Late-onset Alzheimer's disease (AD) can, in part, be considered a metabolic disease. Besides age, female sex and APOE ε4 genotype represent strong risk factors for AD that also give rise to large metabolic differences. We systematically investigated group-specific metabolic alterations by conducting stratified association analyses of 139 serum metabolites in 1,517 individuals from the AD Neuroimaging Initiative with AD biomarkers. We observed substantial sex differences in effects of 15 metabolites with partially overlapping differences for APOE ε4 status groups. Several group-specific metabol

Steroid biosynthesis and metabolism are reflected by the serum steroid metabolome and, in even more detail, by the 24-hour urine steroid metabolome, which can provide unique insights into alterations of steroid flow and output indicative of underlying conditions. Mass spectrometry-based steroid metabolome profiling has allowed for the identification of unique multisteroid signatures associated with disorders of steroid biosynthesis and metabolism that can be used for personalized approaches to diagnosis, differential diagnosis, and prognostic prediction. Additionally, steroid metabolome analys

Ozone is an asthma trigger. In mice, the gut microbiome contributes to ozone-induced airway hyperresponsiveness, a defining feature of asthma, but the mechanistic basis for the role of the gut microbiome has not been established. Gut bacteria can affect the function of distal organs by generating metabolites that enter the blood and circulate systemically. We hypothesized that global metabolomic profiling of serum collected from ozone exposed mice could be used to identify metabolites contributing to the role of the microbiome in ozone-induced airway hyperresponsiveness. Mice were treated for

BACKGROUND: Maternal exposure to traffic-related air pollution during pregnancy has been shown to increase the risk of adverse birth outcomes and neurodevelopmental disorders. By utilizing high-resolution metabolomics (HRM), we investigated perturbations of the maternal serum metabolome in response to traffic-related air pollution to identify biological mechanisms.
METHODS: We retrieved stored mid-pregnancy serum samples from 160 mothers who lived in the Central Valley of California known for high air particulate levels. We estimated prenatal traffic-related air pollution exposure (carbon mon

Paternal environment can induce detrimental developmental origins of health and disease (DOHaD) effects in resulting offspring and even future descendants. Such paternal-induced DOHaD effects might originate from alterations in a possible seminal fluid microbiome (SFM) and composite metabolome. Seminal vesicles secrete a slightly basic product enriched with fructose and other carbohydrates, providing an ideal habitat for microorganisms. Past studies confirm the existence of a SFM that is influenced by genetic and nutritional status. Herein, we sought to determine whether treatment of male mice

Trypanosoma cruzi parasites are the causative agents of Chagas disease. These parasites infect cardiac and gastrointestinal tissues, leading to local inflammation and tissue damage. Digestive Chagas disease is associated with perturbations in food absorption, intestinal traffic and defecation. However, the impact of T. cruzi infection on the gut microbiota and metabolome have yet to be characterized. In this study, we applied mass spectrometry-based metabolomics and 16S rRNA sequencing to profile infection-associated alterations in fecal bacterial composition and fecal metabolome through the a

Dihydropyrimidinase deficiency is a rare autosomal recessive disease affecting the second step of pyrimidine degradation. It is caused by mutations in the DPYS gene. Only approximately 30 cases have been reported to date, with a phenotypical variability ranging from asymptomatic to severe neurological illness. We report a case of dihydropyrimidinase deficiency incidentally detected by urine metabolome analysis. Gas chromatography-mass spectrometry-based urine metabolomics demonstrated significant elevations of dihydrouracil and dihydrothymine, which were subsequently confirmed by a quantitativ

OBJECTIVE: Charcot-Marie-Tooth 1A (CMT1A) disease is the most common inherited neuropathy that lacks of therapy and of molecular markers to assess disease severity. Herein, we have pursued the identification of potential biomarkers in plasma samples and skin biopsies that could define the phenotype of CMT1A patients at mild (Mi), moderate (Mo) and severe (Se) stages of disease as assessed by the CMT neuropathy score to contribute to the understanding of CMT pathophysiology and eventually inform of the severity of the disease.
METHODS: We have used: (i) a high-throughput untargeted metabolomic

Nowadays, pediatric cardiology is still a very complex topic. Children are not small adults and very little is known about how heart diseases can affect their metabolic profile both in the acute phase and in the long term. In this context, metabolomics analysis could provide multiple information in the field of pediatric cardiology. For instance, it could be used to predict the occurrence of fetal cardiac abnormalities or predict the outcomes of complex open-heart surgeries for each patient to improve the survival rate and provide better treatment. In this review, the authors describe all the

This chapter reviews the emerging role of metabolomics in nephrotic syndrome (NS), a kidney disorder characterized by proteinuria, hypoalbuminemia, edema, and hyperlipidemia. Metabolomics provides valuable insights into the complex metabolic changes associated with NS, including disruptions in lipid, amino acid, and energy metabolism and oxidative stress markers. Through untargeted and targeted approaches, metabolomics enables the discovery of novel potential biomarkers that could enhance diagnosis, monitor disease progression, and personalize treatment strategies. Despite challenges such as m

Metabolomics is an expanding field of systems biology that is gaining significant attention in respiratory research. As a unique approach to understanding and diagnosing diseases, metabolomics provides a snapshot of all metabolites present in biological samples such as exhaled breath condensate, bronchoalveolar lavage, plasma, serum, urine, and other specimens that may be obtained from patients with respiratory diseases. In this article, we review the rapidly expanding field of metabolomics in its application to respiratory diseases, including asthma, chronic obstructive pulmonary disease (COP

With great social and economic importance, black wattle bark (Acacia mearnsii De Wild.) stands out due to its high content of condensed tannins. Although it is a known and exploited source of proanthocyanidins, little is known about the other compounds that may be present in this raw material and that may further contribute to its bioactive properties. Given this context, this work focused on applying untargeted metabolomics to investigate the metabolic diversity of black wattle bark using different extraction solvents. Furthermore, it aimed to investigate the proanthocyanidin profile of the r

Ulcerative colitis (UC) pathogenesis involves complex interactions between inflammatory reactions, gut dysbiosis, metabolic disruption, and barrier dysfunction. Current therapies primarily target inflammation but fail to correct the underlying dysbiotic ecosystem. We hypothesized that methylated tirilazad (MT), a synthetic 21-aminosteroid with the antioxidant properties of tirilazad, would alleviate colitis by orchestrating a beneficial restructuring of the gut microbiome and its metabolic output. Using a DSS-induced murine colitis model in C57BL/6 mice, we evaluated MT via integrated multi-om

AIMS: Use of sodium-glucose cotransporter 2 inhibitors (SGLT2i) in patients with type 1 diabetes (T1D) is limited by increased risk of ketoacidosis. We aimed to determine metabolic pathways affected by SGLT2i treatment in adults with T1D during ketosis induced by insulin withdrawal.
MATERIALS AND METHODS: Plasma metabolomic and lipidomic analysis by ultra-high-performance liquid chromatography/mass spectrometry was performed at baseline and after supervised insulin withdrawal in 17 adults with T1D treated with usual care (UC) and (DP) dapagliflozin (10 mg daily for 14 days).
RESULTS: Plasma

Chronic kidney disease (CKD) has been associated with alterations in plasma-free aromatic amino acids (AAA)-tryptophan (Trp), phenylalanine (Phe), and tyrosine (Tyr)-and some downstream metabolites. However, AAA metabolism comprises a wider set of compounds, including microbiome-derived and sulfate-conjugated metabolites excreted in urine with potential biological implications. Alport syndrome (AS), a genetic condition defined by progressive renal impairment, frequently advances to CKD, suggesting that disturbances in AAA-related metabolism may also be relevant in this disorder. Nevertheless,

Accurate determination of cause of death and estimation of postmortem interval (PMI) are critical yet challenging tasks in forensic science, particularly in cases with rapid demise and absence of obvious morphological abnormalities. We employed an integrative multi-omics approach to characterize postmortem microbial succession and metabolic alterations on facial skin in mouse models of hemorrhagic shock (HS) and organophosphorus poisoning (OP) across three decomposition stages: bloating (2 days), active decay (8 days), and advanced decay (16 days). Metagenomic profiling revealed significantly

The gut microbiota is increasingly recognized as a regulator of host metabolism and bone physiology. However, how microbial colonization integrates systemic metabolic cues with skeletal remodeling under metabolic stress remains unclear. We used germ-free (GF) and gnotobiotic C57BL/6J mice colonized with the defined 12-member Oligo-Mouse-Microbiota (Oligo-MM12) to dissect microbiota-dependent bone adaptation during high-fat diet (HFD)-induced obesity. Micro-CT analysis revealed that only colonized mice exhibited structural adaptations, namely increased cortical thickness and trabecular area, in

UNLABELLED: Vulvar lichen sclerosus (VLS) is a chronic anogenital disease with long-term risks of genital disfigurement and squamous cell cancer. The involvement of the gut-skin axis in prepubertal VLS remains unclear. We conducted a cross-sectional study of 43 prepubertal VLS patients and 46 healthy controls, performing fecal 16S rRNA sequencing and serum LC-MS to characterize gut microbiome-metabolome associations and identify altered microbial and metabolic signatures. We further assessed their exploratory classification performance using random forest models. No significant differences in
