Hyperplasia and hypertrophy of Leydig cells associated with testicular germ cell tumours containing syncytiotrophoblastic giant cells.

In patients with seminomas and non-seminomatous germ cell tumours of the testis syncytiotrophoblastic giant cells (STGC) are largely responsible for elevated levels of chorionic gonadotropin. In searching for these cells, the question arises whether hyperplasia and/or hypertrophy of Leydig cells in the adjacent testicular tissue is a relevant finding. To elucidate this problem we analysed the tumour-free testicular tissue of 20 seminomas and 20 combined seminomatous and non-seminomatous germ cell tumours with or without immunohistochemically demonstrable STGC morphometrically. ATG5 gene regulates testosterone synthesis of testicular Leydig cells in Hezuo pig.

Autophagy-related gene 5 (ATG5) plays a crucial role in autophagosome formation. Recent studies have investigated the role of autophagy in regulating testosterone production; however, its expression in testicular tissues and Leydig cells of Hezuo pig remains less understood. In this study, we cloned the coding sequence (CDS) region of the ATG5 gene and assessed its expression using qPCR across various tissues and testes at different developmental stages in Hezuo pigs. Subsequently, we constructed silencing and overexpression vectors for ATG5 and transfected them into Leydig cells. Cell prolife

Monotropein improves late-onset hypogonadism in TM3 Leydig cells and aged rats.

BACKGROUND AND AIM: Late-onset hypogonadism (LOH) is an age-related condition characterized by declining testosterone levels and impaired reproductive and metabolic functions. Fermented Morinda citrifolia extract (FME) has demonstrated therapeutic potential against LOH; however, its active constituents and underlying mechanisms remain unclear. The aim of this study was to identify the key bioactive compounds in FME and evaluate their efficacies and mechanisms of action in alleviating LOH. METHODS: High-performance liquid chromatography (HPLC) was used to quantify the iridoids and coumarins in

Role of p57KIP2 in Stem and Progenitor Leydig Cells of Mouse Testes.

PURPOSE: Precise control of proliferation and differentiation of Leydig cells is important for gonadal androgenesis and spermatogenesis. Though cyclin-dependent kinase inhibitors are crucial for cell proliferation and differentiation, their role in the development of early adult Leydig cells (ALCs) remained unanswered. To understand mechanism for ALC development, functional expression of p57KIP2 (cdkn1c) was investigated in the stem Leydig cells (SLCs) and progenitor Leydig cells (PLCs) in mice. MATERIALS AND METHODS: The roles of p57KIP2 in the proliferation, differentiation, apoptosis, and

Sirtuin 1-mediated autophagy regulates testosterone synthesis in Leydig cells of piglets.

Testosterone is secreted by Leydig cells (LCs), which play an important physiological role in preserving male secondary sex characteristics, protecting male reproductive function, and establishing the blood-testis barrier. Studies have shown that autophagy is particularly active in LCs; however, its involvement in testosterone synthesis in porcine LCs has not been fully explored. Therefore, this experiment aimed to investigate the influence of autophagy on testosterone secretion in porcine LCs and its potential regulatory mechanism. Our results demonstrated that both testicular autophagy and s

Citrinin inhibits the function of Leydig cells in male rats in prepuberty.

Citrinin, a mycotoxin existing in fruits, has nephrotoxicity, hepatotoxicity and embryotoxicity. The effects of citrinin on Leydig cell development in prepuberty remains unclear. Male Sprague-Dawley rats were gavaged with 0, 1, 2.5, and 5 mg/kg citrinin from postnatal days 21-28. Citrinin at 5 mg/kg significantly decreased serum testosterone levels, while increasing serum LH and FSH levels. Citrinin at 1-5 mg/kg markedly downregulated Hsd17b3 and HSD17B3 expression, while upregulating Srd5a1 (SRD5A1) and Akr1c14 (AKR1C14) expression at 2.5 and/or 5 mg/kg. Citrinin at 5 mg/kg also significantly

Hyperhomocysteinemia lowers serum testosterone concentration via impairing testosterone production in Leydig cells.

Hyperhomocysteinemia (HHcy) plays a salient role in male infertility. However, whether HHcy interferes with testosterone production remains inconclusive. Here, we reported a lower serum testosterone level in HHcy mice. Single-cell RNA sequencing revealed that genes related to testosterone biosynthesis, together with nuclear receptor subfamily 5 group A member 1 (Nr5a1), a key transcription factor for steroidogenic genes, were downregulated in the Leydig cells (LCs) of HHcy mice. Mechanistically, Hcy lowered trimethylation of histone H3 on lysine 4 (H3K4me3), which was bound on the promoter reg

Ssc-miR-429 expression proliles and functions on inducing Leydig cells apoptosis.

Leydig cells (LCs) play an indispensable role in testosterone synthesis, and their dysfunction can result in male reproductive disorders. Previous transcriptome sequencing revealed differential expression of MicroRNA-429 (miR-429) in both Leydig stem cells (SLCs) and LCs, indicating its potential regulatory function in LCs. In this study, we examined the expression of miR-429 in seven pig tissues (heart, liver, spleen, lung, kidney, testis, epididymis, brain) and investigated its impact on the proliferation and apoptosis of testicular interstitial cells using various techniques such as CCK-8,

Bisphenol A (BPA) induces apoptosis of mouse Leydig cells via oxidative stress.

As one of the most frequently produced synthetic compounds worldwide, bisphenol A (BPA) has been widely used in many kinds of products such as appliances, housewares, and beverage cans. BPA has been shown to cause damage to male reproductive system; however, the potential mechanism remains to be investigated. In the present study, BPA exposure decreased the testis and epididymis coefficient, caused a disintegration of germinal epithelium, decreased the density and motility of sperm in the epididymis tissue, and increased the number of abnormal sperm morphology, which indicated that BPA exposur

FGF9 promotes cell proliferation and tumorigenesis in TM3 mouse Leydig progenitor cells.

Fibroblast growth factor 9 (FGF9) modulates cell proliferation, differentiation and motility for development and tissue repair in normal cells. Growing evidence shows that abnormal activation of FGF9 signaling is associated with tumor malignancy. We have previously reported that FGF9 increases MA-10 mouse Leydig tumor cell proliferation, in vitro, and tumor growth, in vivo. Also, FGF9 promotes the tumor growth and liver metastasis of mouse Lewis lung cancer cells, in vivo. However, the effects of FGF9 in the early stage of tumorigenesis remains elusive. In this study, TM3 mouse Leydig progenit

Resveratrol protects Leydig cells from nicotine-induced oxidative damage through enhanced autophagy.

Some studies have revealed that nicotine can damage the male reproductive system through various means including oxidative stress, which is a primary factor in the pathogenesis of male infertility. The strong anti-oxidative capacity of resveratrol has been demonstrated previously, but its role in the context of male reproduction remains inconclusive. To explore the biological role of resveratrol in protecting male reproductive function and the potential underlying mechanism, nicotine-induced Leydig cells were used as a cell model of oxidative damage. The data showed that resveratrol treatment

Mumps virus-induced innate immune responses in mouse Sertoli and Leydig cells.

Mumps virus (MuV) infection frequently causes orchitis and impairs male fertility. However, the mechanisms underlying the innate immune responses to MuV infection in the testis have yet to be investigated. This study showed that MuV induced innate immune responses in mouse Sertoli and Leydig cells through TLR2 and retinoic acid-inducible gene I (RIG-I) signaling, which result in the production of proinflammatory cytokines and chemokines, including TNF-α, IL-6, MCP-1, CXCL10, and type 1 interferons (IFN-α and IFN-β). By contrast, MuV did not induce the cytokine production in male germ cells. In

Morphological and functional maturation of Leydig cells: from rodent models to primates.

BACKGROUND: Leydig cells (LC) are the sites of testicular androgen production. Development of LC occurs in the testes of most mammalian species as two distinct growth phases, i.e. as fetal and pubertal/adult populations. In primates there are indications of a third neonatal growth phase. LC androgen production begins in embryonic life and is crucial for the intrauterine masculinization of the male fetal genital tract and brain, and continues until birth after which it rapidly declines. A short post-natal phase of LC activity in primates (including human) termed 'mini-puberty' precedes the peri

Changes in mouse Leydig cells ultrastructure and testosterone secretion after diethylcarbamazine administration.

Diethylcarbamazine (DEC) has been proven to be highly effective against lymphatic filariasis, although its effect on vertebrate cells remains uncertain. Mice Leydig cells after treatment with 200mg/kg of DEC for 12 days showed numerous lipid droplets, degenerated mitochondria, residual bodies and several giant whorl-like smooth endoplasmic reticulum, some of them encircling large lipids droplets. Treatment with lower dosages showed similar alterations on Leydig cells and the morphological effects decreased directly proportional to the drug concentration. Serum testosterone levels were signific

Effects of melatonin on Leydig cells in pinealectomized rat: an immunohistochemical study.

We have investigated immunohistochemically the effects of melatonin on Leydig cells in rat. Three groups of Wistar rats were used. Rats in group I and II were sham-pinealectomized (control) and pinealectomized, respectively, whereas rats in group III were pinealectomized and injected daily with melatonin for 2 months. At the end of the experiment, all animals were killed by decapitation and blood samples were obtained. Serum testosterone levels were determined with the use of a chemiluminescent enzyme immunoassay. Testicular tissue was collected and processed for semiquantitative evaluation of

Inhibitory actions of pineal indoles on steroidogenesis in isolated rat Leydig cells.

The inhibition elicited by pineal indoles on testosterone production by isolated rat Leydig cells could not be overcome by a maximally active dose of luteinizing hormone (LH), and dibutyryl-cAMP-induced steroidogenesis was also suppressed, suggesting that the indoles did not exert their effect through an interaction with LH receptors on Leydig cells. Pregnenolone-induced progesterone secretion was unaffected, indicating that the activity of 3 beta-dehydrogenase was not altered. Methoxytryptamine (MTN) at a dose of 1 mM decreased progesterone-induced 17 alpha-hydroxyprogesterone secretion by 50

Quantitative evaluation of Leydig cells in testicular biopsies of men with varicocele.

A quantitative analysis of Leydig cells was performed in 23 testicular biopsies of men with left varicocele and sperm count ranging from zero to 95,000 sperm/mm3. The oligozoospermic patients had more Leydig cells and higher FSH and LH serum levels than the patient group with more than 10,000 sperm/mm3. The Leydig cell density appeared tightly correlated (p less than 0.01) with the serum level of LH. In oligozoospermic subjects, an altered Leydig cell function could trigger an increased LH secretion; this seems likely to be responsible for the stimulation of interstitial cells resulting in an
