Nitric oxide effects on human erythrocytes structural and functional properties--an in vitro study.

NO is present in the blood at 10(-7) M under physiological conditions, but at concentrations higher than 10(-6) M during inflammatory disease states. The aim of this study was to characterize what are the effects of these different NO concentrations on erythrocyte structural and functional properties. Blood was collected from eleven healthy men and incubated with SpermineNONOate in order to expose it during incubation time to NO concentrations between 10(-7) M and 10(-3) M. We measured erythrocyte aggregation and deformability, membrane lipid peroxidation and fluidity, p50, hemoglobin, oxyhemo

Melatonin-tuned mRNA-loaded lipid nanoparticles enhance NRF2-mediated mesenchymal stem cell therapy for pulmonary fibrosis.

Pulmonary fibrosis is characterized by persistent oxidative stress and excessive extracellular matrix deposition, yet effective therapeutic strategies remain limited. Here, we report a nanoengineering strategy that integrates melatonin and Nrf2 mRNA within lipid nanoparticle (LNPs) to functionally engineer mesenchymal stem cells (MSCs) for redox-targeted antifibrotic therapy. Molecular dynamics simulations and membrane fluidity analyses revealed that melatonin modulates lipid packing in cholesterol-rich LNP membranes, increasing membrane fluidity and facilitating endosomal escape, thereby impr

Effect of chlorpromazine on the permeability of beta-lactam antibiotics across rat intestinal brush border membrane vesicles.

The effect of chlorpromazine on the membrane permeability of beta-lactam antibiotics (benzylpenicillin, ampicillin, cephradine and cephalexin) and actively transported substances (glycylglycine and D-glucose) has been studied using rat intestinal brush border membrane vesicles. Except for cephalexin, the initial uptakes at 25 degrees C of these antibiotics were significantly enhanced in the presence of chlorpromazine. In contrast, the transport of glycylglycine and D-glucose was significantly inhibited. These results suggest that the two groups, drugs and actively transported substances, have

Distinct membrane interaction mechanisms of gramicidin A and S in mammalian cells: A combined experimental and molecular dynamics study.

Gramicidins, a class of ionophores, with a well-known antimicrobial activity against bacteria, and more recently against cancer cells, present two distinct structure types: linear peptides for Gramicidin A, B and C and cyclic peptide for Gramicidin S. Independent of their structure, the first stage of their action mechanism involves their interaction with the plasma membrane of the cells. Considering this, we were interested to study how the peptide structure affects cell properties (cell viability, LDH release, etc) and how it correlates with membrane fluidity parameters (GP , ΔSr, Rs) and mo

Drug delivery system (DDS)-inspired cosmetic liposomes: Unveiling physicochemical determinants of skin penetration under practical application conditions.

Although liposomes are widely studied in pharmaceutical drug delivery systems (DDS), their penetration behavior under practical cosmetic application conditions remains poorly understood. To establish design guidelines based on DDS concepts, this study elucidated how the physicochemical properties of cosmetic liposomes govern human skin penetration. Five types of liposomes were prepared with systematically varied particle sizes, membrane fluidities, and surface charges using a modified polyol dilution method. Skin penetration evaluation using excised human skin revealed distinct trends relative

Reversal of diet-induced changes in adenylate cyclase activity and fatty acid composition of rat submandibular salivary gland lipids.

This study sought to determine if the diet-induced changes in submandibular salivary glands can be reversed. Two groups of male weanling Sprague-Dawley rats were fed purified diets containing 9% butter + 1% corn oil (group I, control) or 9% ethyl ester concentrate of n-3 fatty acids + 1% corn oil (group II, experimental). After 5 weeks of feeding the respective diets, rats in group I were divided into two subgroups: Ia, which was maintained on the control diet, and Ib, which was shifted to the experimental diet for the reversal study. The rats in the experimental group were kept on their origi

Confined Water Organization in the Fluid Phase of Phospholipid Self-Assemblies: The Three-Water Layer Model.

Phospholipid-based self-assemblies are frequently studied in cell membrane research and are used in numerous medical and biotechnological applications. Despite current extensive knowledge on membrane nanostructure and its mechanical properties under various environmental conditions, there is still a lack of understanding on interfacial lipid-water interactions. In this chapter, the nature of the confined water is described in the light of the recently proposed Three-Water Layer (TWL) model, discerning (i) 'headgroup water', (ii) 'perturbed water' near the membrane/water interface, and (iii) a

Membrane destabilization by monomeric hIAPP observed by imaging fluorescence correlation spectroscopy.

Monomeric hIAPP significantly destabilizes both model and live cell membranes by increasing membrane fluidity. This interaction with membranes happens via carpet formation followed by lipid extraction in a concentration dependent manner and thus we propose that hIAPP aggregation prior to membrane interaction may not be necessary for its cytotoxicity.

Ischemic pre-conditioning preserves brain mitochondrial functions during the middle cerebral artery occlusion in rat.

It is well known that a brief period of ischemia increases tolerance to a subsequent severe ischemic episode. In the present study, bilateral carotid arteries occlusion (BCAO) was applied as pre-conditioning to testify whether this kind of ischemia could preserve the function of mitochondria with the impairment induced by middle cerebral artery occlusion (MCAO) in brain. The activities of respiratory enzyme complex I to IV, mitochondria swelling, membrane potential, and membrane fluidity were investigated. The results showed that the percentage of infarct area decreased greatly due to the isch

Comparison of the effect of scorpion venom (Buthus martensii Kashi) on the rat brain and heart mitochondria.

A partially purified fraction SVc and a purified homogeneous polypeptide SVIII were isolated from the scorpion (Buthus martensii Kashi) venom, collected in Shan Dong Province of China. SVc decreased the RCR, ADP/O and Qo2 of the rat brain mitochondria. It also decreased the cytochrome oxidase activity and increased the membrane lipid fluidity of the mitochondria. Effect of scorpion venom on the rat heart mitochondria was somewhat different from that of rat brain mitochondria. SVc also decreased RCR, ADP/O and increased the membrane lipid fluidity of heart mitochondria. However, the Qo2 and cyt

AA and Pregnancy.

AA is a crucial fatty acid in pregnancy, especially for the growth and development of the fetus and its central nervous system (CNS). As already discussed in the previous chapters, AA regulates inflammation, immune response, and cell signaling. AA has a modulatory action on gene expression, serves as a mechanotransducer, and regulates cell membrane fluidity. By regulating cell membrane fluidity, AA modulates the expression and binding of several receptors located on the cell membrane. Eicosanoids derived from AA have both beneficial and harmful effects on pregnancy. For instance, (i) AA metabo

Protein-enhanced small molecule disruptors of ordered membrane domains.

Membrane order and fluidity influence many biological processes. However, tools to manipulate membranes under physiological conditions have been limited. In the process of high-throughput screening for molecules that shift the phase partitioning between ordered and disordered membrane phases of the tetraspan membrane protein peripheral myelin protein 22 (PMP22), we identified two chemically similar compounds, VU0615562 and VU0619195, that shift PMP22 toward the disordered phase and destabilize the "lipid raft"-like ordered phase. Follow-up experiments showed that this latter activity is, count

Alcohol and the heart: theoretical considerations.

It is now well established that consumption of ethyl alcohol, both acute and chronic, exerts deleterious effects on the heart. Evidence is presented that the initial event that precipitates both acute and chronic changes reflects the physical effects of alcohol on membrane phospholipids and perhaps proteins. The presence of alcohol increases membrane fluidity, a condition that leads to an adaptive change in the phospholipid composition of the membranes, with resultant greater rigidity of the membranes. The effects of alcohol on the lipid bilayer of the plasma membrane, when combined with other

Beyond electrostatics: Antimicrobial peptide selectivity and the influence of cholesterol-mediated fluidity and lipid chain length on protegrin-1 activity.

Antimicrobial peptides (AMPs) are a promising class of innate host defense molecules for next-generation antibiotics, as they uniquely target and permeabilize membranes of pathogens. This selectivity has been explained by the electrostatic attraction between these predominantly cationic peptides and the bacterial membrane, which is heavily populated with anionic lipids. However, AMP-resistant bacteria have non-electrostatic countermeasures that modulate membrane rigidity and thickness. We explore how variations in physical properties affect the membrane affinity and disruption process of prote

[Is fluidization of the phospholipid bilayer by general anesthetics a reason for general anesthesia?].

It was found that chlorpromazine and pipolfene potentiating general anaesthesia increase viscosity of phospholipid membrane in the region of polar headgroups and in the hydrophobic area near glycerol sceleton. These results (independently of potentiating mechanism) confuse a mechanism of anaesthetics action as increased fluidity of membrane phospholipids.

Peroxynitrite affects lidocaine by acting on membrane-constituting lipids.

Inflammation frequently decreases local anesthetic effects, especially in dental anesthesia in patients with pulpitis and periodontitis. The pharmacokinetics and the mode of action of local anesthetics are closely associated with the hydrophobic interactions between these drugs and lipid bilayers that change the membrane physicochemical property, fluidity. A lipid oxidant, peroxynitrite, is produced by inflammatory cells, and it may act on nerve cell membranes and affect anesthetic efficacy. With respect to this speculated action, we addressed whether peroxynitrite acted on membrane-constituti

[Effect of xanthone from Canscora lucidissima on cultured myocytes anoxia-reoxygenation injuries].

The anoxia/reoxygenation model of cultured neonatal rats myocardial cells were developed, according to Laarse's method: anoxia and glucose deficiency for 60 min followed by reoxygenation and re-exposure to glucose for 30 min. The results showed that the survival rate of myocardial cells in the anoxia group was significantly decreased, release of lactate dehydrogenase (LDH) from myocytes was increased and membrane fluidity was decreased, all the changes were much more severe in the reoxygenation group. 30 min before anoxia, addition of xanthones: 1,8-dihydroxy-3, 5-dimethoxyxanthone, 1-hydroxy-

Renal Fanconi syndrome: developmental basis for a new animal model with relevance to human disease.

Using succinylacetone (SA), a metabolite of tyrosine excreted in excess by infants and children with hereditary tyrosinemia and the renal Fanconi syndrome (FS), we have investigated developmentally-related membrane transport events leading to emergence of the generalized renal tubular dysfunction seen in human FS. SA was found to impair sugar and amino acid uptake by both newborn renal tubules and 7-day renal brush-border membrane vesicles (BBMV). This impairment by SA was due in part to a slowing of substrate cotransport rate of 22Na+-entry into BBMV. Concentration-dependent uptake studies in

Mepivacaine and bupivacaine, frequently used local anesthetics, differ in a variety of clinical effects but show only small differences in chemical structure. In order to learn more about molecular mechanisms of action, we investigated the effects of these two local anesthetics on artificial membranes and liposomes. Methods. Artificial membranes were formed from dipalmitoylphosphatidylcholine - cholesterol or from a phospholipid mixture extracted from rabbit heart to separate into two aqueous phases (100 mM NaCl, 1 mM glycine, pH 7.4). In vitro effects of short-chain aliphatic alcohols, benzyl alcohol and chlorpromazine on the transport of precursors of monoamines across the human erythrocyte membrane.

In previous papers we reported a deficit of tyrosine (TYR) and tryptophan (TRP) transport across the erythrocyte membrane in depressed patients. To investigate further the transport mechanism of both monoamine precursors, we tested in healthy subjects the role played by membrane fluidity, using different fluidizing agents such as alcohols and the neuroleptic chlorpromazine. We found that the transport of both amino acids depended on the length of the chain of each alcohol tested (number of carbon atoms = C). No inhibition was observed after methanol (C1) preincubation, in contrast to benzyl al
