Mycobacterium genavense infection in canaries.

A case of mycobacteriosis in a collection of canaries (Serinus canarius) is described. The affected birds showed nonspecific clinical signs and macroscopic lesions (slight splenomegaly). Histologic lesions found in liver, spleen, lungs, and kidneys consisted of noncaseous nodules containing accumulations of large macrophages that showed a highly vacuolated cytoplasm in which numerous acid-fast organisms were detected. Attempts to isolate and culture the organisms using Löwenstein-Jensen and Coletsos media proved unsuccessful. Microorganisms belonging to the species Mycobacterium genavense were

Granulomas Induced by Mycobacterium leprae.

Leprosy, a chronic infectious disease caused by Mycobacterium leprae, affects primarily the skin, mucous membranes, and peripheral nerves. It is characterized by a spectrum of symptoms, largely a result of the immunological response of the host to the antigens of M. leprae. At one pole [lepromatous leprosy (LL)] there is an enormous growth of the bacilli in the tissues resulting from a specific and profound lack of cell-mediated immunity (CMI) of the host to M. leprae. At the opposite pole [tuberculoid leprosy (TT)], patients are responsive to M. leprae antigens, and there are few bacilli pres

Mycobacterium haemophilum: an emerging pathogen.

Mycobacterium haemophilum is emerging as a pathogen of immunocompromised patients particularly those with AIDS and organ transplants. Infection has also occurred in healthy children. Adults usually present with cutaneous manifestations, septic arthritis or occasionally pneumonia. Children have perihilar, cervical or submandibular adenitis. The organism grows on mycobacterial media supplemented with ferric ammonium citrate or hemin, incubated at 30 degrees C to 32 degrees C, two to three weeks after inoculation. The most active antimicrobial agents in vitro are amikacin, ciprofloxacin, clarithr

Immunobiology of Mycobacterium avium infection.

Infection caused by organisms of the Mycobacterium avium complex is diagnosed in 50% to 60% of AIDS patients with the advanced stage of disease. Mycobacterium avium is an environmental bacterium that gains access to the host through both the gastrointestinal tract and the respiratory tract. After crossing the mucosal barrier Mycobacterium avium disseminates, infecting chiefly mononuclear phagocytes of the reticuloendothelial system. A number of cells of the immune system such as CD4+ T cells, natural killer cells and macrophages have been shown to be involved in the host response to Mycobacter

Aquatic hazard Mycobacterium marinum infection.

We describe two patients with Mycobacterium marinum infection and review the pertinent literature. M. marinum infection follows trauma, often trivial, in water or from marine life. Clinical manifestations include superficial cutaneous lesions which are either solitary or multiple in a sporotrichoid distribution, involvement of the deeper structures of the hand and wrist and disseminated disease. Biopsy of infected tissue reveals a mixed suppurative-granulomatous reaction with sparse to absent acid-fast bacilli. Definitive diagnosis is achieved by growing the organism from appropriate specimens

Glutamine synthetase from Mycobacterium avium.

Mycobacterium avium was previously shown to be dependent upon ammonia or glutamine as a nitrogen source. In an effort to assess the physiology of ammonia assimilation by M. avium, a characterization of its glutamine synthetase was performed. The enzyme from M. avium was purified by streptomycin sulfate treatment, ammonium sulfate precipitation, and affinity chromatography. The enzyme was unusual in that it had a pH optimum of 6.4 and maximum enzyme activity was obtained between 50 and 60 degrees C as shown by the transferase assay. The glutamine synthetase activity from batch-cultured cells de

[Endophthalmitis caused by mycobacterium tuberculosis].

A 60-year-old woman developed an anterior uveitis of unknown etiology in her right eye. Topical treatment was corticosteroids led to a regression of the inflammation, but three months later a severe relapse occurred, followed by endophthalmitis and blindness. At this time a previously unknown pulmonary tuberculosis was diagnosed. In histologic sections of the enucleated eye numerous acid-fast bacilli were demonstrated. Diagnostic and therapeutic considerations are discussed.

Antigenic analysis of Mycobacterium leprae.

About twenty distinct antigenic components have been demonstrated in Mycobacterium leprae (M. leprae) by crossed immunoelectrophoresis against a rabbit antiserum produced by immunization with concentrated M. leprae antigen. This system allows a more detailed analysis of the antigenic relationship between M. leprae and other mycobacteria and a better characterization of the antigenic content of various M leprae preparations than with previously available anstisera of the antigenic content of various M. leprae preparations than with previously available antisera which reacted with far fewer comp

Prime-boost with Mycobacterium smegmatis recombinant vaccine improves protection in mice infected with Mycobacterium tuberculosis.

The development of a new vaccine as a substitute for Bacillus Calmette-Guerin or to improve its efficacy is one of the many World Health Organization goals to control tuberculosis. Mycobacterial vectors have been used successfully in the development of vaccines against tuberculosis. To enhance the potential utility of Mycobacterium smegmatis as a vaccine, it was transformed with a recombinant plasmid containing the partial sequences of the genes Ag85c, MPT51, and HspX (CMX) from M. tuberculosis. The newly generated recombinant strain mc(2)-CMX was tested in a murine model of infection. The rec

Mycobacterium bovis BCG immunization induces protective immunity against nine different Mycobacterium tuberculosis strains in mice.

Recent preclinical and epidemiologic studies have suggested that certain Mycobacterium tuberculosis genotypes (in particular, Beijing lineage strains) may be resistant to Mycobacterium bovis BCG vaccine-induced antituberculosis protective immunity. To investigate the strain specificity of BCG-induced protective responses in a murine model of pulmonary tuberculosis, C57BL/6 mice were vaccinated with BCG vaccine and then challenged 2 months later with one of nine M. tuberculosis isolates. Four of these strains were from the W-Beijing lineage (HN878, N4, NHN5, and ChS) while four were non-Beijing

Immunogenicity of Mycobacterium tuberculosis antigens in Mycobacterium bovis BCG-vaccinated and M. bovis-infected cattle.

The development of novel vaccine strategies supplementing Mycobacterium bovis BCG (BCG) constitutes an urgent research challenge. To identify potential subunit vaccine candidates, we have tested a series of eight recently identified Mycobacterium tuberculosis antigens in M. bovis-infected and BCG-vaccinated cattle. These antigens were characterized on the basis of their ability to induce in vitro gamma interferon responses in infected or BCG-vaccinated calves. We were able to establish a hierarchy of these antigens based on how frequently they were recognized in both groups of animals. In part

Evaluation of the Mycobacterium RealTime Kit Vircell (MRTVircell) Assay for Detecting Mycobacterium Species in Clinical Specimens.

Rapid and accurate diagnosis of mycobacterial infections is crucial for guiding therapeutic decisions. This study presents the first evaluation of a novel molecular assay, the Mycobacterium RealTime PCR Kit Vircell (MRTVircell), a real-time PCR-based test designed for the specific detection of Mycobacterium tuberculosis complex (MTBC), Mycobacterium avium complex (MAC), Mycobacterium abscessus complex (MABC), and other nontuberculous mycobacteria (NTM) in both respiratory and non-respiratory samples. The evaluation was conducted under routine workflow conditions using 721 clinical specimens, i

In vitro validation of in silico-selected targets for PCR detection of genus Mycobacterium and species Mycobacterium marinum.

Mycobacterial infections pose significant diagnostic challenges due to the genetic diversity of species, limitations of current detection methods, and the need for rapid and accurate identification tools. In this study, we developed and validated a novel molecular approach for the specific detection of the Mycobacterium genus and the Mycobacterium marinum species based on the identification of unique DNA sequences. Using comparative genomic alignments and in silico screening of curated genomic databases, we identified a 391 bp region of the mmpl gene specific to the Mycobacterium genus, and a

Differences in drug susceptibility pattern between Mycobacterium avium and Mycobacterium intracellulare isolated in respiratory specimens.

Mycobacterium avium complex (MAC) is the most common etiologic organisms of nontuberculous mycobacteria (NTM) lung disease. In this study, we aimed to retrospectively investigate the differences in drug susceptibility patterns of two major MAC species; Mycobacterium avium and Mycobacterium intracellulare. A total of 1883 major two MAC isolates (1060 M. avium and 823 M. intracellulare) from respiratory specimens were included in this study during the period 2011─2016. The minimum inhibitory concentrations (MICs) were determined by broth microdilution method and MIC50/MIC90 values were derived f
