Chemically Defined Organoid Culture System for Cholangiocyte Differentiation.

Cholangiocyte organoids provide a powerful platform for applications ranging from in vitro modeling to tissue engineering for regenerative medicine. However, their expansion and differentiation are typically conducted in animal-derived hydrogels, which impede the full maturation of organoids into functional cholangiocytes. In addition, these hydrogels are poorly defined and complex, limiting the clinical applicability of organoids. In this study, a novel medium composition combined with synthetic polyisocyanopeptide (PIC) hydrogels to enhance the maturation of intrahepatic cholangiocyte organo

Eradicating the mediators of neuronal death with a fine-tooth comb.

Ischemic tolerance is an evolutionarily conserved form of cerebral plasticity in which a brief period of cerebral ischemia (called ischemic preconditioning) confers transient tolerance to a subsequent ischemic challenge in the brain. Polycomb group proteins are gene-silencing factors that are abundant and widely distributed during embryogenesis and are essential to epigenetic cellular memory, pluripotency, and stem cell self-renewal. New insight into the molecular mechanisms underlying ischemic tolerance is highlighted by the finding that ischemic preconditioning activates polycomb proteins in

The Aiolos transcription factor is up-regulated in chronic lymphocytic leukemia.

The Aiolos transcription factor, member of the Ikaros family of zinc finger proteins, plays an important role in the control of mature B lymphocyte differentiation and proliferation, and its function appears to be modulated through alternative splicing. To assess Aiolos isoform role in humans' pathologies, we studied Aiolos variant distribution and expression in mature B lymphoproliferative disorders (chronic lymphocytic leukemia [CLL] and other B-cell lymphomas). We demonstrated that more than 80% of expressed Aiolos in normal as well as in malignant B cells is of the hAio1 type, and we showe

Defective Guanine Nucleotide Exchange in the Elongation Factor-like 1 (EFL1) GTPase by Mutations in the Shwachman-Diamond Syndrome Protein.

Ribosome biogenesis is orchestrated by the action of several accessory factors that provide time and directionality to the process. One such accessory factor is the GTPase EFL1 involved in the cytoplasmic maturation of the ribosomal 60S subunit. EFL1 and SBDS, the protein mutated in the Shwachman-Diamond syndrome (SBDS), release the anti-association factor eIF6 from the surface of the ribosomal subunit 60S. Here we report a kinetic analysis of fluorescent guanine nucleotides binding to EFL1 alone and in the presence of SBDS using fluorescence stopped-flow spectroscopy. Binding kinetics of EFL1

Characterization and functional evaluation of goat PDX1 regulatory modules through comparative analysis of conserved interspecies homologs.

PDX1 is a crucial transcription factor in pancreas development and mature β-cell function. However, the regulation of PDX1 expression in larger animals mirroring human pancreas morphogenesis and endocrine maturation remains poorly understood. Therefore, we conducted a comparative analysis to characterize regulatory regions of goat PDX1 gene and assessed their transcriptional activity by transient transfection of several transgenic EGFP constructs in β- and non-β cell lines. We recognized several highly conserved regions encompassing the promoter and cis-regulatory elements (Area I-IV) at 5' fl

Stiffness-Activated Stellate Cells Drive Pancreatic Cancer Liver Colonization via GMFG-TNS4 Signaling.

Liver metastasis remains the primary cause of death in pancreatic cancer. Collagen deposition by activated hepatic stellate cells (HSCs) generates a stiff fibrotic niche that favors metastatic colonization, yet the underlying mechanisms remain incompletely understood. Using stiffness-tunable hydrogels, it is shown that elevated substrate stiffness activates HSCs and establishes a self-reinforcing loop of matrix stiffening. Mechanistically, stiffness triggers Piezo1-mediated Ca2 + influx, induces endoplasmic reticulum stress (ERS), and activates the IRE1α-XBP1 pathway to upregulate glia maturat

Single-Cell Immune Profiling Reveals Neutrophils Promote Myasthenia Gravis Exacerbation Through BAFF Secretion.

Neutrophils play a critical role in the pathogenesis of autoimmune diseases, including myasthenia gravis (MG), but their specific function in MG exacerbations remains unclear. This study utilizes single-cell RNA sequencing (scRNA-seq) of bone marrow and peripheral blood from MG patients during acute exacerbations, combined with experimental autoimmune myasthenia gravis (EAMG) mouse models and clinical cohort analyses, to investigate the potential involvement of a neutrophil-B-cell activating factor (BAFF) -plasma cell axis. The results reveal that, during MG acute exacerbation, bone-marrow neu

Biological and Structural Analyses of New Potent Allosteric Inhibitors of HIV-1 Integrase.

HIV-1 integrase-LEDGF allosteric inhibitors (INLAIs) share the binding site on the viral protein with the host factor LEDGF/p75. These small molecules act as molecular glues promoting hyper-multimerization of HIV-1 IN protein to severely perturb maturation of viral particles. Herein, we describe a new series of INLAIs based on a benzene scaffold that display antiviral activity in the single digit nanomolar range. Akin to other compounds of this class, the INLAIs predominantly inhibit the late stages of HIV-1 replication. A series of high-resolution crystal structures revealed how these small m

The clinical, morphologic, and molecular changes in the ileum associated with early postnatal dexamethasone administration: from the baby's bowel to the researcher's bench.

Focal small bowel perforation (FSBP) is a life-threatening event that predominantly affects extremely low birth weight (ELBW) infants. Histopathology from surgical specimens of ileum with FSBP shows a healthy mucosa overlying a thinned muscularis with segmental degeneration. Clinical data strongly support an association between early postnatal administration of dexamethasone (EPD) and FSBP. Additional risk factors, including gestational age, administration of prophylactic indomethacin, and severity of illness, may be synergistic with EPD for the pathogenesis of perforations. Animal models of d

Growth-rate coordination across the width of a leaf preserves its flatness.

The growth and division of cells in plant leaves is highly dynamic in time and space, even though cells cannot move relative to their neighbors. Thus, organ shape must emerge from carefully coordinated growth, especially in leaves that remain relatively flat as they grow. Here we explored the phenotype of the jagged and wavy (jaw-D) mutant in Arabidopsis thaliana, in which the leaves do not remain flat. It has previously been shown that the jaw-D mutant phenotype is caused by the overexpression of miR319, which represses TCP transcription factors, thus delaying maturation of the leaf. We analy

Canine oocyte maturation in culture: significance of estrogen and EGF receptor gene expression in cumulus cells.

We examined the role of cumulus cells regarding in vitro maturation of canine oocytes, and investigated estrogen and epidermal growth factor (EGF) receptor gene expression and action on nuclear maturation. Canine cumulus-oocyte complexes (COC) were collected from anestrous and diestrous bitches; only COC with vitelline diameter >100 microm were used. In Experiment 1, expression of estrogen receptor (ER) alpha, ERbeta and EGF-receptor (EGF-R) were determined by reverse transcription-polymerase chain reaction (RT-PCR), using mRNA from the oocyte or cumulus cell. Transcripts for the ERbeta and EG

DDR1 Modulates Cytoskeletal Remodeling and Podosome Formation in Renal Fibroblasts.

Discoidin domain receptor 1 (DDR1) has been implicated in fibrotic progression in multiple organs, including the kidney. However, its role in regulating cytoskeletal organization and matrix remodeling in renal fibroblasts remains unclear. Here, we investigated how DDR1 expression is regulated by profibrotic stimulation and extracellular matrix stiffness, and how DDR1 influences cytoskeletal organization and collagen remodeling. Single-cell RNA sequencing of murine kidneys subjected to unilateral ureteral obstruction (UUO) revealed enrichment of Ddr1 expression in transitional fibroblast popula

Znf687 recruits Brd4-Smrt complex to regulate gfi1aa during neutrophil development.

Neutrophils are key component of the innate immune system in vertebrates. Diverse transcription factors and cofactors act in a well-coordinated manner to ensure proper neutrophil development. Dysregulation of the transcriptional program triggering neutrophil differentiation is associated with various human hematologic disorders such as neutropenia, neutrophilia, and leukemia. In the current study we show the zinc finger protein Znf687 is a lineage-preferential transcription factor, whose deficiency leads to an impaired neutrophil development in zebrafish. Mechanistically, Znf687 functions as a

Interleukin-1β: Friend or foe for gastrointestinal cancers.

Gastrointestinal (GI) cancer is a malignancy arising in the digestive system and accounts for approximately a third of increasing global cancer-related mortality, especially in the colorectum, esophagus, stomach, and liver. Interleukin-1β (IL-1β) is a leukocytic pyrogen recognized as a tumor progression-related cytokine. IL-1β secretion and maturation in inflammatory responses could be regulated by nuclear factor-kappaB-dependent expression of NLR family pyrin domain containing 3, inflammasome formation, and activation of IL-1 converting enzyme. Several studies have documented the pro-tumorige

TGF-beta inhibits proliferation of and promotes differentiation of human promonocytic leukemia cells.

Transforming growth factor-beta 1 (TGF-beta 1) has been implicated in a variety of responses associated with wound healing and inflammation. Thus, TGF-beta 1 enhances production of several extracellular matrix proteins both in vitro and in vivo, is chemotactic for monocytes, and alters the functioning of lymphocytes. We have examined the ability of TGF-beta 1 to affect the behavior of human THP-1 promonocytic leukemia cells, a cell line with the capacity to differentiate into macrophage-like cells. TGF-beta 1 reduces the growth rate of these cells, induces morphologic changes, and promotes adh

Chitosan hydrogel loaded with mesenchymal stem cell-derived exosomes significantly accelerated wound closure and enhanced tissue regeneration in a full thickness porcine skin defect model.

There is an urgent need for more effective wound healing therapies, particularly wound dressings that not only promote wound closure and reduce scar formation but also retain antibacterial properties. Chitosan, a natural copolymer with inherent hemostatic and antibacterial properties, and mesenchymal stem cell derived exosomes, which promote rapid re-epithelialization, collagen maturation, and scar reduction, were integrated into a single biofunctional dressing. This study evaluated the therapeutic efficacy and underlying mechanisms of this combined approach using a porcine full thickness skin

Cardiac Progenitor Cell Recruitment Drives Fetal Cardiac Regeneration by Enhanced Angiogenesis.

BACKGROUND: In contrast to adults, the fetal response to myocardial infarction (MI) is regenerative, requiring the recruitment of cardiac progenitor cells to replace infarcted myocardium. Macrophage contribution to tissue repair depends on their phenotype: M1 are proinflammatory and initiate angiogenesis; M2a are profibrotic and contribute to blood vessels maturation; and M2c are proremodeling and proangiogenesis. The goal of the present study was to expand on this work by examining cardiac progenitor cells recruitment, and the role of macrophages in promoting angiogenesis and cardiac regenera

Precise genetic control of ATOH1 enhances maturation of regenerated hair cells in the mature mouse utricle.

Vestibular hair cells are mechanoreceptors critical for detecting head position and motion. In mammals, hair cell loss causes vestibular dysfunction as spontaneous regeneration is nearly absent. Constitutive expression of exogenous ATOH1, a hair cell transcription factor, increases hair cell regeneration, however, these cells fail to fully mature. Here, we profiled mouse utricles at 14 time points, and defined transcriptomes of developing and mature vestibular hair cells. To mimic native hair cells which downregulate endogenous ATOH1 as they mature, we engineered viral vectors carrying the sup
