Paracoccidioidesbrasiliensis induces IL-32 and is controlled by IL-15/IL-32/vitamin D pathway in vitro.

Paracoccidioidomycosis (PCM) is a systemic fungal disease caused by Paracoccidioides spp., whose clinical outcome depends on immune response. Interleukin 32 (IL-32) is a cytokine present in inflammatory and infectious diseases, including bacterial, virus and protozoan infections. Its role in fungal disease remains unclear. The axis IL-15, IL-32 and vitamin D leads to microbicidal capacity against intracellular pathogens. Thus, the aims of this study were to investigate the production of IL-32 during Paracoccidioides spp. infection and whether this cytokine and IL-15 can increase P. brasiliensi

Structural insights into the co-evolution of IL-2 and its private receptor in fish.

Interleukin (IL) -2, a member of the four α-helical cytokine family, has broad regulatory roles in mediating vertebrate immune response. In mammals, IL-2 and IL-15 share a common evolutionary origin and possess overlapping but distinct functions. IL-2 and IL-15 bind to distinct private receptors for signaling. However, fish appear to possess a single IL-15Rα like gene whilst lack additional gene(s) coding for IL-2Rα. Whether the IL-2 and IL-15 interact with the same receptor in fish and how their functions and receptors have evolved are not fully understood. In this study, homologues of IL-2 a

Modulation of immune genes in the mucosal-associated lymphoid tissues of cobia by Sarcodia suae extract.

Rachycentron canadum (cobia) is a marine fish species of high economic value in aquaculture due to its fast growth rate and good feed conversion efficacy. Regrettably, the industry has been affected by significant setbacks from high mortality due to diseases. Consequently, an improved perception of innate immunity correlated to each mucosal-associated lymphoid tissue (MALT) in teleost fish is necessary to understand hosts' response towards infections better. The utilization of polysaccharides in seaweed to stimulate the immune system has gathered unprecedented attention. The present study exam

Role of anti-tumorigenic cytokines in gastrointestinal cancers.

Gastrointestinal (GI) cancers are among the deadliest worldwide, with millions of cases and deaths annually. In the U.S., they account for over 25 % of cancer-related deaths, with colorectal cancer (CRC) leading in incidence and mortality, followed by pancreatic, liver, gastric, and esophageal cancers. These malignancies vary in cellular origin and etiology but share common risk factors, which contribute to chronic inflammation and immune dysfunction, influencing cytokine-driven pathways that shape the tumor environment. This review outlines and discusses the anti-tumorigenic properties of key

The contrasting roles of IL-2 and IL-15 in the life and death of lymphocytes: implications for the immunotherapy of rheumatological diseases.

Interleukin-15 (IL-15) is a 14-15-kDa member of the 4alpha helix bundle family of cytokines that stimulate T and NK (natural killer) cells. IL-15 and IL-2 utilize heterotrimeric receptors that include the cytokine-specific private receptors IL-2Ralpha and IL-15Ralpha, as well as two receptor elements that they share, IL-2Rbeta and gammac. Although IL-2 and IL-15 share two receptor subunits and many functions, at times they provide contrasting contributions to T-cell-mediated immune responses. IL-2, through its pivotal role in activation-induced cell death (AICD), is involved in peripheral tole

B cells drive CCR5+CD4+ tissue-resident memory T-cell cytotoxicity via IL-15Rα-IL-15 signaling in primary biliary cholangitis.

BACKGROUND & AIMS: IL-15Rα mediates immune responses by binding and trans-presenting IL-15 to tissue-resident memory T (TRM) cells, enabling sustained activation. Given the pivotal role of IL-15Rα/IL-15 signaling in autoimmune diseases, elucidating its organ-specific expression and pathogenic functions in primary biliary cholangitis (PBC) remains crucial. METHODS: Immunohistochemical staining was performed on liver tissues from healthy controls (n = 10) and patients with PBC (n = 94), autoimmune hepatitis (n = 32), chronic hepatitis B (n = 10), and metabolic dysfunction-associated steatohepat

γc-cytokines in HIV Cure Research.

PURPOSE OF REVIEW: Well tolerated, scalable strategies capable of targeting latent HIV reservoirs are essential for the development of cure approaches that can be implemented globally. In recent years, common γ-chain cytokines (γc-cytokines) have emerged as promising candidates as standalone interventions or in combination with complementary therapeutic strategies. γc-cytokines exhibit latency-reversing activity and immunomodulatory effects. In this review, we summarize recent in-vitro, ex-vivo, and in-vivo studies evaluating the use of γc-cytokines in HIV cure research. RECENT FINDINGS: γc-c

Specific miRNAs are associated with human cancer cachexia in an organ-specific manner.

BACKGROUND: Cancer cachexia (CCx) is a complex and multi-organ wasting syndrome characterized by substantial weight loss and poor prognosis. An improved understanding of the mechanisms involved in the onset and progression of cancer cachexia is essential. How microRNAs contribute to the clinical manifestation and progression of CCx remains elusive. The aim of this study was to identify specific miRNAs related to organ-specific CCx and explore their functional role in humans. METHODS: miRNA patterns in serum and in cachexia target organs (liver, muscle and adipose tissue) from weight stable (N

Characterizing the mechanisms underpinning interleukin-15Rα-mediated protection against sepsis and candidiasis.

The interleukin (IL)-15Rα receptor has crucial, protective roles in sepsis and candidiasis via binding to its ligand, IL-15. However, the underlying mechanisms remain largely unexplored. In our study, we first confirmed the protective effects of IL-15 using clinical samples from patients with sepsis and candidiasis, and also in animal models with those conditions. We therapeutically administered IL-15 to IL-15Rα-deficient mice to elucidate the roles of IL-15Rα in sepsis and candidiasis treatment. Bacterial and fungal infections expedite mortality, caused organ damage, elevated the microbial bu

Engineered VNP20009 expressing IL-15&15Rα augments anti-tumor immunity for bladder cancer treatment.

Surgical resection combined with intravesical instillation of chemotherapeutics or Bacillus Calmette-Guerin (BCG) to remove residual cancer cells is the gold standard for the clinical treatment of patients with bladder cancer. In a recent clinical trial, a new super-agonist complex of IL-15 - N803, has shown promising results when used in combination with BCG to treat patients with bladder cancer who do not respond to BCG. Herein, we used temperature-controlled pBV220 plasmid encoding Interleukin-15 and its receptor alpha subunit (IL-15&15Rα) to transform VNP20009, an attenuated salmonella typ

Characteristics of NK cells activation following IL-12, IL-15, IL-18 cytokines stimulation for preclinical purposes.

Adoptive cell therapy is a promising strategy in cancer treatment, among which NK cell therapies represent an encouraging approach. Freshly isolated NK cells exhibit low cytolytic activity. Therefore, efficient activation of NK cells is essential to enhance their anti-cancer properties. The aim of this study was to compare ex vivo NK cells activation with commonly used cytokines, including IL-2, IL-12, IL-15, IL-18, or STING agonist, and to determine the most effective activation method. Our results indicate higher viability and expression of activating receptors and granzyme B in IL-15-treate

IL-2 receptors preassemble and signal in the ER/Golgi causing resistance to antiproliferative anti-IL-2Rα therapies.

Interleukin-2 (IL-2) and IL-15 play pivotal roles in T cell activation, apoptosis, and survival, and are implicated in leukemias and autoimmune diseases. Their heterotrimeric receptors share their β- and γc-chains, but have distinct α-chains. Anti-IL-2Rα (daclizumab) therapy targeting cell surface-expressed receptor subunits to inhibit T cell proliferation has only brought limited success in adult T cell leukemia/lymphoma (ATL) and in multiple sclerosis. We asked whether IL-2R subunits could already preassemble and signal efficiently in the endoplasmic reticulum (ER) and the Golgi. A combinati

Immunologic Biomarkers in Peripheral Blood of Persons With Tuberculosis and Advanced HIV.

INTRODUCTION: Tuberculosis (TB) is a common opportunistic infection among people living with HIV. Diagnostic tests such as culture, Xpert-MTB-RIF, and ULTRA have low sensitivity in paucibacillary TB disease; a blood biomarker could improve TB diagnostic capabilities. We assessed soluble factors to identify biomarkers associated with TB among persons with advanced HIV. METHODS: A case-control (1:1) study was conducted, with participants from Rio de Janeiro and Manaus, Brazil. People living with HIV presenting with CD4 count ≤100 cells/mm3 were eligible to participate. Cases had culture-confirm

The Roles of IL-7 and IL-15 in Niches for Lymphocyte Progenitors and Immune Cells in Lymphoid Organs.

Lymphoid organs consist of immune cells and stromal cells. The stromal cells produce various cytokines that support the development, maintenance, and response of the immune cells. IL-7 and IL-15 are the major cytokines produced by stromal cells and are essential for the development and maintenance of lymphocytes and innate lymphoid cells (ILCs). In addition, IL-7 is indispensable for the organogenesis of lymphoid organs. However, because the amount of these two cytokines is relatively low, it has been difficult to directly detect their expression. Recently, several groups succeeded in establis

CAR NK cell production from human cord blood NK progenitor cells is enhanced by stimulation of the IL-15 receptor.

Cord blood (CB)-derived chimeric antigen receptor (CAR) natural killer (NK) cells have demonstrated significant antitumor efficacy. We recently reported that CB-derived CAR NK cells predominantly originate from CD7+CD56-CD34-HLA-DR-Lin- NK cell precursors in CB. Here, we demonstrate that stimulating the interleukin (IL)-15 receptor on these NK precursors enhances the production of CAR NK cells from CB cells. In CB CD56-CD34-HLA-DR-Lin- cells, the IL-15 receptor was exclusively expressed on CD7+ NK cell precursors. Using K562 feeder cells that express not only 4-1BB ligand and membrane-bound (m
