Response patterns and neuronal networks of photosensory pineal organs.

Pineal organs of poikilotherm animals have photoreceptor cells with cone type structures of disk membranes of the outer segment. In the lamprey, Lampetra japonica, photoreceptors responded to light at 6 X 10(-4) lux as a threshold with hyperpolarization, which modulated the spike discharge of the ganglion cells. Electrophysiological data and high performance liquid chromatography analysis of visual pigments of the pineal suggested the presence of several types of photoreceptor cells. Actually, two different immuno-reactivities were observed in photoreceptors by using antibodies against rod spe

Evolution of the expression and regulation of the nuclear hormone receptor ERR gene family in the chordate lineage.

The Estrogen Related Receptor (ERR) nuclear hormone receptor genes have a wide diversity of roles in vertebrate development. In embryos, ERR genes are expressed in several tissues, including the central and peripheral nervous systems. Here we seek to establish the evolutionary history of chordate ERR genes, their expression and their regulation. We examine ERR expression in mollusc, amphioxus and sea squirt embryos, finding the single ERR orthologue is expressed in the nervous system in all three, with muscle expression also found in the two chordates. We show that most jawed vertebrates and l

The hemisegmental locomotor network revisited.

The organization of the minimal neuronal substrate capable of generating locomotor rhythmicity in vertebrates is investigated in several species, with an emphasis on identifying evolutionary-conserved features. In lamprey, an eel-like lower vertebrate that swims by undulatory movements of the body, the network has been identified as a recurrent network of excitatory interneurons localized in each spinal hemisegment. This conclusion rested upon the observation that each side of the spinal cord is able to express rhythmic locomotor-related bursting after being surgically separated along the midl

Lampricide 3-trifluoromethyl-4-nitrophenol (TFM) impacts whole-organism oxygen consumption in non-target fishes.

The application of the lampricide TFM (3-trifluoromethyl-4-nitrophenol) to streams infested with larval sea lamprey constitutes the backbone of the Sea Lamprey Control Program in the Laurentian Great Lakes, but native species have greatly varying sensitivities to TFM. Earlier work by D'Souza and colleagues revealed that the whole-animal oxygen consumption of larval sea lamprey responded in a dose-dependent manner to increasing TFM concentrations. We set out to determine 1) if TFM exposure leads to stepwise increases in whole-animal oxygen consumption in non-target fishes, and 2) if exposure to

Fish species sensitivity classification for environmental impact assessment, conservation and restoration planning.

Species conservation, river rehabilitation, stock enhancement, environmental impact assessment and related planning tools require indicators to identify significant impacts but also mitigation success. Since river systems are shaped by disturbances from floods and droughts, typical riverine fish species should have evolved life history traits providing resilience against such disturbances. This study compiled and analyzed resilience traits of European lampreys and fish species to derive a novel sensitivity classification of species to mortality. We assembled life history traits like maximum le

Anti-CD38 VLRB-Fc antibodies from transgenic tobacco exhibit potent anti-myeloma activity in vitro and in vivo.

Multiple myeloma (MM) is an incurable hematological malignancy characterized by the abnormal proliferation of malignant plasma cells. While current therapies, particularly anti-CD38 monoclonal antibodies such as daratumumab, have improved patient outcomes, they are often associated with high costs and potential adverse effects. Therefore, there is a growing need for alternative, cost-effective therapeutic strategies. In this study, we report for the first time the successful generation of a plant-derived lamprey variable lymphocyte receptor B (VLRB) recombinant antibody, targeting CD38-express

Activation of Transducin by Bistable Pigment Parapinopsin in the Pineal Organ of Lower Vertebrates.

Pineal organs of lower vertebrates contain several kinds of photosensitive molecules, opsins that are suggested to be involved in different light-regulated physiological functions. We previously reported that parapinopsin is an ultraviolet (UV)-sensitive opsin that underlies hyperpolarization of the pineal photoreceptor cells of lower vertebrates to achieve pineal wavelength discrimination. Although, parapinopsin is phylogenetically close to vertebrate visual opsins, it exhibits a property similar to invertebrate visual opsins and melanopsin: the photoproduct of parapinopsin is stable and reve

Ancient evolutionary origin of vertebrate enteric neurons from trunk-derived neural crest.

The enteric nervous system of jawed vertebrates arises primarily from vagal neural crest cells that migrate to the foregut and subsequently colonize and innervate the entire gastrointestinal tract. Here we examine development of the enteric nervous system in the basal jawless vertebrate the sea lamprey (Petromyzon marinus) to gain insight into its evolutionary origin. Surprisingly, we find no evidence for the existence of a vagally derived enteric neural crest population in the lamprey. Rather, labelling with the lipophilic dye DiI shows that late-migrating cells, originating from the trunk ne

Disturbances to energy metabolism in juvenile lake sturgeon (Acipenser fulvescens) following exposure to niclosamide.

Since the 1960s, invasive sea lamprey (Petromyzon marinus) populations in the Laurentian Great Lakes have been controlled by applying two chemicals, 3-trifluoromethyl-4-nitrophenol (TFM) and 2',5-dichloro-4'-nitrosalicylanilide (niclosamide, aka. Bayluscide®), to streams infested with larval sea lamprey. These "lampricide" applications primarily rely on TFM, and are often combined with 1-2% niclosamide, which increases treatment effectiveness. Niclosamide is also used alone to treat lentic habitats and in rivers with high discharge. However, little is known about niclosamide's possible adverse

Evolution of hormone selectivity in glucocorticoid and mineralocorticoid receptors.

Mineralocorticoid receptors (MR) and glucocorticoid receptors (GR) are descended from an ancestral corticoid receptor (CR). To date, the earliest CR have been found in lamprey and hagfish, two jawless fish (cyclostomes) that evolved at the base of the vertebrate line. Lamprey CR has both MR and GR activity. Distinct orthologs of the GR and MR first appear in skates and sharks, which are cartilaginous fishes (Chondrichthyes). Aldosterone, the physiological mineralocorticoid in terrestrial vertebrates, first appears in lobe-finned fish, such as lungfish and coelacanth, forerunners of terrestrial

Cranial nerves of the coelacanth, Latimeria chalumnae [Osteichthyes: Sarcopterygii: Actinistia], and comparisons with other craniata.

We reconstructed the cranial nerves of a serially sectioned prenatal coelacanth, Latimeria chalumnae. This allowed us to correct several mistakes in the literature and to make broad phylogenetic comparisons with other craniates. The genera surveyed in our phylogenetic analysis were Eptatretus, Myxine, Petromyzon, Lampetra, Chimaera, Hydrolagus, Squalus, Mustelus, Polypterus, Acipenser, Lepisosteus, Amia, Neoceratodus, Protopterus, Lepidosiren, Latimeria and Ambystoma. Cladistic analysis of our data shows that Latimeria shares with Ambystoma two characters of the cranial nerves. Our chief findi

A Brainstem Neural Substrate for Stopping Locomotion.

Locomotion occurs sporadically and needs to be started, maintained, and stopped. The neural substrate underlying the activation of locomotion is partly known, but little is known about mechanisms involved in termination of locomotion. Recently, reticulospinal neurons (stop cells) were found to play a crucial role in stopping locomotion in the lamprey: their activation halts ongoing locomotion and their inactivation slows down the termination process. Intracellular recordings of these cells revealed a distinct activity pattern, with a burst of action potentials at the beginning of a locomotor b

Muscarinic receptor activation elicits sustained, recurring depolarizations in reticulospinal neurons.

In lampreys, brain stem reticulospinal (RS) neurons constitute the main descending input to the spinal cord and activate the spinal locomotor central pattern generators. Cholinergic nicotinic inputs activate RS neurons, and consequently, induce locomotion. Cholinergic muscarinic agonists also induce locomotion when applied to the brain stem of birds. This study examined whether bath applications of muscarinic agonists could activate RS neurons and initiate motor output in lampreys. Bath applications of 25 microM muscarine elicited sustained, recurring depolarizations (mean duration of 5.0 +/-

The pineal organ as a folded retina: immunocytochemical localization of opsins.

The most simple pineal complex (the pineal and parapineal organs of lampreys), consists of saccular evaginations of the diencephalic roof, and has a retina-like structure containing photoreceptor cells and secondary neurons. In more differentiated vertebrates, the successive folding of the pineal wall multiplies the cells and reduces the lumen of the organ, but the pattern of the histological organization remains similar to that of lampreys; therefore, we consider the histological structure of the pineal organ of higher vertebrates as a 'folded retina'. The cell membrane of several pineal phot

Brainstem neural mechanisms controlling locomotion with special reference to basal vertebrates.

Over the last 60 years, the basic neural circuitry responsible for the supraspinal control of locomotion has progressively been uncovered. Initially, significant progress was made in identifying the different supraspinal structures controlling locomotion in mammals as well as some of the underlying mechanisms. It became clear, however, that the complexity of the mammalian central nervous system (CNS) prevented researchers from characterizing the detailed cellular mechanisms involved and that animal models with a simpler nervous system were needed. Basal vertebrate species such as lampreys, xen

From pineal photoreception to optogenetics: functional colour opponency based on a pineal bistable opsin.

Most animals sense light through opsins, which are photosensitive G protein-coupled receptors that utilize light information to regulate various physiological functions. In vertebrates, a representative example of such functions is vision, which is based on photoreception by visual opsins expressed in rod and cone photoreceptor cells of the retina. Accumulating photobiological evidence has gradually revealed the significance of the molecular properties of these opsins, as well as those of signal transduction-related molecules such as transducin, rhodopsin kinases, and arrestins, and their coll

Upstream passage windows for anadromous fish at a large barrier in the Rhine-Meuse estuary, 1996-2018.

Passage behaviour at barriers of migratory fish is well studied in rivers, whereas little is known about fish movement at estuarine barriers. In response to climate change, more estuarine barriers are likely to be erected in the future, further compromising connectivity for diadromous fish. As these fish species are already severely endangered, there is an urgent need to better understand passage behaviour at barriers to facilitate connectivity. These fish face challenges at estuarine sluice gates, where suitable passage windows occur only infrequently. To enhance fish migration, sluice manage

The evolutionary old forebrain as site of action to develop new psychotropic drugs.

BACKGROUND: Previously, the authors have developed a model of how reward-seeking and distress- avoiding behaviour is regulated by the human brain. The forebrain's evolution in vertebrates was taken as a starting point. AIMS: The authors want to inspire colleagues to study in particular the pharmacological effects on the described ancient forebrain structures in order to modify specific symptoms of mental disorders. METHODS: Compilation of data and ideas of previous articles, with examples to illustrate. RESULTS: A primary (lamprey-like), secondary (frog-like) and tertiary (mammal-like) fore

Use of chemical communication in the management of freshwater aquatic species that are vectors of human diseases or are invasive.

Chemical communication occurs when both originator (signaller) and one or more receiver(s) possess specializations for chemical exchange of information. Chemical information can be used by a wide variety of species to locate food and mates, avoid predators and engage in social interactions. In this review, we focus on chemical signalling between mates or cues from nest sites or hosts by selected aquatic pest species and indicate how chemical information can be used to manage pests. The pests are vectors of disease (blood-sucking insects) or invasive species (crayfishes and fishes) that have ex

Evolution of endothelin receptors in vertebrates.

Endothelin receptors are G protein coupled receptors (GPCRs) of the β-group of rhodopsin receptors that bind to endothelin ligands, which are 21 amino acid long peptides derived from longer prepro-endothelin precursors. The most basal Ednr-like GPCR is found outside vertebrates in the cephalochordate amphioxus, but endothelin ligands are only present among vertebrates, including the lineages of jawless vertebrates (lampreys and hagfishes), cartilaginous vertebrates (sharks, rays, and chimaeras), and bony vertebrates (ray-finned fishes and lobe-finned vertebrates including tetrapods).
