Using an antimalarial in mosquitoes overcomes Anopheles and Plasmodium resistance to malaria control strategies.

The spread of insecticide resistance in Anopheles mosquitoes and drug resistance in Plasmodium parasites is contributing to a global resurgence of malaria, making the generation of control tools that can overcome these roadblocks an urgent public health priority. We recently showed that the transmission of Plasmodium falciparum parasites can be efficiently blocked when exposing Anopheles gambiae females to antimalarials deposited on a treated surface, with no negative consequences on major components of mosquito fitness. Here, we demonstrate this approach can overcome the hurdles of insecticid

Mathematical model to assess the control of Aedes aegypti mosquitoes by the sterile insect technique.

We propose a mathematical model to assess the effects of irradiated (or transgenic) male insects introduction in a previously infested region. The release of sterile male insects aims to displace gradually the natural (wild) insect from the habitat. We discuss the suitability of this release technique when applied to peri-domestically adapted Aedes aegypti mosquitoes which are transmissors of Yellow Fever and Dengue disease.

Rational design and multidimensional profiling of polyfluorinated pyrethroids: a resistance-breaking strategy for next-generation mosquito vector control.

BACKGROUND: Pyrethroids based on chrysanthemic acid are widely used for mosquito control due to their high potency and low mammalian toxicity. However, growing resistance and environmental concerns necessitate new analogues. This study designed novel polyfluorinated pyrethroids derived from chrysanthemic acid scaffolds to overcome resistance, reduce ecotoxicity, and improve photostability. RESULTS: Fourteen of 54 synthesized chrysanthemate-based compounds exceeded bifenthrin (LC50 = 1.96 mg/L) against sensitive mosquitoes. Lead compounds BFA-3-1, BFA-3-2, and BFA-3-4 showed LC50 values of 0.5

An Ecological Impact Assessment Framework and Integrated Management Pathway for Large-Scale Chemical Mosquito Control in Urban Areas.

Global warming and rapid urbanization are intensifying the threat of mosquito-borne diseases, prompting many cities to default to large-scale chemical spraying as an emergency response. However, this quick-fix approach may conceal profound ecological collateral damage that has long been overlooked. Here, we systematically map the impacts of mass larviciding and adulticiding on urban ecosystems and present a novel ecological risk indicator framework that tracks acute nontarget effects, the environmental fate and transport of active ingredients, impairment of ecosystem functions, and human healt

Uneven mosquito control knowledge, attitudes, and practices in Malaysia shape sustainable dengue prevention across urban and rural settings.

Dengue remains a persistent public health challenge in Malaysia, where rapid urbanization, ecological transformation, and behavioral gaps sustain Aedes mosquito proliferation despite decades of control programs. Understanding geographic and urban-rural disparities in community knowledge and preventive practices is essential for developing sustainable, evidence-based strategies. This cross-sectional study, conducted from February to May 2024, involved 866 respondents from 12 states in Peninsular Malaysia representing diverse urban and rural populations. Using a validated Knowledge, Attitudes an

Neem-based products as potential eco-friendly mosquito control agents over conventional eco-toxic chemical pesticides-A review.

Mosquitoes cause serious health hazards for millions of people across the globe by acting as vectors of deadly communicable diseases like malaria, filariasis, dengue and yellow fever. Use of conventional chemical insecticides to control mosquito vectors has led to the development of biological resistance in them along with adverse environmental consequences. In this light, the recent years have witnessed enormous efforts of researchers to develop eco-friendly and cost-effective alternatives with special emphasis on plant-derived mosquitocidal compounds. Neem oil, derived from neem seeds (Azadi

Dopamine receptor antagonists as new mode-of-action insecticide leads for control of Aedes and Culex mosquito vectors.

BACKGROUND: New mode-of-action insecticides are sought to provide continued control of pesticide resistant arthropod vectors of neglected tropical diseases (NTDs). We previously identified antagonists of the AaDOP2 D1-like dopamine receptor (DAR) from the yellow fever mosquito, Aedes aegypti, with toxicity to Ae. aegypti larvae as leads for novel insecticides. To extend DAR-based insecticide discovery, we evaluated the molecular and pharmacological characteristics of an orthologous DAR target, CqDOP2, from Culex quinquefasciatus, the vector of lymphatic filariasis and West Nile virus. Approaches to vector control: new and trusted. 1. Humoral immune responses in blackfly and mosquito vectors of filariae.

The vectors of filariasis, mosquitoes and blackflies, are capable of mounting a defence response to the infection. This selective review describes the molecules that are involved in these immune systems. Several antibacterial peptides are known to be induced and secreted into the haemolymph by the fat body and the circulating haemocytes. In addition, haemagglutinating lectins with carbohydrate specificities to the surface of the developing filarial larvae appear. Activation of a range of proteases occurs rapidly as does activation of the prophenoloxidase pathway. The possible roles of these an

A diffusion model for mosquito control trials with spillover: application to calculations of power, sample size, and bias.

BACKGROUND: Mosquito movement is ubiquitous and complicates the design of cluster randomized trials (CRTs) of vector control interventions because it can cause spillover between trial arms. There has been no analytical approach to quantify the impact of this spillover on trial power, required sample sizes, or bias. This precludes formal allowance for spillover in formulae for study power, and a precautionary principle is generally used in trial design, leading to CRTs with very large clusters and extensive buffer zones between arms. METHODS: The diffusion equation was solved to give a mathema

Simulated adult control on laboratory larval mesocosms demonstrates overcompensation in the yellow fever mosquito, Aedes aegypti (Diptera: Culicidae).

Mortality events can affect populations in counterintuitive ways leading to compensation or overcompensation when density-dependent effects are present. These phenomena occur in a variety of taxa and are particularly important to understand for the management of commercial species, the conservation of endangered species, and the control of arthropod pests. Using a mesocosm experimental design, we simulated different levels of adult control by altering larval densities (i.e. via the simulated mortality event) to assess mortality effects in Aedes aegypti L. (Diptera: Culicidae). Various larval d

Effectiveness of stand-alone anti-adult mosquito control strategies against Aedes-borne diseases: scoping review with meta-analysis.

BACKGROUND: The world is witnessing the emergence of infections transmitted by Aedes mosquitoes. However, preventing large outbreaks challenges the health systems of endemic countries. Targeting infected adult Aedes mosquitoes may be a better means for resource-constrained health systems where integrated vector control may be less feasible. We reviewed the evidence of the effectiveness of stand-alone adult mosquito control measures against Aedes mosquito-borne diseases in this scoping review and meta-analysis. METHODS: The MEDLINE, Embase, Scopus and Cochrane Library (Cochrane Database of Sys

Advancing the art of mosquito control: the journey of the sterile insect technique against Aedes aegypti in Cuba.

BACKGROUND: Aedes aegypti, the primary vector of dengue, chikungunya, and Zika viruses, poses a significant public health threat worldwide. Traditional control methods using insecticides are increasingly challenged by resistance and environmental concerns. The sterile insect technique (SIT) offers an eco-friendly alternative that has been successfully applied to other insect pests. This article aims to briefly review Ae. aegypti management in Cuba, highlighting the accomplishments, challenges, and future directions of the SIT. MAIN BODY: Here we provide a brief summary of the extensive histor

Prevention and control of mosquito-borne arboviral diseases: lessons learned from a school-based intervention in Brazil (Zikamob).

BACKGROUND: Since the 1980s, when dengue was reintroduced in Brazil, outbreaks and epidemics caused by different arbovirus strains transmitted by vector mosquitoes such as Aedes aegypti have been an annual occurrence. The aim of this study was to evaluate the behavioural change of high school students and teachers who participated in an educational intervention for the prevention and vector control of arboviral diseases. METHODS: In this school-based intervention, a self-reported questionnaire was used in a pre-post intervention methodology to assess environmental risk factors, sociodemograph

Mosquito larvae control by photodynamic inactivation of their intestinal flora - a proof of principal study on Chaoborus sp.

Mosquitoes are carriers of dangerous infectious disease pathogens all over the world. Owing to travelling and global warming, tropical disease-carrying species such as Aedes, Anopheles and Culex spread beyond tropical and subtropical zones, even to Europe. The aim of this study is to investigate the potential of photodynamic agents to combat mosquito larvae. Three different photosensitizers were tested on Chaoborus sp. larvae: TMPyP and TPPS as antimicrobial photosensitizers, and mTHPC as a PDT drug against eukaryotic animal and human cells. Chaoborus sp. is a commercially available harmless s

MicroRNA-275 targets sarco/endoplasmic reticulum Ca2+ adenosine triphosphatase (SERCA) to control key functions in the mosquito gut.

The yellow fever mosquito Aedes aegypti is the major vector of arboviruses, causing numerous devastating human diseases, such as dengue and yellow fevers, Chikungunya and Zika. Female mosquitoes need vertebrate blood for egg development, and repeated cycles of blood feeding are tightly linked to pathogen transmission. The mosquito's posterior midgut (gut) is involved in blood digestion and also serves as an entry point for pathogens. Thus, the mosquito gut is an important tissue to investigate. The miRNA aae-miR-275 (miR-275) has been shown to be required for normal blood digestion in the fema

Life-table analysis of Anopheles malaria vectors: generational mortality as tool in mosquito vector abundance and control studies.

BACKGROUND & OBJECTIVES: Vector control will for sometime remain a primary weapon in the war against vector borne diseases. Malaria is of paramount importance in this with its associated high morbidity and mortality especially in sub-Saharan Africa. This study on generational mortality associated factors in Anopheles mosquitoes life-table analysis was designed to investigate the fecundity, levels of mortality and mortality associated factors at the aquatic stages of anopheline malaria vectors. METHODS: Mortality associated factors were investigated at the eggs, I and II instar larval, III and

Malaria outbreak control in an African village by community application of 'deet' mosquito repellent to ankles and feet.

The malaria vector Anopheles arabiensis Patton (Diptera: Culicidae) shows a marked predilection (> 80%) for biting the ankles and feet of human subjects, as revealed by our previous observations at Malahlapanga in the Kruger National Park, South Africa. Topical application of insect repellent, 15% deet (N,N-diethyl-3-methylbenzamide), to feet and ankles reduced the overall biting rate of An. arabiensis by 69%. A focal malaria epidemic in Albertsnek village (25 degrees 33'S, 31 degrees 59' E) near the Mozambique border, following flooding during February 2000, provided an opportunity to apply t

A meta-analysis reveals that dragonflies and damselflies can provide effective biological control of mosquitoes.

Dragonfly/damselfly naiads have the potential to control mosquitoes, and indirectly the diseases they carry, due to their extensive predation on mosquito larvae. Experimental studies have measured the effectiveness of individual dragonfly/damselfly naiads in controlling mosquitoes by introducing them to mosquito larvae and counting the number of larvae eaten in a given time period (i.e. predation success). Without a quantitative synthesis, however, such individual measures are unable to provide a generalized estimation about the effectiveness of dragonflies/damselflies as biological mosquito c
