Age-related learning deficits in transgenic mice expressing the 751-amino acid isoform of human beta-amyloid precursor protein.

The beta-amyloid precursor protein (beta-APP), from which the beta-A4 peptide is derived, is considered to be central to the pathogenesis of Alzheimer disease (AD). Transgenic mice expressing the 751-amino acid isoform of human beta-APP (beta-APP751) have been shown to develop early AD-like histopathology with diffuse deposits of beta-A4 and aberrant tau protein expression in the brain, particularly in the hippocampus, cortex, and amygdala. We now report that beta-APP751 transgenic mice exhibit age-dependent deficits in spatial learning in a water-maze task and in spontaneous alternation in a

Stem cell transplantation for enhancement of learning and memory in adult neurocognitive disorders.

The role of adult hippocampal neurogenesis in learning and memory is still incompletely understood. Ablation of neurogenesis with different methods produced equivocal results with respect to working memory in Morris water maze and radial arm maze experiments. Therefore, it is remarkable that in the past few years several investigators have found a positive impact on working memory after adding stem or progenitor cells to the hippocampus in various disease models. The literature on stem cell transplantation for adult neurocognitive disorders is reviewed in this article and attempted to be recon

Emotional and learning behaviour in mice overexpressing heat shock protein 70.

The effects of inducible heat shock protein 70 (HSP70) on emotional and learning behaviour as well as hippocampal long-term potentiation was investigated in transgenic HSP70 overexpressing mice. In active two-way avoidance learning (shuttle box) as well as spatial 8-arm radial maze learning, the HSP70 overexpressing mice showed diminished learning performance. In several tests there was no indication of differences in anxiety behaviour between transgenic mice and wild-type mice. This suggests that impairment in learning behaviour is unrelated to the learning task and motivational aspects of be

Cognitive effects of methamphetamine and amphetamine withdrawal in rodents: a systematic review.

BACKGROUND: Methamphetamine (METH) and amphetamine (AMPH) are widely misused psychostimulants that induce enduring alterations in brain function and behavior, including cognitive impairment. To date, literature in this area has not been sufficiently reviewed and summarized to account for how methodological variables, such as dosage and administration contingency, influence cognitive outcomes. OBJECTIVE: This systematic review evaluates the effects of METH and AMPH withdrawal on rodent cognition, with particular attention to how outcomes are influenced by dose, sex, strain, and withdrawal dura

Effects of liposome-encapsulated hemoglobin on learning ability in tokai high-avoider rat after total brain ischemia and reperfusion.

Liposome-encapsulated hemoglobin with low O2 -affinity (l-LEH) was shown to be protective in focal brain ischemia and reperfusion (I/R) in rats and primates. We tested l-LEH in the transient whole brain ischemia in the Tokai high-avoider rat (THA), which has been selected, mated, and bred over 77 generations for a high and consistent learning ability determined by the Sidman avoidance test (SAT). Young/naïve (before SAT) and adult/parent (after SAT) THA rats underwent acute and complete four-vessel occlusion in the chest for 3 or 5 min, administration of 2 mL/kg of l-LEH, saline, or homologous

Effects of inhibiting neonatal methamphetamine-induced corticosterone release in rats by adrenal autotransplantation on later learning, memory, and plasma corticosterone levels.

RATIONALE: Neonatal rat methamphetamine (MA) exposure has been shown to cause long-term behavioral impairments similar to some of those observed following neonatal stress. The mechanism by which MA induces impairments is unknown but may be related to early increases in corticosterone release. We previously developed a method to attenuate MA-induced corticosterone release using adrenal autotransplantation (ADXA) in neonatal rats. This exposure period corresponds to the second-half of human pregnancy. OBJECTIVE: To determine whether inhibition of neonatal MA-induced increases in corticosterone

Visual spatial learning outcomes for clinical trials in neurofibromatosis type 1.

Cognitive problems are common in children with neurofibromatosis type 1, representing a significant source of lifelong morbidity. Assessment of cognitive function has been challenging in the setting of clinical trials. Spatial learning deficits may be an important target for cognitive interventions. We leveraged a large, international cognitive study in affected children with NF1 treated with lovastatin to assess spatial learning using the "Arena Maze", a portable, computerized task that allows for retesting in the same environment. As with the parent study, spatial learning assessed with this

Prenatal Iron Deficiency in Guinea Pigs Increases Locomotor Activity but Does Not Influence Learning and Memory.

The objective of the current study was to determine whether prenatal iron deficiency induced during gestation in guinea pigs affected locomotor activity and learning and memory processes in the progeny. Dams were fed either iron-deficient anemic or iron-sufficient diets throughout gestation and lactation. After weaning, all pups were fed an iron-sufficient diet. On postnatal day 24 and 40, the pups' locomotor activity was observed within an open-field test, and from postnatal day 25 to 40, their learning and memory processes were assessed within a Morris Water Maze. The behavioural and cogniti

Propentofylline improves learning and memory deficits in rats induced by beta-amyloid protein-(1-40).

We have reported that continuous infusion of beta-amyloid protein-(1-40) into the cerebral ventricle produces learning and memory deficits in rats. Propentofylline has potent stimulatory effects on nerve growth factor (NGF) synthesis/secretion in mouse astrocytes in vitro and increases cerebral NGF content in aged rats. In the present study, we examined the effects of propentofylline on learning and memory deficits in beta-amyloid protein-infused rats. The rats were given propentofylline orally once a day throughout the period of behavioral examination. In the beta-amyloid protein-infused rats

Sulfate-reducing bacteria impairs working memory in mice.

The ability of gut microbes to bi-directionally communicate with the brain and vice versa form the basis of the gut microbiome-central nervous system axis. It has been shown that inoculation with pathogenic gut bacteria alters the behavior of mice; however, it is not known whether or not non-pathogenic resident microbes have similar effects. In this study, we tested the hypothesis that the administration of sulfate-reducing bacteria (SRB), a specific group of resident gut bacteria that generate hydrogen sulfide (H2S), impair learning and memory performance in mice tested in an 8-arm radial maz

Tetrahydroaminoacridine attenuates dizocilpine-induced behavioural changes.

The acetylcholinesterase inhibitor tetrahydroaminoacridine (THA) is known to interact with neurotransmitter systems other than the cholinergic, e.g. several studies have shown interaction of THA with the N-methyl-D-aspartate (NMDA) receptor subtype of the glutamatergic system. We therefore investigated whether behavioral changes in rats, caused by the non-competitive NMDA-antagonist dizocilpine, were altered by THA. Spontaneous locomotion was measured in an open field and learning behaviour was evaluated in a spatial learning task in the 8-arm radial maze. Hyperactivity in the open field cause

Inhibition of oestrogen biosynthesis induces mild anxiety in C57BL/6J ovariectomized female mice.

OBJECTIVE Letrozole, a next-generation aromatase inhibitor, has become a favored drug for the treatment of breast cancer in postmenopausal women. Although letrozole is generally well tolerated, its adverse effects on the central nervous system have been reported. The present study aimed to assess the behavioural outcomes of letrozole administration in mice to determine its side effects. METHODS C57BL/6J female ovariectomized mice received administration of letrozole (2.5 mg/kg per day) or vehicle by gavage for 3 weeks. Behavioural tasks were used to assess anxiety, depression, as well as learn

Nesfatin-1, a potent anorexic agent, decreases exploration and induces anxiety-like behavior in rats without altering learning or memory.

The anorectic neuropeptide nesfatin-1 has recently been characterized as a potential mood regulator, but the accurate effect of nesfatin-1 on anxiety and learning and memory behavior and the possible mechanisms remains unknown. In the present study, to test the hypothesis that nesfatin-1 might affect the anxiety-like and learning and memory behaviors in rats via ERK/CREB/BDNF pathway, nesfatin-1 was administered intraperitoneally to rats with the doses (10, 20, 40μg/kg), and the behavioral performance was tested using the open field task, the Morris water maze (MWM), and the Y maze. Neural functional and morphological consequences of retinal degeneration in C3H Lurcher mutant and wild type mice.

Lurcher mutant mice represent a model of olivocerebellar degeneration associated with the total functional elimination of the cerebellar cortex. The affected animals suffer from cerebellar ataxia and worsening of cognitive functions. Healthy littermates of Lurchers-wild type mice serve as controls. Except mentioned patterns some animals derived from the C3H strain exhibit signs of a hereditary retinal degeneration. The impact of the retinal degeneration on visuospatial abilities and on the neuronal morphology in visual projection of both C3H Lurcher mutant and wild type mice has been studied i

Spatial delayed nonmatching-to-sample performances in long-living Ames dwarf mice.

Ames dwarf mice have an extended lifespan by comparison with normal mice. Behavioral testing has revealed that sometimes Ames dwarf mice also evince superior performances relative to normal mice, but in other cases they do not. In this experiment, Ames dwarf and normal mice were compared on a T-maze test and on a delayed nonmatching-to-sample variant of a T-maze test. On the simple T-maze, Ames dwarf and normal mice committed comparable numbers of errors. On the nonmatching-to-sample task, normal mice mastered the discrimination by the end of the experiment while Ames dwarf mice did not. The a

Impaired learning and memory and altered hippocampal neurodevelopment resulting from interleukin-2 gene deletion.

Interleukin-2 (IL-2), the protypical T cell growth factor and immunoregulatory cytokine produced by lymphocytes, has been implicated as a brain neurotrophic factor and neuromodulator. The consequences of the absence of endogenous IL-2 on brain development and function were unknown. Brain IL-2 receptors are enriched in the hippocampal formation, an area critical for the acquisition and consolidation of spatial learning and memory. Thus, we tested the hypothesis that mice lacking IL-2 would exhibit alterations in hippocampal-dependent learning and neurodevelopment. Compared with C57BL/6-IL-2+/+

Gene × environment interaction between latrophilin-3 (Lphn3 or Adgrl3) and developmental permethrin exposure in Sprague Dawley rats.

Attention deficit hyperactivity disorder (ADHD) occurs in 9.8% of U.S. children and has a large hereditary component arising from multiple gene variants. One of these is Latrophiln-3 (LPHN-3). Using CRISPR/Cas9 we deleted exon 3 in Sprague Dawley rats to create a global Lphn3 knockout (gKO). The gKO rats are hyperactive, startle hyper-reactive, impulsive, and have impaired working, spatial, and egocentric learning and memory. Permethrin (PRM) is a widely used pyrethroid insecticide. Acute exposure to PRM alters acoustic startle but its long-term effects from developmental exposure are unknown.

Profound retrograde but absence of anterograde amnesia for cued place learning in rats with hippocampal lesions.

Previous studies in our lab have shown that slight modifications in the spatial reference memory procedure can overcome the deficit in spatial learning typically observed in rats with hippocampal damage. However, it is unknown if memory acquired under such training circumstances is spared after hippocampal lesions. With this aim a four-arm plus-shaped maze and a spatial reference memory paradigm were used, in which the goal arm was doubly marked: by an intramaze cue (a piece of sandpaper positioned on the floor of the arm) and by the extramaze constellation of stimuli around the maze. Effects of prenatal alcohol exposure on the hippocampus: spatial behavior, electrophysiology, and neuroanatomy.

Prenatal exposure to alcohol can result in fetal alcohol syndrome (FAS), characterized by growth retardation, facial dysmorphologies, and a host of neurobehavioral impairments. Neurobehavioral effects in FAS, and in alcohol-related neurodevelopmental disorder, include poor learning and memory, attentional deficits, and motor dysfunction. Many of these behavioral deficits can be modeled in rodents. This paper reviews the literature suggesting that many fetal alcohol effects result, at least in part, from teratogenic effects of alcohol on the hippocampus. Neurobehavioral studies show that animal
