Composite Coating for the Food Industry Based on Fluoroplast and ZnO-NPs: Physical and Chemical Properties, Antibacterial and Antibiofilm Activity, Cytotoxicity.

Bacterial contamination of meat products during its preparation at the enterprise is an important problem for the global food industry. Cutting boards are one of the main sources of infection. In order to solve this problem, the creation of mechanically stable coatings with antibacterial activity is one of the most promising strategies. For such a coating, we developed a composite material based on "liquid" Teflon and zinc oxide nanoparticles (ZnO-NPs). The nanoparticles obtained with laser ablation had a rod-like morphology, an average size of ~60 nm, and a ζ-potential of +30 mV. The polymer

A Streamlined Workflow for the Isolation of Bovine Satellite Cells From Muscle Tissue.

BACKGROUND: Satellite cells are the primary drivers of skeletal muscle growth and regeneration and are a major cell source in both muscle tissue engineering and cultivated meat. However, current bovine satellite cell isolation can be technically demanding or yield heterogeneous cell populations. METHODS: This study presents a workflow optimized for simplicity and cost-efficiency by integrating rapid muscle processing using electric mincing with magnetic-activated cell sorting targeting Integrin-α7 to reliably enrich satellite cells. The isolated cells were characterized by Pax7 immunostaining

Fish hypersensitivity. I. In vitro and oral challenge results in fish-allergic patients.

The purpose of this study was to determine whether patients allergic to one fish species can safely eat other fish species. Eleven atopic, food-allergic children and young adults with histories consistent with IgE-mediated fish hypersensitivity were skin prick tested to 10 fish species. Skin prick tests (SPTs) were positive to all 10 fish in eight of the 11 patients, and the remaining three patients had at least two positive fish SPTs. Positive oral challenges occurred to only one fish in seven of the patients, to two fish species in one patient, and to three fish species in two patients. One

Bison and bovine rectoanal junctions exhibit similar cellular architecture and Escherichia coli O157 adherence patterns.

BACKGROUND: Escherichia coli O157 (E. coli O157) has been isolated from bison retail meat, a fact that is important given that bison meat has been implicated in an E. coli O157-multistate outbreak. In addition, E. coli O157 has also been isolated from bison feces at slaughter and on farms. Cattle are well documented as E. coli O157 reservoirs, and the primary site of E. coli O157 persistence in such reservoirs is the rectoanal junction (RAJ), located at the distal end of the bovine gastrointestinal tract. Since bison and cattle share many genetic similarities manifested as common lineage, susc

Disclose ATP-synthase as a protein target of the antimicrobial peptide RiLK1 in Escherichia coli: An alternative receptor-mediated bactericidal mechanism.

In this study, we investigated the mechanism of action of the 10-aminoacid RiLK1 peptide against Escherichia coli (strain ATCC 25922), both in vitro and in contaminated meat matrices. Therefore, a mass spectrometry-based functional proteomics platform was employed to identify specific molecular targets of RiLK1 in a membrane protein-enriched E. coli lysate and to obtain information on their interaction mechanism. This target deconvolution approach combines MS-limited proteolysis techniques, like Drug Affinity Responsive Target Stability (DARTS) and targeted-limited Proteolysis coupled with Mas

Inhibitory effects of the herbal quorum sensing quencher isoorientin on spoilage characteristics of Serratia liquefaciens S01: In vitro and in a smoked chicken leg model.

The study aimed to screen herbal-derived quorum sensing (QS) quenchers and investigate their inhibitory effects on the Serratia liquefaciens S01, a specific spoilage bacterium in smoked chicken legs. Targeting the QS proteins LuxI and LuxR of S. liquefaciens S01, ten compounds were screened as potential QS quenchers by molecular docking. Among these compounds, isoorientin exhibited a relatively low minimum inhibitory concentration (MIC, 250 μg/mL), potent quorum quenching activity, and the strongest binding affinity. In vitro experiments, sub-MIC concentrations of isoorientin significantly inh

Digestive Fate of a Seaweed-Yak Meat Protein-Polysaccharide Matrix under Simulated Elderly Gastrointestinal Dysfunction: Nutrient Release, Antioxidant Response, and Peptidomic Remodeling.

Age-related gastrointestinal dysfunction may alter the digestive fate of composite foods. Using a static in vitro model, we compared a seaweed-yak meat matrix under four simulated older-adult gastrointestinal conditions: normal gastric and intestinal function, isolated intestinal dysfunction, isolated gastric dysfunction, and combined dysfunction. Intestinal digestion decreased particle size from 2325.03 nm to 731.74-1088.07 nm and significantly increased apparent protein digestibility from 9.72-29.05% to 73.27-83.73% (P < 0.05). Combined dysfunction significantly reduced terminal digestibilit

Synergistic in-vitro activity of Fosinopril alone and in combination with imidocarb dipropionate against Babesia bovis.

Bovine babesiosis, caused by Babesia bovis is a major cattle disease worldwide, particularly in tropical and subtropical regions. A commonly used drug for the control and treatment of babesiosis is imidocarb dipropionate (ID), which has proved effective against B. bovis. Nevertheless, its toxicity to ruminants and other mammals, along with the persistence of drug residues in meat and milk for several months after treatment, has led to restriction on its use. Furthermore, several reports indicate that complete parasite elimination is not obtained in some cases. Interestingly, the angiotensin-co

Conservation of Markers and Stemness in Adipose Stem and Progenitor Cells between Cattle and Other Species.

Adipose stem and progenitor cells (ASPCs) have been isolated from humans and animals for use in regenerative medicine and therapy. However, knowledge of ASPCs in other species is limited. Particularly, ASPCs in livestock are expected to enhance the fat content and meat composition. In this study, we isolated bovine ASPCs using cell surface markers. Specifically, we focused on ASPC markers in humans and experimental animals, namely CD26, CD146, and CD54. Stromal vascular fraction cells from bovine fat were separated using flow cytometry before primary culture. We evaluated the self-renewal and

Development and Characterization of a Collagen-Based Three-Dimensional In Vitro Model to Mimic Biofilm Formation in a Wound Bed.

Current studies using in vitro biofilm culturing systems have been instrumental at elucidating wound biofilm formation but fail to account for the diverse environment that bacteria are exposed to within the host. In the current study, we recapitulated this wound biofilm microenvironment by creating a hydrogel composed of collagen, thrombin, fibrinogen, meat broth, and FBS and subsequently infected the scaffolds with UAMS-1. We characterized the material properties of the hydrogel (noninfected) and found no significant differences in the storage modulus when fibrin was added to the collagen hyd

Safety Evaluation of Soy Leghemoglobin Protein Preparation Derived From Pichia pastoris, Intended for Use as a Flavor Catalyst in Plant-Based Meat.

The leghemoglobin protein (LegH) from soy ( Glycine max) expressed in Pichia pastoris (LegH preparation, LegH Prep) imparts a meat-like flavor profile onto plant-based food products. The safety of LegH Prep was evaluated through a series of in vitro and in vivo tests. The genotoxic potential of LegH Prep was assessed using the bacterial reverse mutation assay (Ames test) and the in vitro chromosome aberration test. LegH Prep was nonmutagenic and nonclastogenic in each test, respectively. Systemic toxicity was assessed in a 28-day dietary study in male and female Sprague Dawley rats. There were

Integrated transcriptomic and WGCNA analyses identify RBP7 as a regulator of skeletal muscle development in chickens.

Skeletal muscle development determines meat yield and quality in chickens. We performed transcriptomic profiling of breast muscles from fast-growing Arbor Acres (AA) and slow-growing Lushi blue-shelled-egg (LS) chicken - an indigenous dual-purpose type breed - at 1, 3, and 5 weeks. A total of 1727 differentially expressed genes (DEGs) were identified. Functional enrichment indicated that AA-specific DEGs were enriched in MAPK signaling, whereas LS-specific DEGs were associated with Hedgehog signaling and branched-chain amino acid degradation. Weighted gene co-expression network analysis identi

Insulin-secreting L-cells for the treatment of insulin-dependent diabetes.

Cell-based treatments for insulin-dependent diabetes (IDD) may provide more physiologic regulation of blood glucose levels than daily insulin injections, thereby reducing the occurrence of secondary complications associated with diabetes. An autologous cell source is especially attractive for regulatory and ethical reasons in addition to eliminating the need for immunosuppression. This study uses non-beta-cells, genetically modified for physiologic insulin secretion. Enteroendocrine L-cells, exhibit regulated secretion in response to physiologic stimuli and their endogenous products are fully

Repurposing Cryopreserved Slaughterhouse Waste for Vascular Replacement Therapy.

Current vascular replacement therapy (VRT) standards primarily rely on autografting and allografting. However, autografting is associated with substantial donor site comorbidities, while the limited availability of graft tissues constrains allografting. Although synthetic alternatives are widely accessible, they are prone to long-term failure. This study explores an alternative approach to help address the critical supply-demand mismatch for VRT by repurposing cryopreserved slaughterhouse-derived waste aortas. Using an in vitro perfusion-based model, we demonstrated that these tissues maintain

Essential Oils against Sarcoptes scabiei.

Herbal remedia are widely employed in folk medicine, and have been more and more often studied and considered in the treatment of several infections. Sarcoptic mange (scabies, when referring to human patients) is a highly contagious skin disease caused by Sarcoptes scabiei (sarcoptiformes, Sarcoptinae), an astigmatid mite which burrows into the epidermis, actively penetrating the stratum corneum. This parasitosis negatively affects livestock productions and represents a constraint on animal and human health. The treatment relies on permethrine and ivermectine but, since these molecules do not
