Early death of neural crest cells is responsible for total enteric aganglionosis in Sox10(Dom)/Sox10(Dom) mouse embryos.

Intestinal aganglionosis results from homologous genetic defects in humans and mice, including mutations of Sox10, which encodes a transcription factor expressed in neural crest cells. To gain insight into the embryological basis for this condition, the phenotype and pathogenesis of intestinal aganglionosis in Sox10(Dom)/Sox10(Dom) embryos were studied. The distribution of enteric neural precursors and other neural crest derivatives in Sox10(Dom)/Sox10(Dom) embryos was analyzed with immunochemical and transgenic markers. The ability of wild-type neural crest cells to colonize Sox10(Dom)/Sox10(

Childhood maltreatment impacts the activity of the neural empathy but not theory of mind network: findings from a transdiagnostic sample.

BACKGROUND: Childhood maltreatment (CM) is a risk factor for psychopathology, including posttraumatic stress disorder (PTSD), major depressive disorder (MDD) and somatic symptom disorder (SSD). One possible mechanism linking CM with psychopathology is social cognition, comprising empathy and theory of mind (ToM). While prior research has focused on the association of CM with behavioral empathy and ToM capacities, not much is known about its association with functional neural correlates. METHODS: The sample included N = 101 adults with PTSD, MDD, and SSD, as well as healthy volunteers (HV) wit

Neural transdifferentiation of human bone marrow mesenchymal stem cells on hydrophobic polymer-modified surface and therapeutic effects in an animal model of ischemic stroke.

Human bone marrow-derived mesenchymal stem cells (MSCs) have multi-lineage differentiation potential and can become cells of mesodermal and neural lineages. These stem cells thus hold considerable clinical promise for the treatment of neurodegenerative diseases. For successful regeneration of damaged neural tissues, directed differentiation of neural or neuronal precursor cells from MSCs and integration of transplanted cells are pivotal factors. We induced MSCs into neurogenesis using a modified protocol. The therapeutic potency of the resulting neural progenitor cells in a rat model of ischem

An Injectable Hydrogel Loaded with GMSCs-Derived Neural Lineage Cells Promotes Recovery after Stroke.

Ischemic stroke is a devastating medical condition with poor prognosis due to the lack of effective treatment modalities. Transplantation of human neural stem cells or primary neural cells is a promising treatment approach, but this is hindered by limited suitable cell sources and low in vitro expansion capacity. This study aimed (1) use small molecules (SM) to reprogram gingival mesenchymal stem cells (GMSCs) commitment to the neural lineage cells in vitro, and (2) use hyaluronic acid (HA) hydrogel scaffolds seeded with GMSCs-derived neural lineage cells to treat ischemic stroke in vivo. Scale-Dependent Development of Neural Signal Complexity and its Association with Working Memory in Childhood.

Neural development during childhood involves progressive reorganization of brain dynamics across multiple temporal scales. The present study examined developmental differences in neural signal complexity, indexed by modified multiscale entropy (mMSE), in 253 typically developing children aged 3-9 years. Resting-state EEG was analyzed across 20 temporal scales spanning approximately 4-80 ms, and age-related differences were assessed using linear mixed-effects models with sex included as a covariate. Results showed systematic age-related increases in neural signal complexity, with developmental

Milk fat globule-EGF factor VIII attenuates CNS injury by promoting neural stem cell proliferation and migration after cerebral ischemia.

The mediators in activating neural stem cells during the regenerative process of neurogenesis following stroke have not been fully identified. Milk fat globule-EGF Factor VIII (MFG-E8), a secreted glycoprotein serves several cellular functions by binding to its receptor, αv β3-integrin. However, its role in regulating neural stem cells after stroke has not been determined yet. We therefore, aim to reveal whether MFG-E8 promotes neural stem cell proliferation and migration during stroke. Stroke was induced in wild-type (Wt) and MFG-E8-deficinet (Mfge8-/-) mice by transient middle cerebral arter

The many faces of anxiety-neurobiological correlates of anxiety phenotypes.

Anxiety is an all-inclusive concept incorporating somatic symptoms (palpitations, dizziness, dyspnea), emotional and cognitive elements (negative affect, fear, worry, rumination) and behavioral components (e.g., avoidance). The aim of this study was to examine the specific neural correlates associated with anxiety phenotypes (worry, rumination, somatic anxiety) and negative affect (neuroticism). Twenty-nine anxious participants and 30 healthy controls were included in the study. We analyzed seed-based intrinsic connectivity and used correlation maps in a multivariable regression model to descr

Neural Correlates of Music Listening and Recall in the Human Brain.

Previous neuroimaging studies have identified various brain regions that are activated by music listening or recall. However, little is known about how these brain regions represent the time course and temporal features of music during listening and recall. Here we analyzed neural activity in different brain regions associated with music listening and recall using electrocorticography recordings obtained from 10 epilepsy patients of both genders implanted with subdural electrodes. Electrocorticography signals were recorded while subjects were listening to familiar instrumental music or recalli

Actin capping protein CAPZB regulates cell morphology, differentiation, and neural crest migration in craniofacial morphogenesis†.

CAPZB is an actin-capping protein that caps the growing end of F-actin and modulates the cytoskeleton and tethers actin filaments to the Z-line of the sarcomere in muscles. Whole-genome sequencing was performed on a subject with micrognathia, cleft palate and hypotonia that harbored a de novo, balanced chromosomal translocation that disrupts the CAPZB gene. The function of capzb was analyzed in the zebrafish model. capzb(-/-) mutants exhibit both craniofacial and muscle defects that recapitulate the phenotypes observed in the human subject. Loss of capzb affects cell morphology, differentiatio

A novel methodological approach to understanding the cortical and subcortical effects of aerobic exercise in Parkinson's disease.

INTRODUCTION: Aerobic exercise mitigates symptoms of Parkinson's disease (PD) and may slow disease progression; however, the neural mechanisms underlying these improvements are not well understood. In this study, we discuss the methodology for simultaneously recording local field potentials (LFP) from the subthalamic nucleus (STN), cortical activity using scalp electroencephalography (EEG), and exercise performance metrics during a 40-min aerobic cycling session. Data from a single patient with PD are presented to illustrate the utility, feasibility, and data integrity of the experimental set
