Mycobacteriophages and Their Applications.

Mycobacterial infections caused by tuberculous and non-tuberculous strains pose significant treatment challenges, especially among immunocompromised patients. Conventional antibiotic therapies often fail due to bacterial resistance, highlighting the need for alternative therapeutic strategies. Mycobacteriophages are emerging as promising candidates for the treatment of mycobacteria. This review comprehensively explores phage isolation, characterization, and clinical applications. Despite the need for more extensive in vitro and in vivo studies, existing evidence shows their efficacy against bo

Facing antibiotic resistance: Staphylococcus aureus phages as a medical tool.

Staphylococcus aureus is a common and often virulent pathogen in humans. This bacterium is widespread, being present on the skin and in the nose of healthy people. Staphylococcus aureus can cause infections with severe outcomes ranging from pustules to sepsis and death. The introduction of antibiotics led to a general belief that the problem of bacterial infections would be solved. Nonetheless, pathogens including staphylococci have evolved mechanisms of drug resistance. Among current attempts to address this problem, phage therapy offers a promising alternative to combat staphylococcal infect

Enhanced multi-omic viral profiling from microbial community sequencing with BAQLaVa.

Viruses are crucial components of microbial communities, both phage that infect bacterial community members as well as pathogenic and other eukaryotic viruses. However, they remain unobserved by most current technologies, due to combinations of experimental and analytical factors. To address the latter, we developed the BAQLaVa algorithm for high-resolution profiling of >120,000 viral species (viral genome bins, VGBs) via reference-based metagenome (MGX) or metatranscriptome (MTX) alignment to complementary nucleotide markers and proteome sets. In comprehensive benchmarking, BAQLaVa substantia

Genomic and biological characterization of lytic phages infecting Pseudomonas syringae associated with almond bacterial blast.

Pseudomonas species are Gram-negative bacterial pathogens that affect a wide range of economically important crops, including almond. Increasing resistance of Pseudomonas syringae to conventional management strategies highlights the need for alternative disease control options. In this study, we isolated and characterized three lytic bacteriophages, including vB_PsyP_Mobley, vB_PsyP_Plaza, and vB_PsyP_Mission, targeting almond-infecting Pseudomonas strains. Host-range analysis across 36 isolates revealed partially overlapping infectivity profiles, with strongest activity against phylogroup 2 (

Enterococcus Phage Abitsa: Biological Characterization, Antibiofilm Activity and Evolutionary Insights into the Genus Efquatrovirus.

Enterococcus faecalis is an important opportunistic pathogen associated with persistent oral infections, biofilm formation, and antimicrobial tolerance, which makes phages targeting this species of both therapeutic and evolutionary interest. Here, we describe the isolation and characterization of Abitsa, a novel lytic Enterococcus phage representing a new species within the genus Efquatrovirus. Abitsa formed small plaques and displayed siphovirus-like morphology. It showed an optimal multiplicity of infection of 0.01, a short latent period of 10 min and a burst size of 28 ± 5 virions per infec

A new Kayvirus vB_SauM-MUHD-1 combats Methicillin-resistant Staphylococcus aureus wound infections.

Methicillin-resistant Staphylococcus aureus (MRSA), a leading cause of chronic and post-surgical wound infections, is a hard-to-treat pathogen. In this study, we isolated and characterized a lytic MRSA bacteriophage, vB_SauM-MUHD-1, and evaluated its therapeutic potential in a murine wound infection model. Among 107 clinical wound samples, MRSA was reported in 48.6% of cases. Phage vB_SauM-MUHD-1, isolated from sewage, demonstrated lytic activity against 70.6% of the tested MRSA isolates. The phage exhibited efficient replication kinetics and remained stable under physiologically relevant cond

SLPW: A Virulent Bacteriophage Targeting Methicillin-Resistant Staphylococcus aureus In vitro and In vivo.

Staphylococcus aureus (S. aureus) is a Gram-positive pathogen causing a variety of infections in humans and animals. Extensive use of antibiotics has led to the emergence of methicillin-resistant S. aureus (MRSA). As an alternative antibacterial agent against drug-resistant S. aureus, a lytic phage, designated SLPW, was isolated from fecal sewage in a pig farm. The SLPW was morphologically classified under Podoviridae and contains a double-stranded DNA genome. The genome of SLPW was 17,861 bp (29.35% G+C) containing 20 open reading frames and lacked regions encoding lysogeny-related integrase

Isolation of specialized transducing bacteriophages for gluconate 6-phosphate dehydrogenase (gnd) of Escherichia coli.

Specialized transducing phages for gluconate 6-phosphate dehydrogenase (gnd), a constitutive enzyme in Escherichia coli, have been isolated using a method previously described for other genes. The gnd-his region, carried on an F' episome, was first transposed to tonB. Rare phages carrying gnd were selected, by transduction, from phi80 lysogens of these strains; one phage also carried his (phi80gndhis). From the transductants, high-frequency transducing lysates were obtained; low multiplicity of infection then yielded defective lysogens. tonB deletion analysis of the phi80dgndhis lysogen shows

Expression of phage transcription in P2 lysogens infected with helper-dependent coliphage P4.

The DNA of helper-dependent coliphage P4 and the DNA of its helper-P2-show no detectable sequence homology as measured by DNA.DNA hybridization. The lack of cross-hybridization permits direct analysis of P4 as well as of P2 transcription in P4-infected P2 lysogens by RNA.DNA hybridization. P4-transactivated P2 transcription can be detected around 20 min after P4 infection of the P2 lysogen and the rate (per infected cell) of that transcription becomes equal to that of the P4 transcription at the end of the latent period of P4. Furthermore, P4 transcription appears to be stimulated by the prese

Genomic and functional characterization of a lytic Klebsiella phage UHKP with antibiofilm activity.

Klebsiella pneumoniae is an opportunistic pathogen causing severe hospital-acquired infections, and it rapidly acquires multidrug resistance. Its robust biofilm formation further complicates treatment and drives interest in phage therapy. A phage UHKP was isolated from hospital sewage using an MDR K. pneumoniae strain (KP-03). UHKP formed clear plaques and having phage titer 2.3 × 109 PFU/mL. Host-range testing on 19 clinical isolates showed a narrow spectrum. Only four MDR strains (KP-03, KP-05, KP-08, KP-11) and one K-17 serotype were lysed, with no activity on other strains or species. One-

Two classes of Mu lig mutants: the thermosensitives for integration and replication and the hyperproducers for ligase.

We have previously shown that Mu restores an active DNA replication at non-permissive temperature in E. coli K12 ligts7 strains. In this paper we describe two new types of phage mutants for the Mu lig function. The Mu ligts mutants are conditional lethals defective for both integration and replication of DNA, unable to 'complement' the bacterial lig mutation; the map between B and lys. The mutants of the other type, on the other hand, are able to restore to a maximum level the activity impaired by the ligts7 mutation in the host. We suggest the hypothesis that the product of Mu lig gene could

Isolation, characterization and genomic analysis of three lytic bacteriophages (PBM) targeting Avian Pathogenic Escherichiacoli.

Avian pathogenic Escherichia coli (APEC) cause significant economic losses in the poultry industry, further exacerbated by the growing number of multi-drug resistance pathogens that make treating infections more difficult. To combat this, the study presents two novel Schitoviridae phages (PBM-2 and PBM-3) and a novel Straboviridae phage (PBM-1) that show significant and highly specific lytic activity against APEC strains. Moreover, the phages show significant lytic activity at MOI as low as 0.1 and show substantial thermal (4-45 °C) and acidic tolerance (pH 3-7). The PBM phages also show suita

Biocoatings with Enhanced Bacterial Viability via Coagulant Dipping and Wet Sintering by Immersion.

Biocoatings are typically colloidal polymer films confining metabolically active, nongrowing bacteria. Depending on the species of confined bacteria, biocoatings find applications in wastewater treatment, biofuel production, carbon fixation, environmental remediation, biosensing, and more. However, the successful use of biocoatings faces numerous challenges, including a low permeability to reactants and metabolized products, osmotic stress on bacteria during drying of the coatings, and cell dehydration leading to bacteria death. Here, we address these challenges through two interlinked process

Isolation and Characterization of the New Lytic Bacteriophage KEC4 Against Escherichia coli MDR Strains.

Being first applied for the treatment of infectious diseases of the gut at the start of the 20th century, bacteriophages are again now considered as alternative antimicrobial tools for targeting antibiotic-resistant enterobacteria. Here, we report the new bacteriophage Escherichia phage KEC4 isolated from the Kukshum River (Chuvash Republic, Russia), lysing Escherichia coli and belonging to the Septuagintavirus genus. The genome consists of 145,125 bp with a GC content of 41.3% and contains 6 tRNA and 303 protein-coding sequences. Among them, only 72 encode proteins with known functions, while
