The endogenous β-galactosidase expressed in intestinal microbes is demonstrated to help humans in lactose usage, and treatment associated with the promotion of beneficial microorganism in the gut is correlated with lactose tolerance. From this point, a kind of recombinant live β-galactosidase delivery system using food-grade protein expression techniques and selected probiotics as vehicle was promoted by us for the purpose of application in lactose intolerance subjects. Previously, a recombinant Lactococcus lactis MG1363 strain expressing food-grade β-galactosidase, the L. lactis MG1363/FGZW,

A gene associated with lipopolysaccharide (LPS) transport was cloned from a local clinical Vibrio cholerae O1 strain of the Ogawa serotype by using the Lactococcus lactis nisin-controlled expression (NICE) system. The V. cholerae wzm gene, which codes for an integral membrane transporter protein, was expressed and targeted to the cytoplasmic membrane, and was crudely isolated through simple centrifugation and SDS solubilization. To examine seroreactivity of this construct, rabbits were orally fed with 10(9) cfu/ml of live, recombinant L. lactis carrying the wzm gene, induced with nisin prior t

Sixteen exopolysaccharide (EPS)-producing Lactococcus lactis strains were analyzed for the chemical compositions of their EPSs and the locations, sequences, and organization of the eps genes involved in EPS biosynthesis. This allowed the grouping of these strains into three major groups, representatives of which were studied in detail. Previously, we have characterized the eps gene cluster of strain NIZO B40 (group I) and determined the function of three of its glycosyltransferase (GTF) genes. Fragments of the eps gene clusters of strains NIZO B35 (group II) and NIZO B891 (group III) were clon

Campylobacter jejuni, the most common foodborne pathogen, relies on adhesion, invasion, and biofilm formation for successful virulence and persistence in the food-processing environments. In this study, we investigated the effects of the transient bacterium with probiotic properties Lactococcus cremoris on C. jejuni virulence via an adhesion and invasion assay using human colon adenocarcinoma Caco-2 cells, culturability via colony-forming units, and proteomic adaptation in co-culture via label-free quantitative mass spectrometry. Caco-2 cell adhesion and invasion assay revealed that L. cremori

BACKGROUND: Immunotherapy has emerged as a promising strategy for pancreatic cancer. Radiotherapy not only mediates direct tumor cell killing but also provides a source of tumor antigens for dendritic cells (DCs) uptake and presentation via the induction of immunogenic cell death (ICD). However, the limited antigen presentation efficiency following radiotherapy and the rapid enzymatic degradation of adjuvants within the tumor microenvironment hinder the subsequent efficacy of immunotherapy.
METHODS: We developed an in situ hydrogel vaccine, Gel-FOLactis. Combined with 8Gy radiotherapy for tre

Continuous screening of suitable starter candidates for fermented foods necessary based on safety and technological evaluations. For this purpose, 34 Lactococcus lactis strains isolated from kimchi were evaluated to select potential starter candidates. All tested strains were susceptible to ampicillin, chloramphenicol, clindamycin, erythromycin, gentamicin, kanamycin, and vancomycin. However, 32 strains exhibited resistance to streptomycin, while 2 showed resistance to tetracycline. None of the strains demonstrated hemolytic activity. All strains exhibited protease activity and acid production

Lactococcosis is a major bacterial disease impacting rainbow trout production in South Africa and Southeast Asia, particularly during summer. In this study, 15 isolates from affected aquaculture facilities were characterised, revealing Lactococcus petauri (n = 12) as the predominant species, rather than the traditionally recognised L. garvieae (n = 3). This indicates a potential shift in the aetiology of lactococcosis with implications for diagnosis and management. Genomic screening identified multiple virulence factors, including adhesins in 14 isolates, capsular polysaccharide biosynthesis g

Lactococcus lactis is a gram-positive, facultative anaerobic coccus that is occasionally isolated from human mucocutaneous surfaces such as the intestines. It is used in the dairy industry for milk acidification and is mostly nonpathogenic in immunocompetent humans, however a number of cases of infection with L. lactis have been reported in recent years. In this article, we describe two cases of infection due to L. lactis in patients with chronic diarrhea. The first case is a five-month-old boy who was operated on for volvulus on his first day of life and had ileostomy with subsequent diagnosi

Lactococcus lactis CRL 1584 isolated from a Lithobates catesbeianus hatchery inhibits the growth of Citrobacter freundii (a bullfrog pathogen) and Listeria monocytogenes by a synergistic effect between lactic acid, hydrogen peroxide and a bacteriocin-like molecule. The chemical characterization of the bacteriocin in cell-free supernatants indicates that it has a proteinaceous nature. Hexadecane and ethyl acetate did not modify the bacteriocin activity, while 10 and 20 % (v/v) chloroform decreased the activity by 29 and 43 %, respectively. The antimicrobial peptide was heat stable since 85 % of

The Brucella abortus ribosomal protein L7/L12 is an immunodominant antigen and an interesting candidate for the development of oral live vaccines against brucellosis. Here, a recombinant Lactococcus lactis strain producing L7/L12 under the control of nisin inducible promoter was orally administered to BALB/c mice. Significant levels of anti-L7/L12 specific IgA detected in feces revealed an induced local humoral immune response. However, serum analysis did not reveal any anti-L7/L12 antibodies suggesting the absence of a systemic response. Nevertheless, the vaccinated mice showed a partial prot

Oral microorganisms contribute to the progression of oral squamous cell carcinoma (OSCC), and the gut microbiome may also influence OSCC by modulating systemic immunity. This study investigated oral and gut microbial changes in a 4-nitroquinoline N-oxide (4-NQO)-induced OSCC mouse model. After 16 weeks of 4-NQO exposure, significant alterations were observed in the beta diversity of both oral and gut microbiomes. Notably, the relative abundance of Lactococcus increased, especially in oral microbiomes, from week 6 to 16, followed by a decline at week 22, suggesting a 4-NQO-induced niche favorab

The present investigation provides the first report of Lactococcus lactis isolated from the intestinal tract of Anabas testudineus (Bloch, 1792), emphasizing its probiotic attributes and biosafety for aquaculture applications. From 100 bacterial isolates, three strains (GUNA, GUN-1, GUN-2) were identified as L. lactis via 16 S rRNA sequencing. These isolates displayed pronounced antagonism against Aeromonas hydrophila and A. jandaei, along with marked tolerance to gastrointestinal stressors, including low pH (71-80% survival at pH 2.0-3.0), bile salts (69-78% at 0.3-0.5% oxgall), and phenol (6

Bacteriophages infecting Lactococcus spp. are a major cause of fermentation failures in the dairy industry. Among them, c2-type phages remain insufficiently studied, and rapid detection methods are limited. This study aimed to develop a loop-mediated isothermal amplification (LAMP) assay for sensitive detection of genetically diverse c2 phages. As part of this work, 12 prevalent c2 phages isolated from Russian dairy plants and 22 additional c2 phages were sequenced and analyzed. Conserved genomic regions were identified, and two primer sets were designed to account for the high genetic variabi

BACKGROUNDS/AIMS: Obstructive jaundice, resulting from bile duct obstruction, is associated with increased morbidity and mortality due to impaired bile flow, dysbiosis of the gut microbiota, enhanced bacterial translocation, and hepatocellular injury. Persistent biliary obstruction can further progress to hepatic fibrosis and ultimately cirrhosis. Probiotics might help modulate microbiota and reduce liver injury. This study investigates the effect of Lactococcus lactis D4 on Ki-67 expression and liver fibrosis in rats with obstructive jaundice.
METHODS: Fifteen male Wistar rats (10-16 weeks o

Salmonella infection in broiler chickens poses a significant threat to poultry health and food safety, especially with the rise of multidrug-resistant strains. This work assessed the prevalence, antimicrobial resistance profiles, and resistance genes of Salmonella isolates from 30 broiler farms, and evaluated the protective effects of probiotics against Salmonella challenge. Salmonella spp. were isolated from 30% (9/30) of farms, with Salmonella Enteritidis accounting for 60% (15/25) and Salmonella Give for 40% (10/25) of the recovered isolates. Mortality rates were 10.13±0.89% in Salmonella E

OBJECTIVE: Development of an effective mucosal vaccine to induce specific immune responses against Foot-and-mouth disease virus (FMDV).
RESULTS: For this purpose, the FMDV VP1 gene (SPVP1) was optimized and synthesized based on the codon bias of Lactococcus lactis (L. lactis), and then incorporated in the plasmid pNZ8148. L. lactis NZ9000 containing the pNZ8148-SPVP1 recombinant plasmid was used as an oral delivery vehicle to induce anti-FMDV mucosal and systemic immune responses in mice. After confirmation that the SPVP1 protein was expressed successfully in the recombinant L. latic, the mic

Some lactic acid bacteria (LAB) are known to improve atopic dermatitis (AD) through the regulation and stimulation of the host immune system. In this study, we found that ingestion of yogurt containing Lactococcus lactis 11/19-B1 strain (L. lactis 11/19-B1) daily for 8 weeks significantly improved the severity scoring of atopic dermatitis (SCORAD) system score from 38.8 ± 14.4 to 24.2 ± 12.0 in children suffering from AD. We tried to identify which LAB species among the five species contained in the test yogurt contributed to the improvement in AD pathology using an AD mouse model induced by r

The composition of intestinal microbiota is widely believed to not only affect gut health but also influence behaviour. This study aimed to evaluate the probiotic characteristics, antioxidant activity, and antidepressant- and anxiolytic-like activities of Lactococcus lactis subsp. cremoris LL95. This strain showed probiotic properties such as resistance in a simulated gastric tract model and survival at different concentrations of NaCl and bile salts. Moreover, antioxidant activity of LL95 was demonstrated through DPPH radical scavenging activity, scavenging of ABTS•+ radical and ferric ion re

We report the case of an immunocompetent man who presented with fever and abdominal pain and was found to have a hepatic abscess and a peri-renal abscess with a computerized tomography scan. The hepatic abscess was drained percutaneously and cultures revealed the presence of Lactococcus lactis that was sensitive to penicillin. The patient was successfully treated with ceftriaxone and metronidazole with resolution of the abscesses. Further work-up revealed atrophic gastritis, vitamin B12 deficiency, periodontitis and gingivitis, suggesting a possible site of entry for the development of the abs
