The role of IgG antibodies from irradiated cercaria-immunized rabbits in the passive transfer of immunity to Schistosoma mansoni-infected mice.

Antibodies of the IgG subclass isolated from the sera of rabbits immunized with cercariae subjected to 50 kilorads of gamma irradiation passively provided partial immunity against Schistosoma mansoni challenge in C57B1/6J mice. These mice exhibited reductions in adult worm burdens of 43-61% compared with recipients of normal rabbit antibodies. Passively transferred IgG antibodies were most effective when given 4-7-days postchallenge; they were less effective when given just before challenge, and were totally ineffective when given 15 days postchallenge. It was also shown that the Fc portion of

Preclinical Evaluation of Chicken Egg Yolk Antibody (IgY) Anti-RBD Spike SARS-CoV-2-A Candidate for Passive Immunization against COVID-19.

The coronavirus disease 2019 (COVID-19) has become a substantial threat to the international health sector and the global economy. As of 26 December 2021, the number of mortalities resulting from COVID-19 exceeded 5.3 million worldwide. The absence of an effective non-vaccine treatment has prompted the quest for prophylactic agents that can be used to combat COVID-19. This study presents the feasibility of chicken egg yolk antibody (IgY) anti-receptor-binding domain (RBD) spike SARS-CoV-2 as a strong candidate to neutralize the virus for application in passive immunization. For the purpose of

Systemic antibodies administered by passive immunization prevent generalization of the infection by foot-and-mouth disease virus in cattle after oronasal challenge.

The role of passively transferred sera in the protection against aerogenous foot-and-mouth disease (FMD) virus infection in cattle was evaluated using vaccine-induced immune serum preparations obtained at 7 and 26 days post-vaccination (dpv). We showed that circulating antibodies were sufficient to prevent disease generalization after oronasal infection in animals passively transferred with 26-dpv serum but not with the 7-dpv serum. Conversely, conventional FMD vaccination provided clinical protection at 7 dpv, promoting fast and robust antibody responses upon challenge and even though antibod

Passive immunity to porcine epidemic diarrhea virus following immunization of pregnant gilts with a recombinant orf virus vector expressing the spike protein.

Passive immunity is critical for protection of neonatal piglets against porcine epidemic diarrhea virus (PEDV). Here, we investigated the immunogenicity of an orf virus (ORFV) vector expressing the full-length spike (S) protein of PEDV (ORFV-PEDV-S) in pregnant gilts and its ability to confer passive immunity and protection in piglets. Three doses of ORFV-PEDV-S were given to two groups of PEDV-negative pregnant gilts, with the last dose being administered two weeks prior to farrowing. One of the two groups immunized with the ORFV-PEDV-S recombinant virus was also exposed to live PEDV orally o

The active immune role of keratinocytes: from passive targets to key hubs in psoriatic inflammation.

Psoriasis is a chronic immune-mediated inflammatory skin disease historically viewed as a disorder of keratinocyte hyperplasia, with subsequent recognition of central immune cell involvement. However, accumulating multi-omics and functional genomic evidence has reshaped this paradigm, positioning keratinocytes not merely as passive targets of immune attack but as active participants within a bidirectional epithelial-immune network that drives inflammatory initiation, maintenance, and amplification. This critical narrative review synthesizes the molecular mechanisms by which keratinocytes integ

Evaluating the feasibility of using digital Brix refractometry to determine the passive immune status of lambs.

BACKGROUND: Determining serum immunoglobulin G (IgG) concentrations is critical for the assessment of passive immune status in lambs. This study aimed to assess the accuracy of the Brix refractometer for the determination of passive immune status in lambs. METHODS: Blood samples were collected from 417 randomly selected healthy 1‒7-day-old lambs from nine farms in western Türkiye. Digital Brix refractometry was used as the index test to determine the lambs' serum IgG concentrations, and the radial immunodiffusion (RID) technique was used as the reference test. RESULTS: A strong correlation w

Comparison of the Effectiveness of Colostrum, Colostrum Powder and Combined Feeding on Transfer of Passive Immunity in Calves.

BACKGROUND: High-quality colostrum is essential to prevent failed transfer of passive immunity (FTPI) in calves, and research on colostrum substitutes has continued over time when quality colostrum is unavailable. OBJECTIVES: This study evaluated the effects of fresh colostrum (COL), colostrum powder (POW) and a combination (MIX) on TPI in neonatal calves. METHODS: Forty-five neonatal Holstein-Friesian calves were divided into COL (4 L fresh colostrum), POW (1080 g colostrum powder in 4 L water) and MIX (540 g powder in 4 L colostrum) groups. Total protein (TP), Brix%, and IgG concentrations

Risk Factors for and Prevalence of Failure of Passive Transfer of Immunity in Calves on Dairy Farms in Türkiye.

BACKGROUND: Failure of passive transfer of immunity (FPTI) is perhaps the most important factor in the health of neonatal calves. Given the variation between countries in FPTI, national data need to be documented to add to the international database for future meta-analyses. OBJECTIVES: The aim of this study was to evaluate the prevalence of and risk factors for FPTI in dairy herds in Türkiye. METHODS: Blood serum samples from 1026 calves in 76 dairy herds in 16 provinces of Türkiye were analysed using a digital Brix refractometer to assess FPTI, defined as a Brix value < 8.5%. Mathematical modeling and simulation of hepatitis B transmission dynamics with passive immunity and control strategies.

Hepatitis B Virus (HBV) is a continued threat to mankind's health killing hundreds of thousands of people every year and thus calls for control and prevention measures to be put in place. This research constructs a deterministic mathematical model to capture the transmission dynamics of HBV and embracing the control measures including universal immunization of the infants at birth, screening, as well as treatment of the both the acute and chronic cases. We employ the NGM technique to estimate the control reproduction number Rc and also study the stability of the IFE. The study findings reveale

Evaluation of field-based diagnostic methods for assessing passive immune transfer in neonatal lambs.

Passive immune transfer (PIT) is crucial for the survival and health of neonatal lambs, and its assessment is vital in early-life management. This study evaluated the effectiveness of three field-based diagnostic methods-analog Brix refractometry, digital Brix refractometry, and immunocrit-for estimating PIT in lambs, using serum total protein concentration as the reference standard. Thirteen lambs born from eutocic deliveries were assessed. The mean serum protein concentration was 7.50 ± 0.70 g/dL, exceeding the 5.0 g/dL threshold for adequate PIT. All alternative methods showed strong positi

Metabolic profile of transition period in ewes and its influence on passive immunity transference in lambs.

The intensification of ewe production, focusing on increasing prolificity, results in a higher incidence of metabolic disorders, especially in the transition period, which can lead to production losses with consequences for the health of the animals. The objective of this study was to evaluate the metabolic profile in transition period Lacaune ewes according to the parity and litter size born and its influence on the transference of passive immunity. Nineteen Lacaune ewes, with initial mean body weight of 76.36 ± 10.37 kg (SD), kept in a semi-extensive system and receiving the same diet, were

Passive immunity to West Nile virus provides limited protection in a common passerine species.

Passerine birds have played an important role in the establishment, maintenance, and spread of West Nile virus (WNV) in North America, and some are susceptible to WNV-associated mortality. Characterization of passive transfer of anti-WNV antibodies in passerines is important to understanding transmission and demographic effects of WNV on wild birds. We showed passively acquired maternal antibodies to WNV in the house sparrow (Passer domesticus). Although all seropositive females (N = 18) produced antibody-positive egg yolks, only 20% of seropositive mothers (3/15) produced seropositive chicks.

Dynamics of passive immunity to West Nile virus in domestic chickens (Gallus gallus domesticus).

Birds are the principle amplifying hosts for West Nile virus (WNV), and understanding the acquisition and decay of passive immunity is important to avian surveillance and diagnostics. We characterized passive transfer of WNV-neutralizing antibody from chicken (Gallus gallus domesticus) hens to eggs and chicks and the protective efficacy and decay of maternally acquired antibody over time. We also characterized age-associated changes in magnitude of viremia and examined the possibility of vertical transmission of WNV. All egg yolks and chicks from seropositive hens were maternal antibody positi

Perinatal management of idiopathic thrombocytopenic purpura in pregnancy: risk factors for passive immune thrombocytopenia.

Thirty-nine pregnant women with idiopathic thrombocytopenic purpura (ITP) were studied in order to evaluate the influence of therapies for maternal ITP on fetal passive immune thrombocytopenia (PIT). Neonatal platelet counts were also compared with platelet counts, amount of PAIgG, and presence of circulating antiplatelet antibody in maternal blood. Eight of 41 neonates (19.5%) presented PIT without neonatal mortality. A higher incidence of PIT was observed in women with prior splenectomy than in women without splenectomy (66.7% vs 11.4%). Neither a therapeutic effect nor an increased risk of

Passive pre-exposure immunization by tixagevimab/cilgavimab in patients with hematological malignancy and COVID-19: matched-paired analysis in the EPICOVIDEHA registry.

Only few studies have analyzed the efficacy of tixagevimab/cilgavimab to prevent severe Coronavirus disease 2019 (COVID-19) and related complications in hematologic malignancies (HM) patients. Here, we report cases of breakthrough COVID-19 after prophylactic tixagevimab/cilgavimab from the EPICOVIDEHA registry). We identified 47 patients that had received prophylaxis with tixagevimab/cilgavimab in the EPICOVIDEHA registry. Lymphoproliferative disorders (44/47, 93.6%) were the main underlying HM. SARS-CoV-2 strains were genotyped in 7 (14.9%) cases only, and all belonged to the omicron variant.

Maternal immunization with a DNA vaccine candidate elicits specific passive protection against post-natal Zika virus infection in immunocompetent BALB/c mice.

Zika virus (ZIKV) infection is closely associated in the fetus with microcephaly and in the adults with Guillain-Barré syndrome and even male infertility. It is an urgent international priority to develop a safe and effective vaccine that offers protection to both women of childbearing age and their children. In this study, female immunocompetent BALB/c mice were immunized with a DNA-based vaccine candidate, pVAX1-ZME, expressing the prM/E protein of ZIKV, and the immunogenicity for maternal mice and the post-natal protection for suckling mice were evaluated. It was found that administration w

Active and passive immunization strategies based on the SDPM1 peptide demonstrate pre-clinical efficacy in the APPswePSEN1dE9 mouse model for Alzheimer's disease.

Recent clinical and pre-clinical studies suggest that both active and passive immunization strategies targeting Aβ amyloid may have clinical benefit in Alzheimer's disease. Here, we demonstrate that vaccination of APPswePSEN1dE9 mice with SDPM1, an engineered non-native Aβ amyloid-specific binding peptide, lowers brain Aβ amyloid plaque burden and brain Aβ1-40 and Aβ1-42 peptide levels, improves cognitive learning and memory in Morris water maze tests and increases the expression of synaptic brain proteins. This was the case in young mice immunized prior to development of significant brain amy
