Mycobacterium tuberculosis uses intrinsically disordered, fast evolving proteins to interact with conserved host factors.

BACKGROUND: Intrinsically disordered protein regions (IDRs) are implicated in diverse cellular processes in eukaryotes and, in these organisms, they cover up to 40% of the proteome. Surprisingly little is known about IDRs in bacterial proteomes. Specifically, a number of questions remain unanswered, such as the role of these regions in host-pathogen interactions, their adaptive potential and evolutionary trajectories, as well as their biophysical properties. Here we focus on Mycobacterium tuberculosis and take advantage of the fact that, due to its extreme epidemiological relevance, several la

Intrinsic and extrinsic factors interact during development to influence telomere length in a long-lived reptile.

The mechanisms connecting environmental conditions to plasticity in biological aging trajectories are fundamental to understanding individual variation in functional traits and life history. Recent findings suggest that telomere biology is especially dynamic during early life stages and has long-term consequences for subsequent reproduction and survival. However, our current understanding is mostly derived from studies investigating ecological and anthropogenic factors separately, leaving the effects of complex environmental interactions unresolved. Effects of intrinsic and extrinsic factors on ruminating, grazing, and bedding time in bighorn sheep (Ovis canadensis).

Rumination is the repeated process of regurgitation of a food bolus, followed by chewing, swallowing, and regurgitation, which enhance nutrient assimilation. Time spent in lateral recumbency (i.e., bedded, lying) has often been used as a proxy for time spent ruminating due to difficulties of observing detailed rumination behavior in the field. The actual proportion of time spent ruminating, or other activities, will in turn be affected by the age and sex of an individual but also with changes in food quality. We studied the effects of intrinsic and extrinsic factors on time spent ruminating, b

Extrinsic and intrinsic risk factors associated with injuries in young dancers aged 8-16 years.

In the present study, we tried to determine the association between joint ranges of motion, anatomical anomalies, body structure, dance discipline, and injuries in young female recreational dancers. A group of 1336 non-professional female dancers (age 8-16 years), were screened. The risk factors considered for injuries were: range of motion, body structure, anatomical anomalies, dance technique, and dance discipline. Sixty-one different types of injuries and symptoms were identified and later classified into four major categories: knee injuries, foot or ankle tendinopathy, back injuries, and n

Transforming growth factor-beta type-II receptor signalling: intrinsic/associated casein kinase activity, receptor interactions and functional effects of blocking antibodies.

The transforming growth factor beta (TGF-beta) family of growth factors control proliferation, extracellular matrix synthesis and/ or differentiation in a wide variety of cells. However, the molecular mechanisms governing ligand binding, receptor oligomerization and signal transduction remain incompletely understood. In this study, we utilized a set of antibodies selective for the extracellular and intracellular domains of the TGF-beta type-II receptor as probes to investigate the intrinsic kinase activity of this receptor and its physical association in multimeric complexes with type-I and ty

The effect of cyanide on serum cobalamin and on cobalamin analogues when measured by radioassay using R-protein and intrinsic factor.

Concentrations of cobalamin measured in human serum by radioassay differ when R-protein or intrinsic factor is used as the binding determinant. Experiments were carried out to determine whether the concentration of cyanide used to extract the serum cobalamin could account for this difference. When either R-protein or intrinsic factor was used, the cobalamin measured in serum extracted in the absence of cyanide was low. As the concentration of cyanide was raised in the extraction mixture, the cobalamin concentration that was measured increased when either protein binder was used, but the absolu

An evaluation of simultaneously administered free and intrinsic factor bound radioactive cyanocobalamin in the diagnosis of pernicious anaemia in the elderly.

The definitive diagnosis of pernicious anaemia (PA) in the elderly is by no means always straightforward, particularly when inappropriate medication has been introduced before the institution of specific investigatory procedures. A detailed haematological study was carried out on 301 patients aged 60-95 with a serum B(12) concentration at the laboratory's lower level of normal of 150 ng per litre (Euglena gracilis assay). The diagnosis of PA was based on strict predetermined haematological criteria. All patients were subsequently studied by the simultaneous oral administration of the dual isot

Exploring the Association of Helicobacter pylori With Anti-intrinsic Factor and Anti-parietal Cell Antibodies in Pernicious Anemia: A Systematic Review.

Pernicious anemia, historically tied to vitamin B12 malabsorption due to intrinsic factor secretion impairment, has evolved in understanding, especially concerning its association with autoimmune gastritis. This systematic review dives deep into the multifaceted relationship between Helicobacter pylori (H. pylori) infection, autoimmune gastritis, and the presence of anti-intrinsic factors and anti-parietal cell antibodies. Comprehensive database searches revealed a higher prevalence of H. pylori infection in pernicious anemia patients, with some studies suggesting a consequential increase in t

Interaction of von Willebrand factor domain A1 with platelet glycoprotein Ibalpha-(1-289). Slow intrinsic binding kinetics mediate rapid platelet adhesion.

We investigated the crucial hemostatic interaction between von Willebrand factor (VWF) and platelet glycoprotein (GP) Ibalpha. Recombinant VWF A1 domain (residues Glu(497)-Pro(705) of VWF) bound stoichiometrically to a GPIbalpha-calmodulin fusion protein (residues His(1)-Val(289) of GPIbalpha; GPIbalpha-CaM) immobilized on W-7-agarose with a K(d) of 3.3 microM. The variant VWF A1(R545A) bound to GPIbalpha-CaM 20-fold more tightly, mainly because the association rate constant k(on) increased from 1,100 to 8,800 M(-1) s(-1). The GPIbalpha mutations G233V and M239V cause platelet-type pseudo-von

Role of efflux pump(s) in intrinsic resistance of Pseudomonas aeruginosa: active efflux as a contributing factor to beta-lactam resistance.

Wild-type strains of Pseudomonas aeruginosa are more resistant to various beta-lactam antibiotics as well as other agents than most enteric bacteria. Although resistance to compounds of earlier generations is explained by the synergism between the outer membrane barrier and the inducible beta-lactamase, it was puzzling to see significant levels of resistance to compounds that do not act as inducers or are not hydrolyzed rapidly by the chromosomally encoded enzyme. This intrinsic-resistance phenotype becomes enhanced in those strains with the so-called intrinsic carbenicillin resistance. In the

Intrinsic clotting factors in dependency of age, sex, body mass index, and oral contraceptives: definition and risk of elevated clotting factor levels.

Elevated clotting factors have been demonstrated to be a risk factor for venous thromboembolism (VTE). The aim of our study was to investigate the impact of age, sex, body mass index, and oral contraceptives on the clotting factor activities of factors VIII, IX, XI, and XII and their impact on the cutoff definition and risk of VTE associated with elevated clotting factors. Factor VIII, IX, XI, and XII activities were measured in 499 blood donors and 286 patients with VTE. Age and body mass index predicted significantly and independently the clotting factor activities of factors VIII, IX, and X

Bone stress injuries in elite runners: intrinsic and extrinsic risk factors in Italian track and field athletes.

BACKGROUND: Bone stress injuries (BSIs) are overuse injuries that occur when a normal bone is no longer able to withstand repetitive mechanical loading. Given the high occurrence of BSIs in track and field athletes, this study aimed to evaluate risk factors associated with BSI development in elite athletes competing in different track events. METHODS: In this retrospective observational study, 172 Italian elite track and field athletes (mean age 23.7 ± 3.5) were interviewed using a closed-ended questionnaire that encompassed participant characteristics, history of BSI, and potential contributi

Anterior-Posterior Shear Stiffness of C6/C7 Functional Spinal Units: Effects of Mechanical and Intrinsic Factors.

PURPOSE: Determine the effect of mechanical and intrinsic (demographic and anthropometric) variables on anteroposterior shear stiffness in cervical functional spinal units (FSUs), and provide response corridors for computer model validation. METHODS: Thirty-nine human FSUs were loaded non-destructively in statically rotationally unconstrained pure shear up to 200N, in anterior and posterior directions, at rates of 1, 10, and 100 mm/s. Due to the inherent non-linear behavior, the resulting stiffness response was analyzed as bilinear (lower Phase I and higher Phase II). The effect of mechanical
