[Deciphering the links between hepatic viruses and interferons: molecular interactions and therapeutic implications].

Interferons (IFNs) are cytokines that play a key role in the innate immune response, possessing antiviral, antiproliferative, and immunomodulatory properties. Currently, IFNs are used clinically as treatments for lymphomas, multiple sclerosis, and chronic viral infections such as hepatitis B, C, D, and E virus infections. HEV infection can lead to fulminant hepatitis, while chronic infections with HBV, HCV, and HDV are the leading global causes of hepatocellular carcinoma. Despite their limited efficacy and numerous side effects, IFNs remain essential in the treatment of these infections. In t

Transcription factor MafB-mediated inhibition of type I interferons in plasmacytoid dendritic cells.

Type I IFNs (IFN-α and IFN-β), immunomodulatory cytokines secreted from activated plasmacytoid dendritic cells (pDCs), contribute to the innate defense against pathogenic infections and the pathogenesis of the autoimmune disease psoriasis vulgaris. A previous study has shown that an E26 transformation-specific (Ets) family transcription factor Spi-B can transactivate the type I IFN promoter in synergy with IFN regulatory factor (IRF)-7 and is required for type I IFN production in pDCs. However, the mechanism of negative regulation of type I IFNs by pDCs remains unknown. In this study, we repor

The role of interferons type I, II and III in myositis: A review.

The classification of idiopathic inflammatory myopathies (IIM) is based on clinical, serological and histological criteria. The identification of myositis-specific antibodies has helped to define more homogeneous groups of myositis into four dominant subsets: dermatomyositis (DM), antisynthetase syndrome (ASyS), sporadic inclusion body myositis (sIBM) and immune-mediated necrotising myopathy (IMNM). sIBM and IMNM patients present predominantly with muscle involvement, whereas DM and ASyS patients present additionally with other extramuscular features, such as skin, lung and joints manifestatio

Listeria monocytogenes infection rewires host metabolism with regulatory input from type I interferons.

Listeria monocytogenes (L. monocytogenes) is a food-borne bacterial pathogen. Innate immunity to L. monocytogenes is profoundly affected by type I interferons (IFN-I). Here we investigated host metabolism in L. monocytogenes-infected mice and its potential control by IFN-I. Accordingly, we used animals lacking either the IFN-I receptor (IFNAR) or IRF9, a subunit of ISGF3, the master regulator of IFN-I-induced genes. Transcriptomes and metabolite profiles showed that L. monocytogenes infection induces metabolic rewiring of the liver. This affects various metabolic pathways including fatty acid

Type 1 Diabetes: Interferons and the Aftermath of Pancreatic Beta-Cell Enteroviral Infection.

Enteroviruses (EVs) have long been implicated in the pathogenesis of type 1 diabetes (T1D), and accumulating evidence has associated virus-induced autoimmunity with the loss of pancreatic beta cells in T1D. Inflammatory cytokines including interferons (IFN) form a primary line of defence against viral infections, and their chronic elevation is a hallmark feature of many autoimmune diseases. IFNs play a key role in activating and regulating innate and adaptive immune responses, and to do so they modulate the expression of networks of genes and transcription factors known generically as IFN stim

Interferons: A molecular switch between damage and repair in ageing and Alzheimer's disease.

Alzheimer's disease was first described over 100 years ago, yet it remains incurable and affects 44 million people worldwide. Traditionally, research has largely focused on the amyloid cascade hypothesis, but interest in the importance of inflammation in the progression of the disease has recently been increasing. Interferons, a large family of cytokines that trigger the immune system, are believed to play a crucial role in the pathology of Alzheimer's disease. This review focuses on how interferons affect the brain during ageing and whether they could be candidate therapeutic targets for the

Effects of interferons and double-stranded RNA on human prostate cancer cell apoptosis.

The structured processes of clinical medicine and epidemiology are closely aligned, with disease reporting serving as the critical interface between individual patient care and population-level public health action. In Switzerland, mandatory reporting of communicable diseases forms the backbone of national infectious disease surveillance and relies on coordinated contributions from clinicians and laboratories. The Swiss reporting system is regulated by an ordinance that defines notifiable diseases, reporting criteria and strict timelines. It encompasses severe diseases, outbreak-prone infectio

Viral entry route determines how human plasmacytoid dendritic cells produce type I interferons.

Although plasmacytoid dendritic cells (pDCs) represent a rare immune cell type, they are the most important source of type I interferons (IFNs) upon viral infection. Phagocytosed RNA viruses and RNA virus-infected cells are detected by pDCs with the endosomal pattern recognition receptor (PRR) toll-like receptor 7 (TLR7). We showed that replication of the yellow fever live vaccine YF-17D in human pDCs and pDC-like cell lines stimulated type I IFN production through RIG-I (retinoic acid-inducible gene I), a member of the RIG-I-like receptor (RLR) family of cytosolic PRRs. Thus, human pDCs sense

Interferons and viruses: an interplay between induction, signalling, antiviral responses and virus countermeasures.

The interferon (IFN) system is an extremely powerful antiviral response that is capable of controlling most, if not all, virus infections in the absence of adaptive immunity. However, viruses can still replicate and cause disease in vivo, because they have some strategy for at least partially circumventing the IFN response. We reviewed this topic in 2000 [Goodbourn, S., Didcock, L. & Randall, R. E. (2000). J Gen Virol 81, 2341-2364] but, since then, a great deal has been discovered about the molecular mechanisms of the IFN response and how different viruses circumvent it. This information is o

Novel functions of type I interferons revealed by infection studies with Listeria monocytogenes.

Infection of cells and mice with Listeria monocytogenes stimulates production of type I interferons (IFN). These in turn sensitise the Listeria host to lethal sequelae of infection with these bacteria. Here, we summarise recent findings on the production and biological effects of type I IFN in the course of L. monocytogenes infection.

Evolution of previous sarcoidosis under type 1 interferons given for severe associated disease.

Sarcoidosis is a granulomatous disorder with a well-known T helper (Th) type 1 cell commitment and a key pathogenic role of interferon (IFN)-gamma. However, little is known about the influence of type 1 IFNs, such as IFN-alpha and IFN-beta, on the course of previous sarcoidosis. The aim of this study was to determine whether type 1 IFNs can safely be used in patients with sarcoidosis for severe associated disease. The present study examined a series of four patients with sarcoidosis, treated by IFN-alpha or IFN-beta for viral hepatitis (three cases) or multiple sclerosis (one case). IFN was in

Interferons and IRF-1 induce expression of the survival motor neuron (SMN) genes.

BACKGROUND: Spinal muscular atrophy (SMA) is a common recessive disorder, characterized by degeneration of motor neurons of the spinal cord. Deletions, conversions, or mutations of the survival motor neuron gene (SMN) are responsible for SMA. A highly homologous centromeric copy of the SMN gene (SMNc) remains intact in SMA patients. However, there is an inverse correlation between the amount of the SMNc gene product and the clinical severity of the disease. An understanding of SMN and SMNc gene regulation is, therefore, an important step towards therapy for SMA. RESULTS: We identified a candi

[The immunomodulating action of interferons in their combined use with a typhus vaccine].

The data of the comparative study of the immunomodulating action of donor interferon and reaferon in experiments with typhus vaccine, used as an experimental model, on monkeys of different species and guinea pigs are presented. One of the main properties of interferons, besides their antiviral activity, is their capacity for producing an immunomodulating effect.

Activation of neonatal and adult human macrophages by alpha, beta, and gamma interferons.

Increasing evidence suggests that gamma interferon (IFN-gamma) is the major or sole factor in human lymphokines which activates blood monocyte-derived macrophages (M phi) to inhibit or kill Toxoplasma gondii and certain other intracellular pathogens. In the current studies, we found that IFN-gamma effectively activated tissue M phi from adults (peritoneal M phi) and from newborns (placental M phi) as well as blood-derived M phi from adults and from newborns to kill or to inhibit the replication of T. gondii. Results with purified and recombinant IFN-gamma and with adult and newborn M phi were

Multiple interferons in the circulation of patients with systemic lupus erythematosus and vasculitis.

Recently, we found interferon in the sera of patients with systemic lupus erythematosus, rheumatoid arthritis, scleroderma, and Sjögren's syndrome. In this study, we surveyed a variety of other immunologically mediated diseases. We did not find interferon in the sera of patients with Wegener's granulomatosis, sarcoidosis, infectious mononucleosis, minimal change nephritis, kidney transplants, myasthenia gravis, or uveitis, but we did find this protein in the sera of patients with active systemic and cutaneous vasculitis. Attempts to characterize the interferon in the sera of patients with syst

Interferons and systemic sclerosis: correlation between interferon gamma and interferon-lambda 1 (IL-29).

BACKGROUND: Interferon (IFN)-λ1 is a newly described cytokine, member of type III interferons family, which is known for its antiviral, anti-proliferative and antitumor activity. Recent studies indicated that this cytokine has also immune-regulatory function, but its role in the pathogenesis of autoimmune diseases is not established yet. We evaluated serum levels of IFN-λ1 in systemic sclerosis (SSc) patients and healthy controls and its association with IFN-γ and clinical manifestations. METHODS: IFN-λ1 and IFN-γ serum levels were measured by ELISA from 52 patients with SSc and 53 healthy co

Inhibition of interferon-gamma by an interferon-gamma receptor immunoadhesin.

Interferon-gamma (IFN-gamma) is an important cytokine which regulates inflammatory and immune response mechanisms. IFN-gamma enhances the presentation and recognition of antigens by inducing the expression of major histocompatibility complex (MHC) proteins, by activating effector T cells and mononuclear phagocytes, and by modulating immunoglobulin production and class selection in B cells. Inappropriate production of IFN-gamma has been implicated in the pathogenesis of several autoimmune and inflammatory diseases and in graft rejection. Here, we describe a recombinant inhibitor of IFN-gamma, t

Human interferon-ω: an underappreciated type I interferon.

Interferon-ω (IFN-ω) is a member of the human type I interferon family that has historically been overshadowed by IFN-α and IFN-β. Recent human "natural perturbations", most notably selective neutralization of IFN-ω by autoantibodies in life-threatening viral infections, have renewed interest in this comparatively understudied cytokine and indicate that its antiviral activity may not always be fully compensated in defined clinical settings. This renewed focus has prompted reassessment of its single-gene organization, distinct antigenicity, intermediate receptor-binding kinetics, cellular sourc

Cases of interferon-alpha and interferon-beta-induced thyroiditis.

Interferons are currently the major treatment modality for several malignant and non-malignant diseases such as chronic hepatitis C and B, multiple sclerosis, hematological malignancies, malignant melanoma, renal cell carcinoma, etc. Thyroid disorders develop in some of the interferon-treated patients with the incidence ranging from 1% to 35%. These complications may often result in dose reduction or discontinuation of interferon therapy. Interferon induced thyroid disorders can be classified as autoimmune and non-autoimmune thyroiditis. There are many studies on the development of thyroid dys
