Coronaviruses pose a serious threat to public health, driving the need for antiviral therapeutics and vaccines. Therefore, it is paramount to understand how this family of viruses evades cellular antiviral responses and establishes productive infection. The conserved coronavirus nonstructural protein 1 (nsp1) has been shown to inhibit host protein synthesis and, in some coronaviruses, promote host messenger RNA (mRNA) degradation while viral mRNAs are protected. We showed previously that severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) induces activation of host integrated stress r

Lower back pain (LBP), which is a primary cause of disability, is largely attributed to intervertebral disc degeneration (IDD). Macrophages (MΦs) in degenerated intervertebral discs (IVDs) form a chronic inflammatory microenvironment, but how MΦs are recruited to degenerative segments and transform into a proinflammatory phenotype remains unclear. We evaluated chemokine expression in degenerated nucleus pulposus cells (NPCs) to clarify the role of NPCs in the establishment of an inflammatory microenvironment in IDD and explored the mechanisms. We found that the production of C-C motif chemokin

Regardless of adherence to combined antiretroviral therapy, white matter and myelin pathologies persist in patients with HIV-associated neurocognitive disorders, a spectrum of cognitive, motor, and behavioral impairments. We hypothesized that antiretroviral therapy alters the maturation of oligodendrocytes which synthesize myelin. We tested whether specific frontline integrase strand transfer inhibitors would alter oligodendrocyte differentiation and myelination. To model the effect of antiretrovirals on oligodendrocytes, we stimulated primary rat oligodendrocyte precursor cells to differentia

Abiotic challenges such as nutrient shortages and heavy metal toxicity dramatically limit soybean yields worldwide. While many transcriptomic investigations have examined responses to single stresses, few have identified regulatory nodes that function across multiple adverse conditions. In this study, we conducted a multi-dataset transcriptomic meta-analysis integrating six RNA-seq experiments comprising 66 samples across nutrient deficiency and heavy-metal stress conditions. Following batch-effect correction using ComBat and variance-based filtering, weighted gene co-expression network analys

Background and Objectives: The integrated stress response (ISR) is a convergent node in neurodegeneration. We systematically mapped open-access mammalian in vivo evidence for synthetic ISR modulators, comparing efficacy signals, biomarker engagement, and safety across mechanisms and disease classes. Methods: Following PRISMA 2020, we searched PubMed (MEDLINE), Embase, and Scopus from inception to 22 September 2025. Inclusion required mammalian neurodegeneration models; synthetic ISR modulators (eIF2B activators, PERK inhibitors or activators, GADD34-PP1 ISR prolongers); prespecified outcomes;

Doxorubicin is an effective chemotherapeutic agent; however, its use is limited by cardiotoxicity. Mitochondrial dysfunction is a central driver of doxorubicin-mediated cardiotoxicity. The role of the integrated stress response (ISR), a mitochondria-to-nucleus signaling pathway and crucial cellular defense mechanism in doxorubicin-induced cardiotoxicity, remains unclear. We investigated the pharmacological ISR activator salubrinal, a selective inhibitor of eukaryotic initiation factor 2α dephosphorylation with potential cardioprotective properties, to elucidate the molecular mechanisms underly

The integrated stress response (ISR) protects cells from a variety of insults. Once elicited (e.g., by virus infections), it eventually leads to the block of mRNA translation. Central to the ISR are the interactions between translation initiation factors eIF2 and eIF2B. Under normal conditions, eIF2 drives the initiation of protein synthesis through hydrolysis of GTP, which becomes replenished by the guanine nucleotide exchange factor eIF2B. The antiviral branch of the ISR is activated by the RNA-activated kinase PKR which phosphorylates eIF2, thereby converting it into an eIF2B inhibitor. Her

An endophytic bacterium YG-3 with high cadmium (Cd) resistance was isolated from poplar grown in a composite mine tailing. It was identified as Burkholderia cenocepacia based on genomic, physiological and biochemical analyses. The Cd removal rate by YG-3 could reach about 60.0% in Cd aqueous solution with high concentrations of both 100 and 500 mg L-1. Meanwhile, various absorption and adsorption strategies were found in the two different Cd concentrations. The global resistance mechanisms of YG-3 were investigated in several levels, i.e., physiological observation, such as scanning electron m

Temperature represents one of the most critical environmental factors constraining the aquaculture of cold-water fish species. As the primary respiratory organ directly interfacing with the external environment, the gill is among the tissues most sensitive to thermal stress. To elucidate the physiological and molecular mechanisms underlying the thermal response of the plateau cold-water fish Gymnocypris eckloni, this study aimed to investigate the effects of acute heat stress of varying intensities on gill tissue damage and its intrinsic response mechanisms. After acclimation at 16 °C (control

Acute myeloid leukemia (AML) persistence is sustained by leukemic stem cells (LSCs) that survive metabolic deprivation, oxidative stress, hypoxia, proteotoxic burden, and therapeutic pressure. The integrated stress response (ISR) has emerged as a central adaptive network in this process. Through phosphorylation of a subunit of eukaryotic initiation factor 2 (eIF2α) and selective translation of activating transcription factor 4 (ATF4), the ISR coordinates stress-responsive transcriptional programs that may either preserve cellular fitness or promote apoptotic commitment, depending on the intens

Trachinotus ovatus is an economically important mariculture fish, and hypoxia has become a critical threat to this hypoxia-sensitive species. However, the molecular adaptation mechanism of T. ovatus liver to hypoxia remains unclear. In this study, we investigated the effects of acute hypoxic stress (1.5 ± 0.1 mg·L-1 for 6 h) and re-oxygenation (5.8 ± 0.3 mg·L-1 for 12 h) in T. ovatus liver at both the transcriptomic and metabolic levels to elucidate hypoxia adaptation mechanism. Integrated transcriptomics and metabolomics analyses identified 36 genes and seven metabolites as key molecules that

BACKGROUND: Carotid arterial atherosclerotic stenosis is a well-recognized pathological basis of ischemic stroke; however, its underlying molecular mechanisms remain unknown. Vascular smooth muscle cells (VSMCs) play fundamental roles in the initiation and progression of atherosclerosis. Organelle dynamics have been reported to affect atherosclerosis development. However, the association between organelle dynamics and various cellular stresses in atherosclerotic progression remain ambiguous.
METHODS: In this study, we conducted transcriptomics and bioinformatics analyses of stable and vulnera

Translation initiation is the rate-limiting step of protein synthesis that is downregulated during the Integrated Stress Response (ISR). Previously, we demonstrated that most human mRNAs that are resistant to this inhibition possess translated upstream open reading frames (uORFs), and that in some cases a single uORF is sufficient for the resistance. Here we developed a computational model of Initiation Complexes Interference with Elongating Ribosomes (ICIER) to gain insight into the mechanism. We explored the relationship between the flux of scanning ribosomes upstream and downstream of a sin

Type I interferons (IFNs) induce a detrimental response during Listeria monocytogenes (L. monocytogenes) infection. We were interested in identifying mechanisms linking IFN signaling to negative host responses against L. monocytogenes infection. Herein, we found that infection of myeloid cells with L. monocytogenes led to a coordinated induction of type I IFNs and activation of the integrated stress response (ISR). Infected cells did not induce Xbp1 splicing or BiP upregulation, indicating that the unfolded protein response was not triggered. CHOP (Ddit3) gene expression was upregulated during

This study characterizes genomic and phenotypic diversity among Vibrio parahaemolyticus isolates from shrimp aquaculture by integrating phenotypic profiling with pangenomic analysis. Growth was quantified under temperature, salinity, pH, nutrient, and ethanol stresses, together with motility, hemolysis, and antibiotic susceptibility. Core-genome SNP phylogeny identified genetic differentiation among representative isolates, with relationships not consistently associated with sampling location, sample type, or collection period. Comparative genomics showed variation in accessory-gene compositio

Hypoxia critically impacts grass carp (Ctenopharyngodon idella) aquaculture, causing oxidative damage, mortality and economic losses. This study revealed the physiological and genetic response mechanisms of grass carp under hypoxic stress by integrating histology, biochemistry and miRNA-mRNA network analysis. The experimental individuals were confined in sealed containers with pre-aerated water, allowing dissolved oxygen (DO) to decline naturally, and three groups were established: Normoxia (DO: 7.99 ± 0.43 mg∙L-1; control), Hypoxia (DO: 0.88 ± 0.16 mg∙L-1; upward swimming and gulping), and As

Adenosine has emerged as a central metabolic signal linking cellular stress to systemic physiological adaptation. Under conditions such as hypoxia, ischemia, inflammation, and tissue injury, extracellular adenosine triphosphate (eATP) released from stressed cells is sequentially metabolized by the ectonucleotidases CD39 and CD73, generating adenosine that accumulates in the extracellular microenvironment. This stress-responsive nucleoside activates four G-protein-coupled receptors (A1, A2A, A2B, and A3), triggering intracellular signaling networks including the cyclic adenosine monophosphate-p

Maintaining the cytosolic K+/Na+ homeostasis is crucial for plants' salt tolerance. Therefore, elucidating the regulatory mechanism governing K+/Na+ transport is pivotal to engineering salt-tolerant crops. Although many regulators have been shown specifically to modulate either Na+ or K+ transporters, little is known about integrative regulators that orchestrate the transport of both cations. In this study, we reveal that a rice basic helix-loop-helix (bHLH) transcription factor, BES1-INTERACTING MYC-LIKE1 (OsBIM1), negatively regulates K+/Na+ homeostasis under salt stress. Loss of OsBIM1 func
