Penicillamine-induced elastosis perforans serpiginosa with abnormal "lumpy-bumpy" elastic fibers in lesional and non-lesional skin.

Four types of elastosis perforans serpiginosa (EPS) have been described in literature: 1) idiopathic EPS, 2) reactive perforating elastosis associated with connective tissue disorders, 3) in some instances of pseudoxanthoma elasticum (PXE), disease-specific calcified elastic tissue is extruded, producing a clinical picture indistinguishable from other types, may also be seen in patients undergoing hemodialysis and 4) EPS induced by long-term treatment with D-penicillamine is observed in patients suffering from Wilson's disease. Long term D-penicillamine therapy causes an alteration in the derm

Ibrutinib inhibits the replication of multiple poxviruses by targeting the Bruton tyrosine kinase.

The recent surge in global monkeypox (mpox) outbreaks highlights the critical need for developing antiviral agents targeting orthopoxvirus infections. Ibrutinib, a selective Bruton tyrosine kinase (BTK) inhibitor initially approved by the FDA in 2013 for treating chronic lymphocytic leukemia (CLL) and mantle cell lymphoma (MCL), has emerged as a potential antiviral candidate based on a high-throughput screening for its efficacy against vaccinia virus (VACV). This study evaluated the antiviral capability of ibrutinib against VACV, mpox virus (MPXV), and lumpy skin disease virus (LSDV), demonstr

Capripoxvirus Infections: Advances in Diagnosis and Strategic Control Measures.

Sheeppox virus (SPPV), goatpox virus (GTPV) and lumpy skin disease virus (LSDV) belong to the genus Capripoxvirus (CaPV) within the family Poxviridae. These transboundary and highly infectious viruses cause substantial economic losses by affecting the productivity of both small and large ruminants. Clinical manifestations include cutaneous lesions (skin nodules and pustular lesions), lymphadenopathy, pneumonia, reduced milk yield, mastitis, infertility and abortion. The diagnosis is based on a combination of clinical signs, virus isolation, serology and PCR/real-time PCR. Recent advancements h

Nck- and N-WASP-dependent actin-based motility is conserved in divergent vertebrate poxviruses.

Vaccinia virus enhances its cell-to-cell spread by stimulating actin polymerization via Src- and Abl-mediated phosphorylation of the highly conserved orthopoxvirus protein A36. The Yatapoxvirus, Yaba-like disease virus (YLDV), also induces actin polymerization, although it lacks an obvious A36 ortholog. We found that the YLDV protein YL126 can functionally replace A36 to promote Nck- and N-WASP-dependent actin polymerization. At least five phosphorylated tyrosines in YL126, rather than a single residue as in A36, are able to recruit Nck to promote actin polymerization. As is the case for A36,

Humoral immune response profile of a cattle herd vaccinated with 5- and 10-times Bakirköy strain sheep pox vaccine under field conditions.

Vaccination is the most effective control measure for Lumpy Skin Disease (LSD). The Bakırköy strain-derived sheep pox vaccine (SPPV) has been used against LSD in Türkiye since 2013. In this study, a cattle herd was vaccinated with SPPV and 35 cattle, of which 9 and 26 received 10 and 5 sheep doses, respectively, were followed for 200 days for humoral immune responses. Additionally, maternal antibodies in colostrum-fed calves were investigated. The humoral immune responses of naive and previously vaccinated cattle were compared to determine the effects of annual re-vaccination. Furthermore, the

Effects of imeglimin on liver enzymes and body composition in patients with MASLD and type 2 diabetes: a pilot study.

Skins and hides are perishable resources that can be damaged by parasitic diseases and human error, which result in downgrading or rejection. This study was conducted to identify defect types and to determine their prevalence in pickled sheep and wet blue goat skins and wet blue hides. Each selected skin or hide was examined for defects in natural light and the defects were graded according to established quality criteria in Ethiopian standard manuals. Major defects were captured by digital photography. The major pre-slaughter defects included scratches (64.2%), cockle (ekek) (32.8%), wounds o

Using pangenome variation graphs to improve mutation detection in a large DNA virus.

Accurately quantifying viral genetic diversity is essential for understanding pathogen evolution, transmission and emergence. However, standard approaches that map sequencing reads to a single linear reference genome introduce substantial reference bias, particularly for samples that are divergent and recombinant or belong to rare lineages. Pangenome variation graphs (PVGs) mitigate this issue by representing multiple genomes within a unified graph structure, enabling read mapping across all observed and potential haplotypes. Despite this, PVGs have rarely been applied to viruses. Here, we add

Review: Risks of disease transmission through semen in cattle.

The purpose of this paper is to review scientific evidence concerning pathogens that could potentially be transmitted via bovine semen. As a result of a careful analysis of the characteristics of infections that may cause transmission of disease through semen, effective control procedures can be identified that provide minimal constraint to the introduction of new bulls into herds for natural breeding and importation of valuable novel genetics through artificial insemination. The potential for transmission through bovine semen and corresponding effective control procedures are described for bo

A G-protein-coupled chemokine receptor: A putative insertion site for a multi-pathogen recombinant capripoxvirus vaccine strategy.

Capripoxviruses (CaPVs) have been shown to be ideal viral vectors for the development of recombinant multivalent vaccines to enable delivery of immunogenic genes from ruminant pathogens. So far, the viral thymidine kinase (TK) gene is the only gene used to generate recombinants. A putative non-essential gene encoding a G-protein-coupled chemokine receptor subfamily homologue (GPCR) was targeted as an additional insertion site. Peste des petits ruminants (PPR) was chosen as a disease model. A new recombinant CaPV expressing the viral attachment hemagglutinin (H) of the PPR virus (PPRV) in the G

Disease prevention and preparedness: the Food and Agriculture Organization Emergency Prevention System.

In 1994, the Food and Agriculture Organization undertook to revitalise its activities in the control of transboundary animal disease by establishing a new special programme known as the Emergency Prevention System (EMPRES) against transboundary animal and plant pests and diseases. The emphasis of the EMPRES livestock component is placed on pre-empting outbreaks and losses experienced by agriculture through the enhancement of local capacity to detect and react rapidly to plague events. EMPRES concentrates on the co-ordination of the Global Rinderpest Eradication Programme--a time-bound eradicat

Generation and application of immortalized sertoli cell line from sheep testis.

Primary sheep testicular Sertoli cells (STSCs) are ideal for investigating the molecular and pathogenic processes of capripoxvirus. However, the high cost of isolation and culture of primary STSCs, time-consuming operation, and short lifespan greatly limit their real-world application. In our study, the primary STSCs were isolated and immortalized by transfection of a lentiviral recombinant plasmid containing simian virus 40 (SV40) large T antigen. Androgen-binding protein (ABP) and vimentin (VIM) protein expression, SV40 large T antigen activity, proliferation assays, and apoptosis analysis r

Tetrandrine (TET) inhibits African swine fever virus entry into cells by blocking the PI3K/Akt pathway.

African Swine Fever Virus (ASFV) infection causes an acute and highly contagious disease in swine, resulting in significant economic losses and societal harm worldwide. Currently, there are no effective vaccines or antiviral drugs available for ASFV. Tetrandrine (TET) is extracted from the traditional Chinese herb Stephania tetrandrae, possesses diverse biological functions such as anti-inflammatory, anti-tumor, and antiviral activities. The study comprehensively evaluated the anti-ASFV effect of TET and validated it through biological assays. The dose-dependent inhibition of TET against ASFV

Quantitative risk assessment for infectious disease introduction in animal populations: a comprehensive review.

INTRODUCTION: Quantitative risk assessments (QRA) are valuable decision-support tools for estimating disease introduction risks in animal populations. METHODS: This review analyzed peer-reviewed QRA studies published between 2010 and 2024 that focused on risk of introduction, aiming to identify methodological trends and challenges. RESULTS: From the 1,933 studies screened, only 34 (2%) met inclusion criteria, primarily assessing risk through movements of live animals (n = 20), animal products (n = 7), both live animals and their products (n = 2), or vectors (n = 5), with no studies addressin

Common, emerging, vector-borne and infrequent abortogenic virus infections of cattle.

This review deals with the aetiology and the diagnosis of bovine viral abortion. While the abortion rates on beef and dairy cattle farms usually do not exceed 10%, significant economic losses because of abortion storms may be encountered. Determining the cause of abortions is usually a challenge, and it generally remains obscure in more than 50% of the necropsy submitted foetuses. Bovine viral diarrhoea virus and bovine herpesvirus-1 are the most common viruses causally associated with bovine abortions in farmed cattle globally. Rift Valley fever virus and bluetongue virus are important insect

Poxvirus sensitivity of a novel diploid sheep embryonic heart cell line.

Lumpy skin disease virus (LSDV), camelpox virus (CPV), and orf virus (ORFV) are members of the family Poxviridae. These viruses are usually isolated or produced in embryonated eggs or primary cells because continuous cell lines are less sensitive to infection. Disadvantages of the use of eggs or primary cells include limited availability, potential endogenous contaminants, and a limited ability to perform multiple passages. In this study, we developed a diploid cell culture from sheep embryonic hearts (EHs) and demonstrated its high proliferative and long-term storage capacities. In addition,

Viral Diseases in Water Buffalo (Bubalus bubalis): New Insights and Perspectives.

The water buffalo (Bubalus bubalis) has great adaptability to rustic environments and more variable conditions than cattle, who generally share the habitat. Diseases carried by buffaloes are relatively unknown and ignored and could be transmissible; an imbalance occurs between pathogens, environment, and susceptible hosts, generating a severe animal health problem. Also relevant is the effect of climate change on the populations of vectors that transmit viral diseases. The discovery of new virus variants that can pass from bovine (Bos) to buffalo or vice versa or to humans has highlighted the

Impact of infectious diseases on wild bovidae populations in Thailand: insights from population modelling and disease dynamics.

The wildlife and livestock interface is vital for wildlife conservation and habitat management. Infectious diseases maintained by domestic species may impact threatened species such as Asian bovids, as they share natural resources and habitats. To predict the population impact of infectious diseases with different traits, we used stochastic mathematical models to simulate the population dynamics over 100 years for 100 times in a model gaur (Bos gaurus) population with and without disease. We simulated repeated introductions from a reservoir, such as domestic cattle. We selected six bovine infe

Would you bet on the vet? Influences on dairy farmers' vaccination choices, with a spotlight on the Veterinarian impact.

Ensuring effective vaccination is crucial for epidemic control, particularly in voluntary vaccination scenarios. Though highly important for planning voluntary vaccination programs, we lack insights into the effectiveness of veterinarian communication and the impact of disease-specific traits on farmer vaccination intentions. To fill this void, our study compared five diseases affecting Israeli dairy cattle (Botulism, Bovine Ephemeral Fever (BEF), Brucellosis, Lumpy Skin Disease (LSD), and Rabies). Using questionnaires grounded in the theory of planned behavior, we surveyed 340 Israeli dairy f
