L-ICON3 Suppresses Endometriosis in a Murine Model.

Endometriosis is a chronic, systemic gynecological disorder that affects 190 million women worldwide. Current treatments consist of hormonal suppression and surgical excision; however, both have considerable side effects and a high risk of recurrence. Here, we report a systemic therapy to treat endometriosis by L-ICON3, a novel chimeric immunoconjugate molecule that binds to tissue factor (TF), which is upregulated in endometriosis. Endometriosis was induced in mice by implanting endometrium from donors and that developed into experimental endometriosis after 6 weeks. Mice were then randomly a

Single-dose (4.5 mg/m2 ) gemtuzumab ozogamicin in combination with fludarabine, cytarabine and anthracycline as reinduction therapy in relapsed or refractory paediatric acute myeloid leukaemia.

Despite major therapeutic improvements, children with relapsed/refractory Acute Myeloid Leukaemia still have poor outcomes and overall survival does not exceed 40%. New treatments are required to improve their outcome; Gemtuzumab ozogamicin (GO), an anti-CD33 immunoconjugate antibody, is a potent cytotoxic agent whose efficacy has been demonstrated mainly in adults. The main objective of this retrospective multicentre study was to assess the outcome of children treated, between February 2008 and August 2019, with GO at a single 4.5 mg/m2 dose, in combination with Fludarabine, Cytarabine and an

Heroin vaccine: Using titer, affinity, and antinociception as metrics when examining sex and strain differences.

Anti-drug vaccines have potential as new interventions against substance use disorder (SUD). However, given the challenges seen with inter-individual variability in SUD vaccine trials to date, new interventions should ensure a robust immune response and safety profile among a diverse population. This requires accounting for sex and heritable genetic differences in response to both abused substances as well as the vaccination itself. To test response variability to our heroin-tetanus toxoid (Her-TT) immunoconjugate vaccine, we vaccinated male and female mice from several mouse strains including

Chimerization of LL2, a rapidly internalizing antibody specific for B cell lymphoma.

LL2 is a murine monoclonal antibody (MAb) that has been shown to be effective for the diagnosis and treatment of patients with non-Hodgkin's B cell lymphoma. Studies have also shown that radiolabeled murine LL2 (mLL2) or mLL2 and fragments thereof coupled to Pseudomonas exotoxin (PE) can effectively target human B cell lymphoma in mice. We have obtained the DNA sequences encoding the VK and VH domains of mLL2, an IgG2a MAb, which were combined with their respective human kappa and IgG1 constant region domains and expressed in SP2/0 cells. Like its murine counterpart, the chimeric LL2 (cLL2) an

Antibody-drug conjugates: a practical review on clinical management of toxicities.

INTRODUCTION: Antibody-drug conjugates (ADCs) are rapidly expanding across solid tumors, but their toxicity profiles remain heterogeneous and clinically challenging. This review addresses the need for practical, standardized approaches to identifying and managing ADC-related adverse events. AREAS COVERED: We summarize the mechanisms underlying ADC toxicity, focusing on EMA-approved agents and their most frequent or severe adverse events, including ILD/pneumonitis, ocular toxicity, neutropenia, diarrhea, skin reactions, and hyperglycemia. We combine evidence from pivotal trials, regulatory doc

Antibody-Drug Conjugates in the Management of Breast Cancer.

Breast cancer remains a predominant contributor to global cancer-related morbidity and mortality among women, with its incidence steadily rising and presenting complex therapeutic challenges. Conventional treatment modalities, including chemotherapy, endocrine therapy, and molecularly targeted interventions, have demonstrated substantial clinical efficacy. The emergence of therapeutic resistance, systemic cytotoxicity, and metastatic progression continues to impede optimal treatment outcomes, however. Antibody-drug conjugates (ADCs) represent an advanced class of targeted therapeutics engineer

Ocular Side Effects of Antibody-Drug Conjugates for Cancer Treatment.

BACKGROUND: Antibody-drug conjugates (ADCs) are approved for use in treating certain types of cancer and are now in clinical development for many others, as they are therapeutically effective. Some of these agents commonly cause ocular side effects, typically manifesting themselves as blurred vision or a foreign-body sensation. METHODS: For this narrative review of the literature, we carried out a database search and a cross-reference search at the German Federal Institute for Drugs and Medical Devices (BfArM) to identify publications on the diagnosis, prevention, and treatment of ocular side

[Oncology. Antibody-drug conjugates : a challenging innovation].

New therapeutic agents in oncology are emerging rapidly, both in terms of the number of approved drugs and the technological and biological innovation of new treatments. Antibody-drug conjugates (ADC) offer a promising cancer therapy by specifically targeting tumor cells. ADC are composed of a monoclonal antibody recognizing the tumor cell via specific antigens, coupled with a potent cytotoxic agent that resembles classical chemotherapy. This mechanism of action aims to deliver the cytotoxic agent directly to the tumor cell and spare healthy cells. However, important toxicities have been repor

Ocular adverse events associated with antibody-drug conjugates used in cancer: Focus on pathophysiology and management strategies.

Antibody-drug conjugates (ADCs) are designed to maximize cancer cell death with lower cytotoxicity toward noncancerous cells and are an increasingly valuable option for targeted cancer therapies. However, anticancer treatment with ADCs may be associated with ocular adverse events (AEs) such as dry eye, conjunctivitis, photophobia, blurred vision, and corneal abnormalities. While the pathophysiology of ADC-related ocular AEs has not been fully elucidated, most ocular AEs are attributed to off-target effects. Product labelling for approved ADCs includes drug-specific guidance for dose modificati

Middle-up characterization of monoclonal antibodies by online reduction liquid chromatography-mass spectrometry.

This study describes the fully automated middle-up characterization of monoclonal antibodies (mAbs) and next-generation variants by online reduction liquid chromatography-mass spectrometry (LC-MS). Proteins were trapped on-column and subjected to online desalting, denaturation and reduction prior to reversed phase elution of the created subunits in the MS. The evaluation of more than 20 different therapeutic proteins including full length mAbs (subclasses IgG1, IgG2 and IgG4), bispecific antibodies, antibody fragments, fusion proteins and antibody-drug conjugates (ADC) revealed that the online

Therapeutic targeting of CD19 in hematological malignancies: past, present, future and beyond.

Abstract During the past few decades, CD19 has been at the center of various scientific/translational endeavors to develop targeted therapeutics against B-cell malignancies. Due to the expression pattern of CD19 throughout the B-cell lineage, and on most B-cell malignancies, it became a preferred target for the development of experimental therapeutic agents during the first years of the monoclonal antibodies era. Successful preclinical experiments led to the first generation of clinical trials, based predominantly on toxin/anti-CD19 murine immunoconjugates. These, however, mostly failed due to

Targeting MDR1-P-glycoprotein (MDR1-Pgp) in immunochemotherapy of acute myeloid leukemia (AML).

BACKGROUND: Monoclonal antibodies represent the fastest growing sector of pharmaceutical biotechnology and a number of antibody-based biopharmaceuticals have been approved for cancer treatment. However, in many cases the antibodies used for the treatment of tumors offer only a modest survival benefit to cancer patients. AIMS: In the present review-article we intend to analyze: i) the curative regimen gemtuzumab ozogamicin (GO) -mediate characterized by the absence of cytotoxic drugs MDR1-Pgp substrates to overcome the mechanism of action of this multidrug transporter, ii) the safety and effic

Proposal for translational analysis and development of clinical radiolabeled immunoglobulin therapy.

BACKGROUND AND PURPOSE: Radiolabeled immunoglobulin therapy (RIT) can be a selective, effective, low-toxicity outpatient cancer therapy. A consensus on the best approach for the preclinical and clinical development of RIT reagents needs to be developed. We report the M.D. Anderson Cancer Center prior experience in translating RIT from laboratory to clinic for the treatment of Hodgkin's disease and propose a flow diagram for the development of RIT for other malignancies. Disseminated or localized growth of a human B-cell tumor (Daudi) in SCID mice.

A human Burkitt lymphoma (Daudi) has been grown in the mutant mouse called C.B-17 SCID. Twenty-eight days after s.c. injection of Daudi cells, a palpable tumor grew only at the site of injection in all injected mice. In contrast, after intravenous (i.v.) or intraperitoneal (i.p.) injection, macroscopic, disseminated tumors developed. Following i.v. inoculation, tumors grew in the lungs, kidneys, ovaries and adipose tissue, and microscopic tumor infiltrates were observed in the spleen, bone marrow, spinal column and femur, whereas after i.p. injection, the tumors were localized in the abdomen,

Anti-phosphatidylcholine antibodies in patients with brucellosis.

An indirect ELISA was adapted to measure individual classes of anti-phosphatidylcholine (PC) antibodies in patients with brucellosis; a comparison was made with patients with other infectious diseases and healthy human controls. Immunoconjugates of alpha, gamma or mu chain specificity were used. The results were compared with those of conventional tests for brucella antibodies, Rose Bengal, microagglutination (MAT) and Coombs tests, as well as with VDRL. The results indicated that although all sera from patients with brucellosis were VDRL negative, 97% of them (30 of 31) had higher levels of I
