Circulation of three genotypes and identification of unique mutations in neutralizing epitopes of infectious bronchitis virus in chickens in Bangladesh.

Infectious bronchitis (IB) is a highly contagious respiratory disease caused by a gammacoronavirus that has been circulating for many years in chickens in Bangladesh, resulting in significant economic losses. The aim of this study was to detect and characterize infectious bronchitis virus (IBV) from clinical outbreaks and surveillance samples. Real-time RT-PCR was used to detect IBV in pooled lung and tracheal tissue samples (n = 78), oropharyngeal swabs (n = 19), and pooled fecal samples (n = 13) from live-bird markets. Both respiratory and nephropathogenic forms of IB were suspected at necro

Nephropathogenic Infectious Bronchitis Virus Infection Altered the Metabolome Profile and Immune Function of the Bursa of Fabricius in Chicken.

Infectious bronchitis is a highly contagious, acute viral respiratory disease of chickens, regardless of the strain, and its infection may lead to considerable economic losses to the poultry industry. New nephropathogenic infectious bronchitis virus (NIBV) strains have increasingly emerged in recent years; hence, evaluating their infection-influenced immune function changes and the alteration of metabolite profiling is important. Initially, chickens were randomly distributed into two groups: the control group (Con) and the disease group (Dis). Here, the partial cytokines were examined, and the

Molecular Monitoring of D1466 Genotype of Avian Infectious Bronchitis Virus in Iran: A Retrospective Study in 2013-2017.

Infectious bronchitis is one of the most common diseases in the poultry industry in many countries, especially in the regions with a dense poultry farming industry. Gammacoronavirus is the etiologic agent of this disease, with the chickens and poultries as the natural reservoirs of the virus. Various strains of infectious bronchitis virus have been reported in poultry around the world. In terms of pathogenicity, this virus can induce a spectrum of diseases ranging from the moderate respiratory tract to kidney and reproductive diseases. The serotypes of this virus do not cause cross-immunity ag

Insights on genetic characterization and pathogenesis of a GI-19 (QX-like) infectious bronchitis virus isolated in China.

Infectious bronchitis virus (IBV) causes respiratory diseases in chickens, incurring great losses to the poultry industry worldwide. In this study, we isolated an IBV strain, designated as AH-2020, from the chickens vaccinated with H120 and 4/91 in Anhui, China. The sequence homology analysis based on the S1 gene revealed that AH-2020 shares low similarities with the 3 vaccine strains, namely, H120, LDT3-A, and 4/91 (78.19, 80.84, and 81.6%, respectively). Phylogenetic analysis based on the S1 gene revealed that AH-2020 clustered with the GI-19 type. Furthermore, protein modeling revealed that

Protection conferred by a vaccine derived from an inactivated Egyptian variant of infectious bronchitis virus: a challenge experiment.

The current study investigated the protective efficacy of a formalin-inactivated infectious bronchitis virus (IBV) vaccine derived from the field strain KP729422, which exhibits low S1 spike protein sequence homology (77.1-79.8%) with the currently used vaccine strains in Egypt. Two-week-old, specific-pathogen-free chickens were subcutaneously inoculated with a single dose of the vaccine containing 106.7 50% embryo infective dose (EID50) of the inactivated virus. At 6 weeks of age, the chickens were challenged with 104 EID50 of the same virus strain via the oculonasal route. In comparison with

Inflammatory and cell-mediated immune responses in the respiratory tract of chickens to infection with avian infectious bronchitis virus.

Tracheal mucosa is the primary site of replication of avian infectious bronchitis virus (IBV), which leads to both morphologic and immune modulatory changes in this organ. To increase the understanding of the mechanisms involved in these processes, we focused on the evaluation of local inflammatory and cell-mediated immune responses after challenge with the M41 strain of IBV, associating these responses with pathologic changes in the tracheal mucosa. At 24 h post-infection, inflammatory cytokines related genes were significantly upregulated, including peaks of TNFSF15 and TGFβ mRNA production,

Susceptibility of three genetic lines of chicks to infection with a nephropathogenic T strain of avian infectious bronchitis virus.

Mortality rates were compared in three genetic lines of specific pathogen-free chicks inoculated with one of two doses of a nephropathogenic strain of avian infectious bronchitis (IB) virus. The mortality rates were influenced primarily by the chick strain, but also by age and dose of virus. Chicks of the inbred S line were highly susceptible. After inoculation with a low dose of virus at 2 and 4 weeks of age, mortality was 90 and 45%, respectively. Chicks of the HWL non-inbred line were also susceptible, with mortality rates after inoculation at 2 and 4 weeks of age of 70 and 25%, respectivel

Immunopathogenesis of infection in SPF chicks and commercial broiler chickens of a variant infectious bronchitis virus of economic importance.

The immunopathogenesis of an economically important variant of infectious bronchitis virus (IBV) has been studied in day-old specific pathogen-free and 6-week-old broiler chickens. Experimental infection caused very mild respiratory disease and no mortalities. Diarrhoea was seen in the broilers. Virus distribution and titre in respiratory, urinary and enteric tissues were assessed. In SPF chicks, maximum virus isolations were from oesophagus, followed by trachea, kidney, lung and rectum. In contrast, in the broilers, maximum isolations were from ileum, followed by caecal tonsils, rectum, bursa

Hatchery Spray Cabinet Administration Does Not Damage Avian Coronavirus Infectious Bronchitis Virus Vaccine Based on Analysis by Electron Microscopy and Virus Titration.

studies in our laboratory showed that the Arkansas-Delmarva Poultry Industry (Ark-DPI) vaccine given to 1-day-old chickens by hatchery spray cabinet replicated poorly and failed to adequately protect broilers against homologous virus challenge, whereas the same vaccine given by eye-drop did replicate and the birds were protected following homologous virus challenge. To determine if mechanical damage following spray application plays a role in failure of the Ark-DPI vaccine, we examined the morphology of three Ark-DPI vaccines from different manufacturers using an electron microscope and includ

Qingfei Huatan Formula alleviates growth suppression and inflammation-driven metabolic dysregulation in avian infectious bronchitis virus-infected broilers.

Avian infectious bronchitis virus (IBV) infection causes growth suppression in broilers, which has been hypothesized to involve a diversion of metabolic resources from growth toward immune defense. This study evaluated the efficacy and molecular mechanisms of Qingfei Huatan Formula (QHF) in IBV-challenged broilers. A total of 180 Yellow-feathered broilers were randomly assigned to a blank control, IBV-challenged (M41 strain), Ribavirin, and QHF dose-gradient groups. High-dose QHF intervention significantly increased body weight gain (BWG) at 14 days post-infection compared with the IBV model g

Genetic and pathogenic characteristics of a novel recombinant GI-19 infectious bronchitis virus strain isolated from northeastern China.

Since the 1980s, despite vaccination, the infectious bronchitis virus (IBV) infection rate in commercial broilers and layers in China has continued to rise significantly, causing substantial economic losses to the poultry industry. In this study, an IBV strain was isolated from a layer farm in northeast China and named CK/CH/LN/2302. The whole genome sequence analysis revealed that CK/CH/LN/2302 shared a high level of homology (96.41 %) with the GI-19 strain SC/SDL/19. The phylogenetic tree based on the S1 gene indicates that CK/CH/LN/2302 belongs to the GI-19 lineage. Notably, recombination a

Preparation of polyclonal antibodies to chicken P62 protein and its application in nephropathogenic infectious bronchitis virus-infected chickens.

p62, also known as SQSTM1, has been shown to be closely related to the coronavirus. However, it remains unclear on the relationship between p62 and NIBV infection. Moreover, there are no available antibodies against the chicken p62 protein. Thus, this study aimed to prepare p62 polyclonal antibody and investigate the correlation between the p62 protein and NIBV infection. Here, PET-32a-p62 prokaryotic fusion expression vector was constructed for prokaryotic protein expression, and then p62 polyclonal antibody was prepared by immunizing rabbits. Lastly, these antibodies were then utilized in We

Attenuated live infectious bronchitis virus QX vaccine disseminates slowly to target organs distant from the site of inoculation.

Infectious bronchitis (IB) is a highly contagious respiratory disease of poultry, caused by the avian coronavirus infectious bronchitis virus (IBV). Currently, one of the most relevant genotypes circulating worldwide is IBV-QX (GI-19), for which vaccines have been developed by passaging virulent QX strains in embryonated chicken eggs. Here we explored the attenuated phenotype of a commercially available QX live vaccine, IB Primo QX, in specific pathogens free broilers. At hatch, birds were inoculated with QX vaccine or its virulent progenitor IBV-D388, and postmortem swabs and tissues were col

Inactivated infectious bronchitis virus vaccine encapsulated in chitosan nanoparticles induces mucosal immune responses and effective protection against challenge.

Avian infectious bronchitis virus (IBV) is one of the most important viral diseases of poultry. The mucosa of upper respiratory tract, specially the trachea, is the primary replication site for this virus. However, conventional inactivate IBV vaccines usually elicit reduced mucosal immune responses and local protection. Thus, an inactivated IBV vaccine containing BR-I genotype strain encapsulated in chitosan nanoparticles (IBV-CS) was produced by ionic gelation method to be administered by oculo-nasal route to chickens. IBV-CS vaccine administered alone resulted in markedly mucosal immune resp
