Exploring the effects of Golgi Reassembly and Stacking Proteins in lipid membranes.

Golgi Reassembly and Stacking Proteins (GRASP) have been associated with Golgi-ribbon structure and unconventional vesicular protein secretion. In performing these functions, GRASPs consistently interact with membranes. The presence of lipid modifications, such as myristoylation, is a crucial consideration for obtaining detailed information about the interactions between GRASPs and membranes. Nonetheless, it has been overlooked in the literature so far. Here, we describe a reconstitution protocol for myristoylated human GRASP65 and GRASP55 in lipid model membranes, enabling investigation of th

Recent developments in fluorescence lifetime imaging of lipid membrane dynamics.

Lipid packing in cellular membranes exerts direct effect on membrane tension and microviscosity, and plays a central role in cellular adaptation, homeostasis and disease developement. According to conventional mechanical descriptions, viscosity and tension are directly interconnected, with increased tension leading to decreased membrane microviscosity. However, intricate molecular interactions that contribute to provide the structure and function of cellular membranes suggest a more complex relationship between these parameters. In this perspective, we review recent progress in using environme

Modular PEG coatings for engineering a biomimetic glycocalyx on lipid membranes.

Engineering synthetic cells with biomimetic surface architectures requires precise control over the spatial organization and mechanical properties of the membrane surface. Inspired by the protective and regulatory functions of the natural glycocalyx, we present a modular platform for constructing hierarchical, crosslinked PEG networks on fluid lipid membranes. Through spatially controlled, sequential strain-promoted azide-alkyne cycloaddition (SPAAC), we assemble a tunable, multilayered PEG mesh that mimics the dynamic, dense, and mechanically resilient architecture of the glycocalyx. By incor

Elucidating the pH Effects on Oleic Acid and Interactions with Lipid Membranes.

The interplay between fatty acids (FAs) and lipid membranes under varying pH conditions is important for deciphering fundamental cellular processes and advancing the design of responsive applications. Endowed with membrane incorporation, fatty acids (FAs) can induce curvature on membranes, resulting in alterations in fluidity, permeability, and stability. Despite the wide amount of research on membrane effects induced by FAs, the pH effect on their interaction behaviors remained unclear. Considering the varying pH environments in vivo, the study investigates how pH modulates the aggregation be

What Are the Essential Water Molecules in Regulating Lipid Membrane Fluidity?

Living organisms often endure extreme dehydration, which disrupts the cell membrane structure and function. Dehydration can induce a fluid-to-gel phase transition in lipid bilayers, compromising their fluidity and biological role. Lipid bilayer self-assembly is governed by intricate interactions between water and lipid head groups, but which water molecules are crucial for maintaining fluidity? We address this fundamental question using molecular dynamics (MD) simulations of a liquid-ordered (Lo) lipid bilayer over a hydration range of h = 2-30. We have identified a critical threshold at hydra

Molecular Insights into Osmolyte-Mediated Adaptation of Lipid Membrane of Extremophiles.

Extremophiles survive and thrive in extreme conditions on Earth, which pose challenges to their survivability. Osmotic stress, caused by different extreme conditions, such as water loss due to evaporation in hot and dry conditions, osmotic imbalance due to hypersalinity or excessive pressure, and freeze-drying, causes water loss, which severely affects cell membranes by altering the physicochemical properties of the lipid bilayer. While the organisms employ various adaptive strategies, osmolyte-mediated adaptation (OMA) plays a crucial role in maintaining membrane stability under desiccation.

Membrane lipid composition directs the cellular selectivity of antimicrobial metallohelices.

Two enantiomeric pairs of iron(ii) metallohelices, available as water-soluble, stable, and optically pure bimetallic complexes, differ principally in the length of the central hydrophobic region between two cationic domains, and have distinct activity and cell selectivity profiles against Gram-positive and Gram-negative microbes. The effects of dose concentration and temperature on levels of intracellular accumulation in E. coli and S. aureus, studied via isotopic labelling, indicate that the metallohelices enter the microbial cells via passive diffusion, whereupon (as previously determined) t

Bacterial Membrane Coating Potentiates Lipid Nanoparticles for mRNA Delivery.

Bacterial outer membrane vesicles (OMVs) possess intrinsic immunostimulatory properties by carrying native antigens and pathogen-associated molecular patterns (PAMPs). Lipid nanoparticles (LNPs) have become the leading platform for messenger ribonucleic acid (mRNA) delivery, underpinning the clinical success of mRNA vaccines. Here, we developed a hybrid nanoparticle formulation, OMV-coated LNPs (OMV-LNPs), that combines the high delivery efficiency of LNPs with the immunogenicity of the OMVs to enhance gene delivery. Using Escherichia coli-derived OMVs and dengue virus (DENV) E80 protein as a

Effect of Satureja montana Essential Oil on Model Lipid Membranes.

Satureja montana essential oil is a natural substance able to inhibit the growth of several pathogens. This antimicrobial effect is often attributed to its ability to penetrate cellular structures and disrupt them. Although these properties are recognized as playing a key role in the mechanism of action of this substance, many unresolved issues still exist, and fundamental studies focused on such aspects are scarce. In this framework, we investigated the interaction of SEO with lipid monolayers, which represent simplified models of cell membranes, using the Langmuir monolayer technique, comple

Fumonisin distorts the cellular membrane lipid profile: A mechanistic insight.

Monitoring modifications in membrane lipids in association with external stimuli/agents, including fumonisins (FUMs), is a widely employed approach to assess cellular metabolic response/status. FUMs are prevalent fusariotoxins worldwide that have diverse structures with varying toxicity across species; nevertheless, they can induce metabolic disturbances and disease, including cancer. The capacity of FUMs to disrupt membrane lipids, demonstrated across numerous species and organs/tissues, is ascribed to a multitude of factors/events, which range from direct to indirect effects. Certain events

Understanding Aβ Peptide Binding to Lipid Membranes: A Biophysical Perspective.

Aβ peptides are known to bind neural plasma membranes in a process leading to the deposit of Aβ-enriched plaques. These extracellular structures are characteristic of Alzheimer's disease, the major cause of late-age dementia. The mechanisms of Aβ plaque formation and deposition are far from being understood. A vast number of studies in the literature describe the efforts to analyze those mechanisms using a variety of tools. The present review focuses on biophysical studies mostly carried out with model membranes or with computational tools. This review starts by describing basic physical aspec

Characterization and regulation of 2D-3D convertible lipid membrane transformation.

Micro-nanomaterials that can adopt different structures are powerful tools in the fields of biological and medical sciences. We previously developed a lipid membrane that can convert between 2D nanosheet and 3D vesicle forms using cationic copolymer polyallylamine-graft-polyethylene glycol and the anionic peptide E5. The properties of the membrane during conversion have been characterized only by confocal laser scan microscopy. Furthermore, due to the 2D symmetry of the lipid nanosheet, the random folding of the lipid bilayer into either the original or the reverse orientation occurs during sh

Orientation of the Dysferlin C2A Domain is Responsive to the Composition of Lipid Membranes.

Dysferlin is a 230 kD protein that plays a critical function in the active resealing of micron-sized injuries to the muscle sarcolemma by recruiting vesicles to patch the injured site via vesicle fusion. Muscular dystrophy is observed in humans when mutations disrupt this repair process or dysferlin is absent. While lipid binding by dysferlin's C2A domain (dysC2A) is considered fundamental to the membrane resealing process, the molecular mechanism of this interaction is not fully understood. By applying nonlinear surface-specific vibrational spectroscopy, we have successfully demonstrated that

Single-molecule Diffusion and Assembly on Polymer-crowded Lipid Membranes.

Cellular membranes are highly crowded environments for biomolecular reactions and signaling. Yet, most in vitro experiments probing protein interaction with lipids employ naked bilayer membranes. Such systems lack the complexities of crowding by membrane-embedded proteins and glycans and exclude the associated volume effects encountered on cellular membrane surfaces. Also, the negatively charged glass surface onto which the lipid bilayers are formed prevents the free diffusion of transmembrane biomolecules. Here, we present a well-characterized polymer-lipid membrane as a mimic for crowded lip

Molecular study of endo and phytocannabinoids on lipid membranes of different composition.

The discovery of the endocannabinoid system (ECS) dates back only 30 years. Although many research groups have been elucidating its components, location, functions and metabolism, the peculiarities of the compounds considered "neurotransmitters" of ECS generate questions that have not yet been answered or controversies in the literature. In this context, we studied the molecular behaviour of the main endocannabinoid compounds and the main phytocannabinoids in eukaryotic outer and inner model membranes. The high lipophilicity of these compounds gives place to the hypothesis that cannabinoids ma

The molecular mechanisms of listeriolysin O-induced lipid membrane damage.

Listeria monocytogenes is an intracellular food-borne pathogen that causes listeriosis, a severe and potentially life-threatening disease. Listeria uses a number of virulence factors to proliferate and spread to various cells and tissues. In this process, three bacterial virulence factors, the pore-forming protein listeriolysin O and phospholipases PlcA and PlcB, play a crucial role. Listeriolysin O belongs to a family of cholesterol-dependent cytolysins that are mostly expressed by gram-positive bacteria. Its unique structural features in an otherwise conserved three-dimensional fold, such as

Probing key elements of teixobactin-lipid II interactions in membranes.

Teixobactin (Txb) is a recently discovered antibiotic against Gram-positive bacteria that induces no detectable resistance. The bactericidal mechanism is believed to be the inhibition of cell wall biosynthesis by Txb binding to lipid II and lipid III. Txb binding specificity likely arises from targeting of the shared lipid component, the pyrophosphate moiety. Despite synthesis and functional assessment of numerous chemical analogs of Txb, and consequent identification of the Txb pharmacophore, the detailed structural information of Txb-substrate binding is still lacking. Here, we use molecular

Application of Biosensors Based on Lipid Membranes for the Rapid Detection of Toxins.

Lipid assemblies in the form of two dimensional films have been used extensively as biosensing platforms. These films exhibit certain similarities with cell membranes, thus providing a suitable means for the immobilization of proteinaceous moieties and, further, a number of intrinsic signal amplification mechanisms. Their implementation in the detection of toxins yielded reliable and fast detectors for in field analyses of environmental and clinical samples. Some examples are presented herein, including aflatoxin and cholera toxin detection. The conditions and parameters that determine the ana

Membrane lipid alterations in the metabolic syndrome and the role of dietary oils.

The metabolic syndrome is a cluster of pathological conditions, including hypertension, hyperglycemia, hypertriglyceridemia, obesity and low HDL levels that is of great concern worldwide, as individuals with metabolic syndrome have an increased risk of type-2 diabetes and cardiovascular disease. Insulin resistance, the key feature of the metabolic syndrome, might be at the same time cause and consequence of impaired lipid composition in plasma membranes of insulin-sensitive tissues like liver, muscle and adipose tissue. Diet intervention has been proposed as a powerful tool to prevent the deve
