Aryl Hydrocarbon Receptor Activation by TCDD Modulates Expression of Extracellular Matrix Remodeling Genes during Experimental Liver Fibrosis.

The aryl hydrocarbon receptor (AhR) is a soluble, ligand-activated transcription factor that mediates the toxicity of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). Increasing evidence implicates the AhR in regulating extracellular matrix (ECM) homeostasis. We recently reported that TCDD increased necroinflammation and myofibroblast activation during liver injury elicited by carbon tetrachloride (CCl4). However, TCDD did not increase collagen deposition or exacerbate fibrosis in CCl4-treated mice, which raises the possibility that TCDD may enhance ECM turnover. The goal of this study was to deter

The use of nano-quercetin to arrest mitochondrial damage and MMP-9 upregulation during prevention of gastric inflammation induced by ethanol in rat.

Gastric ulcer is a multifaceted process that involves reactive oxygen species (ROS) generation, extracellular matrix degradation and mitochondrial damage. Mitochondria play a crucial role for homeostasis of ROS and cell survival. In our study, we investigated the efficacy and mechanism of polymeric nanocapsuled quercetin (NQC) over the free quercetin (QC) molecule in prevention of ethanol-induced gastric ulcer in rat. NQC possessed significantly higher efficacy (~20 fold) than free QC while preventing gastric ulcers. Our data show that prior administration of NQC and/or QC significantly blocke

Enhanced T-cell expression of RANK ligand in acute coronary syndrome: possible role in plaque destabilization.

OBJECTIVE: Based on its role in inflammation and matrix degradation, we hypothesized a role for osteoprotegerin (OPG), RANK, and RANK ligand (RANKL) in coronary artery disease. METHODS AND RESULTS: We examined the expression of various members of the OPG/RANKL/RANK axis in patients with stable and unstable angina and in the atherosclerotic lesions of apolipoprotein E-deficient (apoE(-/-)) mice. Our findings were: (1) Serum levels of OPG were raised in patients with unstable angina (n=40), but not in those with stable angina (n=40), comparing controls (n=20); (2) mRNA levels of RANKL were incr

Ginsenoside Rb1 attenuates angiotensin II-induced abdominal aortic aneurysm through inactivation of the JNK and p38 signaling pathways.

BACKGROUND: Abdominal aortic aneurysm (AAA), a life-threatening vascular disease, accounts for approximately 10% of the morbidity in people over 65 years old. No satisfactory approach is available to treat AAA. Ginsenosides Rb1 and Rg1 are primary ingredients of Panax notoginseng for the treatment of cardiovascular diseases, but their impact on AAA is unknown. METHODS AND RESULTS: An AAA model was established using an Ang II infusion in ApoE(-/-) mice. After continuous stimulation of Ang II for 28 days, suprarenal aortic aneurysms developed in 77% mice and 12% mice died suddenly due to AAA ru

Role of glutamine in promoting the proliferation and chondrogenic differentiation of ATDC5 cells: based on transcriptomic analysis.

BACKGROUND: Osteoarthritis (OA) is a commonly occurring degenerative joint disorder. Presently, primary clinical interventions, including nonsteroidal anti-inflammatory drugs (NSAIDs), primarily provide symptomatic alleviation. Glutamine (Gln) has been shown to have protective effects on chondrocytes, which may postpone OA progression. Nevertheless, its effectiveness in the ATDC5 chondrogenic cell line has yet to be explored. OBJECTIVE: The objective of this research was to explore the ability of Gln to increase the proliferation and chondrogenic differentiation of ATDC5 cells and to clarify t

Alterations in the molecular determinants of bladder compliance at hydrostatic pressures less than 40 cm. H2O.

PURPOSE: Bladder outlet obstruction with intravesical pressures exceeding 40 cm. H2O often results in irreversible renal damage. Bladder outlet obstruction also results in alterations in bladder physiology, including wall thickening, reduced compliance and decreased capacity. If unchecked these changes may lead to the subsequent need for bladder augmentation. From a biomechanical standpoint, compliance is primarily related to extracellular matrix deposition, which in turn is dependent on the balanced activity of proteolytic enzymes (that is matrix metalloproteinases [MMPs]) and their endogenou

Role of inflammatory cytokines in tissue remodeling and disease progression.

BACKGROUND: The persistence of the activation of pro-inflammatory cytokines, which facilitates the pathological tissue remodeling by disrupting the extracellular matrix (ECM), has been identified as a risk factor for chronic inflammation. A disproportion in the proinflammatory cytokines and anti-inflammatory cytokines enhances tissue destruction and disease etiology. AIM: This study aimed to assess the role of inflammatory cytokines and tissue remodeling markers in disease progression and determine the association of these factors with systemic inflammation and structural tissue changes. Metalloproteinases in Restorative Dentistry: An In Silico Study toward an Ideal Animal Model.

In dentistry, various animal models are used to evaluate adhesive systems, dental caries and periodontal diseases. Metalloproteinases (MMPs) are enzymes that degrade collagen in the dentin matrix and are categorized in over 20 different classes. Collagenases and gelatinases are intrinsic constituents of the human dentin organic matrix fibrillar network and are the most abundant MMPs in this tissue. Understanding such enzymes' action on dentin is important in the development of approaches that could reduce dentin degradation and provide restorative procedures with extended longevity. This in si

Immunohistochemical expression of matrix metalloproteinase 13 in chronic periodontitis.

The extracellular matrix is a complex integrated system responsible for the physiologic properties of connective tissue. Collagen is the major extracellular component that is altered in pathologic conditions, mainly periodontitis. The destruction involves proteolytic enzymes, primarily matrix metalloproteinases (MMPs), which play a key role in mediating and regulating the connective tissue destruction in periodontitis. The study group included 40 patients with clinically diagnosed chronic periodontitis. The control group included 20 patients with clinically normal gingiva covering impacted thi

MMP2/TIMP2 ratio as a measurable indicator for differentiating diffuse adenomyosis, ovarian endometriosis, and their co-existence.

Adenomyosis and endometriosis, two common estrogen-dependent benign gynecological disorders usually present with overlapping symptoms, such as pain and infertility. These two conditions often co-exist and are associated with poor reproductive outcome. Both adenomyosis and endometriosis are associated with extracellular matrix (ECM) remodeling, regulated by matrix metalloproteinases (MMPs) and their tissue inhibitors (TIMPs). MMP2/TIMP2 balance is known to be crucial for endometrial ECM homeostasis. This study, for the first time, compares MMP2 and TIMP2 activity in adenomyosis, endometriosis,

Inhibition of matrix metalloproteinases during chronic allograft nephropathy in rats.

BACKGROUND: Chronic allograft nephropathy (CAN) belongs to the major causes of long-term kidney allograft failure. One of the histologic hallmarks of CAN is interstitial fibrosis, influenced by matrix metalloproteinases (MMPs) that are controlling extracellular matrix (ECM) degradation. Whether MMPs affect the development and progression of CAN is not clear so far. To analyze the role of MMPs in CAN, we investigated the effects of an early and a late application of BAY 12-9566, an inhibitor of MMP-2, -3, and -9 on the development and progression of CAN in a rat kidney-transplantation model. Genetic variation associated with preterm birth in African-American women.

BACKGROUND: Preterm birth is considered a multifactorial condition; however, emerging evidence suggests that genetic variation among individuals may have an important role. Prior studies have suggested that single-nucleotide polymorphisms associated with genes related to the immune system, and particularly the maternal inflammatory response, may be associated with an increased risk of preterm delivery. OBJECTIVE: The objective of the study was to identify single-nucleotide polymorphisms associated with spontaneous preterm birth <37 weeks within a cohort of African-American women. STUDY DESIG

Expression profile of microRNAs and mRNAs in human placentas from pregnancies complicated by preeclampsia and preterm labor.

MicroRNAs (miRNAs) have emerged as key regulators of gene expression stability implicated in cell proliferation, apoptosis, and development, whereas their altered expression has been associated with various pathological disorders. The objective of this study was to assess the expression profile of miRNAs and their predicted target genes in placentas from patients with preeclampsia (PC) and preterm (PT) labor as compared to normal term (NT) pregnancies. Using microarray profiling of 820 miRNAs and 18,630 mRNA transcripts, the analysis indicated that 283 of these miRNAs and 9119 mRNAs were expre

Matrix metalloproteinases-2 and -9 activities in bovine follicular fluid of different-sized follicles: relationship to intra-follicular inhibin and steroid concentrations.

Matrix metalloproteinases (MMPs) play very important roles in extracellular matrix (ECM) remodeling during ovarian follicular development, ovulation and atresia. The aim of the present study was to determine the content of gelatinases in follicular fluid in various sized bovine follicles. Bovine ovaries were collected from local slaughterhouse and follicular fluid from follicles of 2 to over 25 mm in diameter was collected. Gelatinase activity within the follicular fluid was analyzed by gelatin zymography. The concentration of inhibin in the follicular fluid was also measured by immunoblot ana

Matrix Protective and Anti-Inflammatory In Vitro Potential of Adipose Mesenchymal Stromal Cell-Derived EVs Coupled With Platelet-Derived Fibrin Gel.

BACKGROUND: The limited intrinsic repair capacity of articular cartilage impairs tissue regeneration in osteoarthritis (OA). Platelet-rich plasma (PRP) fibrin gels offer a viable autologous scaffold for cartilage repair but often promote fibrous tissue rather than native hyaline structures. Mesenchymal stromal cell-derived extracellular vesicles (MSC-EVs) can overcome this limitation by actively promoting hyaline cartilage formation and suppressing matrix degradation. This study describes and validates a protocol for embedding adipose-derived MSC (ASC)-EVs into PRP gels (PRP-EVs) and evaluates
