Experimental evidence of fluid secretion of rabbit lacrimal gland duct epithelium.

PURPOSE: To investigate the osmotic water permeability of lacrimal gland (LG) duct epithelium by means of calculation of filtration permeability and to investigate LG ductal fluid secretion. METHODS: Experiments were performed on isolated rabbit LG duct segments maintained in short-term culture. Osmotically determined fluid movement or fluid secretion into the closed intraluminal space of cultured LG interlobular ducts was analyzed using video microscopic technique. RESULTS: The end of the LG ducts sealed after overnight incubation forming a closed luminal space. For the calculation of osmot

Low osmolality and shear stress during liposuction impair cell viability in autologous fat grafting.

BACKGROUND: Liposuction and subsequent autologous fat grafting have become essential techniques for fat augmentation in plastic surgery. However, standard harvesting techniques that ensure the survival of adipocytes and stromal vascular fraction (SVF) cells and thus preserve the transplanted fat volume are lacking. In particular, the effect of different parameters of the tumescent solution has not been studied in this context. We hypothesized that the osmolality of the tumescent solution could have a significant effect on the survival of adipocytes and SVF cells. METHODS: We developed two dis

Glycine-induced hypo-osmolar hyponatremia.

BACKGROUND: Hyponatremia is commonly observed following transurethral resection of the prostate or endometrial resection when the operative field is irrigated with hypotonic glycine. Although glycine-induced hyponatremia has been associated with brain damage, the mortality is low, and it has been suggested that the condition might not be hypo-osmolar and thus might not cause brain edema. OBJECTIVE: To determine if glycine-induced hyponatremia is a hypo-osmolar condition. METHODS: The study was a retrospective evaluation of 13 men who underwent transurethral resection of the prostate and 5 wo

A flow EPR study of deformation and orientation characteristics of erythrocyte ghosts: effects of lysing and resealing conditions.

The effects of various conditions in lysing and resealing the red cell membrane on the degree of ghost deformation and orientation in flow are investigated using the flow EPR and spin-label method. The relatively low deformability of the standard ghost, which is lysed and resealed, respectively, in hypotonic and isotonic NaCl-Tris buffer, is markedly enhanced by the presence of Mg-ATP, chlorpromazine, or Ca2+ ion during resealing. The effect is concentration dependent, and there is an optimal level for each treatment. Chlorpromazine and Ca2+ are also effective when added to the resealed ghosts

Human erythrocytes as drug carriers: loading efficiency and side effects of hypotonic dialysis, chlorpromazine treatment and fusion with liposomes.

Human red blood cells (RBCs) are emerging as a highly biocompatible microparticulate drug delivery system. So far, drugs have commonly been loaded into freshly isolated RBCs using rather disruptive methods based on hypotonic shock, and assessment of damage was restricted to hemolysis. Here, we investigated loading of RBCs from blood bank units with enzymes of various molecular weights using hypotonic dialysis (HD), pretreatment with chlorpromazine (CPZ) and fusion with liposomes. The latter two techniques have received little attention in RBC loading so far. Along with loading efficiency, all

Volume regulation in cultured cells derived from human retinal pigment epithelium.

To characterize volume regulatory mechanisms, unidirectional Rb+ efflux and influx, unidirectional Cl- influx, and cell volume were measured in cultured human retinal pigment epithelium (HRPE). The HRPE was found to be capable of both regulatory volume increase (RVI), in response to a hypertonic challenge, and regulatory volume decrease (RVD), in response to a hypotonic challenge. Bumetanide-sensitive Rb+ influx increased almost threefold on incubation in a hypertonic (390 mosmol/kgH2O) medium. Bumetanide-insensitive Rb+ influx was activated by hypotonic (190 mosmol/kgH2O) challenge as well as

[Non-ketosic hyperosmolar hyperglycemic coma. Case reports and therapeutic consideration].

Non-ketonic hyperosmolar hyperglycaemic coma (N.K.H.H.C.) is by no means uncommon in diabetes. Its picture includes sensorial depression, hyperglycaemia, hyperazotemia, marked dehydration and plasma hyperosmolarity. It is mostly found in elderly subjects with non-serious diabetes. Reference is made to 6 personal cases observed during a period of 14 months. The incidence of N.K.H.H.C. noted during this period was 2.2%; this was higher than that of ketoacidotic coma. Two patients died from hypovolaemic shock and one from septic complications. Three survived the episode. Treatment was based on th

Low-detergent sonication decellularization preserves extracellular matrix architecture and enhances cell infiltration in human acellular dermal matrix.

INTRODUCTION: Acellular dermal matrix (ADM) scaffolds support tissue regeneration by providing structural integrity, biological cues, and a permissive microenvironment; optimal ADMs integrate with native tissue while preserving extracellular matrix (ECM) composition, microarchitecture, and biomechanics. Decellularization is a critical step in ADM development, as incomplete cellular removal can induce inflammation, whereas excessive chemical treatment can disrupt ECM structure and impair scaffold biofunctionality. METHODS: Here, we optimized human dermal decellularization to develop ADMs that

Activation of a chondrocyte volume-sensitive Cl(-) conductance prior to macroscopic cartilage lesion formation in the rabbit knee anterior cruciate ligament transection osteoarthritis model.

OBJECTIVE: The anterior cruciate ligament transection (ACLT) rabbit osteoarthritis (OA) model confers permanent knee instability and induces joint degeneration. The degeneration process is complex, but includes chondrocyte apoptosis and OA-like loss of cartilage integrity. Previously, we reported that activation of a volume-sensitive Cl(-) current (ICl,vol) can mediate cell shrinkage and apoptosis in rabbit articular chondrocytes. Our objective was therefore to investigate whether ICl,vol was activated in the early stages of the rabbit ACLT OA model. DESIGN: Adult Rabbits underwent unilateral

Rapid, effective, and long-lasting behavioral recovery produced by microsutures, methylene blue, and polyethylene glycol after completely cutting rat sciatic nerves.

Behavioral function lost in mammals (including humans) after peripheral nerve severance is slowly (weeks to years) and often poorly restored by 1-2-mm/day, nonspecifically directed outgrowths from proximal axonal stumps. To survive, proximal stumps must quickly repair (seal) plasmalemmal damage. We report that, after complete cut- or crush-severance of rat sciatic nerves, morphological continuity, action potential conduction, and behavioral functions can be consistently (>98% of trials), rapidly (minutes to days), dramatically (70-85% recovery), and chronically restored and some Wallerian dege

Control of deaths from diarrheal disease in rural communities. I. Design of an intervention study and effects on child mortality.

From May through October 1980, the "Strengthening Rural Health Delivery" project (SRHD) under the Rural Health Department of the Ministry of Health of Egypt had conducted an investigation into prevention of child mortality from diarrheal disease through testing various modules of Oral Rehydration Therapy delivery mechanisms. In a six-cell design counting a total of almost 29,000 children, ORT was provided both as hypotonic sucrose/salt solution prepared and administered by mothers and normotonic, balanced electrolyte solution in the hands of both mothers and health care providers and the effec

Involvement of suppressor cells induced with membrane fractions of trypanosomes in immunosuppression of trypanosomiasis.

We showed that infection with Trypanosoma congolense in mice led to suppression of listeria-induced delayed type hypersensitivity (DTH). Mice were pre-treated with irradiated T. evansi, which caused rapid and effective suppression of DTH. A membrane fraction obtained by homogenizing T. evansi variant in a hypotonic buffer solution and centrifuging it at 150,000g produced suppression of listeria-induced DTH when injected i.p. into mice as early as 1 day before listeria immunization. Furthermore, we demonstrated by an adoptive transfer system that the suppressor cells involved in this suppressio

[Focus on Albumin: new perspectives for the clinical application in intensive care medicine : From the quantitative routine to qualitative individualized theranostics].

Human albumin plays a double role in intensive care medicine. It is therapeutically administered as a colloid and diagnostically employed as a laboratory parameter for risk and monitoring classification. For both fields of application, a quantitative logic has so far been dominant. Large randomized studies consistently show that human albumin is overall safer than crystalloids but does not provide a general benefit in most intensive care medical scenarios. Subgroup-specific risks must be considered: in cases of traumatic brain injuries the administration of hypotonic 4% albumin solution was as

Fluid resuscitation in diabetic emergencies--a reappraisal.

The first objective in diabetic ketoacidosis is to restore the circulating volume and improve tissue perfusion. In any form of hypovolaemic shock the most efficient way of restoring circulating volume is to be use colloid solutions rather than crystalloids. At least three times the amount of crystalloid must be used to achieve the same effect. The historical reason for using isotonic saline in diabetic ketoacidosis is related not to its similarity to the fluid lost, but to its supposed efficiency in correcting the circulating volume. Excess crystalloid expands the interstitial space which resu

Biconcave shape of human red-blood-cell ghosts relies on density differences between the rim and dimple of the ghost's plasma membrane.

The shape of the human red blood cell is known to be a biconcave disk. It is evident from a variety of theoretical work that known physical properties of the membrane, such as its bending energy and elasticity, can explain the red-blood-cell biconcave shape as well as other shapes that red blood cells assume. But these analyses do not provide information on the underlying molecular causes. This paper describes experiments that attempt to identify some of the underlying determinates of cell shape. To this end, red-blood-cell ghosts were made by hypotonic hemolysis and then reconstituted such th

Transepithelial fluid shift generated by osmolarity gradients in unstimulated perfused rat submandibular glands.

The effects of osmotic gradients on transepithelial water movements were examined in unstimulated perfused submandibular glands of the rat. Osmotic gradients were applied transepithelially by adding sucrose to or removing it from the perfusate. An infusion of hypotonic perfusate shifted fluid from the interstitium to the lumen (luminal fluid shift) transiently, whereas an infusion of hypertonic perfusate shifted fluid from the lumen to the interstitium (interstitial fluid shift) transiently. The amount of fluid shifted from lumen to interstitium increased as the luminal fluid osmolarity was ra

ClC-2 Contributes to Hypotonicity-Induced Adrenal Aldosterone Secretion.

AIM: The zona glomerulosa (ZG) of the adrenal cortex regulates blood pressure and electrolyte homeostasis through aldosterone production. In ZG cells, potassium and angiotensin II (Ang II) trigger calcium oscillations that drive aldosterone synthesis. Changes in serum osmolality also modulate aldosterone production in a chloride-dependent fashion, but the involved proteins remain unclear. Because the chloride channel ClC-2 is activated by hypoosmolality, we investigated its role in ZG osmoregulation. METHODS: We used Clcn2 knockout (KO) and wild-type (WT) mice. Explanted adrenal glands were i
