation of persistent organic contaminants. However, current understanding of ˙OH chemistry remains fragmented because radical generation is typically interpreted within technology-specific frameworks, limiting cross-system mechanistic understanding and catalyst development. This review establishes a unified framework for ˙OH generation across AOPs and demonstrates that diverse technologies can be interpreted through three recurring mech

Hydroxyl radical from iridium(III)-based photosensitizer triggers PANoptosis and ferroptosis for hypoxia-tolerant photodynamic immunotherapy.

Exploring photosensitizers that efficiently generate oxygen-independent hydroxyl radical (•OH) and activate highly immunogenic PANoptosis and ferroptosis shows promise in addressing the limited efficacy of photodynamic immunotherapy (PDI) in the hypoxic and immunosuppressive tumor microenvironment (TME). However, challenges still exist due to the unsatisfactory •OH production efficiency in most organic photosensitizers and the unresolved relationship between •OH and PANoptosis. Here, we designed and synthesized two novel iridium(III) complexes, Ir1 and Ir2. Notably, Ir1 produced more type I/II

Side-Chain Sulfonation to Modulate Hydroxyl Radical Generators for Efficient Suppression of Hepatoma Cells under Hypoxia.

The development of type I photosensitizer (PS) targeting subcellular organelles that directly destroy key subcellular compartments and overcome the hypoxic environment has become a critical breakthrough for improving therapeutic effects on tumors. Herein, we proposed a simple tactic of side-chain sulfonation to construct the subcellular organelles targeting type I PS. Initially, the traditional lipophilic donor-π-acceptor (D-π-A) system (3SSYDI) was constructed, which exhibited type II reactive oxygen species generation capability and lipid droplets (LDs)-targeting property. Innovatively, the

Overlooked pyrite-mediated heterogeneous Fenton processes: Mechanisms of surface hydroxyl radical generation and associated decontamination performance.

Pyrite-based Fenton-like processes have been extensively studied for wastewater decontamination; however, most relevant studies placed excessive emphasis on the homogeneous Fenton reaction mediated by aqueous Fe2+, resulting in the proposed technologies facing issues such as additional acid requirements for pH adjustment and excessive iron sludge production. Herein, through in situ shell-isolated nanoparticle-enhanced Raman spectroscopy (SHINERS), custom dual-chamber reactor experiments, and a series of control experiments, significant hydroxyl radical generation was identified during the pyri

Promote hydroxyl radical and key intermediates formation for deep toluene mineralization via unique electron transfer channel.

The degradation and mineralization of volatile organic compounds (VOCs) in gas-solid phase photocatalytic systems suffer great challenges due to the low electron transfer efficiency and slow benzene ring-opening kinetics. Hence, a heterojunction photocatalyst of Bi2SiO5/TiO2 has been synthesized by a facile method. Bi2SiO5/TiO2 shows the ability of mineralizing toluene to CO2 with a degradation rate of 85.5%, while TiO2 is 49.0% and presents a continuous deactivation. Experimental characterizations and theoretical calculations indicate that a unique electron transfer channel of Bi/Si-O-Ti can

Preferential recognition of hydroxyl radical-modified superoxide dismutase by circulating autoantibodies in patients with alopecia areata.

BACKGROUND: Alopecia areata (AA) is a common form of localized, non-scarring hair loss. The cause of AA is unknown but reports suggest an autoimmune etiology, where oxygen free radicals play an important role. OBJECTIVE: The aim of this study was to investigate the role of a hydroxyl radicals (·OH)-modified antioxidant enzyme, superoxide dismutase (SOD), in AA autoimmunity. METHODS: SOD was modified by ·OH radicals. Binding characteristics of autoantibodies in AA patients (n=26) against ·OH-modified SOD (·OH-SOD) were evaluated by immunoassays and the results were compared with those of heal

Quantifying protein interface footprinting by hydroxyl radical oxidation and molecular dynamics simulation: application to galectin-1.

Biomolecular surface mapping methods offer an important alternative method for characterizing protein-protein and protein-ligand interactions in cases in which it is not possible to determine high-resolution three-dimensional (3D) structures of complexes. Hydroxyl radical footprinting offers a significant advance in footprint resolution compared with traditional chemical derivatization. Here we present results of footprinting performed with hydroxyl radicals generated on the nanosecond time scale by laser-induced photodissociation of hydrogen peroxide. We applied this emerging method to a carb

A hydroxyl radical-like species oxidizes cynomolgus monkey artery wall proteins in early diabetic vascular disease.

Recent evidence argues strongly that the marked increase in risk for atherosclerotic heart disease seen in diabetics cannot be explained by a generalized increase in oxidative stress. Here, we used streptozotocin to induce hyperglycemia in cynomolgus monkeys for 6 months and tested whether high glucose levels promote localized oxidative damage to artery wall proteins. We focused on three potential agents of oxidative damage: hydroxyl radical, tyrosyl radical, and reactive nitrogen species. To determine which pathways operate in vivo, we quantified four stable end products of these reactants --

Hydroxyl Radical-Driven Oxidation as a Key Pathway for Greenhouse Gas Production During Soil Drying-Rewetting.

The drying-rewetting of soil can increase the release of greenhouse gases over a short time period and is one of the key pathways for greenhouse gas emissions in many terrestrial ecosystems, particularly in drylands. The mechanisms underlying this pulse of greenhouse gas emissions remain nearly unknown. Here, we conducted simulated soil rewetting experiments using typical cover of dryland soils (bareland, cyanobacteria/lichen-covered soil, and moss-covered soil). The 13C, 15N, and 18O labeling techniques allowed to explore the intrinsic mechanisms of rapid carbon (C) and nitrogen (N) release f

Dissolved organic matter promoted hydroxyl radical formation and phenanthrene attenuation during oxygenation of iron-pillared montmorillonites.

The oxygenation of Fe(II)-bearing minerals for hydroxyl radicals (HO•) formation and contaminant attenuation receives increasing attentions. However, information on dissolved organic matter (DOM) with different types, concentrations, and molecular weights (MWs) in manipulating HO• formation and contaminant attenuation during mineral oxygenation remain unclear. In this study, four iron-pillared montmorillonites (IPMs) and two DOM samples [e.g., humic acids (HA) and fulvic acids (FA)] were prepared to explore the HO• formation and phenanthrene attenuation during the oxygenation of IPMs in the pr

Cupric ion reducing antioxidant capacity assay for antioxidants in human serum and for hydroxyl radical scavengers.

Tests measuring the combined antioxidant effect of the nonenzymatic defenses in biological fluids may be useful in providing an index of the organism's capability to counteract reactive species known as pro-oxidants, resist oxidative damage, and combat oxidative stress-related diseases. The selected chromogenic redox reagent for the assay of human serum should be easily accessible, stable, selective, and respond to all types of biologically important antioxidants such as ascorbic acid, alpha-tocopherol, beta-carotene, reduced glutathione (GSH), uric acid, and bilirubin, regardless of chemical

Development of a group contribution method to predict aqueous phase hydroxyl radical (HO*) reaction rate constants.

The hydroxyl radical (HO*) is a strong oxidant that reacts with electron-rich sites of organic compounds and initiates complex chain mechanisms. In order to help understand the reaction mechanisms, a rule-based model was previously developed to predict the reaction pathways. For a kinetic model, there is a need to develop a rate constant estimator that predicts the rate constants for a variety of organic compounds. In this study, a group contribution method (GCM) is developed to predict the aqueous phase HO* rate constants for the following reaction mechanisms: (1) H-atom abstraction, (2) HO*

Selective brain hypothermia protects against hypoxic-ischemic injury in newborn rats by reducing hydroxyl radical production.

We hypothesized that selective brain hypothermia (SBHT) decreases production of hydroxyl radicals (*OH) induced by hypoxia-ischemia (H-I) and reperfusion and attenuates neuronal damage in neonatal rat brain. Anesthetized 7-day-old rats were divided into a normothermia (NT) group (n=6) and a SBHT group (n=7) and subjected to 90-min H-I, followed by a 90-min recovery period. Brain temperature (BT) was regulated by a water-cooled metallic plate placed under the head. The BT of the SBHT group was set at 31.0+/-1.0 degrees C during the H-I and recovery period. A microdialysis study investigating the mechanisms of hydroxyl radical formation in rat striatum exposed to glutamate.

Considerable evidence has linked hydroxyl radicals (.OH) to excitotoxicity. Glutamate infused through a microdialysis probe into rat striatum induced a massive .OH production, which was completely blocked by PBN and attenuated by dizocilpine, 2-amino-5-phosphonopentanoic acid (AP-5), NG-nitro-L-arginine methyl ester (L-NAME) and mepacrine. Thus, we suggest that the neurotoxic effects of glutamate in vivo may derive from an increased formation of .OH resulting from excessive activation of NMDA receptors and downstream enzymes such as NOS and PLA2.

1-Trichloromethyl-1,2,3,4-tetrahydro-beta-carboline increases extracellular serotonin and stimulates hydroxyl radical production in rats.

1-Trichloromethyl-1,2,3,4-tetrahydro-beta-carboline (TaClo), a neurotoxin structurally similar to the dopaminergic neurotoxin MPTP, may be formed in humans treated with chloral hydrate or exposed to trichloroethylene, a widely used industrial solvent. Systemically administered TaClo (0.4 mg/kg, i.p.) induced an immediate and transient release of dopamine (DA) and serotonin (5-HT) measured using microdialysis. However, only 5-HT was increased significantly (area under the curve, AUC, for the 1-2 h-period following TaClo administration: 400% compared with the respective control value; 2-3 h-peri

Oxidative stress and the mobilisation of arachidonic acid in stimulated human platelets: role of hydroxyl radical.

Platelet functions, including eicosanoid biosynthesis, can be significantly altered by exposure to reactive oxygen species. We utilised the redox properties of the phenazine derivative, pyocyanin, to generate low micromolar levels of reactive oxygen species in order to investigate the metabolism of arachidonic acid by human platelets under oxidative stress. Eicosanoid production by platelets, pre-labelled with [3H]arachidonic acid (AA) and stimulated with the calcium ionophore A23187, was inhibited in the presence of pyocyanin. In contrast, platelets pre-treated with pyocyanin and concurrently
